1Transcriptomic analysis in Anemone flaccida rhizomes reveals ancillary pathway for triterpene saponins biosynthesis and differential responsiveness to phytohormones显示文摘Anemone flaccida Fr. Schmidt is a perennial medicinal herb that contains pentacyclic triterpenoid saponins as the major bioactive constituents. In China, the rhizomes are used as treatments for a variety of ailments including arthritis. However, yields of the saponins are low, and little is known about the plant's genetic background or phytohormonal responsiveness. Using one-quarter of the 454 pyrosequencing information from the Roche GS FLX Titanium platform, we performed a transcriptomic analysis to identify 157 genes putatively encoding 26 enzymes involved in the synthesis of the bioactive compounds. It was revealed that there are two biosynthetic pathways of triterpene saponins in A. flaccida. One pathway depends on β-amyrin synthase and is similar to that found in other plants. The second, subsidiary('backburner') pathway is catalyzed by camelliol C synthase and yields β-amyrin as minor byproduct. Both pathways used cytochrome P450-dependent monooxygenases(CYPs) and family 1 uridine diphosphate glycosyltransferases(UGTs) to modify the triterpenoid backbone. The expression of CYPs and UGTs were quite different in roots treated with the phytohormones methyl jasmonate, salicylic acid and indole-3-acetic acid. 19Molecular rotors as a class of generally highly active ion transporters显示文摘We describe here a class of unconventional ion transporters,molecular rotors that transport ions through a rotating function rather than via traditional carrier or channel mechanisms.Mimicking macroscopic rotors,these molecular rotors consist of three modularly tunable components,i.e.,a membrane-anchoring stator,a crown ether-containing rotator for ion binding and transport,and a triple bond-based axle that allows the rotator to freely rotate around the stator in the lipid membrane.Lipid bilayer experiments reveal the generally high ability of all molecular rotors in promoting the highly efficient transmembrane K^(+)flux(EC50 values=0.49-1.37 mol%relative to lipid).While molecular rotors differing only in the ion-binding unit exhibit similar ion transport activities,those differing in the rotator’s length display activity differences by up to 174%.Jie Shen Joan Jia Ying Han Ruijuan Ye Huaqiang Zeng 2021Science China Chemistry2021,64,12:0
20Detection of antibiotic resistance toxigenic Clostridium difficile in processed retail lettuce显示文摘Objectives:Clostridium difficile is the major cause of infectious diarrhoea in humans after antimicrobial treatment.Clostridium difficile has been isolated from food animals and meat.The main purpose of this study was to characterize C.difficile isolated from retail lettuce and determine the antibiotic resistance using five common clinical-selected antibiotics(metronidazole,vancomycin,clindamycin,erythromycin,and cefotaxime).Materials and Methods:Lettuce samples(grown in California,Arkansas,and Louisiana)were purchased from retail stores.Results:Toxigenic C.difficile was isolated from 13.8 per cent(41/297)of the lettuce samples.Among the toxigenic isolates,only 82.9 per cent(34/41)produced toxin B,17.1 per cent(7/41)produced both toxin A and toxin B,and two of the Louisiana C.difficile isolates were identified as ribotype 027.Under the treatment of the five antibiotics,the virulence C.difficile isolates were identified as having antibiotic resistance to metronidazole,vancomycin,and erythromycin.Conclusion:The present study reports the highest prevalence of toxigenic C.difficile in US retail lettuce.The antibiotic resistance to metronidazole,vancomycin,and erythromycin of the isolated C.difficile from retail lettuces could lead to public health concerns.Yi Han Joan King Marlene E.Janes 2018Food Quality and Safety2018,2,1:0
