2Long-term clinical observation of patients with acute and chronic complete spinal cord injury after transplantation of Neuro Regen scaffold显示文摘Spinal cord injury(SCI)often results in an inhibitory environment at the injury site.In our previous studies,transplantation of a scaffold combined with stem cells was proven to induce neural regeneration in animal models of complete SCI.Based on these preclinical studies,collagen scaffolds loaded with the patients’own bone marrow mononuclear cells or human umbilical cord mesenchymal stem cells were transplanted into SCI patients.Fifteen patients with acute complete SCI and 51 patients with chronic complete SCI were enrolled and followed up for 2 to 5 years.No serious adverse events related to functional scaffold transplantation were observed.Among the patients with acute SCI,five patients achieved expansion of their sensory positions and six patients recovered sensation in the bowel or bladder.Additionally,four patients regained voluntary walking ability accompanied by reconnection of neural signal transduction.Among patients with chronic SCI,16 patients achieved expansion of their sensation level and 30 patients experienced enhanced reflexive defecation sensation or increased skin sweating below the injury site.Nearly half of the patients with chronic cervical SCI developed enhanced finger activity.These long-term follow-up results suggest that functional scaffold transplantation may represent a feasible treatment for patients with complete SCI.Fengwu Tang Jiaguang Tang Yannan Zhao Jiaojiao Zhang Zhifeng Xiao Bing Chen Guang Han Na Yin Xianfeng Jiang Changyu Zhao Shixiang Cheng Ziqiang Wang Yumei Chen Qiaoling Chen Keran Song Zhiwei Zhang Junjie Niu Lingjun Wang Qin Shi Liang Chen Huilin Yang Shuxun Hou Sai Zhang Jianwu Dai 2022Science China(Life Sciences)2022,65,5:3
3Loss of Lkb1 impairs Treg function and stability to aggravate graft-versus-host disease after bone marrow transplantation显示文摘Accumulating evidence suggests that a reduction in the number of Foxp3^(+) regulatory T cells(Tregs)contributes to the pathogenesis of acute graft-versus-host disease(aGVHD),which is a major adverse complication that can occur after allogeneic hematopoietic stem cell transplantation(allo-HSCT).However,the precise features and mechanism underlying the defects in Tregs remain largely unknown.In this study,we demonstrated that Tregs were more dramatically decreased in bone marrow compared with those in peripheral blood from aGVHD patients and that bone marrow Treg defects were negatively associated with hematopoietic reconstitution.Tregs from aGVHD patients exhibited multiple defects,including the instability of Foxp3 expression,especially in response to IL-12,impaired suppressor function,decreased migratory capacity,and increased apoptosis.Transcriptional profiling revealed the downregulation of Lkb1,a previously identified critical regulator of murine Treg identity and metabolism,and murine Lkb1-regulated genes in Tregs from aGVHD patients.Foxp3 expression in human Tregs could be decreased and increased by the knockdown and overexpression of the Lkb1 gene,respectively.Furthermore,a loss-of-function assay in an aGVHD murine model confirmed that Lkb1 deficiency could impair Tregs and aggravate disease severity.These findings reveal that Lkb1 downregulation contributes to multiple defects in Tregs in human aGVHD and highlight the Lkb1-related pathways that could serve as therapeutic targets that may potentially be manipulated to mitigate aGVHD.Xiuhua Su Qianqian Wang Wei Guo Xiaolei Pei Qing Niu Maolan Liu Yuanyuan Liu Song Chen Sizhou Feng Yi He Donglin Yang Rongli Zhang Qiaoling Ma Weihua Zhai Aiming Pang Jialin Wei Yong Huang Yuechen Luo Mingzhe Han Xiaoming Feng Erlie Jiang 2020Cellular & Molecular Immunology2020,17,5:3
4Targeted therapeutics and novel signaling pathways in nonalcohol-associated fatty liver/steatohepatitis(NAFL/NASH)显示文摘Non-alcohol-associated fatty liver/steatohepatitis(NAFL/NASH)has become the leading cause of liver disease worldwide.NASH,an advanced form of NAFL,can be progressive and more susceptible to developing cirrhosis and hepatocellular carcinoma.Currently,lifestyle interventions are the most essential and effective strategies for preventing and controlling NAFL without the development of fibrosis.While there are stll limited appropriate drugs specifically to treat NAFL/NASH,growing progress is being seen in elucidating the pathogenesis and identifying therapeutic targets.In this review,we discussed recent developments in etiology and prospective therapeutic targets,as well as pharmacological candidates in pre/clinical trials and patents,with a focus on diabetes,hepatic lipid metabolism,inflammation,and fibrosis.Importantly,growing evidence elucidates that the disruption of the gut-liver axis and microbederived metabolites drive the pathogenesis of NAFL/NASH.Extracellular vesicles(EVs)act as a signaling mediator,resulting in lipid accumulation,macrophage and hepatic stellate cell activation,further promoting inflammation and liver fibrosis progression during the development of NAFL/NASH.Targeting gut microbiota or EVs may serve as new strategies for the treatment of NAFL/NASH.Finally,other mechanisms,such as cell therapy and genetic approaches,also have enormous therapeutic potential.Incorporating drugs with different mechanisms and personalized medicine may improve the efficacy to better benefit patients with NAFL/NASH.Xiaohan Xu Kyle L.Poulsen Ljuan Wu Shan Liu Tatsunori Miyata Qiaoling Song Qingda Wei Chenyang Zhao Chunhua Lin Jinbo Yang 2022Signal Transduction and Targeted Therapy2022,7,9:2
5STX5 Inhibits Hepatocellular Carcinoma Adhesion and Promotes Metastasis by Regulating the PI3K/mTOR Pathway显示文摘Background and Aims:Syntaxin 5(STX5)is a member of the syntaxin or target-soluble SNAP receptor(t-SNARE)fam-ily and plays a critical role in autophagy.However,its function and molecular mechanism in tumor cell migration are still un-known.The role of STX5 in influencing hepatocellular carci-noma(HCC)is an important topic in our research.Methods:By using quantitative reverse transcription polymerase chain reaction(qPCR),western blotting,and immunohistochemical analysis of RNA and protein in tissues,we comprehensively evaluated data sets from public databases and clinical patient cohorts for STX5.The correlation of STX5 expression with the clinicopathological characteristics of HCC patients were assessed.In addition,we predicted signal pathways from dif-ferentially expressed genes(DEGs)and the Cancer Genome Atlas(TCGA)databases,and confirmed the prediction using integrated transcriptome and RNA-seq.We further investi-gated the underlying mechanisms of STX5 in the migration and adhesion of HCC cells both in vitro and in vivo.Results:In the TCGA dataset and our patient cohort,STX5 levels were significantly higher in HCC tissues than in adjacent normal liver tissues.At the same time,high expression of STX5 pre-dicted worse prognosis in patients with liver cancer.High ex-pression of STX5 indicates the decrease of adhesion and the increase of migration of HCC cells,and the conversion of epi-thelial-mesenchymal transition(EMT)in vitro via PI3K/mTOR pathway activation.Conversely,when Sirolimus,a phospho-inositide 3-kinase(PI3K)/AKT/mechanistic target of rapa-mycin(mTOR)inhibitor acts on cells simultaneously,STX5 overexpression-mediated enhancement of HCC metastasis is reversed.Double-negative regulation of STX5 and mTOR further enhanced the inhibitory effect of STX5 on HCC me-tastasis.In vivo,STX5 knockdown inhibited the metastasis of HCC cells.Conclusions:Our study demonstrates a novel research result that STX5 promotes HCC metastasis through PI3K/mTOR pathway.We believe that combined inhibition of STX5 and mTOR is a potential treatment for effectively pro-longing patient survival and inhibiting HCC metastasis.Bin Zhang Ziyin Zhao Youpeng Wang Tingting Guo Mingyang He Ge Guan Pai Peng Jinzhen Cai Bingyuan Zhang Xutao Liu Qiaoling Song 2023Journal of Clinical and Translational Hepatology2023,11,3:1

8E2F is required for STAT3-mediated upregulation of cyclin B1 and Cdc2 expressions and contributes to G2–M phase transition显示文摘Activation of transcription factor STAT3 is involved in cell proliferation,differentiation,and cell survival.Constitutive activation of STAT3 pathway has been associated with the oncogenesis of various types of cancers.It has been reported that STAT3 plays a key role in the G1 to S phase cell cycle transition induced by the cytokine receptor subunit gp130,through the upregulation of cyclins D1,D2,D3,A,and Cdc25A and the concomitant downregulation of p21 and p27.However,its role in mediating G2–M phase transition has not been studied.The cyclin B1/Cdc2 complex is widely accepted as the trigger of mitosis in all organisms and is believed to be necessary for progression through S phase and keep active during the G2–M transition and progression.In the present study,we found that activation of STAT3 stimulates cyclin B1 and Cdc2 expressions.Deletion and site-directed mutations on cyclin B1 and Cdc2 promoters indicated that E2F element mediates the upregulation of these two promoters in a STAT3-dependent manner.The findings reported here demonstrated that STAT3 participates in modulating G2–M phase checkpoint by regulating gene expressions of cyclin B1 and Cdc2 via E2F.Jingjie Sun Yuping Du Qiaoling Song Jing Nan Peizhu Guan Jihui Guo Xiao Wang Jinbo Yang Chenyang Zhao 2019Acta Biochimica et Biophysica Sinica2019,51,3:1

11Overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films with enhanced electrocatalytic ability for rutin and luteolin determination显示文摘In this study, a simple and effective method was proposed to improve the electrocatalytic ability of overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films modified on glassy carbon electrode for rutin and luteolin determination. The composite electrode was prepared by cyclic voltammetry copolymerization with LiClO_(4)-water as the supporting electrolyte. The peak current of rutin and luteolin on the composite electrode gradually decreased or even disappeared with the increase in the positive potential limit. After incubation in NaOH–ethanol solution with a volume ratio of 1:1, the composite electrodes prepared at positive potential limit greater than 1.5 V exhibited enhanced differential pulse voltammetry peak currents, reduced charge transfer resistance, larger effective specific surface area and higher electron transfer rate constant. The composite electrode prepared in the potential range of 0–1.7 V showed optimal electrocatalytic performance. The X-ray photoelectron spectroscopy results indicated that the content of –SO_(2)/–SO and –C=N– groups in the composite film increased significantly after incubation. Further, the Raman spectra and Fourier transform infrared spectra revealed that the thiophene ring structure changed from benzene-type to quinone-type, and the quinone-type pyrrole ring was formed. 13Roles of abscisic acid and gibberellins in maintaining primary and secondary dormancy of Korean pine seeds显示文摘Primary dormancy of seeds of Korean pine(Pinus koraiensis Sieb.et Zucc.)after dispersal in the autumn and the induction of secondary dormancy the fi rst summer following seed dispersal limit the regeneration of mixed broadleaved Korean pine forests in Northeast China.This study was to determine how changes in the levels of abscisic acid(ABA)and gibberellic acid(GA)maintain primary and secondary dormancy of Korean pine seeds under germination conditions.We transferred seeds with one of fi ve primary dormancy states or three secondary dormancy states to germination conditions and measured changes in the levels of ABA,GA 1+3(GA 1 and GA 3)and GA 4+7(GA 4 and GA 7)in the seed coat,megagametophyte and embryo during incubation.Seed coat ABA levels in primary dormant seeds(PDS)and ABA levels in various parts of secondary dormant seeds(SDS)gradually declined during incubation but were still higher than in seeds for which dormancy was progressively released.GA 4+7 and GA 1+3 levels in embryos greatly decreased 35%and 24%,respectively,during incubation of SDS,and thus,the ratio of ABA to GA 4+7 in embryos and megagametophytes signifi cantly increased.The ratio of ABA to GA 1+3 in various parts of SDS increased slightly during incubation.In contrast,in seeds for which secondary dormancy was already released,GA 4+7 and GA 1+3 levels in the embryo,GA 4+7/ABA ratio in the embryo and seed coat,and the GA 1+3/ABA in the embryo and megagametophyte signifi cantly increased during incubation.There was no trend in the changes in the levels of ABA,GA 4+7 or GA 1+3 in embryos and megagametophytes of PDS or the levels of GA 4+7 or GA 1+3 in megagametophytes of SDS during incubation.The results suggest that high ABA levels in the seed coat maintain primary dormancy of Korean pine seeds.Maintenance of secondary dormancy involves a reduction of GA 4+7,GA 1+3,GA 4+7/ABA,and GA 1+3/ABA and the retention of high ABA levels.Yuan Song Jiaojun Zhu Qiaoling Yan 2020Journal of Forestry Research2020,31,6:0
