| 1 | Grazing alters environmental control mechanisms of evapotranspiration in an alpine meadow of the tibetan Plateau显示文摘Aims Evapotranspiration(Et)is an important component of the terrestrial water cycle and is easily affected by external disturbances,such as climate change and grazing.Identifying Et responses to grazing is instructive for determining grazing activity and informative for understanding the water cycle.Methods this study utilized 2 years(2014 and 2017)of eddy covariance data to test how grazing regulated Et for an alpine meadow ecosystem on the tibetan Plateau(tP)by path analysis.Important Findings Radiation dominated Et with a decision coefficient of 64-74%.the soil water content(SWc)worked as the limiting factor in the fenced site.However,in the grazing site,the limiting factor was the vapor pressure deficit(VPD).Grazing had large effects on Et because it greatly affected the water conditions.the SWc and VPD were enhanced by 14.63%and 4.36%in the grazing site,respectively.therefore,sufficient water was supplied to Et,especially during drought,and strengthened the transpiration pull.As a result,a favorable micrometeorological environment was created for Et.Grazing shifted the limiting factor of Et from the SWc to VPD,which weakened the limiting effect of the water conditions on Et and advanced the Et peak time.In addition,grazing altered the compositions of Et by changing the community structure,which directly resulted in an increased Et.In summary,grazing enhanced Et through altering the community structure and micrometeorological environments.the findings of this study further improve our understanding of the driving mechanisms of grazing on Et and will improve our predictions for the global water cycle. | Tingting An Mingjie Xu Tao Zhang Chengqun Yu Yingge Li Ning Chen Jiaxing Zu Junxiang Li Juntao Zhu Yi Sun Tingting Zhao Guirui Yu | 2019 | Journal of Plant Ecology2019,12,5: | 3 |
| 2 | Protection of Immuno-Compromised Mice from Lethal Infection of Klebsiella pneumonia by rAAV2-BPI23-Fcγ1 Gene Transfer显示文摘In previous research, chimerical BPI23-Fcγ1 gene which consisted of human bactericidal/permeability increasing protein (BPI) gene of encoding the functional N terminus (amino acid residues 1 to 199) of human BPI and Fcγ1 gene of encoding the Fc segment of human immunoglobulin G1 was successfully reconstructed within a recombinant adeno-associated virus serotype 2 (rAAV2) vector as rAAV2-BPI23-Fcγ1. Here, to evaluate the potentiality of applying gene therapy to gram negative bacterial (GNB) infection in high-risk patients, we investigated protection of immuno-compromised mice and immunocompetent mice from challenge with minimal lethal dose (MLD) Klebsiella pneumonia infection after rAAV2-BPI23-Fcγ1 gene transferred. The results showed that the survival rate of rAAV2-BPI23-Fcγ1 transferred immunocompetent mice as well as immuno-compromised mice (40.0% and 44.4%, respectively) were significantly higher than that of corresponding control mice (6.7% and 4.4%, respectively); the bacteria counting, level of endotoxin and proinflammatory cytokines in the rAAV2-BPI23- Fcγ1 transferred immuno-compromised mice were markedly lower than that of rAAV2-EGFP and rAAV2-Null transferred immuno-compromised mice. | 5 | Synergy of in-situ heterogeneous interphases tailored lithium deposition显示文摘The implementation of a robust artificial solid electrolyte interphase(ASEI)to replace the unstable natural SEI can regulate lithium deposition behaviors and avoid the safety hazards caused by dendrites permeation in lithium metal batteries.Despite of devoted efforts in tailoring components of ASEI,the intrinsic mechanism of interfacial synergy within the heterogeneous interphases has not been well elucidated yet.Herein,we show that the lithium plating/striping behaviors can be substantially enhanced(over 900 h with an overpotential of less than 20 mV at 1 mA·cm^(−2)in Li|Li symmetric cells and 146 cycles in anode-free cells)by regulating the heterogeneous interphases.This favorable ASEI composed of LiF and Li_(3)N components can be in-situ generated during cycling by large-scale fabricated fluorinated boron nitride coatings.Further,the synergy of each heterogeneous component within ASEI was explored theoretically and experimentally.Li_(3)N has high adsorption energy and low ion diffusion barrier,which facilitates the transport of lithium ions and avoids its local accumulation to evolve into dendrites.Both the substrate and LiF are interfacially stable with high electron tunneling barriers,preventing the electrolyte decomposition and parasitic reactions.Finally,the high stiffness of the boron nitride also ensures lithium dendrites are suppressed once they grow,providing a stable environment for long-term cycling of lithium metal batteries. | Yinuo Li Anjun Hu Xingdong Gan Miao He Jun Zhu Wei Chen Yin Hu Tianyu Lei Fei Li Yaoyao Li Yuxin Fan Fan Wang Mingjie Zhou An Wen Baihai Li | 2023 | Nano Research2023,16,6: | 0 |

| 7 | SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC. | Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li | 2023 | Signal Transduction and Targeted Therapy2023,8,12: | 0 |
| 8 | An HGF-dependent positive feedback loop between bladder cancer cells and fibroblasts mediates lymphangiogenesis and lymphatic metastasis显示文摘Background Cancer-associated fibroblasts (CAFs) play a vital role in facilitating tumor progression through extensive reciprocal interplay with cancer cells. Tumor-derived extracellular vesicles (EVs) are the critical mediators involved in the crosstalk between cancer cells and stromal cells, contributing to the metastasis of cancers. Yet, the biological mechanisms of tumor-derived EVs in triggering CAFs phenotype to stimulate the lymph node (LN) metastasis of bladder cancer (BCa) are largely unknown. Here, we aimed to explore the effects and molecular mechanisms of tumor-derived EV-mediated CAFs phenotype in regulating BCa LN metastasis. Methods The high-throughput sequencing was utilized to identify the crucial long non-coding RNA (lncRNA) associated with CAF enrichment in BCa. The functional role of the transition of fibroblasts to CAFs induced by LINC00665-mediated EVs was investigated through the in vitro and in vivo assays. Chromatin isolation by RNA purification assays, fluorescence resonance energy transfer assays, cytokine profiling and patient-derived xenograft (PDX) model were performed to explore the underlying mechanism of LINC00665 in the LN metastasis of BCa. Results We found that CAFs are widely enriched in the tumor microenvironment of BCa, which correlated with BCa lymphangiogenesis and LN metastasis. We then identified a CAF-associated long non-coding RNA, LINC00665, which acted as a crucial mediator of CAF infiltration in BCa. Clinically, LINC00665 was associated with LN metastasis and poor prognosis in patients with BCa. Mechanistically, LINC00665 transcriptionally upregulated RAB27B expression and induced H3K4me3 modification on the promoter of RAB27B through the recruitment of hnRNPL. Moreover, RAB27B-induced EVs secretion endowed fibroblasts with the CAF phenotype, which reciprocally induced LINC00665 overexpression to form a RAB27B-HGF-c-Myc positive feedback loop, enhancing the lymphangiogenesis and LN metastasis of BCa. Importantly, we demonstrated that blocking EV-transmitted LINC00665 or HGF broke this loop and impaired BCa lymphangiogenesis in a PDX model.
