| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 3 | Gut Microbiota Dysbiosis Strengthens Kupffer Cell-mediated Hepatitis B Virus Persistence through Inducing Endotoxemia in Mice显示文摘Background and Aims:Change of gut microbiota com-position is associated with the outcome of hepatitis B virus(HBV)infection,yet the related mechanisms are not fully characterized.The objective of this study was to investigate the immune mechanism associated with HBV persistence induced by gut microbiota dysbiosis.Methods:C57BL/6 mice were sterilized for gut-microbiota by using an antibi-otic(ABX)mixture protocol,and were monitored for their serum endotoxin(lipopolysaccharide[LPS])levels.An HBV-replicating mouse model was established by performing HBV-expressing plasmid pAAV/HBV1.2 hydrodynamic injec-tion(HDI)with or without LPS,and was monitored for se-rum hepatitis B surface antigen,hepatitis B e antigen,HBV DNA,and cytokine levels.Kupffer cells(KCs)were purified from antibiotic-treated mice and HBV-replicating mice and analyzed for IL-10 production and T cell suppression ability.Results:ABX treatment resulted in increased serum LPS levels in mice.The KCs separated from both ABX-treated and LPS-treated HBV-replicating mice showed significantly increased IL-10 production and enhanced ability to suppress IFN-γproduction of TCR-activated T cells than the KCs sep-arated from their counterpart controls.HDI of pAAV/HBV1.2 in combination with LPS in mice led to a delayed HBV clear-ance and early elevation of serum IL-10 levels compared to pAAV/HBV1.2 HDI alone.Moreover,IL-10 function blockade or KC depletion led to accelerated HBV clearance in LPS-treated HBV-replicating mice.Conclusions:Our results suggest that dysbiosis of the gut microbiota in mice leads to endotoxemia,which induces KC IL-10 production and strengthens KC-mediated T cell suppression,and thus fa-cilitates HBV persistence. | Wenqing Zhou Jinzhuo Luo Xiaohong Xie Shangqing Yang Dan Zhu Hongming Huang Dongliang Yang Jia Liu | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 4 |

Recent studies on antibiotic therapy for infection after the surgical installation of implants to internal fractures were reviewed and analyzed.In general, systematic antibiotics are selected based on the results of bacterial culture. The duration of antibiotic treatment lasts for no more than 4 to 6 weeks. Orally administered and intravenously injected antibiotics have similar efficacies. Orally administered antibiotics exhibit a lower incidence of complications and are less costly than intravenously injected antibiotics. In addition, the efficacy of daptomycin in the treatment of bone infection is problematic. Rifampicin or fluoroquinolone antibiotics should be jointly administered when infection with bacterial biofilms is likely to occur. Calcium sulfate is a typical topically applied antibiotic delivery vehicle that can be completely degraded, with good biocompatibility, bone conduction, and drug release. | 13 | Macrophage membrane-biomimetic adhesive polycaprolactone nanocamptothecin for improving cancer-targeting efficiency and impairing metastasis显示文摘The recent remarkable success and safety of mRNA lipid nanoparticle technology for producing severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccines has stimulated intensive efforts to expand nanoparticle strategies to treat various diseases.Numerous synthetic nanoparticles have been developed for pharmaceutical delivery and cancer treatment.However,only a limited number of nanotherapies have enter clinical trials or are clinically approved.Systemically administered nanotherapies are likely to be sequestered by host mononuclear phagocyte system(MPS),resulting in suboptimal pharmacokinetics and insufficient drug concentrations in tumors.Bioinspired drug-delivery formulations have emerged as an alternative approach to evade the MPS and show potential to improve drug therapeutic efficacy.Here we developed a biodegradable polymer-conjugated camptothecin prodrug encapsulated in the plasma membrane of lipopolysaccharide-stimulated macrophages.Polymer conjugation revived the parent camptothecin agent(e.g.,7-ethyl-10-hydroxy-camptothecin),enabling lipid nanoparticle encapsulation.Furthermore,macrophage membrane cloaking transformed the nonadhesive lipid nanoparticles into bioadhesive nanocamptothecin,increasing the cellular uptake and tumor-tropic effects of this biomimetic therapy.When tested in a preclinical murine model of breast cancer,macrophage-camouflaged nanocamptothecin exhibited a higher level of tumor accumulation than uncoated nanoparticles.Furthermore,intravenous administration of the therapy effectively suppressed tumor growth and the metastatic burden without causing systematic toxicity.Our study describes a combinatorial strategy that uses polymeric prodrug design and cell membrane cloaking to achieve therapeutics with high efficacy and low toxicity.This approach might also be generally applicable to formulate other therapeutic candidates that are not compatible or miscible with biomimetic delivery carriers. | Kangkang Ying Yifeng Zhu Jianqin Wan Chenyue Zhan Yuchen Wang Binbin Xie Peirong Xu Hongming Pan Hangxiang Wang | 2023 | Bioactive Materials2023,,2: | 1 |

| 16 | Meta Ordinal Regression Forest for Medical Image Classification With Ordinal Labels显示文摘The performance of medical image classification has been enhanced by deep convolutional neural networks(CNNs),which are typically trained with cross-entropy(CE)loss.However,when the label presents an intrinsic ordinal property in nature,e.g.,the development from benign to malignant tumor,CE loss cannot take into account such ordinal information to allow for better generalization.To improve model generalization with ordinal information,we propose a novel meta ordinal regression forest(MORF)method for medical image classification with ordinal labels,which learns the ordinal relationship through the combination of convolutional neural network and differential forest in a meta-learning framework.The merits of the proposed MORF come from the following two components:A tree-wise weighting net(TWW-Net)and a grouped feature selection(GFS)module.First,the TWW-Net assigns each tree in the forest with a specific weight that is mapped from the classification loss of the corresponding tree.Hence,all the trees possess varying weights,which is helpful for alleviating the tree-wise prediction variance.Second,the GFS module enables a dynamic forest rather than a fixed one that was previously used,allowing for random feature perturbation.During training,we alternatively optimize the parameters of the CNN backbone and TWW-Net in the meta-learning framework through calculating the Hessian matrix.Experimental results on two medical image classification datasets with ordinal labels,i.e.,LIDC-IDRI and Breast Ultrasound datasets,demonstrate the superior performances of our MORF method over existing state-of-the-art methods. | Yiming Lei Haiping Zhu Junping Zhang Hongming Shan | 2022 | IEEE/CAA Journal of Automatica Sinica2022,9,7: | 0 |

| 19 | Predictive values of plasma TNFαand IL-8 for intracranial hemorrhage in patients with acute promyelocytic leukemia显示文摘In patients with acute promyelocytic leukemia(APL),intracranial hemorrhage(ICH),if not identified promptly,could be fatal.It is the leading cause of failure of induction and early death.Thus,biomarkers that could promptly predict severe complications are critical.Here,cytokine differences between patients with APL with and without ICH were investigated to develop predictive models for this complication.The initial cytokine profiling using plasma samples from 39 patients and 18 healthy donors found a series of cytokines that were remarkedly different between patients with APL and healthy controls.The APL patients were subsequently divided into high and low white blood cell count groups.Results showed that tumor necrosis factor a and interleukin 8(IL-8)were vital in distinguishing patients with APL who did or did not develop ICH.In addition,verification in 81 patients with APL demonstrated that the two cytokines were positively correlated with the cumulative incidence of ICH.Finally,in-vitro and in-vivo experimental evidence were provided to show that IL-8 influenced the migration of APL-derived NB4 cells and impaired the blood-brain barrier in PML/RARαpositive blast-transplanted FVB/NJ mice.These assessments may facilitate the early warning of ICH and reduce future mortality levels in APL. | Fangyi Dong Li Chen Chaoxian Zhao Xiaoyang Li Yun Tan Huan Song Wen Jin Hongming Zhu Yunxiang Zhang Kai Xue Junmin Li Kankan Wang | 2022 | Frontiers of Medicine2022,16,6: | 0 |
