1Effect of Liuweibuqi capsule, a Chinese patent medicine, on the JAK1/STAT3 pathway and MMP9/TIMP1 in a chronic obstructive pulmonary disease rat model显示文摘OBJECTIVE: To observe effect of Liuweibuqi Capsule, a Traditional Chinese Medicine(TCM), on the janus kinase(JAK)/signal transducer and activator of transcription(STAT) pathway and matrix metalloproteinases(MMPs) in a chronic obstructive pulmonary disease(COPD) rat model with lung deficiency in terms of TCM's pattern differentiation.METHODS: Rats were randomly divided into a normal group, model group, Liuweibuqi group, Jinshuibao group, and spleen aminopeptidase group(n=10). Aside from the normal group, all rats were ex-posed to smoke plus lipopolysaccharide tracheal instillation to establish the COPD model with lung deficiency. Models were established after 28 days and then the normal and model groups were given normal saline(0.09 g/kg), Liuweibuqi group was given Liuweibuqi capsule(0.35 g/kg), Jinshuibao group was given Jinshuibao capsules(0.495 g/kg), and the spleen group was given spleen aminopeptidase(0.33 mg/kg), once a day for 30 days. Changes in symptoms, signs, and lung histology were observed. Lung function was measured with a spirometer. Serum cytokines were detected using enzyme-linked immunosorbent assay, and changes in the JAK/STAT pathway, MMP-9, and MMPs inhibitor1(TIMP1) were detected by immunohistochemistry, RT-PCR, and western blotting, respectively.RESULTS: Compared with the normal group, lung tissue was damaged, and lung function was reduced in the model control group. Additionally, the levels of interleukin(IL)-1β, γ interferon(IFN-γ), and IL-6 were higher, while IL-4 and IL-10 were lower in the model control group than those in the normal group. The expressions of JAK1, STAT3, p-STAT3,and MMP-9 m RNA and protein in lung tissue were higher, and TIMP1 m RNA and protein was lower in the model group compared with the normal group.After treatment, compared with the model group,the expression of inflammatory cytokines was lower in each treatment group, and expressions of JAK/STAT pathway, MMPs were lower. Compared with the positive control groups, the Jinshuibao and spleen aminopeptidase groups, lung function was better, and JAK1, STAT3, and p-STAT3 protein were lower and TIMP1 was higher in the Liuweibuqi group.CONCLUSION: Liuweibuqi capsules can improve the symptoms of COPD possibly by regulating the expression of the JAK1/STAT3 pathway and MMP9/TIMP1.Wang Chengyang Li Zegen Liu Xiangguo Peng Qinghe Li Fang Li Da Wang Chuanbo 2015Journal of Traditional Chinese Medicine2015,35,1:23
2ZmCOL3, a CCT gene represses flowering in maize by interfering with the circadian clock and activating expression of ZmCCT显示文摘Flowering time is a trait vital to the adaptation of flowering plants to different environments.Here, we report that CCT domain genes play an important role in flowering in maize(Zea mays L.).Among the 53 CCT family genes we identified in maize, 28 were located in flowering time quantitative trait locus regions and 15 were significantly associated with flowering time, based on candidate-gene association mapping analysis.Furthermore, a CCT gene named ZmCOL3 was shown to be a repressor of flowering.Overexpressing ZmCOL3 delayed flowering time by approximately 4 d, in either long-day or short-day conditions.The absence of one cytosine in the ZmCOL3 3'UTR and the presence of a 551 bp fragment in the promoter region are likely the causal polymorphisms contributing to the maize adaptation from tropical to temperate regions.We propose a modified model of the maize photoperiod pathway, wherein ZmCOL3 acts as an inhibitor of flowering either by transactivating transcription of ZmCCT, one of the key genes regulating maize flowering, or by interfering with the circadian clock.Minliang Jin Xiangguo Liu Wei Jia Haijun Liu Wenqiang Li Yong Peng Yanfang Du Yuebin Wang Yuejia Yin Xuehai Zhang Qing Liu Min Deng Nan Li Xiyan Cui Dongyun Hao Jianbing Yan 2018Journal of Integrative Plant Biology2018,60,6:14
3Effect of Wuziyanzong Pill on sperm quality and calcium ion content in oligoasthenospermia rats显示文摘OBJECTIVE:To study the effect of Wuziyanzong treatment on the sperm quality and content of calcium ions(Ca 2+) in oligoasthenospermia rats.METHODS:A model of oligoasthenospermia was induced in 50 Sprague Dawley rats by treatment with tripterygium glycosides at 30 mg/kg per day for 8 weeks.They were divided randomly into a model group,a positive group(Huangjingzanyu capsule,3.01 g/kg),and low,medium and high dose Wuziyanzong treatment groups(2.30,4.60,9.20 g/kg crude drug respectively) with 10 in each group.Another 10 rats were used as a control group.The rats in the control and model groups were administered distilled water,while the rats in the remaining groups were administered Wuziyanzong for 30 d.The epididymides were removed,spermatozoa recovered and the sperm density and viability were measured.The spermatozoa were purified and the contents of Ca 2+ in the cytoplasm and mitochondria were detected by flow cytometry and atomic absorption spectrometry,respectively.RESULTS:After 8 weeks of treatment with tripterygium glycosides,the sperm density,sperm activity and the Ca 2 + content of spermatozoa in the model rats were all significantly decreased compared with the control group(all P<0.05).After 30 d treatment,the sperm density and activity improved and the Ca 2 + content of sperm were increased significantly in the medium and high dose Wuziyanzong treatment groups in comparison with the model group(all P<0.05).CONCLUSION:The Wuziyanzong treatment increased sperm density,improved sperm viability and enhanced the content of Ca 2+ in the sperm cytoplasm and mitochondria in this rat model of oligoasthenospermia.Tongsheng Wang Jinling Huang Deling Wu Qing Li Xiangguo Liu Han Chen Lei Qiao Dongdong Li Jun Li 2012Journal of Traditional Chinese Medicine2012,32,4:11
4Enhanced Vitamin C Production Mediated by an ABA-Induced PTP-like Nucleotidase Improves Plant Drought Tolerance in Arabidopsis and Maize显示文摘Abscisic acid(ABA)is a key phytohormone that mediates environmental stress responses.Vitamin C,or L-ascorbic acid(AsA),is the most abundant antioxidant protecting against stress damage in plants.How the ABA and AsA signaling pathways interact in stress responses remains elusive.In this study,we characterized the role of a previously unidentified gene,PTPN(PTP-like Nucleotidase)in plant drought tolerance.In Arabidopsis,(AtPTPN was expressed in multiple tissues and upregulated by ABA and drought treatments.Loss-of-function mutants oiAtPTPN were hyposensitive to ABA but hypersensitive to drought stresses,whereas plants with enhanced expression AtPTPN showed opposite phenotypes to.Overexpression of maize PTPN(ZmPTPN)promoted,while knockdown oiZmPTPN inhibited plant drought tolerance,indicating conserved and positive roles of PTPN in plant drought tolerance.We found that both AtPTPN and ZmPTPN release Pi by hydrolyzing GDP/GMP/dGMP/IMP/dIMP,and that AtPTPN positively regulated AsA production via endogenous Pi content control.Consistently,overexpression of VTC2,the rate-limiting synthetic enzyme in AsA biosynthesis,promoted AsA production and plant drought tolerance,and these effects were largely dependent on AtPTPN activity.Furthermore,we demonstrated that the heat shock transcription factor HSFA6a directly binds the AtPTPN promoter and activates AtPTPN expression.Genetic analyses showed that AtPTPN is required for HSFA6a to regulate ABA and drought responses.Taken together,our data indicate that PTPN-mediated crosstalk between the ABA signaling and AsA biosynthesis pathways positively controls plant drought tolerance.Hui Zhang Yanli Xiang Neng He Xiangguo Liu Hongbo Liu Liping Fang Fei Zhang Xiaopeng Sun Delin Zhang Xingwang Li William Terzaghi Jianbing Yan Mingqiu Dai 2020Molecular Plant2020,13,5:7
5Effect of Competitive Adsorption between Sodium Tripolyphosphate and Naphthalene Superplasticizer on Fluidity of Cement Paste显示文摘The adsorption amount, ζ-potential of cement particles and fluidity of cement paste were tested to research the competitive adsorption between naphthalene superplasticizer(FDN) and STPP. The experimental results showed that the presence of STPP could significantly improve the fluidity of cement paste and reduce the fluidity loss with FDN. There existed a competitive adsorption between STPP and FDN. STPP and calcium ions formed complexes; they preferentially adsorbed onto surface of cement particles and preempt adsorption points of FDN; and it reduced adsorption amount of FDN. In the absence of STPP, saturation adsorption amount of FDN was 5.93 mg/g; but when the dosage of STPP was 0.1%, it reduced to 4.3 mg/g(about 72.5%). The adsorption amount of FDN was reduced by STPP, but ζ-potential of cement particles enhanced and fluidity of cement paste increased because of strong negative charge effect of the complexes. Adsorption of the complexes would delay Ca2+ into liquid and inhibit formation of active adsorption points. Then, content of FDN in liquid increased with the addition of STPP and ζ-potential of cement particles became stable. 7Properties and Hydration Mechanism on High-strength Anchorage Grouting Material for Highway Slope显示文摘The rheological and mechanical properties of high-strength anchorage grouting materials for highway slope were investigated to optimize the mix proportion. The experimental results showed that the optimized mix proportion of high-strength anchorage grouting material(HAGM) was C3(FA:SP:SF= 1:2:2; AG1:AG2=3:7 and 0.03% FC), which is agreement with the limitation of JCT 986-2005. Moreover, the XRD and FTIR results showed the addition of expansive components was in favor of the formation of ettringite. 8A Three-Dimensional Dynamic Constitutive Model and Its Finite Element Implementation for NiTi Alloy Based on Irreversible Thermodynamics显示文摘In the present work, the governing equations based on theory of irreversible thermodynamics is deduced by introducing two internal variables to characterize the phase transformation and finite plastic deformation evolution for NiTi shape memory alloy. Thus a three-dimensional dynamic constitutive model is established by summarizing the governing equations for phase transformation and plastic deformation under high strain rate loading conditions. By adopting a stress compensation algorithm to update the stress tensor, the phenomenologicalbased constitutive model is embedded into ABAQUS finite element software as user material subroutine with FORTRAN code. Thus the numerical simulation of dynamic responses of NiTi alloy is successfully implemented. The numerical simulation results are in good agreement with the experimental data, validating the feasibility of this proposed model. 9Influence of Nano-SiO2 Addition on Properties of Sulphoaluminate Cement Based Material显示文摘The influences of different nano-SiO_2(NS) contents on the mechanical properties and rheological behavior of sulfoaluminate cement(SAC) based composite materials were studied.Results show that with increasing content of NS,the apparent viscosity,and shearing strength of fresh paste gradually increase but the fluidity decreases.With a dosage of 3.0%NS,the tensile and flexural strengths of mortars at 56 days were increased by 87.0%and 84.6%,respectively,compared with that in the absence of NS,indicating that the toughness of hardened mortars is significantly improved.Besides,the exothermic peaks of hydration are obviously increased and will earlier occur,and the second and the third peaks appear 2.61 hours and 2.56 hours earlier,respectively than that in the absence of NS,and the hydration of SAC before 8 hours is accelerated.The forming mechanism of strengths was revealed by scanning electron microscopy(SEM),hydration heat,X-ray diffraction(XRD) and derivative thermogravimetry(DTG).The micro-aggregate filling effect and nucleation effect at early age and weak pozzolanic effect at late age of NS make the microstructure more compact,which obviously enhances the strength of SAC mortars.马保国 李海南 MEI Junpeng LI Xiangguo 2017Journal of Wuhan University of Technology(Materials Science)2017,32,1:2
10Ultra High Early Strength Self-compacting Mortar Based on Sulfoaluminate Cement and Silica Fume显示文摘The influences of silica fume and aluminum sulfate on hydration process of sulfoaluminate cement were carried out by ring flow,setting time,hydration heat,XRD and DTG analyses.In addition,mortar mixtures with different functional additives have been studied through compressive strength,flexural strength,volume stability at early age and porosity characterization tests.The results show that the addition of silica fume and aluminum sulfate reduces the fluidity and shortens the setting time of sulfoaluminate cement paste,promoting hydration process and increasing hydration products at early age.In the case of appropriate proportion of mortar,the inclusion of hydroxy propyl methyl cellulose,dispersible polypropylene fiber and organic silicon kind of defoamer can control segregation and bleeding,improve mechanical strength and volume stability at early age,and modify the pore distribution of sulfoaluminate cement mortar,respectively.The sulfoaluminate cement mortar can carry out gravitational grouting in the absence of outside force,the compressive strength of 2 hours and 24 hours have reached 26 and 58 MPa respectively,and have good microexpansion and tiny pore distribution characterization.朱艳超 马保国 LI Xiangguo HU Di 2013Journal of Wuhan University of Technology(Materials Science)2013,28,5:1
11Toxicity Test of Wood Vinegar Mixed with Different Fungicides on Rhizoctonia solani显示文摘There are few kinds of fungicides for the prevention and treatment of Rhizoctonia solani,and the excessive use of chemical fungicides can cause many hazards and toxic and side effects,not in line with the requirements of rice planting. The mixed use of wood vinegar and chemical fungicides can reduce the use of chemical fungicides,alleviate pollution and improve rice quality. In this experiment,the Horsfall method was used to study the synergistic inhibiting effect of mixture between wood vinegar and chemical fungicides on the growth of Rhizoctonia solani,as well as the optimal ratio.
