1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20

3Role of PLC-PIP2 and cAMP-PKA Signal Pathways in Radiation-induced Immune-suppressing Effect显示文摘Objective The purpose of the present study was to observe the changes in CD4+CD25+Nrp1+Treg cells after irradiation with different doses and explore the possible molecular mechanisms involved. Methods ICR mice and mouse lymphoma cell line(EL-4 cells) was used. The expressions of CD4,CD25,Nrp1,calcineurin and PKC-α were detected by flow cytometry. The expressions of TGF-β1,IL-10,PKA and cAMP were estimated with ELISA. Results At 12 h after irradiation,the expression of Nrp1 increased significantly in 4.0 Gy group,compared with sham-irradiation group(P<0.05) in the spleen and thymus,respectively,when ICR mice received whole-body irradiation(WBI). Meanwhile the synthesis of Interleukin 10(IL-10) and transforming growth factor-β1(TGF-β1) increased significantly after high dose irradiation(HDR)(> or = 1.0 Gy). In addition,the expression of cAMP and PKA protein increased,while PKC-α,calcineurin decreased at 12h in thymus cells after 4.0 Gy X-irradiation. While TGF-β1 was clearly inhibited when the PLC-PIP2 signal pathway was stimulated or the cAMP-PKA signal pathway was blocked after 4.0 Gy X-irradiation,this did not limit the up-regulation of CD4+CD25+Nrp1+Treg cells after ionizing radiation. 4A cross-sectional study on compliance with topical glaucoma medication and its associated socioeconomic burden for a Chinese population显示文摘AIM: To estimate the overall drug compliance for local Chinese glaucoma patients on long-term topical treatment.METHODS: This was a retrospective cross-sectional study.Fifty-seven primary glaucoma patients from the subspecialty clinic of a publicly-funded tertiary care hospital in Hong Kong completed a questionnaire on compliance with topical glaucoma medication,attitude towards glaucoma and therapy and vision related quality of life.Noncompliance was defined as reporting missing more than or equal to 10% of the prescribed topical glaucoma medication during the 2wk immediately prior to the consultation.Relationships between noncompliance and demographics,attitude,disease and treatment status was studied.Cost was estimated with quality of life and direct medical cost involved with noncompliance.Multivariable logistic regression on noncompliance was performed on selected factors.RESULTS: Compliance was calculated as 75%(95% CI: 64%-87%) among 57 subjects(mean age 69 y,female 51%).No statistical significant relationship was established between noncompliance and any single factors or outcomes.Age(P=0.048) and forgetfulness(P=0.064) were found to be marginally significant predictive factors on noncompliance in multivariable logistic regression.Noncompliance might be related(P=0.130) to poorer self-rated vision-associated quality of life.The societal cost of noncompliance was estimated to be over 2510 life-years and US$ 3.7 million territory-wide.CONCLUSION: The compliance of Chinese glaucoma patients in Hong Kong is comparable to other parts in the world,and carries detrimental impacts on individual and societal levels.Age and forgetfulness are two possible independent predictors for noncompliance.Man-Hin Lui Jason Chun Ho Lam Yi Lee Kwong Tak Siu Wong Pak Long Cheung Sherri Shun Yan Lai Pak Lun Yuen Wai Yiu Lam Ho Yan Chan Yiu Fai Wong Jimmy Shiu Ming Lai Kendrick Co Shih 2017International Journal of Ophthalmology(English edition)2017,10,2:5

6Successive Interference Cancellation and Alignment in K-User MIMO Interference Channels with Partial Unidirectional Strong Interference显示文摘Recently cellular networks have been densely and heterogeneously deployed indoors and outdoors to expand the network capacity,and thus the in-building propagation loss and the transmit power diversity of access points will exacerbate link heterogeneity and result in partial unidirectional strong interference.To make full use of the strong interference feature,we propose the successive interference cancellation and alignment(SICA)scheme in the K-user interference channel with partial unidirectional strong interference.SICA is designed to transmit two kinds of data streams simultaneously,the alignment streams and superposition streams.The alignment streams will follow the interference alignment criterion to maintain the optimal degrees of freedom(DoF)performance;the superposition streams are handled via successive interference cancellation at all the strongly interfered receivers to improve the overall achievable rate.The joint transceiver designs for SICA is modeled as a weighted sum rate(WSR)maximization problem,and then can be alternately solved for a local optimum according to the optimality equivalence between WSR and its corresponding weighted mean square error(WMMSE)problem.Simulation results have confirmed the sum rate improvement and DoF optimality of the proposed SICA scheme.Long Suo Hongyan Li Shun Zhang Jiandong Li 2022China Communications2022,19,2:2

9Real-time measurements of PM_(2.5),PM_(10-2.5),and BC in an urban street canyon显示文摘A continuous dichotomous beta gauge monitor was used to characterize the hourly content of PM_(2.5).PM_(10-2.5).and Black Carbon(BC)over a 12-month period in an urban street canyon of Hong Kong.Hourly vehicle counts for nine vehicle classes and meteorological data were also recorded.The average weekly cycles of PM_(2.5),PM_(10-2.5).and BC suggested that all species are related to traffic,with high concentrations on workdays and low concentrations over the weekends.PM_(2.5) exhibited two comparable concentrations at 10:00-11:00(63.4 μg/m^3) and 17:00-18:00(65.0 μg/m^3) local time(LT) during workdays,corresponding to the hours when the numbers of diesel-fueled and gasoline-fueled vehicles were at their maximum levels:3179 and 2907 h^(-1),respectively.BC is emitted mainly by diesel-fueled vehicles and this showed the highest concentration(31.2 μg/m^3) during the midday period(10:00-11:00 LT) on workdays.A poor correlation was found between PM_(2.5) concentration and wind speed(R = 0.51,P-value> 0.001).In contrast,the concentration of PM_(10-2.5) was found to depend upon wind speed and it increased with obvious statistical significance as wind speed increased(R = 0.98,P-value< 0.0001).Yan Cheng Shun Cheng Lee Yuan Gao Long Cui Wenjing Deng Junji Cao Zhenxing Shen Jian Sun 2015Particuology2015,13,3:1

13Roles of FERONIA-like receptor genes in regulating grain size and quality in rice显示文摘Grain yield and quality are critical factors that determine the value of grain crops.In this study,we analyzed the functions of 12 FERONIA-like receptor(FLR)family members in rice and investigated their effects on grain size and quality.We found that FLR1,FLR2 and FLR8 negatively regulated grain size,and FLR15 positively regulated grain size.flr1 mutants had a higher cell number and an accelerated rate of grain filling compared to wild-type plants,which led to grains with greater widths.A mechanism underlying the regulation of grain size by FLR1 is that FLR1 is associated with OsRac1 Rho-like GTPase,a positive regulator of grain size.Regarding grain quality,the flr1 mutant had a higher percentage of chalkiness compared with wild-type plants,and seeds carrying mutations in flr3 and flr14 had endosperms with white floury cores.To elucidate the possible mechanism underlying this phenomenon,we found that FLR1 was constitutively expressed during endosperm development.RNA-seq analysis identified 2,367 genes that were differentially expressed in the flr1 mutant,including genes involved in starch and sucrose metabolism and carbon fixation.In this study,we identified the roles played by several FLR genes in regulating grain size and quality in rice and provided insights into the molecular mechanism governing the FLR1-mediated regulation of grain size.Long Wang Dandan Wang Zhuhong Yang Shun Jiang Jianing Qu Wei He Zhenming Liu Junjie Xing Youchu Ma Qinlu Lin Feng Yu 2021Science China(Life Sciences)2021,64,2:1
