1Abnormal subchondral bone remodeling and its association with articular cartilage degradation in knees of type 2 diabetes patients显示文摘Type 2 diabetes(T2 D) is associated with systemic abnormal bone remodeling and bone loss. Meanwhile,abnormal subchondral bone remodeling induces cartilage degradation, resulting in osteoarthritis(OA).Accordingly, we investigated alterations in subchondral bone remodeling, microstructure and strength in knees from T2 D patients and their association with cartilage degradation. Tibial plateaus were collected from knee OA patients undergoing total knee arthroplasty and divided into non-diabetic(n = 70) and diabetes(n = 51) groups. Tibial plateaus were also collected from cadaver donors(n = 20) and used as controls.Subchondral bone microstructure was assessed using micro-computed tomography. Bone strength was evaluated by micro-finite-element analysis. Cartilage degradation was estimated using histology. The expression of tartrate-resistant acidic phosphatase(TRAP), osterix, and osteocalcin were calculated using immunohistochemistry. Osteoarthritis Research Society International(OARSI) scores of lateral tibial plateau did not differ between non-diabetic and diabetes groups, while higher OARSI scores on medial side were detected in diabetes group. Lower bone volume fraction and trabecular number and higher structure model index were found on both sides in diabetes group. These microstructural alterations translated into lower elastic modulus in diabetes group. Moreover, diabetes group had a larger number of TRAP^+ osteoclasts and lower number of Osterix^+ osteoprogenitors and Osteocalcin^+ osteoblasts. T2 D knees are characterized by abnormal subchondral bone remodeling and microstructural and mechanical impairments, which were associated with exacerbated cartilage degradation. 2Characteristics of volatile compounds removal in biogas slurry of pig manure by ozone oxidation and organic solvents extraction显示文摘Biogas slurry is not suitable for liquid fertilizer due to its high amounts of volatile materials being of complicated composition and peculiar smell. In order to remove volatiles from biogas slurry efficiently, the dynamic headspace and gas chromatography-mass spectrometry were used to clear the composition of volatiles. Nitrogen stripping and superfluous ozone were also used to remove volatiles from biogas slurry. The results showed that there were 21 kinds of volatile compounds in the biogas slurry, including sulfur compounds, organic amines, benzene, halogen generation of hydrocarbons and alkanes, some of which had strong peculiar smell. The volatile compounds in biogas slurry can be removed with the rate of 53.0% by nitrogen stripping and with rate of 81.7% by the oxidization and stripping of the superfluous ozone. On this basis, the removal rate of the volatile compounds reached 99.2% by chloroform and n-hexane extraction, and almost all of odor was eliminated. 6Excitation energy transfer mechanisms in allophycocyanin(Ⅰ)——Energy transfer mechanisms in monomer显示文摘The mechanisms of excitation energy transfer within allophycocyanin monomer with the theory of generalized master equation (GME) and the technique of time-resolved fluorescence anisotropic spectroscopy are studied. In the case of known information of its structure and spectra, the theory applied is based on the assumption that the coupling interaction between two chromophores is fairly weak. The theory correctly predicts the experimentlly observed rate for excitation energy transfer in allophycocyanin monomer. 13The efficacy assessment of convalescent plasma therapy for COVID-19 patients:a multi-center case series显示文摘Convalescent plasma(CP)transfusion has been indicated as a promising therapy in the treatment for other emerging viral infections.However,the quality control of CP and individual variation in patients in different studies make it rather difficult to evaluate the efficacy and risk of CP therapy for coronavirus disease 2019(COVID-19).We aimed to explore the potential efficacy of CP therapy,and to assess the possible factors associated with its efficacy.We enrolled eight critical or severe COVID-19 patients from four centers.Each patient was transfused with 200–400mL of CP from seven recovered donors.The primary indicators for clinical efficacy assessment were the changes of clinical symptoms,laboratory parameters,and radiological image after CP transfusion.CP donors had a wide range of antibody levels measured by serology tests which were to some degree correlated with the neutralizing antibody(NAb)level.No adverse events were observed during and after CP transfusion.Following CP transfusion,six out of eight patients showed improved oxygen support status;chest CT indicated varying degrees of absorption of pulmonary lesions in six patients within 8 days;the viral load was decreased to a negative level in five patients who had the previous viremia;other laboratory parameters also tended to improve,including increased lymphocyte counts,decreased C-reactive protein,procalcitonin,and indicators for liver function.The clinical efficacy might be associated with CP transfusion time,transfused dose,and the NAb levels of CP.This study indicated that CP might be a potential therapy for severe patients with COVID-19.Hao Zeng Dongfang Wang Jingmin Nie Haoyu Liang Jiang Gu Anne Zhao Lixin Xu Chunhui Lang Xiaoping Cui Xiaolan Guo Changlong Zhou Haibo Li Bin Guo Jinyong Zhang Qiang Wang Li Fang Wen Liu Yishan Huang Wei Mao Yaokai Chen Quanming Zou 2020Signal Transduction and Targeted Therapy2020,5,1:1

15Synthesis and energy transfer of model conjugate of phycobilisome rods显示文摘The conjugate of R-phycoerythrin (R-PE) and C-phycocyanin (C-PC) was synthesized through a heterobifunctional coupling reagent, N-succinimidyl 3-(2-pyridyldithio) propionate. The molar ratio of R-PE to C-PC was determined by absorption spectra, and the result was 2:1. The energy transfer phenomena were observed from steady-state fluorescence spectra. The calculated result showed that the energy transfer efficiency from R-PE to C-PC was 88%. The energy transfer kinetics was determined by picosecond time-resolved fluorescence spectra. 17Superresolution live-cell imaging reveals that the localization of TMEM106B to filopodia in oligodendrocytes is compromised by the hypomyelination-related D252N mutation显示文摘Hypomyelination leukodystrophies constitute a group of heritable white matter disorders exhibiting defective myelin development.Initially identified as a lysosomal protein,the TMEM106B D252N mutant has recently been associated with hypomyelination.However,how lysosomal TMEM106B facilitates myelination and how the D252N mutation disrupts that process are poorly understood.We used superresolution Hessian structured illumination microscopy(Hessian-SIM)and spinning discconfocal structured illumination microscopy(SD-SIM)to find that the wild-type TMEM106B protein is targeted to the plasma membrane,filopodia,and lysosomes in human oligodendrocytes.The D252N mutation reduces the size of lysosomes in oligodendrocytes and compromises lysosome changes upon starvation stress.Most importantly,we detected reductions in the length and number of filopodia in cells expressing the D252N mutant.PLP1 is the most abundant myelin protein that almost entirely colocalizes with TMEM106B,and coexpressing PLP1 with the D252N mutant readily rescues the lysosome and filopodia phenotypes of cells.Therefore,interactions between TMEM106B and PLP1 on the plasma membrane are essential for filopodia formation and myelination in oligodendrocytes,which may be sustained by the delivery of these proteins from lysosomes via exocytosis.Shijia Xing Xiaolu Zheng Huifang Yan Yanquan Mo Ruoyu Duan Zhixing Chen Kunhao Wang Kai Gao Tongsheng Chen Shiqun Zhao Jingmin Wang Liangyi Chen 2023Science China(Life Sciences)2023,66,8:0

19Transcription factor ETV4 promotes the development of hepatocellular carcinoma by driving hepatic TNF-αsignaling显示文摘Background Hepatic inflammation is the major risk factor of hepatocellular carcinoma(HCC).However,the underlying mechanism by which hepatic inflammation progresses to HCC is poorly understood.This study was designed to investigate the role of ETS translocation variant 4(ETV4)in linking hepatic inflammation to HCC.Methods Quantitative real-time PCR and immunoblotting were used to detect the expression of ETV4 in HCC tissues and cell lines.RNA sequencing and luciferase reporter assays were performed to identify the target genes of ETV4.Hepatocyte-specific ETV4-knockout(ETV4fl/fl,alb-cre)and transgenic(ETV4Hep-TG)mice and diethylnitrosamine-carbon tetrachloride(DEN-CCL4)treatment experiments were applied to investigate the function of ETV4 in vivo.The Cancer Genome Atlas(TCGA)database mining and pathological analysis were carried out to determine the correlation of ETV4 with tumor necrosis factor-alpha(TNF-α)and mitogen-activated protein kinase 11(MAPK11).Results We revealed that ETV4 was highly expressed in HCC.High levels of ETV4 predicted a poor survival rate of HCC patients.Then we identified ETV4 as a transcription activator of TNF-αand MAPK11.ETV4 was positively correlated with TNF-αand MAPK11 in HCC patients.As expected,an increase in hepatic TNF-αsecretion and macrophage accumulation were observed in the livers of ETV4Hep-TG mice.The protein levels of TNF-α,MAPK11,and CD68 were significantly higher in the livers of ETV4Hep-TG mice compared with wild type mice but lower in ETV4fl/fl,alb-cre mice compared with ETV4fl/fl mice as treated with DEN-CCL4,indicating that ETV4 functioned as a driver of TNF-α/MAPK11 expression and macrophage accumulation during hepatic inflammation.Hepatocyte-specific knockout of ETV4 significantly prevented development of DEN-CCL4-induced HCC,while transgenic expression of ETV4 promoted growth of HCC.Conclusions ETV4 promoted hepatic inflammation and HCC by activating transcription of TNF-αand MAPK11.Both the ETV4/TNF-αand ETV4/MAPK11 axes represented two potential therapeutic targets for highly associated hepatic inflammation and HCC.ETV4+TNF-αwere potential prognostic markers for HCC patients.Dandan Qi Min Lu Pengfei Xu Xinli Yao Yongchen Chen Lipeng Gan Yong Li Yahua Cui Xiaomei Tong Shuhong Liu Jingmin Zhao Ningning Liu Xin Ye 2023Cancer Communications2023,43,12:0
20Cellular gp96 upregulates AFP expression by blocking NR5A2 SUMOylation and ubiquitination in hepatocellular carcinoma显示文摘Alpha-fetoprotein (AFP) is the most widely used biomarker for the diagnosis of hepatocellular carcinoma (HCC). However, asubstantial proportion of HCC patients have either normal or marginally increased AFP levels in serum, and the underlyingmechanisms are not fully understood. In the present study, we provided in vitro and in vivo evidence that heat shock protein gp96promoted AFP expression at the transcriptional level in HCC. NR5A2 was identified as a key transcription factor for the AFP gene, andits stability was enhanced by gp96. A further mechanistic study by co-immunoprecipitation, GST pull-down, and molecular dockingshowed gp96 and the SUMO E3 ligase RanBP2 competitively binding to NR5A2 at the sites spanning from aa 507 to aa 539. Thebinding of gp96 inhibited SUMOylation, ubiquitination, and subsequent degradation of NR5A2. In addition, clinical analysis of HCCpatients indicated that gp96 expression in tumors was positively correlated with serum AFP levels.
