| 2 | Collagen type Ⅱ suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction显示文摘Hypertrophic differentiation is not only the terminal process of endochondral ossification in the growth plate but is also an important pathological change in osteoarthritic cartilage.Collagen type II(COL2A1)was previously considered to be only a structural component of the cartilage matrix,but recently,it has been revealed to be an extracellular signaling molecule that can significantly suppress chondrocyte hypertrophy.However,the mechanisms by which COL2A1 regulates hypertrophic differentiation remain unclear.In our study,a Col2a1 p.Gly1170Ser mutant mouse model was constructed,and Col2a1 loss was demonstrated in homozygotes.Loss of Col2a1 was found to accelerate chondrocyte hypertrophy through the bone morphogenetic protein(BMP)-SMAD1 pathway.Upon interacting with COL2A1,integrinβ1(ITGB1),the major receptor for COL2A1,competed with BMP receptors for binding to SMAD1 and then inhibited SMAD1 activation and nuclear import.COL2A1 could also activate ITGB1-induced ERK1/2 phosphorylation and,through ERK1/2-SMAD1 interaction,it further repressed SMAD1 activation,thus inhibiting BMP-SMAD1-mediated chondrocyte hypertrophy.Moreover,COL2A1 expression was downregulated,while chondrocyte hypertrophic markers and BMP-SMAD1 signaling activity were upregulated in degenerative human articular cartilage.Our study reveals novel mechanisms for the inhibition of chondrocyte hypertrophy by COL2A1 and suggests that the degradation and decrease in COL2A1 might initiate and promote osteoarthritis progression. | Chengjie Lian Xudong Wang Xianjian Qiu Zizhao Wu Bo Gao Lei Liu Guoyan Liang Hang Zhou Xiaoming Yang Yan Peng Anjing Liang Caixia Xu Dongsheng Huang Peiqiang Su | 2019 | Bone Research2019,7,1: | 17 |
| 3 | Small RNAs, emerging regulators critical for the development of horticultural traits显示文摘Small RNAs(sRNAs)have been recently recognized as key genetic and epigenetic regulators in various organisms,ranging from the modification of DNA and histone methylations to the modulation of the abundance of coding or non-coding RNAs.In plants,major regulatory sRNAs are classified as respective microRNA(miRNA)and small interfering RNA(siRNA)species,with the former primarily engaging in posttranscriptional regulation while the latter in transcriptional one.Many of these characterized sRNAs are involved in regulation of diverse biological programs,processes,and pathways in response to developmental cues,environmental signals/stresses,pathogen infection,and pest attacks.Recently,sRNAs-mediated regulations have also been extensively investigated in horticultural plants,with many novel mechanisms unveiled,which display far more mechanistic complexity and unique regulatory features compared to those studied in model species.Here,we review the recent progress of sRNA research in horticultural plants,with emphasis on mechanistic aspects as well as their relevance to trait regulation.Given that major and pioneered sRNA research has been carried out in the model and other plants,we also discuss ongoing sRNA research on these plants.Because miRNAs and phased siRNAs(phasiRNAs)are the most studied sRNA regulators,this review focuses on their biogenesis,conservation,function,and targeted genes and traits as well as the mechanistic relation between them,aiming at providing readers comprehensive information instrumental for future sRNA research in horticulture crops. | Chengjie Chen Zaohai Zeng Zongrang Liu Rui Xia | 2018 | Horticulture Research2018,5,1: | 12 |

| 5 | Coexpression network analysis reveals an MYB transcriptional activator involved in capsaicinoid biosynthesis in hot peppers显示文摘Plant biosynthesis involves numerous specialized metabolites with diverse chemical natures and biological activities.The biosynthesis of metabolites often exclusively occurs in response to tissue-specific combinatorial developmental cues that are controlled at the transcriptional level.Capsaicinoids are a group of specialized metabolites that confer a pungent flavor to pepper fruits.Capsaicinoid biosynthesis occurs in the fruit placenta and combines its developmental cues.Although the capsaicinoid biosynthetic pathway has been largely characterized,the regulatory mechanisms that control capsaicinoid metabolism have not been fully elucidated.In this study,we combined fruit placenta transcriptome data with weighted gene coexpression network analysis(WGCNA)to generate coexpression networks.A capsaicinoid-related gene module was identified in which the MYB transcription factor CaMYB48 plays a critical role in regulating capsaicinoid in pepper.Capsaicinoid biosynthetic gene(CBG)and CaMYB48 expression primarily occurs in the placenta and is consistent with capsaicinoid biosynthesis.CaMYB48 encodes a nucleus-localized protein that primarily functions as a transcriptional activator through its C-terminal activation motif.CaMYB48 regulates capsaicinoid biosynthesis by directly regulating the expression of CBGs,including AT3a and KasIa.Taken together,the results of this study indicate ways to generate robust networks optimized for the mining of CBG-related regulators,establishing a foundation for future research elucidating capsaicinoid regulation. | Binmei Sun Xin Zhou Changming Chen Chengjie Chen Kunhao Chen Muxi Chen Shaoqun Liu Guoju Chen Bihao Cao Fanrong Cao Jianjun Lei Zhangsheng Zhu | 2020 | Horticulture Research2020,7,1: | 5 |

| 10 | Circulation Features Associated with the Record-breaking Typhoon Silence in August 2014显示文摘Climatologically, August is the month with the most tropical cyclone(TC) formation over the western North Pacific(WNP) during the typhoon season. In this study, the reason for abnormal TC activity during August is discussed—especially August 2014, when no TCs formed. The large-scale background of August 2014 is presented, with low-level large-scale easterly anomalies and anticyclonic anomalies dominating over the main TC genesis region, a weak monsoon trough system,and a strong WNP subtropical high(WPSH), leading to significantly reduced low-level convergence, upper-level divergence,and mid-level upward motion. These unfavorable large-scale conditions suppressed convection and cyclogenesis. In August2014, equatorial waves were inactive within the negative phase of the Madden–Julian Oscillation(MJO), with fewer tropical disturbances. Although the low-level vorticity and convection of those disturbances were partly promoted by the convective envelopes of equatorial waves, the integral evolution of disturbances, as well as the equatorial waves, were suppressed when propagating into the negative MJO phase. Moreover, the upper-level potential vorticity(PV) streamers associated with anticyclonic Rossby wave breaking events imported extratropical cold and dry air into the tropics. | 20 | Mean Flow–Storm Track Relationship and Rossby Wave Breaking in Two Types of El-Nino显示文摘The features of large-scale circulation, storm tracks and the dynamical relationship between them were examined by investigating Rossby wave breaking(RWB) processes associated with Eastern Pacific(EP) and Central Pacific(CP) El-Nin o. During EP El-Nin o, the geopotential height anomaly at 500 hPa(Z500) exhibits a Pacific–North America(PNA) pattern. During CP El-Nin o, the Z500 anomaly shows a north positive–south negative pattern over the North Pacific. The anomalous distributions of baroclinicity and storm track are consistent with those of upper-level zonal wind for both EP and CP El-Nin o, suggesting impacts of mean flow on storm track variability. Anticyclonic wave breaking(AWB) occurs less frequently in EP El-Nin o years, while cyclonic wave breaking(CWB) occurs more frequently in CP El-Nin o years over the North Pacific sector. Outside the North Pacific, more CWB events occur over North America during EP El-Nin o. When AWB events occur less frequently over the North Pacific during EP El-Nin o, Z500 decreases locally and the zonal wind is strengthened(weakened) to the south(north). This is because AWB events reflect a monopole high anomaly at the centroid of breaking events. When CWB events occur more frequently over the North Pacific under CP El-Nin o conditions, and over North America under EP El-Nin o condition, Z500 increases(decreases) to the northeast(southwest), since CWB events are related to a northeast–southwest dipole Z500 anomaly.
