| 1 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 3 | Verification of Mutagen Function of Zeocin in Nannochloropsis oceanica Through Transcriptome Analysis显示文摘Zeocin can cause double strand breaks of DNA and thus is frequently used as a selective antibiotic of eukaryotic Sh ble transformants. In non-transformation system, Zeocin may function as a mutagen if not totally lethal. To verify such function of Zeocin, we mutated Nannochloropsis oceanica by increasing the concentration of Zeocin in medium gradually, and isolated a N. oceanica strain(single cell culture) which survived Zeocin up to 10.0μg mL^(-1). The Zeocin-tolerant strain entered the exponential growth phase later and grew slower than the wild strain. Transcriptome profiling showed that the Zeocin-tolerant N. oceanica strain survived Zeocin mainly by adapting(heritable), rather than acclimating(plastic) to Zeocin. Hence mutating N. oceanica with Zeocin was approved effective. Meanwhile, the physiological characteristics of this Zeocin-tolerant strain were demonstrated. As we proposed, N. oceanica tolerated Zeocin by strengthening its protein degradation and antioxidation. The genes controlling cell division and cellular response to stimuli may also have played important roles in the reduction of growth and the tolerance to Zeocin. Our findings evidenced that Zeocin can serve as an appropriate mutagen of microalgae. | 5 | Efficient antimicrobial properties of layered double hydroxide assembled with transition metals via a facile preparation method显示文摘Mg2+in MgAl-layered double hydroxides nanoparticles was substituted with different divalent transition metal ions(MAl-LDHs,M:Mg2+,Cu2+,Ni2+,Co2+,and Mn2+)via a facile method to be used as antibacterial agents.The phase structural and morphological characterizations of MAl-LDHs were investigated by XRD,FTIR spectroscopy and TEM.The results have shown that all of MAl-LDHs had typical layered structures except MnAl-LDH which contained Mn304 phases.Particular morphology of MnAl-LDH with ellipsoids,spherical and rod-like structure and CuAl-LDH with rod-like shape existed.IC50(the concentrations providing 50%antibacterial activity)values of CuAl-LDH,NiAl-LDH,CoAl-LDH,and MnAlLDH in broth dilution tests were^800-1500μg/mL.Dosages of CuAl-LDH,CoAl-LDH,and MnAl-LDH with>10 mm inhibition zone in disk diffusion tests were^150-300μg/disk.Antibacterial mechanism of MAl-LDHs may be attributed to the synergistic factors including effected surroundings,surface interactions,morphology of particles,ROS and metal ions.The results indicate a facile method to synthesis LDHs based effective antibacterial agents with the potential application in the area of water treatment and antibacterial coating. | Mengxue Li Li Li Sijie Lin | 2020 | Chinese Chemical Letters2020,31,6: | 2 |
| 6 | Optimization of the OCO-2 Cloud Screening Algorithm and Evaluation against MODIS and TCCON Measurements over Land Surfaces in Europe and Japan显示文摘A method to tighten the cloud screening thresholds based on local conditions is used to provide more stringent schemes for Orbiting Carbon Observatory-2(OCO-2)cloud screening algorithms.Cloud screening strategies are essential to remove scenes with significant cloud and/or aerosol contamination from OCO-2 observations,which helps to save on the data processing cost and ensure high quality retrievals of the column-averaged CO2 dry air mole fraction(XCO2).Based on the radiance measurements in the 0.76μm O2A band,1.61μm(weak),and 2.06μm(strong)CO2 bands,the current combination of the A-Band Preprocessor(ABP)algorithm and Iterative Maximum A Posteriori(IMAP)Differential Optical Absorption Spectroscopy(DOAS)Preprocessor(IDP)algorithm passes around 20%-25%of all soundings,which means that some contaminated scenes also pass the screening process.In this work,three independent pairs of threshold parameters used in the ABP and IDP algorithms are sufficiently tuned until the overall pass rate is close to the monthly clear-sky fraction from the MODIS cloud mask.The tightened thresholds are applied to observations over land surfaces in Europe and Japan in 2016.The results show improvement of agreement and positive predictive value compared to the collocated MODIS cloud mask,especially in summer and fall.In addition,analysis indicates that XCO2 retrievals with more stringent thresholds are in closer agreement with measurements from collocated Total Carbon Column Observing Network(TCCON)sites. | Sijie CHEN Shuaibo WANG Lin SU Changzhe DONG Ju KE Zhuofan ZHENG Chonghui CHENG Bowen TONG Dong LIU | 2020 | Advances in Atmospheric Sciences2020,37,4: | 1 |

| 8 | Mechanisms of Pb(Ⅱ) coprecipitation with natrojarosite and its behavior during acid dissolution显示文摘Lead(Pb) coprecipitation with jarosite is common in natural and engineered environments,such as acid mine drainage(AMD) sites and hydrometallurgical industry. Despite the high relevance for environmental impact, few studies have examined the exact interaction of Pb with jarosite and the dissolution behavior of each phase. In the present work, we demonstrate that Pb mainly interacts with jarosite in four modes, namely incorporation, occlusion,physically mixing, and chemically mixing. For comparison, the four modes of Pb-bearing natrojarosite were synthesized and characterized separately. Batch dissolution experiments were undertaken on these synthetic Pb-bearing natrojarosites under pH_(2) to simulate the AMD environments. The introduction of Pb decreases the final Fe releasing efficiency of jarosite-type compounds from 18.18% to 3.45%-5.01%, showing a remarkable inhibition of their dissolution. For Pb releasing behavior, PbSO_(4) dissolves in preference to Pb-substituted natrojarosite, i.e.,(Na, Pb)-jarosite, which primarily results in the sharp increase of Pb releasing concentration(> 40 mg/L). PbSO_(4) occlusion by jarosite-type compounds can significantly reduce the release of Pb. | 10 | Physical and toxicological profiles of human IAPP amyloids and plaques显示文摘Although much has been learned about the fibrillization kinetics, structure and toxicity of amyloid proteins, the properties of amyloid fibrils beyond the saturation phase are often perceived as chemically and biologically inert, despite evidence suggesting otherwise. To fill this knowledge gap, we examined the physical and biological characteristics of human islet amyloid polypeptide(IAPP) fibrils that were aged up to two months. Not only did aging decrease the toxicity of IAPP fibrils, but the fibrils also sequestered fresh IAPP and suppressed their toxicity in an embryonic zebrafish model. The mechanical properties of IAPP fibrils in different aging stages were probed by atomic force microscopy and sonication, which displayed comparable stiffness but age-dependent fragmentation, followed by self-assembly of such fragments into the largest lamellar amyloid structures reported to date. | 17 | Regulatory Effect of JAK2/STAT3 on the Immune Function of Endotoxin-tolerant Dendritic Cells and its Involvement in Acute Liver Failure显示文摘Background and Aims:Acute liver failure(ALF)is a potentially fatal clinical syndrome with no effective treatment.This study aimed to explore the role of Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)pathway in modulating the phenotype and immune function of endotoxin-tolerant dendritic cells(ETDCs).In addition,we explored the use of EDTCs in an experimental model of ALF and investigated the associated mechanisms.Methods:In the in vitro experiment,ETDCs were transfected with adenovirus to induce SOCS1^(+/+)ETDCs and SOCS1^(−/−)ETDCs.Thereafter,costimulatory molecules and mixed lymphocyte reaction were assessed.Experimental mice were randomly divided into normal control,ALF,ALF+mock-ETDCs,ALF+SOCS1^(+/+)ETDCs,ALF+AG490,and ALF+AG490+SOCS1^(+/+)ETDCs groups.We examined the therapeutic effect of adoptive cellular immunotherapy by tail-vein injection of target ETDCs 12 h before ALF modeling.AG490,a JAK2/STAT3 inhibitor,was used in the in vivo experiment to further explore the protective mechanism of SOCS1^(+/+)ETDCs.Results:Compared with control ETDCs,SOCS1^(+/+)ETDCs had lower expression of costimulatory molecules,weaker allostimulatory ability,lower levels of IL-6 and TNF-αexpression and higher IL-10 secretion.SOCS1^(−/−)ETDCs showed the opposite results.In the in vivo experiments,the ALF+SOCS1^(+/+)ETDCs and ALF+AG490+SOCS1^(+/+)ETDCs groups showed less pathological damage and suppressed activation of JAK2/STAT3 pathway.The changes were more pronounced in the ALF+AG490+SOCS1^(+/+)ETDCs group.Infusion of SOCS1^(+/+)ETDCs had a protective effect against ALF possibly via inhibition of JAK2 and STAT3 phosphorylation.Conclusions:The SOCS1 gene had an important role in induction of endotoxin tolerance.SOCS1^(+/+)ETDCs alleviated lipopolysaccharide/D-galactosamine-induced ALF by downregulating the JAK2/STAT3 signaling pathway. | Yukai Chen Chaochen Hou Naibin Yang Yanyan Yang Youran Chen Deyong Kong Yuchun Jiang Minghao Lin Sijie Zheng Shanshan Li Mingqin Lu | 2022 | Journal of Clinical and Translational Hepatology2022,10,5: | 0 |
| 18 | Doping Gd^(3+)Ion in PDA-PHBV Coating on Ti6Al4V Alloy for Enhancing Corrosion Resistance and Proliferation of Human Gingival Fibroblasts and Human Umbilical Vein Endothelial Cells显示文摘In this study,we fabricated poly(3-hydroxybutyrate-3-hydroxyvalerate)(PHBV)coatings doped with Gd^(3+)(1,5,and 10×10^(−4) mol/L)on Ti6Al4V alloy for the first time to promote soft tissue sealing around dental implants.The corrosion resistance of Gd^(3+)-modified PHBV-coated Ti6Al4V was studied by electrochemical and immersion tests,respectively,whereas CCK-8 and RT-PCR evaluated the biocompatibility to human gingival fibroblasts(HGFs)and human umbilical vein endothelial cells(HUVECs).It was found that the Gd^(3+)-modified PHBV coating could enhance the corrosion resistance of Ti6Al4V.In vitro cell tests showed that PHBV coatings with and without Gd^(3+) addition could promote adhesion and proliferation of HGFs and HUVECs,showing a Gd^(3+) content-dependent manner.Moreover,it was found that the PDA-PHBV@1Gd showed the best proliferation to HGFs by up-regulating gene expressions of VINCULIN,ITGB1,and ITGA3,whereas the best response to HUVECs with the highest gene expression of eNOS and HIF-1αgenes was found in the PDA-PHBV@5Gd-coated group. | Sijie Qin Xiongcheng Xu Yanjin Lu Liu Li Tingting Huang Jinxin Lin | 2022 | Acta Metallurgica Sinica(English Letters)2022,35,5: | 0 |
