| 1 | Identification of seed-specific promoter nap300 and its comparison with 7S promoter显示文摘By fusing seed-specific promoter nap300 with β-glucuronidase gene, it was found that this about 300bp DNA fragment was sufficient to direct seed-specific gene expression. The substitution mutation in both distB and proxB elements had a little effect on the expression efficiency and almost no effect on the organ-specific expression pattern. In the experiment designed to compare nap300 with 7S promoter, the result showed that tissue specificity for nap300 was higher than that for 7S, and its expression level was lower than 7S's. | 3 | Lignin biosynthesis by suppression of two O-methyl-transferases显示文摘Caffeic acid O-methyltransferase (COMT) and caffeoyl-CoA-3-O-methyltransferase (CCoAOMT) genes encode two methyltransferases at different substrate levels in lignin biosynthesis. We constructed two single antisense expression vectors containing the cDNA of COMT or CCoAOMT genes and one dual antisense expression vector containing cDNAs of both OMTs genes. The antisense constructs were transferred into tobacco mediated by Agrobac-terium tumefacience. PCR-Southern analysis indicated that antisense cDNAs had been integrated into the genome of the transgenic tobacco. The antisense genes were also expressed at transcriptional level displayed by Northern dot analysis. Klason lignin assay of 3-month-old transgenic tobaccos showed that repression in COMT or CCoAOMT alone could result in reduction of lignin content, more reduction was caused by suppression in CCoAOMT than in COMT. | 4 | Characterization of 500 Chinese patients with cervical esophageal cancer by clinicopathological and treatment outcomes显示文摘Objective: There are no comprehensive studies on survival outcomes and optimal treatment protocols for cervical esophageal cancer(CEC), due to its rare clinical prevalence. Our objective was to determine the relationship between pathological characteristics, treatment protocols, and survival outcomes in Chinese CEC patients.Methods: A total of 500 Chinese CEC patients were selected from our 500,000 esophageal and gastric cardia carcinoma database(1973–2018). There were two main groups: patients treated with surgery, and patients receiving non-surgical treatments(radiotherapy, radiochemotherapy, and chemotherapy). The Chi-square test and Kaplan–Meier method were used to compare the continuous variables and survival.Results: Among the 500 CEC patients, 278(55.6%) were male, and the median age was 60.9 ± 9.4 years. A total of 496 patients(99.2%) were diagnosed with squamous cell carcinoma. In 171(34.2%) patients who received surgery, 22(12.9%) had undergone laryngectomy. In 322(64.4%) patients who received non-surgical treatments, 245(76.1%) received radiotherapy. Stratified survival analysis showed that only T stage was related with survival outcomes for CEC patients in the surgical group, and the outcomes between laryngectomy and non-laryngectomy patients were similar. It was noteworthy that the 5-year survival rate was similar in CEC patients among the different groups treated with surgery, radiotherapy, chemotherapy, or radiochemotherapy(P = 0.244). Conclusions: The CEC patients had similar survival outcomes after curative esophagectomy and radiotherapy, including those with or without total laryngectomy. | 5 | Pulping performance of transgenic poplar with depressed Caffeoyl-CoA O-methyltransferase显示文摘This paper evaluated pulping performance of 3-year-old field-grown transgenic poplar (Populus tremula × Populus alba). The transgenic poplar with anti-sense CCoAOMT had an about 13% decreased lignin content, in which a slight increment was found in S/G ratio. Chemical analysis showed that the transgenic poplar had significantly less benezene-ethanol extractive than that of control wood, but no significant differences were found in contents of ash, cold water extractive, hot water extractive, 1% NaOH extractive, holocellulose, pentosans and cellulose. Fiber assay demonstrated that down-regulation of CCoAOMT expression improved the fiber quality in transgenic poplar. DNA sequencing results show that phbA cloned in pBluescriptSK^+ has an identical sequence with that reported previously except for one base pair. The plant constitutive expression vector has been constructed and tobacco has been transformed in order to examine the phbA gene function and the efficiency of ctp gene product. SDS-polyacrylamide gel electrophoresis result shows that the ctp gene product could direct foreign protein into plastid efficiently and phbA gene could be translated into corresponding protein with correct size. | 16 | Molecular characterization of OsPRP1 from rice,which is expressed preferentially in anthers显示文摘A proline-rich protein-encoding cDNA encoded by a rice gene, OsPRP1, was isolated by PCR-mediated RNA subtraction hybridization strategy and rapid amplification of cDNA ends. The deduced protein consists of 224 amino acids with the highest level of proline residue (14.29%). Following the putative signal peptide, OsPRP1 contains two structural domains, of which the N-terminal domain lacks Pro-rich repetitive sequences, and the C-terminal domain has two repetitive proline-rich sequences of 18 amino acid residues with PEPK motifs. Southern blot and sequence analysis show that OsPRP1 exists as four copies in rice genome and is localized in rice chromosome 10. RT-PCR experiments reveal that OsPRP1 is expressed preferentially in spikelets and buds with lower levels in roots and leaves. In situ hybridization indicates that OsPRP1 transcripts are present at high levels in pollen mother cells (PMCs), meiotic PMCs, tapetal cells and vascular bundle cells of flower organs. The expression of OsPRP1 in anthers has temporal specificity. The transcripts are accumulated at high levels in PMCs, at the highest levels in meiotic PMCs and at undetectable levels in uninucleate pollen. | 17 | Genetic polymorphisms in genes regulating cell death and prognosis of patients with rectal cancer receiving postoperative chemoradiotherapy显示文摘Objective:The identification of biomarkers for predicting chemoradiotherapy efficacy is essential to optimize personalized treatment.This study determined the effects of genetic variations in genes involved in apoptosis,pyroptosis,and ferroptosis on the prognosis of patients with locally advanced rectal cancer receiving postoperative chemoradiotherapy(CRT).Methods:The Sequenom MassARRAY was used to detect 217 genetic variations in 40 genes from 300 patients with rectal cancer who received postoperative CRT.The associations between genetic variations and overall survival(OS)were evaluated using hazard ratios(HRs)and 95%confidence intervals(CIs)computed using a Cox proportional regression model.Functional experiments were performed to determine the functions of the arachidonate 5-lipoxygenase(ALOX5)gene and the ALOX5 rs702365 variant.Results:We detected 16 genetic polymorphisms in CASP3,CASP7,TRAILR2,GSDME,CASP4,HO-1,ALOX5,GPX4,and NRF2 that were significantly associated with OS in the additive model(P<0.05).There was a substantial cumulative effect of three genetic polymorphisms(CASP4 rs571407,ALOX5 rs2242332,and HO-1 rs17883419)on OS.Genetic variations in the CASP4 and ALOX5 gene haplotypes were associated with a higher OS.We demonstrated,for the first time,that rs702365[G]>[C]represses ALOX5 transcription and corollary experiments suggested that ALOX5 may promote colon cancer cell growth by mediating an inflammatory response.Conclusions:Polymorphisms in genes regulating cell death may play essential roles in the prognosis of patients with rectal cancer who are treated with postoperative CRT and may serve as potential genetic biomarkers for individualized treatment. | Hongxia Chen Luxi Yin Jie Yang Ningxin Ren Jinna Chen Qixuan Lu Ying Huang Yanru Feng Weihu Wang Shulian Wang Yueping Liu Yongwen Song Yexiong Li Jing Jin Wen Tan Dongxin Lin | 2023 | Cancer Biology & Medicine2023,20,4: | 0 |

| 19 | Regeneration of Fe_(2)O_(3)-based high-temperature coal gas desulfurization sorbent in atmosphere with sulfur dioxide显示文摘Regeneration of a high-temperature coal gas desulfurization sorbent is a key technology in its industrial applications.A Fe_(2)O_(3)-based high-temperature coal gas desulfurizer was prepared using red mud from steel factory.The influences of regeneration temperature,space velocity and regeneration gas concentration in SO_(2) atmosphere on regeneration performances of the desulfurization sorbent were tested in a fixed bed reactor.The changes of phase and the composition of the Fe_(2)O_(3)-based high-temperature coal gas desulfurization sorbent before and after regeneration were examined by X-ray diffraction(XRD)and X-ray Photoelectron spectroscopy(XPS),and the changes of pore structure were characterized by the mercury intrusion method.The results show that the major products are Fe3O4 and elemental sulfur;the influences of regeneration temperature,space velocity and SO_(2) concentration in inlet on regeneration performances and the changes of pore structure of the desulfurization sorbent before and after regeneration are visible.The desulfurization sorbent cannot be regenerated at 500℃ in SO_(2) atmosphere.Within the range of 600℃-800℃,the time of regeneration becomes shorter,and the regeneration conversion increases as the temperature rises.The time of regeneration also becomes shorter,and the elemental sulfur content of tail gas increases as the SO_(2) concentration in inlet is increased.The increase in space velocity enhances the reactive course;the best VSP is 6000 h^(-1) for regeneration conversion.At 800℃,20 vol-%SO_(2) and 6000 h^(-1),the regeneration conversion can reach nearly to 90%. | Ruizhuang ZHAO Ju SHANGGUAN Yanru LOU Jin SONG Jie MI Huiling FAN | 2010 | Frontiers of Chemical Science and Engineering2010,4,4: | 0 |
