1Pilose antler aqueous extract promotes the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells by stimulating the BMP-2/Smad1, 5/Runx2 signaling pathway显示文摘Peptides from Pilose antler aqueous extract(PAAE) have been shown to stimulate the proliferation and differentiation of bone marrow mesenchymal stem cells(BMSCs). However, the underlying molecular mechanisms are not well understood. Here, PAAE was isolated and purified to explore the molecular mechanisms underlying PAAE’s effects on BMSCs as well as its osteoprotective effects in ovariectomized rats. Our results showed that PAAE promoted proliferation and differentiation of BMSCs to become osteoblasts by enhancing ALP activity and increasing extracellular matrix mineralization. The trabecular microarchitecture of ovariectomized rats was also found to be protected by PAAE. Quantitative reverse transcription-polymerase chain reaction(Quantitative RT-PCR) results suggest that PAAE also increased the expression of osteogenic markers including, alkaline phosphatase(ALP), runt-related transcription factor 2(Runx2), osteocalcin(OCN), bone morphogenetic protein-2(BMP-2), and collagen I(COL-I). Immunoblotting results indicated that PAAE upregulated the levels of BMP-2 and Runx2 and was associated with Smad1/5 phosphorylation. PAAE A at the concentration of 200μg·mL^-1 showed the strongest effect on proliferation and osteogenic differentiation of BMSCs after 48 h. Using matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF MS), we identified the molecular weight of PAAE A and found that it is less than 3000 Da and showed several significant peaks. In conclusion, PAAE activates the BMP-2/Smad1, 5/Runx2 pathway to induce osteoblastic differentiation and mineralization in BMSCs and can inhibit OVX-induced bone loss. 2TANDEM and GMAW Twin Wire Welding of Q690 Steel Used in Hydraulic Support显示文摘Compared with using semi-automatic gas shielded arc welding, using automatic TANDEM twin wire welding and twin wire gas metal arc welding (GMAW) to weld Q690 steel, a low-alloy high-strength structural steel used in the hydraulic support in the fully-mechanized mining face, the welding speed, deposition rate, production environment and welding quality can be obviously improved. Compared with GMAW twin wire welding, a refined microstructure in the weld and heat-affected zone (HAZ), narrow HAZ and improved joint strength were achieved with TANDEM on Q690. Also, due to the push-pull pulsed way in TANDEM welding, the droplet transfer, distribution on heat flow and interaction between two arcs were completely different from those in GMAW twin wire system. The heat input of TANDEM is only about 76.6% of GMAW, and correspondingly, the welding speed and welding seam can be obviously improved. The complete oscillation caused by TANDEM pulsed current occurred in the welding pool, which refined the grains in the microstructure. 6Paleogene Tectonic Evolution Controls on Sequence Stratigraphic Patterns in the Central Part of Deepwater Area of Qiongdongnan Basin, Northern South China Sea显示文摘In active rift basins, tectonism is extremely important for sequence stratigraphic patterns, affecting both the sequence architecture and internal makeup. Sequence stratigraphic framework of a Paleogene rift succession in Qiongdongnan Basin, northern South China Sea, was built using seismic profiles, complemented by well logs and cores. One first-order and three second-order sequences were identified on the basis of basin-scale unconformities, and seven third-order sequences are defined by unconformities along the basin margins and correlative conformities within the central basin. Through unconformity analysis and backstripping procedure, the Paleogene synrift tectonic evolution of deepwater area of Qiongdongnan Basin was proved to be episodic, which can be divided into rifting stage-I, rifting stage-II and rifting stage-III. Episodic rifting resulted in the formation of various types of structural slope break belts, which controlled different architectures and internal makeup of sequences. 9Polysaccharides from Tricholoma matsutake and Lentinus edodes enhance 5-fluorouracil-mediated H22 cell growth inhibition显示文摘OBJECTIVE: Few studies have investigated the effects produced by combinations of polysaccharides and chemotherapeutic drugs in cancer treatment.We hypothesized that a combinationo fpolysaccha rides(COP) from Lentinus edodes and Tricholoma matsutake would improve the efficacy of 5-fluorouracil(5-FU)-mediated inhibition of H22 cell growth.METHODS: Mice were injected H22 cells and then treated with either 5-FU, polysaccharides from Tricholoma matsutake(PTM), polysaccharides from Lentinus edodes(PL), PTM+PL, 5-FU+PTM, 5-FU+PL, or 5-FU+COP. The tumor weight and volume,and splenic CD4 + and CD8 + T cell frequencies,were determined. Additionally, splenic natural killer(NK) cell and cytotoxic Tlymphocyte (CTL) activities were assessed and the serum levels of tumor necrosis factor-α(TNF-α),Interleukin-2(IL-2),and Interferon-γ (IFN-γ) were measured. 5-FU, PL, PTM, PTM+PL, 5-FU+PL, and 5-FU+PTM groups, mice treated with 5-FU+COP showed:(a)significantly reduced tumor weight and volume(P<0.05);(b) significantly higher serum levels ofTNF-α,IL-2, and IFN-γ(P<0.05);(c) significantly increased CD4+andCD8+Tcell frequencies in the spleen(P<0.05); and(d) significantly increased splenic NK cell and CTL activities(P<0.05). The tumor weight and volume in mice treated with 5-FU+PL or 5-FU+PTM were significantly reduced compared with mice treated with 5-FU alone(P<0.05). 10Biomass-derived nitrogen-doped hierarchical porous carbon as efficient sulfur host for lithium–sulfur batteries显示文摘Lithium-sulfur(Li-S) battery is a potential energy storage technology with high energy density and low cost. However, the gap between theoretical expectation and practical performance limits its wide implementation. Herein, we report a nitrogen-doped porous carbon derived from biomass pomelo peel as sulfur host material for Li-S batteries. The hierarchical porous architecture and the polar surface introduced by N-doping render a favorable combination of physical and chemical sulfur confinements as well as an expedite electron/ion transfer, thus contributing to a facilitated and stabilized sulfur electrochemistry. As a result, the corresponding sulfur composite electrodes exhibit an ultrahigh initial capacity of 1534.6 mAh g^-1, high coulombic efficiency over 98% upon 300 cycles, and decent rate capability up to 2 C. 12Micro RNA-1290 promotes esophageal squamous cell carcinoma cell proliferation and metastasis显示文摘AIM:To investigate the biological role of mi R-1290 in esophageal squamous cell carcinoma(ESCC) progression and invasion and the underlying mechanism.METHODS:Quantitative real-time polymerase chain reaction(q RT-PCR) was performed to evaluate mi R-1290 expression in ESCC tissue samples.The roles of mi R-1290 in cell proliferation,migration and invasion were identified using mi R-1290 mimic-transfected cells.In addition,the regulatory effect of mi R-1290 on suppressor of cancer cell invasion(SCAI) was evaluated using q RT-PCR,Western blot analysis and a dual luciferase reporter assay.RESULTS:mi R-1290 was significantly upregulated in ESCC tissue samples compared with normal adjacent tissues(9.213 ± 1.150 vs 1.000 ± 0.0),(P < 0.01).Upregulation of mi R-1290 was associated with tumor differentiation(P = 0.021),N classification(P = 0.006) and tumor-node-metastasis stage(P = 0.021) in ESCC patients.Moreover,ectopic mi R-1290 expression potently promoted ESCC cell growth(P < 0.01),migration(P < 0.01) and invasion(P < 0.01) in vitro.mi R-1290 overexpression in ESCC cell lines decreased SCAI expression at the translational level and reduced SCAI-driven luciferase-reporter activity(P < 0.01).CONCLUSION:Our findings suggested that mi R-1290 may play an oncogenic role in cellular processes of ESCC.Ming Li Xiao-Yan He Zhi-Mei Zhang Shuo Li Li-Hua Ren Ri-Sheng Cao Ya-Dong Feng Yin-Lin Ji Ye Zhao Rui-Hua Shi 2015World Journal of Gastroenterology2015,21,11:13

14Uptake of HIV Self-testing among Men Who have Sex with Men in Beijing, China: a Cross-sectional Study显示文摘Objective To examine HIV self-testing uptake and its determinates among men who have sex with men(MSM) in Beijing, China. Methods A cross-sectional online survey was conducted in Beijing, China in 2016. Participants were users of a popular Chinese gay networking application and had an unknown or negative HIV status. Univariate and multivariate logistic regression analyses were conducted to examine factors associated with HIV self-testing based on adjusted odds ratio(AOR) and 95% confidence interval(CI). Results Among the 5,996 MSM included in the study, 2,383(39.7%) reported to have used HIV self-testing kits. Willingness to use an HIV self-test kit in the future was expressed by 92% of the participants. High monthly income(AOR = 1.49; CI = 1.10-2.02; P = 0.010), large number of male sex partners(≥ 2: AOR = 1.24; CI = 1.09-1.43; P = 0.002), sexual activity with commercial male sex partners(≥ 2: AOR = 1.94; CI = 1.34-2.82; P = 0.001), long-term drug use(AOR = 1.42; CI = 1.23-1.62; P < 0.001), and long-term HIV voluntary counseling and testing(VCT) attendance(AOR = 3.62; CI = 3.11-4.22; P < 0.001) were all associated with increased odds of HIV self-testing uptake. Conclusion The nearly 40% rate of HIV self-testing uptake among MSM in our sample was high. In addition, an over 90% willingness to use kits in the future was encouraging. 18Q-switched and mode-locked Er-doped fiber laser using PtSe2 as a saturable absorber显示文摘We report a passively Q-switched and mode-locked erbium-doped fiber laser(EDFL) based on PtSe_2, a new two-dimensional material, as a saturable absorber(SA). Self-started Q-switching at 1560 nm in the EDFL was achieved at a threshold pump power of 65 mW, and at the maximum pump power of 450 m W, the maximum single Q-switched pulse energy is 143.2 n J. Due to the polarization-dependent characteristics of the PtSe_2-based SA, the laser can be switched from the Q-switched state to the mode-locked state by adjusting the polarization state. A mode-locked pulse train with a repetition rate of 23.3 MHz and a pulse width of 1.02 ps can be generated when the pump power increases to about 80 m W, and the stable mode-locked state is maintained until the pump power reaches its maximum 450 m W. The maximum single mode-locked pulse energy is0.53 nJ. 19ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant. 20Ganoderma lucidum extract ameliorates MPTP-induced parkinsonism and protects dopaminergic neurons from oxidative stress via regulating mitochondrial function, autophagy, and apoptosis显示文摘Neuroprotection targeting mitochondrial dysfunction has been proposed as an important therapeutic strategy for Parkinson's disease. Ganoderma lucidum (GL) has emerged as a novel age nt that protects neurons from oxidative stress. However, the detailed mechanisms underlying GL-induced neuroprotection have not been documented. In this study, we investigated the neuroprotective effects of GL extract (GLE) and the underlying mechanisms in the classic MPTP(1 -methyl-4-phenyl-1,2,3,6- tetrahydropyridine)-induced mouse model of PD. Mice were injected with MPTP to in duce parkins on ism. The n the mice were administered GLE (400 mg kg^-1 d^-1,ig) for 4 weeks. We observed that GLE administration significantly improved locomotor performanee and increased tyrosine hydroxylase expression in the substantia nigra pars compact (SNpc) of MPTP-treated mice. In in vitro study, treatment of neuroblastoma neuro-2a cells with 1 -methyl-4-phenyIpyridinium (MPP^+, 1 mmol/L) caused mitochondrial membrane potential collapse, radical oxygen species accumulation, and ATP depletion. Application of GLE (800 pg/mL) protected n euroblastoma neu ro-2a cells agai nst MPP in suit. Application of GLE also improved mitochondrial movement dysfunction in cultured primary mesencephalic neurons. In addition, GLE counteracted the decline in NIX (also called BNIP3L) expression and increase in the LC3-II/LC3-I ratio evoked by MPP I Moreover, GLE reactivated MPP^+-inhibited AMPK, mTOR, and ULK1. Similarly, GLE was sufficient to counteract MPP1 -induced inhibition of PINK1 and Parkin expression. GLE suppressed MPP^+-induced cytochrome C release and activation of caspase-3 and caspase-9.
