1Comparative transcriptomics analysis of Zygosaccharomyces mellis under high-glucose stress显示文摘The high-glucose tolerance of yeast is the main factor determining the efficiency of high-density alcohol fermentation.Zygosaccharomyces mellis LGL-1 isolated from honey could survive under 700 g/L high-glucose stress and its tolerant characteristics were identified in our previous study.This study was performed to explore and clarify the high-glucose tolerance mechanism of Z.mellis LGL-1.Comparative transcriptomic analysis was used to analyze the genes with differential expression in Z.mellis under high-glucose conditions of 300,500 and 700 g/L.With 300 g/L samples as reference,there were 937 and 2380 differentially expressed genes(DEGs)in the 500 and 700 g/L samples,respectively.Meanwhile,there was 825 significant DEGs in the 700 g/L samples compared with that of the 500 g/L samples.The result revealed that transcriptional changes in multiple metabolic pathways occur in response to high-glucose stress.q-RT PCR analysis further confirmed that several stress response pathways,such as the high osmolarity glycerol mitogen-activated protein kinase(HOG-MAPK)signal transduction pathway,trehalose synthesis pathway and oxidative stress response are closely related to high-glucose tolerance in Z.mellis.This study clarifies mechanisms of Z.mellis in response to high-glucose osmotic stress,providing theoretical basis for the process control of high-density alcohol fermentation.Gongliang Liu Xinyu Bi Changli Tao Yongtao Fei Sujuan Gao Jinglong Liang Weidong Bai 2021Food Science and Human Wellness2021,10,1:2

3Claw-shaped flexible and low-impedance conductive polymer electrodes for EEG recordings: Anemone dry electrode显示文摘Electroencephalogram(EEG) signals provide important brain information in healthcare settings. Wet electrodes have several limitations, including their time-consuming preparation and no long-term acquisition, and dry electrodes have become a promising candidate. However, dry electrodes face several challenges related to signal quality, comfort, conductivity and cost.This study aims to develop and evaluate a novel dry electrode for EEG recordings. The electrode material properties are characterized by electrochemical, mechanical force, and skin measurements. The electrode performance is evaluated by comparing the impedance, signal-to-noise ratio(SNR), and EEG signal features of the proposed electrode with those of a wet electrode and two commercial dry electrodes. The dry electrode design is based on the biological structure of the sea anemone and demonstrates an improved scalp fit and reduced signal noise and motion artifacts. The electrode materials include advanced conducting polymers combined with thermoplastic elastomers(TPEs) and carbon nanotubes(CNTs), which exhibit good electrical conductivity, mechanical properties, and safety. A low-cost injection molding fabrication method is proposed. The dry electrodes show a scalp contact impedance of 7 kΩ at 20 Hz, which is lower than that of wet and commercial dry electrodes.Resting-state EEG and event-related potential signals collected by the anemone dry electrode achieved more than 90% similarity with signals acquired by wet electrodes. Thus, a low-cost, comfortable anemone dry electrode that exhibits excellent EEG recording performance is presented. 5Allogeneic gene-edited HIV-specific CAR-T cells secreting PD-1 blocking scFv enhance specific cytotoxic activity against HIV Envt cells in vivo显示文摘HIV-specific chimeric antigen receptor(CAR)T-cells have been developed to target HIV-1 infected CD4t T-cells that express HIV Env proteins.However,T cell exhaustion and the patient-specific autologous paradigm of CAR-T cell hurdled clinical applications.Here,we created HIV-specific CAR-T cells using human peripheral blood mononuclear cells and a 3BNC117-E27(3BE)CAR construct that enabled the expression of programmed cell death protein(PD-1)-blocking scFv E27 and the single-chain variable fragment of the HIV-1-specific broadly neutralizing antibody 3BNC117 to target native HIV Env.Compared with T cells expressing 3BNC117-CAR alone,3BE CAR-T cells showed greater cytotoxic activity against HIV Envt cells with stronger proliferation capability,higher killing efficiency,and enhanced cytokine secretion in the presence of HIV Env-expressing cells.Furthermore,we manufactured TCR-deficient 3BE CAR-T cells through gene editing and demonstrated that these CAR-T cells could effectively kill HIV Env^(+) cells in vivo without the occurrence of severe graft-versus-host disease(GvHD)in NSG mice.These data suggest that we have provided a feasible approach to the generation of“off-theshelf”anti-HIV CAR-T cells in combination with PD-1 checkpoint blockade immunotherapy,which can be a powerful therapeutic candidate for the functional cure of HIV.Hanyu Pan Xinyi Yang Jing Wang Huitong Liang Zhengtao Jiang Lin Zhao Yanan Wang Zhiming Liang Xiaoting Shen Qinru Lin Yue Liang Jinglong Yang Panpan Lu Yuqi Zhu Min Li Pengfei Wang Jianqing Xu Hongzhou Lu Huanzhang Zhu 2023Virologica Sinica2023,38,2:0
6High speed laser cladding as a new approach to prepare ultra-high temperature ceramic coatings显示文摘Ultra-high temperature ceramic(UHTC)coatings are used to protect the hot-end components of hypervelocity aerocrafts from thermal ablation.This study provides a new approach to fabricate UHTC coatings with high speed laser cladding(HSLC)technology,and places more emphasis on investigating the formation mechanism,phase compositions,and mechanical properties of HSLC-UHTC coatings.Results show that a well-bonded interface between the coating and the tantalum alloy substrate can be formed.The coating is mainly composed of(Zr,Ta)C ceramic solid solution phase with a content of higher than 90% by volume and Ta(W)metal solid solution phase.At a relatively high powder feeding rate,the ZrC ceramic phase appears in the coating while a dense ZrC UHTC top layer with a thickness of up to~50μm is successfully fabricated.As for the mechanical properties of the HSLC coatings,the fracture toughness of the coating decreases with the increase of powder feeding rate.The increase of carbide solid solution phase can significantly improve the high temperature microhardness(552.7±1.8 HV0.5@1000℃).The innovative design of HSLC ZrC-based coatings on refractory alloys accomplishes continuous transitions on microstructure and properties from the substrate to the UHTC top layer,which is a very promising candidate scheme for thermal protection coating.Qiyao Deng Pengfei He Chuan Sun Yang Li Yue Xing Yujie Wang Jinglong Li Xiubing Liang Xin Wang Yujie Zhou Yujun Cai 2024Journal of Advanced Ceramics2024,13,2:0

8Diffusion Characteristics of Swells in the North Indian Ocean显示文摘Research on the diffusion characteristics of swells contributes positively to wave energy forecasting, swell monitoring, and early warning. In this work, the South Indian Ocean westerly index(SIWI) and Indian Ocean swell diffusion effect index(IOSDEI) are defined on the basis of the 45-year(September 1957–August 2002) ERA-40 wave reanalysis data from the European Centre for Medium-Range Weather Forecasts(ECMWF) to analyze the impact of the South Indian Ocean westerlies on the propagation of swell acreage. The following results were obtained: 1) The South Indian Ocean swell mainly propagates from southwest to northeast. The swell also spreads to the Arabian Sea upon reaching low-latitude waters. The 2.0-meter contour of the swell can reach northward to Sri Lankan waters. 2) The size of the IOSDEI is determined by the SIWI strength. The IOSDEI requires approximately 2–3.5 days to fully respond to the SIWI. The correlations between SIWI and IOSDEI show obvious seasonal differences, with the highest correlations found in December–January–February(DJF) and the lowest correlations observed in June–July–August(JJA). 3) The SIWI and IOSDEI have a common period of approximately 1 week in JJA and DJF. 9Comparative Study of 3D-Printed Porous Titanium Alloy with Rod Designs of Three Different Geometric Structures for Orthopaedic Implantation显示文摘Porous titanium alloy is currently widely used in clinical treatment of orthopaedic diseases for its lower elastic modulus and ability to integrate with bone tissue.At the micro-level,cells can respond to different geometries,and at the macro-level,the geometric design of implants will also affect the biological function of cells.In this study,three kinds of porous scaffolds with square,triangular and circle rod shapes were designed and 3D printed.This study observed the proliferation and differentiation of MC3T3-E1 cells during surface culture of the three types of scaffolds.It also evaluated the characteristics of the three scaffolds by means of compression tests and scanning electron microscopy to provide a reference for the design of porous titanium alloy implants for clinical applications.The trends of cell proliferation and gene expression between the three types of scaffolds were observed after treatment with two inhibitors.The results show that the square rod porous scaffolds have the best proliferative and osteogenic activities,and these findings may be due to differences in piezo-type mechanosensitive ion channel component 1(Piezo1)and Yes-associated protein(YAP)expression caused by the macro-geometric topography.Jiaxin Li Haozhang Zhong Bojun Cao Zhaoyang Ran Jia Tan Liang Deng Yongqiang Hao Jinglong Yan 2024Acta Metallurgica Sinica(English Letters)2024,37,1:0
10Electrochemical process for recovery of metallic Mn from waste LiMn_(2)O_(4)-based Li-ion batteries in NaCl−CaCl_(2)melts显示文摘new method is proposed for the recovery of Mn via the direct electrochemical reduction of LiMn_(2)O_(4) from the waste of lithium-ion batteries in NaCl−CaCl_(2) melts at 750°C.The results show that the LiMn_(2)O_(4) reduction process by the electrochemical method on the coated electrode surface occurs in three steps:Mn(IV)→Mn(III)→Mn(II)→Mn.The products of this electro-deoxidation are CaMn2O4,MnO,(MnO)x(CaO)1−x,and Mn.Metal Mn appears when the electrolytic voltage increases to 2.6 V,which indicates that increasing the voltage may promote the deoxidation reaction process.With the advancement of the three-phase interline(3PI),electric deoxygenation gradually proceeds from the outer area of the crucible to the core.At high voltage,the kinetic process of the reduction reaction is accelerated,which generates double 3PIs at different stages.Jinglong Liang Dongbin Wang Le Wang Hui Li Weigang Cao Hongyan Yan 2022International Journal of Minerals,Metallurgy and Materials2022,29,3:0
