1Function of lncRNAs and approaches to lncRNA-protein interactions显示文摘Long non-coding RNAs(lncRNAs),which represent a new frontier in molecular biology,play important roles in regulating gene expression at epigenetic,transcriptional and post-transcriptional levels.More and more lncRNAs have been found to play important roles in normal cell physiological activities,and participate in the development of varieties of tumors and other diseases.Previously,we have only been able to determine the function of lncRNAs through multiple mechanisms,including genetic imprinting,chromatin remodeling,splicing regulation,mRNA decay,and translational regulation.Application of technological advances to research into the function of lncRNAs is extremely important.The major tools for exploring lncRNAs include microarrays,RNA sequencing(RNA-seq),Northern blotting,real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),RNA interference(RNAi),RNA-binding protein immunoprecipitation(RIP),chromatin isolation by RNA purification(ChIRP),crosslinking-immunopurification(CLIP),and bioinformatic prediction.In this review,we highlight the functions of lncRNAs,and advanced methods to research lncRNA-protein interactions.ZHU JuanJuan FU HanJiang WU YongGe ZHENG XiaoFei 2013Science China(Life Sciences)2013,56,10:54
2Acupuncture treatment of lung-spleen Qi deficiency in stable chronic obstructive pulmonary disease: a randomized, open-label, controlled trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of acupuncture that reinforces the spleen to strengthen the lung in patients with stable chronic obstructive pulmonary disease(COPD).METHODS:This was a randomized,open-controlled trial in which the acupuncturist and the participants were not blinded,but the outcome evalu-ators and data analysts were blinded.One-hundred-and-two patients with stable COPD were randomly divided into two groups in a 1∶1 ratio.The acupuncture group received 30-minute sessions of acupuncture therapy at the same acupoints three times weekly for 6 weeks in addition to routine conventional Western Medicine treatment;the control group received routine conventional Western Medicine treatment alone.The primary outcome was the Borg scale score,which was assessed immediately after the 6-minute walk test.The secondary outcomes were the 6-minute walk distance,lung function,and oxygen saturation.Measurements were obtained at baseline and after 6 weeks of treatment.RESULTS:After 6 weeks of treatment,the Borg scale score in the acupuncture group was significantly better than that in the control group(2.02±0.71 versus 5.01±0.34,P<0.05).Furthermore,the post-treatment improvements in the 6-minute walk distance,lung function,and oxygen saturation were significantly greater in the acupuncture group than in the control group,showing that the acupuncture group had better exercise tolerance.CONCLUSION:The findings suggest that acupuncture that aims to reinforce the spleen to strengthen the lung is a safe and effective adjuvant therapy that effectively improves the exercise capacity of patients with stable COPD.Li Yu Xiong Chan Zeng Yan Wei Hua Zhuang Guangtong Zhao Lihong Li Chenyi Li Lin Qin Erqi Chen Xiaoqin Fu Juanjuan 2019Journal of Traditional Chinese Medicine2019,39,6:13
3N-P fertilization did not reduce AMF abundance or diversity but alter AMF composition in an alpine grassland infested by a root hemiparasitic plant显示文摘Fertilization has been shown to have suppressive effects on arbuscular mycorrhizal fungi(AMF) and root hemiparasites separately in numerous investigations, but its effects on AMF in the presence of root hemiparasites remain untested. In view of the contrasting nutritional effects of AMF and root hemiparasites on host plants, we tested the hypothesis that fertilization may not show strong suppressive effects on AMF when a plant community was infested by abundant hemiparasitic plants. Plants and soil samples were collected from experimental field plots in Bayanbulak Grassland, where N and P fertilizers had been applied for three continuous years for control against a spreading root hemiparasite, Pedicularis kansuensis. Shoot and root biomass of each plant functional group were determined. Root AMF colonization levels, soil spore abundance, and extraradical hyphae length density were measured for three soil depths(0 e10 cm, 10 e20 cm, 20 e30 cm). Partial 18 S r RNA gene sequencing was used to detect AMF diversity and community composition. In addition, we analyzed the relationship between relative abundance of different AMF genera and environmental factors using Spearman's correlation method. In contrast to suppressive effects reported by many previous studies, fertilization showed no significant effects on AMF root colonization or AMF species diversity in the soil. Instead, a marked increase in soil spore abundance and extraradical hyphae length density were observed. However, fertilization altered relative abundance and AMF composition in the soil. 16Parental and transgenerational impairments of thyroid endocrine system in zebrafish by 2,4,6-tribromophenol显示文摘Many environmental contaminants could be transmitted fromparents and generate impairments to their progeny.The 2,4,6-tribromophenol(TBP),a novel brominated flame retardant which has been frequently detected in various organisms,was supposed to be bioaccumulated and intergenerational transmitted in human beings.Previous studies revealed that TBP could disrupt thyroid endocrine system in zebrafish larvae.However,there is no available data regarding the parental and transgenerational toxicity of this contaminant.Thus,in this study adult zebrafish were exposed to environmental contaminated levels of TBP for 60 days to investigate the parental and transgenerational impairments on thyroid endocrine system.Chemical analysis verified the bioaccumulation of TBP in tested organs of parents(concentration:liver>gonads>brain)and its transmission into eggs.For adults,increased thyroid hormones,disturbed transcriptions of related genes and histopathological changes in thyroid follicles indicate obvious thyroid endocrine disruptions.Transgenerational effects are indicated by the increased thyroid hormones both in eggs(maternal source)and in developed larvae(newly synthesized),aswell as disrupted transcriptional profiles of key genes in HPT axis.The overall results suggest that the accumulated TBP could be transmitted from parent to offspring and generate thyroid endocrine disruptions in both generations.Yuxi Zhou Juanjuan Fu Min Wang Yongyong Guo Lihua Yang Jian Han Bingsheng Zhou 2023Journal of Environmental Sciences2023,,2:1
17A Review:Toxicity of Ammonia-N to Fish and Detoxification Strategy of Fish显示文摘In natural water, the content of ammonia nitrogen is always kept at a low level. However, the ammonia nitrogen content in water will be elevated or even excessive when the density of aquaculture is too large or exogenous ammonia are discharged into the water, causing a series of ad-verse effects on fish survival. Within a certain range, fishes reduce ammonia production in vivo by converting ammonia into glutamine and urea, and increase NH3 volatilization and NH4+emissions to ease ammonia toxicity, but the ability to alleviate ammonia poisoning vary with fish species and size. 18Determination of the predictive factors for diagnostic positivity of nucleic acid amplification tests for diagnosing pulmonary tuberculosis显示文摘Background:Tuberculosis(TB)remains a major threat to human health,and TB diagnostic methods remain unsatisfactory.Nucleic acid amplification tests(NAATs)show higher sensitivity compared with culture for the diagnosis of pulmonary TB(PTB).However,NAATs are expensive and cannot be easily implemented outside major medical centers.To improve the sensitivity of NAATs for PTB diagnosis,we investigated the predictive factors that might optimize NAAT utilization.Methods:A total of 1263 patients with suspected PTB were enrolled for evaluation.The sensitivity,specificity,and accuracy of methods including smear-microbiology,culture of Mtb and NAAT for Mycobacterium tuberculosis(Mtb)detection in sputum and bronchoalveolar lavage fluid samples were compared.Odds ratios and 95%confidence intervals were used to assess variables that might be associated with positive NAAT results for sputum and bronchoalveolar lavage fluid from patients with suspected PTB.Results:NAAT showed higher sensitivity for Mtb detection(61.1%)when compared with smear(9.0%)and Mtb culture(47.8%).We found that an elevated erythrocyte sedimentation rate,the presence of cavities,and positive interferon-��release assay(IGRA)results were indicative of positive Mtb detection by NAAT.Moreover,individuals who had all three of these characteristics showed an 86%diagnostic positivity for PTB from Mtb detection by NAAT.Conclusions:Our study suggests that an elevated erythrocyte sedimentation rate,a positive IGRA result,and the presence of pulmonary cavities are helpful factors for predicting positive Mtb detection by NAAT.Patients with the three positive clinical markers should undergo NAAT for Mtb detection because they are the most likely individuals to be bacteriologically confirmed as having TB.Xingfang Hou Qinglong Guo Qiao Lin Taosheng Ye Jing Bi Juan Liang Ke Yang Yuan Zhou Juanjuan Zhang Zhihang Liang Xuefeng Zhou Gengwei Zhang Xiangdong Fu Hongjian Zhong Guoliang Zhang 2022Infectious Medicine2022,1,1:0

20Graphene controlled phase evolution in Sr-deficient Sr(Ti, Nb)O_(3) thermoelectric ceramics显示文摘Correlated phase and microstructural evolution are systematically investigated by electron microscopies in Sr-deficient Sr(Ti,Nb)O_(3)(STNO)thermoelectric ceramics incorporated with different fraction of reduced graphene oxide(RGO).It is found that while no impurity except for very few Ti_(3)O_(5) precipitates are observed in monolithic STNO,the Nb-enriched rutile TiO_(2) appears in RGO/STNO composites.With increasing RGO content,the amount of precipitates increase at first and then decrease when RGO content becomes high,which can be ascribed to the formation of local Magneli phase.In addition,the energy-dispersive X-ray spectra combined with cathodoluminescence characterization indicates that the variation of Sr deficiency experiences the opposite trend with respect to the precipitates content.These findings clearly reveal the unique reducing effect of RGO on the microstructure of doped SrTiO_(3) with Sr deficiency,which can greatly facilitate the design of perovskite based thermoelectric materials of hierarchical structure.Jia Li Cao Wu Jilong Huang Juanjuan Xing Yuchi Fan Qingqiao Fu Ying Jiang Hui Gu 2021Journal of Materiomics2021,7,2:0
