1Zacopride selectively activates the Kir2.1 channel via a PKA signaling pathway in rat cardiomyocytes显示文摘We recently reported that zacopride is a selective inward rectifier potassium current (IK1 ) channel agonist, suppressing ventricular arrhythmias without affecting atrial arrhythmias. The present study aimed to investigate the unique pharmacological properties of zacopride. The whole-cell patch-clamp technique was used to study IK1 currents in rat atrial myocytes and Kir2.x currents in human embryonic kidney (HEK)-293 cells transfected with inward rectifier potassium channel (Kir)2.1, Kir2.2, Kir2.3, or mutated Kir2.1 (at phosphorylation site S425L). Western immunoblots were performed to estimate the relative protein expression levels of Kir2.x in rat atria and ventricles. Results showed that zacopride did not affect the IK1 and transmembrane potential of atrial myocytes. In HEK293 cells, zacopride increased Kir2.1 homomeric channels by 40.7%±9.7% at 50 mV, but did not affect Kir2.2 and Kir2.3 homomeric channels, and Kir2.1-Kir2.2, Kir2.1-Kir2.3 and Kir2.2-Kir2.3 heteromeric channels. Western immunoblots showed that similar levels of Kir2.3 protein were expressed in rat atria and ventricles, but atrial Kir2.1 protein level was only 25% of that measured in the ventricle. In addition, 5-hydroxytryptamine (5-HT) 3 receptor was undetectable, whereas 5-HT 4 receptor was weakly expressed in HEK293 cells. The Kir2.1-activating effect of zacopride in these cells was abolished by inhibition of protein kinase A (PKA), but not PKC or PKG. 2Deletion of the waaf gene affects O antigen synthesis and pathogenicity in Vibrio parahaemolyticus from shellfish显示文摘Vibrio parahaemolyticus is the main cause of foodborne gastroenteritis,which is widely distributed in shellfish and other seafood.Most V.parahaemolyticus are nonpathogenic,and only a few types,such as serotype O3:K6,are pathogenic,which is also the most prevalent strain in Asia.However,the relationship between this serotype and pathogenicity has yet to be established.The waaf gene is located in the O antigen synthesis gene cluster.Thus,we constructed a waaf gene deletion mutant(i.e.,Δwaaf)of wild-type(WT)which isolated from shellfish serotype O3:K6 via chitin-mediated transformation technology.We then constructed theΔwaaf complementary strain(i.e.,C-△waaf)via the Escherichia coli S17λpir strain by conjugation.The basic physiological characteristics,adhesion to Caco2 cells,and pathogenicity of the WT,△waaf,and C-△waaf strains were compared.Growth curves showed no remarkable differences between the WT andΔwaaf strains.However,theΔwaaf strain non-reactive to O3 antisera and other 12 O-group antisera of V.parahaemolyticus.Moreover,the number of flgella and extracellular polysaccharides decreased,the adhesion decreased,and the pathogenicity weakened.These characteristics of the C-△waaf strain were similar to those of the WT strain These results indicated that the waaf gene is vital to the serotype in V.parahaemolyticus,and changes in O antigen could affect the pathogenicity of this bacterium.This study will be helpful to understand the pathogenic mechanism of V.parahaemolyticus.Feng Zhao Guoying Ding Qilong Wang Huihui Du Guosheng Xiao Deqing Zhou 2022Food Science and Human Wellness2022,11,2:1

6Dihydroartemisinin beneficially regulates splenic immune cell heterogeneity through the SOD3-JNK-AP-1 axis显示文摘The immunomodulatory potential of dihydroartemisinin(DHA) has recently been highlighted;however, the potential mechanism remains to be clarified. Single-cell RNA sequencing was explored in combination with cellular and biochemical approaches to elucidate the immunomodulatory mechanisms of DHA. In this study, we found that DHA induced both spleen enlargement and rearrangement of splenic immune cell subsets in mice. It was revealed that DHA promoted the reversible expansion of effective regulatory T cells and interferon-γ^(+)cytotoxic CD8^(+)T cells in the spleen via induction of superoxide dismutase 3(SOD3) expression and increased phosphorylation of c-Jun N-terminal kinases(JNK) and its downstream activator protein 1(AP-1) transcription factors. Further, SOD3 knockout mice were resistant to the regulatory effect of DHA. 7Dihydroartemisinin regulates immune cell heterogeneity by triggering a cascade reaction of CDK and MAPK phosphorylation显示文摘Artemisinin(ART)and dihydroartemisinin(DHA),apart from their profound anti-malaria effect,can also beneficially modulate the host immune system;however,the underlying molecular mechanisms remain unclear.Here,we report that DHA selectively induced T-cell activation,with an increased proportion of Ki67^(+)CD4^(+)T cells,CD25^(+)CD4^(+)T cells,interferon(IFN)-γ-producing CD8^(+)T cells,Brdu^(+)CD8^(+)T cells and neutrophils,which was found to enhance cellular immunity to experimental malaria and overcome immunosuppression in mice.We further revealed that DHA upregulated the expression of cell proliferation-associated proteins by promoting the phosphorylation of mitogen-activated protein kinase(MAPK),cyclin-dependent kinases(CDKs),and activator protein 1 in the spleen.This study is the first to provide robust evidence that DHA selectively induced the expansion of subsets of splenic T cells through phosphorylated CDKs and MAPK to enhance cellular immune responses under non-pathological or pathological conditions.The data significantly deepened our knowledge in the mechanism underlying DHA-mediated immunomodulation.Qilong Li Quan Yuan Ning Jiang Yiwei Zhang Ziwei Su Lei Lv Xiaoyu Sang Ran Chen Ying Feng Qijun Chen 2022Signal Transduction and Targeted Therapy2022,7,8:0
8Distances Between Phylogenetic Trees: A Survey显示文摘Phylogenetic trees have been widely used in the study of evolutionary biology for representing the tree-like evolution of a collection of species. However, different data sets and different methods often lead to the construction of different phylogenetic trees for the same set of species. Therefore, comparing these trees to determine similarities or, equivalently, dissimilarities, becomes the fundamental issue. Typically, Tree Bisection and Reconnection(TBR)and Subtree Prune and Regraft(SPR) distances have been proposed to facilitate the comparison between different phylogenetic trees. 9A fixed-parameter algorithm for the maximum agreement forest problem on multifurcating trees显示文摘The Maximum Agreement Forest(MAF) problem on two given phylogenetic trees is an important NP-hard problem in the field of computational biology. In this paper, we study the parameterized version of the MAF problem: given two unrooted(multifurcating) phylogenetic trees T_1 and T_2 with the same leaf-label set L, and a parameter k, either construct an agreement forest of at most k trees for T_1 and T_2, or report that no such a forest exists. Whether there is a fixed-parameter tractable algorithm for this problem was posed as an open problem several times in the literature. 10Multirelationship Aware Personalized Recommendation Model显示文摘The existing methods using social information can alleviate the data sparsity issue in collaborative filtering recommendation,but they do not fully tap the complex and diverse user relationships,so it is difficult to obtain an accurate modeling representation of the user.To solve this,we propose a multirelationship aware personalized recommendation(MrAPR)model,which aggregates the various relationships between social users from two aspects of the user’s personal information and interaction sequence.Based on the comprehensive and accurate relationship graphs established,the graph neural network and attention network are used to adaptively distinguish the importance of different relationships and improve the aggregation reliability of multiple relationships.The MrAPR model better describes the characteristics of user interest and can be compatible with the existing sequence recommendation methods.The experimental results on two real-world datasets clearly show the effectiveness of the MrAPR model.Hongtao Song Feng Wang Zhiqiang Ma Qilong Han 2022国际计算机前沿大会会议论文集2022,,1:0

13Improved kernels for triangle packing in tournaments显示文摘Triangle packing problem has been paid lots of attention to in the literature.In this paper,we study the kernelization of the triangle packing problem in tournaments.For the parameterized arc-disjoint triangle packing problem in tournaments,we find a maximal arc-disjoint triangle packing with the number of vertices bounded by 2.5k.Based on the relation between the maximal arc-disjoint triangle packing and the vertices outside of the packing,a kernel of size 3.5k for the problem is obtained,improving the previous best one 6k.For the parameterized vertex-disjoint triangle packing problem in sparse tournaments,several new properties between the triangles in maximal vertex-disjoint triangle packing and the arcs in the feedback arc set are presented,which result in a kernel of size 7k for the problem,improving the previous best one of 15k.We also give a 7k vertex kernel for the parameterized feedback vertex set problem in sparse tournaments.The kernelization process presented in this paper for the parameterized arc-disjoint triangle packing can be applied to solve the parameterized arc-disjoint triangle packing problem on other restricted directed graph classes.Hanchun YUAN Qilong FENG Jianxin WANG 2023Science China(Information Sciences)2023,66,5:0
