2The R2R3-type MYB transcription factor MdMYB90-like is responsible for the enhanced skin color of an apple bud sport mutant显示文摘The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development stages between the wild-type and the skin color mutant.A novel R2R3-MYB transcription factor,MdMYB90-like,was uncovered as the key regulatory gene for enhanced coloration in the mutant.The expression of MdMYB90-like was 21.3 times higher in the mutant.MdMYB90-like regulates anthocyanin biosynthesis directly through the activation of anthocyanin biosynthesis genes and indirectly through the activation of other transcription factors that activate anthocyanin biosynthesis.MdMYB90-like bound to the promoters of both structural genes(MdCHS and MdUFGT)and other transcription factor genes(MdMYB1 and MdbHLH3)in the yeast one-hybrid system,electrophoretic mobility shift assay,and dual-luciferase assay.Transgenic analysis showed that MdMYB90-like was localized in the nucleus,and its overexpression induced the expression of other anthocyanin-related genes,including MdCHS,MdCHI,MdANS,MdUFGT,MdbHLH3,and MdMYB1.The mutant had reduced levels of DNA methylation in two regions(−1183 to−988 and−2018 to−1778)of the MdMYB90-like gene promoter,which might explain the enhanced expression of the gene and the increased anthocyanin content in the mutant apple skin.Chao Sun Chunming Wang Wang Zhang Shuai Liu Weiyao Wang Xinyi Yu Tao Song Maxwell Yu Weichang Yu Shenchun Qu 2021Horticulture Research2021,8,1:6
3Novel slack-based robust scheduling rule for a semiconductor manufacturing system with uncertain processing time显示文摘The NP-hard scheduling problems of semiconductor manufacturing systems(SMSs) are further complicated by stochastic uncertainties. Reactive scheduling is a common dynamic scheduling approach where the scheduling scheme is refreshed in response to real-time uncertainties. The scheduling scheme is overly sensitive to the emergence of uncertainties because the optimization of performance(such as minimum make-span) and the system robustness cannot be achieved simultaneously by conventional reactive scheduling methods. To improve the robustness of the scheduling scheme, we propose a novel slack-based robust scheduling rule(SR) based on the analysis of robustness measurement for SMS with uncertain processing time. The decision in the SR is made in real time given the robustness. The proposed SR is verified under different scenarios, and the results are compared with the existing heuristic rules. 5A high-Tc dc-SQUID-based single-channel MCG and its performance显示文摘A single-channel high-Tc dc-SQUID magnetometer and gradiometer have been developed to record the magnetic field component perpendicular to the human chest generated by heart-beat. Magnetocardiogram (MCG) measurements have been carried out inside a magnetically shielded room. By sequentially adjusting a non-magnetic patient table with 5 cm pitch in X and Y directions, the field signals on a rectangle grid 5×5 over the chest area were registered in real time trace point by point with a typical dwell time over 30 cardiocycles each. Utilizing standard electrocardiogram (ECG) recordings as timing reference measured simultaneously with the MCG signals, the MCG data were then averaged and combined to form magnetic field patterns every 10 ms or so. Both the current dipole, which is parallel to MCG measuring plane and produces the vertical magnetic field, and its depth were determined as a function of time in a standard way. 9Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology. 10An engineered lamellar bone mimicking full-scale hierarchical architecture for bone regeneration显示文摘Lamellar bone,compactly and ingeniously organized in the hierarchical pattern with 6 ordered scales,is the structural motif of mature bone.Each hierarchical scale exerts an essential role in determining physiological behavior and osteogenic bioactivity of bone.Engineering lamellar bone with full-scale hierarchy remains a longstanding challenge.Herein,using bioskiving and mineralization,we attempt to engineer compact constructs resembling full-scale hierarchy of lamellar bone.Through systematically investigating the effect of mineralization on physicochemical properties and bioactivities of multi-sheeted collagen matrix fabricated by bioskiving,the hierarchical mimicry and hierarchy-property relationship are elucidated.With prolongation of mineralization,hierarchical mimicry and osteogenic bioactivity of constructs are performed in a bidirectional manner,i.e.first rising and then descending,which is supposed to be related with transformation of mineralization mechanism from nonclassical to classical crystallization.Construct mineralized 9 days can accurately mimic each hierarchical scale and efficiently promote osteogenesis.Bioinformatic analysis further reveals that this construct potently activates integrinα5-PI3K/AKT signaling pathway through mechanical and biophysical cues,and thereby repairing critical-sized bone defect.The present study provides a bioinspired strategy for completely resembling complex hierarchy of compact mineralized tissue,and offers a critical research model for in-depth studying the structure-function relationship of bone.Tao Yang Zhichao Hao Zhenzhen Wu Binxin Xu Jiangchen Liu Le Fan Qinmei Wang Yanshan Li Dongying Li Sangzhu Tang Chuanzi Liu Weichang Li Wei Teng 2023Bioactive Materials2023,,9:1
11Hunting Sodium Dendrites in NASICON-BasedSolid-State Electrolytes显示文摘NASICON-(Na superionic conductor-)based solid-state electrolytes(SSEs)are believed to be attracting candidates for solid-state sodium batteries due to their high ionic conductivity and prospectively reliable stability.However,the poor interface compatibility and the formation of Na dendrites inhibit their practical application.Herein,we directly observed the propagation of Na dendrites through NASICON-based Na3.1Zr2Si2.1P0.9O12 SSE for the first time.Moreover,a fluorinated amorphous carbon(FAC)interfacial layer on the ceramic surface was simply developed by in situ carbonization of PVDF to improve the compatibility between Na metal and SSEs.Surprisingly,Na dendrites were effectively suppressed due to the formation of NaF in the interface when molten Na metal contacts with the FAC layer.Benefiting from the optimized interface,both the Na||Na symmetric cells and Na3V2(PO4)3||Na solidstate sodium batteries deliver remarkably electrochemical stability.These results offer benign reference to the maturation of NASICON-based solid-state sodium batteries.Qiangqiang Zhang Yaxiang Lu Weichang Guo Yuanjun Shao Lilu Liu Jiaze Lu Xiaohui Rong Xiaogang Han Hong Li Liquan Chen Yong-Sheng Hu 2021Energy Material Advances2021,,1:1

13Bladder microenvironment actuated proteomotors with ammonia amplification for enhanced cancer treatment显示文摘Enzyme-driven micro/nanomotors consuming in situ chemical fuels have attracted lots of attention for biomedical applications.However,motor systems composed by organism-derived organics that maximize the therapeutic efficacy of enzymatic products remain challenging.Herein,swimming proteomotors based on biocompatible urease and human serum albumin are constructed for enhanced antitumor therapy via active motion and ammonia amplification.By decomposing urea into carbon dioxide and ammonia,the designed proteomotors are endowed with self-propulsive capability,which leads to improved internalization and enhanced penetration in vitro.As a glutamine synthetase inhibitor,the loaded L-methionine sulfoximine further prevents the conversion of toxic ammonia into non-toxic glutamine in both tumor and stromal cells,resulting in local ammonia amplification.After intravesical instillation,the proteomotors achieve longer bladder retention and thus significantly inhibit the growth of orthotopic bladder tumor in vivo without adverse effects.We envision that the as-developed swimming proteomotors with amplification of the product toxicity may be a potential platform for active cancer treatment.Hao Tian Juanfeng Ou Yong Wang Jia Sun Junbin Gao Yicheng Ye Ruotian Zhang Bin Chen Fei Wang Weichang Huang Huaan Li Lu Liu Chuxiao Shao Zhili Xu Fei Peng Yingfeng Tu 2023Acta Pharmaceutica Sinica B2023,13,9:0
14Fluorescent bovine serum albumin-silver nanoclusters loaded with paclitaxel can traverse the blood-brain barrier to inhibit the migration of glioma显示文摘Objective::Glioma is the most common and aggressive primary brain tumor. Here, we aimed to establish a nano-drug carrier system to traverse the blood-brain barrier for the treatment and inhibition of glioma migration.Methods::The synthesis of bovine serum albumin protected-silver nanoclusters (BSA-AgNCs) was performed using chemical reduction. The drug paclitaxel (PTX) can be loaded into BSA-AgNCs through electrostatic and hydrophobic interactions to formulate spherical BSA-AgNC-PTX nanoparticles (BSA-AgNC-PTX NPs). A glioma mouse model was established by injecting U251-GFP-Luc cells into the mouse striatum, and all surgical procedures were approved by the Animal Ethics Committee of Nanchang University (SYXK2019-0003) on December 29, 2019.Results::The BSA-AgNC-PTX NPs were able to efficiently pass through the blood-brain barrier, both in vitro and in vivo, to deliver the drug to the tumor site. 15Cobalt-free Composite Ba_(0.5)Sr_(0.5)Fe_(0.9)Ni_(0.1)O_(3-δ)-Ce_(0.8)Sm_(0.2)O_(2-δ) as Cathode for Intermediate-Temperature Solid Oxide Fuel Cell显示文摘New cobalt-free composites consisting of Ba0.5Sr0.5Fe0.9Ni0.1O3-δ(BSFN) and Ce0.8Sm0.2O2-δ(SDC) were investigated as possible cathode materials for intermediate-temperature solid oxide fuel cell (IT-SOFC). BSFN, which was synthesized by auto ignition process, was chemically compatible with SDC up to 1100℃ as indicated by X-ray diffraction analysis. The electrical conductivity of BSFN reached the maximum value of 57 S·cm-1 at 450℃. The thermal expansion coefficient (TEC) value of BSFN was 30.9×10-6K-1 , much higher than that of typical electrolytes. The electrochemical behavior of the composites was analyzed via electrochemical impedance spectroscopy with symmetrical cells BSFN-SDC/SDC/BSFN-SDC. The area specific interfacial polarization resistance (ASR) decreased with increasing SDC content of the composite. The area specific interfacial polarization resistance (ASR) at 700℃ is only 0.49, 0.34 and 0.31 Ω·cm2 when 30, 40, and 50 wt% SDC was cooperated to BSFN, respectively.
