| 1 | Hepatitis B Immunoprophylactic Failure and Characteristics of the Hepatitis B Virus Gene in Mother-Infant Pairs in Parts of China显示文摘Objective To determine the hepatitis B immunoprophylactic failure rate in infants born to hepatitis B virus(HBV) infected mothers and to characterize HBV genes. Methods HBV-serological testing was conducted for pregnant women and infants. The complete genomes of 30 HBV isolates were sequenced, and genetic characteristics were analyzed using MEGA 5 software. Results The immunoprophylactic failure rate for infants who had completed the scheduled hepatitis B vaccination program was 5.76%(32/556). High sequence homology(99.8%-100%) was observed in 8 of the 10 mother-infant pairs. We identified 19 subgenotype C2 strains, 9 subgenotype B2 strains, and 2 subgenotype C1 strains. Three serotypes were detected: adr(19/30), adw(9/30), and ayw(2/30). The frequency of amino acid mutation of the ‘a' determinant region was 16.67%(5/30), including that of Q129 H, F134 Y, S136 Y, and G145 E. We detected 67 amino acid mutations in the basal core promoter, precore, and core regions of the genome. Conclusion The immunoprophylactic failure rate in infants born to HBV-infected mothers is low in the regions of China examined during this study. | 2 | Three-dimensional spongy framework as superlyophilic, strongly absorbing,and electrocatalytic polysulfide reservoir layer for high-rate and long-cycling lithiumsulfur batteries显示文摘In the development of lithium-sulfur (Li-S)batteries,various approaches have been adopted to enhance the electronic conductivity of the sulfur cathode and alleviate the shuttle effect of polysulfides;however,the strategies providing efficient solutions are still limited.To further improve the electrochemical performance of Li-S batteries,in this work we propose a new strategy involving the incorporation of a three-dimensional functional spongy framework as polysulfide reservoir layer,with strong absorbability and electrocatalytic activity towards sulfur species.The spongy framework has a hierarchical architecture composed of highly conductive Ni foam/graphene/carbon nanotubes/MnO2nanoflakes (NGCM).The strongly interconnected Ni foam,graphene,and carbon nanotubes of the NGCM sponge facilitate electron transfer during discharge/charge processes;moreover,the superlyophilic properties of the NGCM sponge ensure good wettability and interface contact with the Li-S electrolyte,and the porous MnO2nanoflakes provide strong chemisorptive and electrocatalytic effects on polysulfides (as confirmed theoretically and experimentally).The NGCM sponge, serving as a polysulfide reservoir layer attached on a conventional sulfur-mixed carbon nanotubes (S/CNTs)cathode,can provide improved reversible capacity, rate capability (593mAh·g^-1at 3.0C),and cycling stability.In addition,the self-discharge rate is greatly reduced,owing to the efficient conservation of polysulfides in the NGCM spongy framework. | Lianbo Ma Guoyin Zhu Wenjun Zhang Peiyang Zhao Yi Hu Yanrong Wang Lei Wang Renpeng Chen Tao Chen Zuoxiu Tie Jie Liu Zhong Jin | 2018 | Nano Research2018,11,12: | 2 |

| 5 | The ScCas9^(++) variant expands the CRISPR toolbox for genome editing in plants显示文摘The development of clustered regularly interspaced palindromic repeats(CRISPR)-associated protein(Cas) variants with a broader recognition scope is critical for further improvement of CRISPR/Cas systems. The original Cas9 protein from Streptococcus canis(ScCas9) can recognize simple NNG-protospacer adjacent motif(PAM)targets, and therefore possesses a broader range relative to current CRISPR/Cas systems, but its editing efficiency is low in plants. Evolved ScCas9^(+) and ScCas9^(++) variants have been shown to possess higher editing efficiencies in human cells, but their activities in plants are currently unknown. Here, we utilized codon-optimized ScCas9, ScCas9^(+) and ScCas9^(++) and a nickase variant ScCas9n^(++) to systematically investigate genome cleavage activity and cytidine base editing efficiency in rice(Oryza sativa L.). This analysis revealed that ScCas9^(++) has higher editing efficiency than ScCas9 and ScCas9^(+) in rice. Furthermore, we fused the evolved cytidine deaminase PmCDA1 with ScCas9n^(++) to generate a new evoBE4max-type cytidine base editor, termed PevoCDA1-ScCas9n^(++) . This base editor achieved stable and efficient multiplex-site base editing at NNG-PAM sites with wider editing windows(C_(-1)-C_(17)) and without target sequence context preference. Multiplex-site base editing of the rice genes OsWx(three targets) and OsEui1(two targets) achieved simultaneous editing and produced new rice germplasm. | 6 | Coil Ambient Temperature and Its Influence on the Formation of Blocking Layer in the Electromagnetic Induction-Controlled Automated Steel-Teeming System显示文摘Ambient temperature of induction coil is an important factor to influence the implementation of the electromagnetic induction-controlled automated steel-teeming(EICAST) technology. Meanwhile, it also affects the formation of Fe–C alloy blocking layer, which determines the length and installation position of induction coil. An experimental platform was designed to imitate actual working conditions in a ladle with the EICAST system. Ambient temperature of induction coil under high-temperature condition was measured to verify the accuracy of numerical result. After containing molten steel for 120 min and steel teeming for 40 min, the ambient temperature on the upper side of induction coil is 791 °C. In addition, the position of blocking layer in a 110 t ladle was measured by sand-collection and steel-pour methods, and the criterion temperatures of blocking layer in numerical simulation process were corrected. When the refining temperature is1600 °C and the containing time of molten steel is 120 min, the thickness of blocking layer is 130 mm, and the distance between the upper surface of blocking layer and the upper surface of nozzle brick is 154 mm. | 8 | Overexpression of maize GOLDEN2 in rice and maize calli improves regeneration by activating chloroplast development显示文摘Golden2(G2), a member of the GARP transcription factor superfamily, regulates several biological processes and phytohormone signaling pathways in plants. In this study, we used a rice codon-optimized maize G2 gene(rZmG2) to improve the regeneration efficiency of rice and maize calli for genetic transformation. We isolated a promoter driving strong and callus-specific expression from rice to drive rZmG2 transcription from a transgene after transformation of two indica and two japonica rice cultivars. The resulting rZmG2 transgenic calli turned green in advance at the differentiation stage, thus significantly raising the regeneration rates of the transgenic indica and japonica rice plants relative to control transformations. Similar effect of this gene on improving maize transformation was also observed. Transcriptome sequencing and RT-qPCR analyses showed that many rice genes related to chloroplast development and phytohormones are upregulated in rZmG2-transgenic calli. These results demonstrate that rZmG2 can promote embryogenic callus differentiation and improve regeneration efficiency by activating chloroplast development and phytohormone pathways. We also established a heat-inducible Cre/loxP-based gene-excision system to remove rZmG2 and the antibiotic selectable gene after obtaining the transgenic plants. | 9 | Pilot HI survey of Planck Galactic Cold Clumps with FAST显示文摘We present a pilot HI survey of 17 Planck Galactic Cold Clumps(PGCCs)with the Five-hundred-meter Aperture Spherical radio Telescope(FAST).HI Narrow Self-Absorption(HINSA)is an effective method to detect cold HI being mixed with molecular hydrogen H2 and improves our understanding of the atomic to molecular transition in the interstellar medium.HINSA was found in 58%PGCCs that we observed.The column density of HINSA was found to have an intermediate correlation with that of 13CO,following log(N(HINSA))=(0.52±0.26)log(N13CO)+(10±4.1).HI abundance relative to total hydrogen[HI]/[H]has an average value of 4.4×10-3,which is about 2.8 times of the average value of previous HINSA surveys toward molecular clouds.For clouds with total column density NH>5×1020 cm-2,an inverse correlation between HINSA abundance and total hydrogen column density is found,confirming the depletion of cold HI gas during molecular gas formation in more massive clouds.Nonthermal line width of 13CO is about 0-0.5 kms-1 larger than that of HINSA.One possible explanation of narrower non-thermal width of HINSA is that HINSA region is smaller than that of 13CO.Based on an analytic model of H2 formation and H2 dissociation by cosmic ray,we found the cloud ages to be within 106.7-107.0 yr for five sources. | 汤宁宇 左沛 李菂 Lei Qian Tie Liu Yue-Fang Wu Marko Krco Meng-Ting Liu You-Ling Yue Yan Zhu Hong-Fei Liu Dong-Jun Yu Jing-Hai Sun Peng Jiang Gao-Feng Pan Hui Li Heng-Qian Gan Rui Yao Shu Liu FAST Collaboration | 2020 | Research in Astronomy and Astrophysics2020,20,5: | 0 |
