1Inactivation of Cryptosporidium by ozone and cell ultrastructures显示文摘The fluorescence staining method and scanning electron microscopy (SEM) were used to study the effect of ozone (O3) inactivating Cryptosporidium in water and cell ultrastructures variation to shed light on the mechanism of inactivation preliminarily.Results indicated that O3 had a stronger inactivating capability.When the concentration of O3 was above 3.0 mg/L and the contact time was up to 7 min,a significant inactivating effect could be achieved.The turbidity on inactivation effects was also found to be statistically significant in artificial water.With increases in turbidity,the inactivating effect decreased.Inactivation rate improved with a temperature increase from 5 to 25°C,but decreased beyond this.The inactivating capability of O3 was found to be stronger under acidic than that under alkalic conditions.When the concentration of organic matter in the reaction system was increased,the competition between Cryptosporidium and organics with O3 probably took place,thereby reducing the inactivation rate.In addition,the cellular morphology of Cryptosporidium varied with different contact times.At zero contact time,cells were rotundity and sphericity,at 60 sec they became folded,underwent emboly,and burst at 480 sec,the cell membrane of Cryptosporidium shrinked and collapsed completely.Zhilin Ran Shaofeng Li Junli Huang Yixing Yuan Chongwei Cui Craig Denver Williams 2010Journal of Environmental Sciences2010,22,12:11
2The role of RIP3 in cardiomyocyte necrosis induced by mitochondrial damage of myocardial ischemia-reperfusion显示文摘Myocardial damage caused by myocardial ischemia-reperfusion injury (MIRI)is difficult to be alleviated because cardiomyocyte necrosis is an irreversible and unregulated death form.Recently, necroptosis,a necrosis form caused by tumor necrosis factor-α (TNF-α)and Fas ligand (FasL),was found to be regulated by receptor interacting protein 3 (RIP3)and RIP3-receptor interacting protein 1 (RIP1)-mixed lineage kinase domain like protein (MLKL)pathway.But it is unclear whether they also play a regulatory role in MIRI-induced necroptosis.Our previous results showed that in rat MIRI,RIP3 could translocate and express highly in mitochondria.Therefore,it is important to explore proteins that interact with RIP3 which was translocated to mitochondria.The aim of this study was to explore the role of RIP3 in cardiomyocyte necrosis induced by mitochondrial damage of hypoxia/reoxygenation (H/R).Our results showed that H/R could cause RIP3-depended mitochondrial fragmentation and necrosis-based death;and RIP3-promoted H/R-induced necroptosis in H9c2 cells through increasing lactate dehydrogenase release and inhibiting cell viability.This process did not require RIP1 or MLKL but dynamin-related protein 1 (Drp1),which was related to Drpl activation,reactive oxygen species elevation,and △Ψm decline.This study provides novel insights into the role of RIP3 in cardiomyocyte injury during H/R.RIP3 may serve as a potential target for the treatment of MIRI.Huifang Hou Yongling Wang Qiong Li Zaibing Li Yan Teng Jingyu Li Xiaoying Wang Junli Chen Ning Huang 2018Acta Biochimica et Biophysica Sinica2018,50,11:10
3Genome-wide Characterization of cis-acting Elements in the Promoters of Key Carotenoid Pathway Genes from the Main Species of Genus Citrus显示文摘Carotenoids are indispensable for both human health and plant survival.Citrus,is one of the fruit crops richest in carotenoid compounds,with approximately 115 kinds of carotenoids;tremendous diversity in carotenoids composition and concentration exists among various species,showing different colors from nearly white to crimson.The carotenoid biosynthetic pathway and the key carotenogenic genes have been identified in citrus;however,the underlying regulatory mechanisms remain unclear.In this study,among the main species of genus Citrus(primitive,wild,and cultivated),we detected carotenoids in flavedo using High-Performance Liquid Chromatography,and analyzed variations in cis-acting elements in the promoters of key carotenoid pathway genes.Intriguingly,both carotenoid composition and content were generally increased during the evolution of citrus,and the corresponding variations in the promoters were identified,including the gain or loss of critical environmental stress-responsive elements and hormone-responsive elements,which are closely associated with carotenoid enhancement.In addition,pummelo has the most heat-responsive elements,but the Mangshan mandarin does not have this element in the promoters of PSY,which is highly related to their geographical origin and indicate that temperature is a critical environmental signal influencing carotenoid accumulation.Moreover,the abscisic acid-responsive motif was rich in almost all the seven species,but the ethylene-responsive motif was deficient,which demystified the unique phytohormone regulation mechanism of carotenoid accumulation in citrus.Overall,our study provides new insights into the molecular regulatory mechanism of carotenoid enhancement in the evolution of citrus,which can facilitate breeding and cultivation efforts to improve the nutritional quality and esthetic value in citrus and hopefully other fruit crops.Kaijie Zhu Qingjiang Wu Yue Huang Junli Ye Qiang Xu Xiuxin Deng 2020Horticultural Plant Journal2020,6,6:8
4Distinctive effects of CD34- and CD133-specific antibody-coated stents on re-endothelialization and in-stent restenosis at the early phase of vascular injury显示文摘It is not clear what effects of CD34-and CD133-specific antibody-coated stents have on reendothelialization and in-stent restenosis(ISR)at the early phase of vascular injury.This study aims at determining the capabilities of different coatings on stents(e.g.gelatin,anti-CD133 and anti-CD34 antibodies)to promote adhesion and proliferation of endothelial progenitor cells(EPCs).The in vitro study revealed that the adhesion force enabled the EPCs coated on glass slides to withstand flow-induced shear stress,so that allowing for the growth of the cells on the slides for 48 h.The in vivo experiment using a rabbit model in which the coated stents with different substrates were implanted showed that anti-CD34 and anti-CD133 antibody-coated stents markedly reduced the intima area and restenosis than bare mental stents(BMS)and gelatin-coated stents.Compared with the anti-CD34 antibody-coated stents,the time of cells adhesion was longer and earlier present in the anti-CD133 antibody-coated stents and anti-CD133 antibody-coated stents have superiority in re-endothelialization and inhibition of ISR.In conclusion,this study demonstrated that anti-CD133 antibody as a stent coating for capturing EPCs is better than anti-CD34 antibody in promoting endothelialization and reducing ISR.Xue Wu Tieying Yin Jie Tian Chaojun Tang Junli Huang Yinping Zhao Xiaojuan Zhang Xiaoyan Deng Yubo Fan Donghong Yu Guixue Wang 2015Regenerative Biomaterials2015,2,2:7
5High-performance quasi-2D perovskite light-emitting diodes: from materials to devices显示文摘Quasi-two-dimensional(quasi-2D)perovskites have attracted extraordinary attention due to their superior semiconducting properties and have emerged as one of the most promising materials for next-generation lightemitting diodes(LEDs).The outstanding optical properties originate from their structural characteristics.In particular,the inherent quantum-well structure endows them with a large exciton binding energy due to the strong dielectricand quantum-confinement effects;the corresponding energy transfer among different n-value species thus results in high photoluminescence quantum yields(PLQYs),particularly at low excitation intensities.The review herein presents an overview of the inherent properties of quasi-2D perovskite materials,the corresponding energy transfer and spectral tunability methodologies for thin films,as well as their application in high-performance LEDs.We then summarize the challenges and potential research directions towards developing high-performance and stable quasi-2D PeLEDs.The review thus provides a systematic and timely summary for the community to deepen the understanding of quasi-2D perovskite materials and resulting LED devices.Li Zhang Changjiu Sun Tingwei He Yuanzhi Jiang Junli Wei Yanmin Huang Mingjian Yuan 2021Light(Science & Applications)2021,10,4:5
6Liposomes for systematic delivery of vancomycin hydrochloride to decrease nephrotoxicity:Characterization and evaluation显示文摘Vancomycin hydrochloride(VANH),the first glycopeptide antibiotic,is a water-soluble drug for the treatment of acute osteomyelitis.Liposomal formulations of VANH have already been manipulated and characterized,which was a mean of increasing their therapeutic index,reducing their toxicity and altering drug biodistribution.One of the challenges for preparing VANH-Lips is their low encapsulation efficiency(EE).In the present study,we aim to improve the liposomal formulation of VANH for higher EE,longer systemic circulation,reduced nephrotoxicity and enhanced antimicrobial activities.Vancomycin hydrochloride-loaded liposomes(VANH-Lips)were formulated by the method of modified reverse phase evaporation.Based on the optimization of formulation with orthogonal experimental design,the average drug encapsulation efficiency and the mean particle size of VANH-Lips were found to be 40.78±2.56%and 188.4±2.77 nm.In vitro drug release of VANH-Lips possessed a sustained release characteristic and their release behavior was in accordance with the Weibull equation.After intravenous injection to mice,the mean residence time(MRT)of VANH-Lips group was significantly prolonged in vivo and the AUC value was improved as well compared with the vancomycin hydrochloride solution(VANH-Sol)group.Furthermore,the biodistribution results in mice showed that VANH-Lips decreased the accumulation of VANH in kidney after intravenous injection.In conclusion,VANH-Lips may be a potential delivery system for VANH to decrease nephrotoxicity in the treatment of osteomyelitis.Junli Liu Zhonglan Wang Fubing Li Jinhua Gao Longmei Wang Guihua Huang 2015Asian Journal of Pharmaceutical Sciences2015,10,3:4
7Uptake of oxidative stress-mediated extracellular vesicles by vascular endothelial cells under low magnitude shear stress显示文摘Extracellular vesicles(EVs)are increasingly used as delivery vehicles for drugs and bioactive molecules,which usually require intravascular administration.The endothelial cells covering the inner surface of blood vessels are susceptible to the shear stress of blood flow.Few studies demonstrate the interplay of red blood cell-derived EVs(RBCEVs)and endothelial cells.Thus,the phagocytosis of EVs by vascular endothelial cells during blood flow needs to be elucidated.In this study,red blood cell-derived extracellular vesicles(RBCEVs)were constructed to investigate endothelial cell phagocytosis in vitro and animal models.Results showed that low magnitude shear stress including low shear stress(LSS)and oscillatory shear stress(OSS)could promote the uptake of RBCEVs by endothelial cells in vitro.In addition,in zebrafish and mouse models,RBCEVs tend to be internalized by endothelial cells under LSS or OSS.Moreover,RBCEVs are easily engulfed by endothelial cells in atherosclerotic plaques exposed to LSS or OSS.In terms of mechanism,oxidative stress induced by LSS is part of the reason for the increased uptake of endothelial cells.Overall,this study shows that vascular endothelial cells can easily engulf EVs in areas of low magnitude shear stress,which will provide a theoretical basis for the development and utilization of EVs-based nano-drug delivery systems in vivo.Xian Qin Kun Zhang Juhui Qiu Nan Wang Kai Qu Yuliang Cui Junli Huang Li Luo Yuan Zhong Tian Tian Wei Wu Yi Wang Guixue Wang 2022Bioactive Materials2022,7,3:3

9Effects of NPY Gene on Total Number of Eggs at 300 Days of Age in Donglan Black-bone Chicken显示文摘In this study,PCR-RFLP technique was employed to detect the genetic polymorphism of NPY gene and analyze the effects of various genotypes on the total number of eggs at 300 days of age in 135 Donglan black-bone chicken. According to the results,there were three genotypes( AA,AB and BB) of NPY gene in Donglan black-bone chicken group. Different genotypes exhibited significant effects( P < 0. 05) on the total number of eggs at 300 days of age. The total number of eggs at 300 days of age of AA genotype was significantly higher than that of BB genotype( P < 0. 05). 15Structural variation and parallel evolution of apomixis in citrus during domestication and diversification显示文摘Apomixis,or asexual seed formation,is prevalent in Citrinae via a mechanism termed nucellar or adventitious embryony.Here,multiple embryos of a maternal genotype form directly from nucellar cells in the ovule and can outcompete the developing zygotic embryo as they utilize the sexually derived endosperm for growth.Whilst nucellar embryony enables the propagation of clonal plants of maternal genetic constitution,it is also a barrier to effective breeding through hybridization.To address the genetics and evolution of apomixis in Citrinae,a chromosome-level genome of the Hongkong kumquat(Fortunella hindsii)was assembled following a genome-wide variation map including structural variants(SVs)based on 234 Citrinae accessions.This map revealed that hybrid citrus cultivars shelter genome-wide deleterious mutations and SVs into heterozygous states free from recessive selection,which may explain the capability of nucellar embryony in most cultivars during Citrinae diversification.Analyses revealed that parallel evolution may explain the repeated origin of apomixis in different genera of Citrinae.Within Fortunella,we found that apomixis of some varieties originated via introgression.In apomictic Fortunella,the locus associated with apomixis contains the FhRWP gene,encoding an RWP-RK domain-containing protein previously shown to be required for nucellar embryogenesis in Citrus.We found the heterozygous SV in the FhRWP and CitRWP promoters from apomictic Citrus and Fortunella,due to either two or three miniature inverted transposon element(MITE)insertions.A transcription factor,FhARID,encoding an AT-rich interaction domain-containing protein binds to the MITEs in the promoter of apomictic varieties,which facilitates induction of nucellar embryogenesis.This study provides evolutionary genomic and molecular insights into apomixis in Citrinae and has potential ramifications for citrus breeding.Nan Wang Xietian Song Junli Ye Siqi Zhang Zhen Cao Chenqiao Zhu Jianbing Hu Yin Zhou Yue Huang Shuo Cao Zhongjie Liu Xiaomeng Wu Lijun Chai Wenwu Guo Qiang Xu Brandon S.Gaut Anna M.G.Koltunow Yongfeng Zhou Xiuxin Deng 2022National Science Review2022,9,10:1
