| 1 | Isolation and diagnosis of Helicobacter pylori by a new method: Microcapillary culture显示文摘AIM:To investigate the performance of the microcapillary culture method(MCM) in Helicobacter pylori(H.pylori) isolation and diagnosis.METHODS:Microcapillary culture(MC),classical culture(CC),rapid urease(CLO) test,and histopathologic examination(HE) were performed with biopsy samples.Homogenized biopsy samples were loaded into capillary tubes and incubated for 48 h at 37 ℃ without providing a microaerophilic environment.Additionally,three or four loops of the homogenized sample were inoculated in a ready-to-use selective medium(Becton Dickinson,Helicobacter Agar,Modified) specific for the isolation of H.pylori and incubated at 37 ℃ in a microaerophilic atmosphere provided by Campy Gen(Becton Dickinson,Gas Pack).Bacteria reproducing in microcapillary tubes were evaluated in an inverted microscope and also were evaluated after performing a CC with the content.Results obtained by CC,CLO test,and HE were compared with those of MC.The diagnostic performances of the methods used in this study were evaluated for specificity,sensitivity,positive predictive value(PPV),negative predictive value(NPV),and CI.RESULTS:H.pylori was found positive by CLO test +HE and/or CC culture in 26 patient antrum and corpus biopsy samples.In 25(25/26) patient biopsy samples,H.pylori was isolated by MCM,whereas in only 14(14/26) patient biopsy samples,H.pylori was isolatedby CC.CLO test and HE were found positive in 17(17/26) patient biopsy samples.Comparing the results of the isolation of H.pylori by MCM,CC,CLO test,and HE,the sensitivity of the MCM was found as 96%,the specificity as 80%,the PPV as 83%,the NPV as 95%,and the 95%CI as 0.76(χ2 =31.51,P < 0.01) whereas the sensitivity of the CC was found as 54%(χ2 =19.15,P < 0.01),and the sensitivity of the CLO test and HE were found as 65%(χ2 =25.26,P < 0.01).CONCLUSION:This new microcapillary cultivation method for H.pylori has high diagnostic sensitivity compared with CC,HE,and CLO tests. | Adil M Allahverdiyev Melahat Bagirova Reyhan Caliskan Hrisi Bahar Tokman Hayat Aliyeva Gokce Unal Olga Nehir Oztel Emrah Sefik Abamor Hilal Toptas Pelin Yuksel Fatma Kalayci Mustafa Aslan Yusuf Erzin Kadir Bal Bekir S Kocazeybek | 2015 | World Journal of Gastroenterology2015,21,9: | 2 |
| 2 | Proteomic and metabolomic profiling underlines the stage- and time-dependent effects of high temperature on grape berry metabolism显示文摘Climate change scenarios predict an increase in mean air temperatures and in the frequency,intensity,and length of extreme temperature events in many wine-growing regions worldwide.Because elevated temperature has detrimental effects on berry growth and composition,it threatens the economic and environmental sustainability of wine production.Using Cabernet Sauvignon fruit-bearing cuttings,we investigated the effects of high temperature(HT)on grapevine berries through a label-free shotgun proteomic analysis coupled to a complementary metabolomic study.Among the 2,279 proteins identified,592 differentially abundant proteins were found in berries exposed to HT.The gene ontology categories“stress,”“protein,”“secondary metabolism,”and“cell wall”were predominantly altered under HT.High temperatures strongly impaired carbohydrate and energy metabolism,and the effects depended on the stage of development and duration of treatment.Transcript amounts correlated poorly with protein expression levels in HT berries,highlighting the value of proteomic studies in the context of heat stress.Furthermore,this work reveals that HT alters key proteins driving berry development and ripening.Finally,we provide a list of differentially abundant proteins that can be considered as potential markers for developing or selecting grape varieties that are better adapted to warmer climates or extreme heat waves. | David Lecourieux Christian Kappel Stéphane Claverol Philippe Pieri Regina Feil John E.Lunn Marc Bonneu Lijun Wang Eric Gomès Serge Delrot Fatma Lecourieux | 2020 | Journal of Integrative Plant Biology2020,62,8: | 2 |
| 3 | Control of Tetranychus urticae Koch by extracts of three essential oils of chamomile,marjoram and Eucalyptus显示文摘Objective:To evaluate the acaricidal activity of extracts of three essential oils of chamomile,marjoram and Eucalyptwi against Tetranychus urtiaie(T.urticae.)Koch.Methods:Extracts of three essential oils of chamomile,marjoram and Eucalyptus with different concentrations(0.5%,1.0%,2.0%,3.0%and 4.0%)were used to control T.urticae Koch.Results:The results showed that chamomile(Chamomilla recutita)represented the most potent efficient acaricidal agent against Tetranychus followed by marjoram(Marjorana hortensis)and Eucalyptus.The LC_(50)values of chamomile,marjoram and Eucalyptus for adults were 0.65,1.84 and 2.18,respectively and for eggs 1.17,6.26 and 7.33,respectively.Activities of enzymes including glutathione-Stxansferase,esterase(α-esterase andβ-esterase)and alkaline phosphatase in susceptible mites were determined and activities of enzymes involved in the resistance of acaricides were proved.Protease enzyme was significantly decreased at LC_(50)of both chamomile and marjoram compared with positive control.Gas chromatography-mass spectrometer(CC-MS)proved that the major compositions of Chamomilla recutita areα-bisabolol oxide A(35.251%),and trans-3-farersene(7.758%),while the main components of Marjorana hortensis are terpincne-4-ol(23.860%),p-cymene(23.404%)and sabinene(10.904%).Conclusions:It can be concluded that extracts of three essential oils of chamomile,marjoram and Eucalyptus possess acaricidal activity against T.urticae. | Abd El-Moneim MR Afify Fatma S Ali Turky AF | 2012 | Asian Pacific Journal of Tropical Biomedicine2012,2,1: | 2 |

| 5 | Effects of moxifloxacin exposure on the conjunctival flora and antibiotic resistance profile following repeated intravitreal injections显示文摘AIM:To evaluate the effects of moxifloxacin exposure on the conjunctival flora and antibiotic resistance profile following repeated intravitreal injections.METHODS:Seventy-two eyes of 36 patients [36 eyes in control group, 36 eyes in intravitreal injection(IVI) group]were enrolled in the study. All the eyes had at least one IVI and had diabetic macular edema(DME) or age-related macular degeneration(ARMD). Moxifloxacin was prescribed to all the patients four times a day for five days following injection. Conjunctival cultures were obtained from the lower fornix via standardized technique with every possible effort made to minimize contamination from the lids, lashes, or skin. Before the application of any ophthalmic medication, conjunctival cultures were obtained from both eyes using sterile cotton culture. An automated microbiology system was used to identify the growing bacteria and determine antibiotic sensitivity.RESULTS:The bacterial cultures were isolated from 72 eyes of 36 patients, sixteen of whom patients(44.4%)were male and twenty(55.6%) were female. Average age was 68.4 ±9.0(range 50-86). The average number of injections before taking cultures was 3.1+1.0. Forty-eight(66.7%) of 72 eyes had at least one significant organism.There was no bacterial growth in 8(20.5%) of IVI eyes and in 16(44.4%) of control eyes(P =0.03). Of the bacteria isolated from culture, 53.8% of coagulase negative staphylococci(CoNS) in IVI eyes and 47.2%CoNS in control eyes. This difference between IVI eyes and control eyes about bacteria isolated from culture was not statistically significant(P =0.2). Eleven of 25 bacteria(44.0%) isolated from IVI eyes and 11(57.9%) of 19 bacteria isolated from control eyes were resistant to oxacillin. The difference in frequency of moxifloxacine resistance between two groups was not statistically significant(12.0% in IVI eyes and 21.1% in control eyes)(P =0.44). There were no cases of resistance to vancomycin, teicoplanin and linezolid.CONCLUSION:There was no difference in species of bacteria isolated from cultures, or in the frequency of resistance to antibiotics between eyes that had recurrent IVI followed by moxifloxacin exposure compared with control eyes. | 12 | Assessment of hepatic fibrosis in pediatric cases with hepatitis C virus in Egypt显示文摘AIM: To assess hepatic fibrosis and factors associated with its progression in children with HCV infection. METHODS: At the Hepatology Unit,Cairo University Children's Hospital,a single liver biopsy was performed to 43 children with HCV infection after an informed consent between 1998-2004. Their mean age at liver biopsy was 8.67 ± 4.3 years. RESULTS: Among the 43 patients' biopsies,12 (27.9%) were having no fibrosis,20 (46.5%) mild fibrosis and 11 (25.6%) moderate to severe fibrosis. The median time for development of fibrosis was estimated to be 5.5 years. Developing fibrosis was significantly associated with shorter duration from first detected ALT elevation to biopsy (12 mo vs 1.2 mo,P = 0.015) and having higher levels of direct serum bilirubin (0.3 mg/dL vs 0.5 mg/dL,P = 0.048). No association was found between fibrosis stage and the presence of co-morbid conditions (P = 0.33). CONCLUSION: Hepatic fibrosis was present in 72.1% of children with HCV infection. The development of fibrosis was associated with higher levels of direct serum bilirubin.
