1Metabolic dysregulation and emerging therapeutical targets for hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is an aggressive human cancer with increasing incidence worldwide.Multiple efforts have been made to explore pharmaceutical therapies to treat HCC,such as targeted tyrosine kinase inhibitors,immune based therapies and combination of chemotherapy.However,limitations exist in current strategies including chemoresistance for instance.Tumor initiation and progression is driven by reprogramming of metabolism,in particular during HCC development.Recently,metabolic associated fatty liver disease(MAFLD),a reappraisal of new nomenclature for nonalcoholic fatty liver disease(NAFLD),indicates growing appreciation of metabolism in the pathogenesis of liver disease,including HCC,thereby suggesting new strategies by targeting abnormal metabolism for HCC treatment.In this review,we introduce directions by highlighting the metabolic targets in glucose,fatty acid,amino acid and glutamine metabolism,which are suitable for HCC pharmaceutical intervention.We also summarize and discuss current pharmaceutical agents and studies targeting deregulated metabolism during HCC treatment.Furthermore,opportunities and challenges in the discovery and development of HCC therapy targeting metabolism are discussed.Danyu Du Chan Liu Mengyao Qin Xiao Zhang Tao Xi Shengtao Yuan Haiping Hao Jing Xiong 2022Acta Pharmaceutica Sinica B2022,12,2:27
2The recommendations of Chinese Parkinson’s disease and movement disorder society consensus on therapeutic management of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is a chronic,progressive and debilitating disease,which affects over 2.5 million people in China.PD is characterized clinically by resting tremor,muscular rigidity,bradykinesia and postural instability.As the disease progresses,additional complications can arise such as non-motor and neurobehavioral symptoms.Pharmacological treatment and surgical intervention for PD have been implemented in China.Until 10 years ago,there was lack of standardization for the management of PD in different regions and among different physicians,leading to different treatment levels in different regions and different physicians.Since then,the Chinese Parkinson’s Disease and Movement Disorder Society have published three versions of guidelines for the management of PD in China,in 2006,2009 and 2014,respectively.Correspondingly,the overall level of treatment for PD in China improved.Objectives:To update the treatment guidelines based on current foreign and domestic practice guidelines and clinical evidence,and to improve the treatment options available to physicians in the management of PD.Summary:A variety of treatment recommendations in the treatment guidelines have been proposed,including physical activity and disease-modifying medication,which should be initiated at the early-stage of the disease.The principles of dosage titration should be followed to avoid acute adverse reactions to the drugs,to achieve a satisfactory clinical effect with a low dose and to reduce the incidence of long-term motor complications.Moreover,different treatment strategies should be considered at different stages of the disease.Importantly,treatment guidelines and personalized treatments should be valued equally.A set of treatment recommendations has been developed to assist physicians to improve and optimize clinical outcomes for patients with PD in China.Shengdi Chen Piu Chan Shenggang Sun Haibo Chen Baorong Zhang Weidong Le Chunfeng Liu Guoguang Peng Beisha Tang Lijuan Wang Yan Cheng Ming Shao Zhenguo Liu Zhenfu Wang Xiaochun Chen Mingwei Wang Xinhua Wan Huifang Shang Yiming Liu Pingyi Xu Jian Wang Tao Feng Xianwen Chen Xingyue Hu Anmu Xie Qin Xiao 2016Translational Neurodegeneration2016,5,1:23
3Clinical and Familial Characteristics of Ten Chinese Patients with Fatal Family Insomnia显示文摘Objective Fatal familial insomnia(FFI) is an autosomal dominant prion disease characterized clinically by inattention,sleep loss,dysautonomia,and motor signs.This study is aimed to investigate clinical and familial characteristics of ten Chinese Patients with FFI.Methods We identified ten FFI cases from the surveillance network for Creutafeldt-Jakob disease(CJD) in China.Final diagnosis of FFI cases was made in accordance with the WHO criteria for CJD.The main clinical features and family histories of these ten FFI cases were analyzed.Results The median age of ten cases at onset was 38 years(from 19 to 55).The foremost symptoms seemed to be various,including sleep disturbances,vision disorder,dizziness and anorexia.Sleep disturbances appeared in all cases and lasted in the whole clinical courses.Progressive sympathetic symptoms,memory loss,movement disturbances,myoclonus and hypertension were also frequently observed.The median duration of the disease was 9.5 months.EEG and MRI did not figure out special abnormality.14-3-3 protein in CSF was positive in five out of eight tested patients.Clear family histories were identified in 8 patients.Conclusion The data from our study confirm that the Chinese FFI cases have similar clinical characteristics as that of the Caucasian cases.Compared with other genetic CJD associated mutations,the genetic frequencies of D178N in PRNP are apparently high among the Chinese cases.SHI Qi CHEN Cao GAO Chen TIAN Chan ZHOU Wei ZHANG BaoYun HAN Jun DONG Xiao Ping 2012Biomedical and Environmental Sciences2012,25,4:8

8Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized,whether and how liver-intrinsic factors drive metastatic spread is incompletely understood.Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver,which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models,respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling,which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors.In summary,this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance.Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention.Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng 2021Cellular & Molecular Immunology2021,18,4:5

10Moisture Origins and Transport Processes for the 2020 Yangtze River Valley Record-Breaking Mei-yu Rainfall显示文摘The summer of 2020 recorded a record-breaking flood due to excessive mei-yu rain falling over the Yangtze River Valley(YRV).Using the Lagrangian model FLEXPART,this paper investigates moisture sources and transport processes behind this extreme event.Based on climate data from 1979 to 2019,the air-particle(an infinitesimally small air parcel)trajectories reaching the YRV show sectors that correspond to five main moisture sources:the Indian monsoon region(IND,27.5%of the total rainfall),the local evaporation(27.4%),the Western Pacific Ocean(WPO,21.3%),the Eurasian continent(8.5%)and Northeast Asia(4.4%).In the 2020 mei-yu season,moisture from all source regions was above normal except that from Northeast Asia.A record-breaking moisture source from the IND and WPO dominated this extreme mei-yu flood in 2020,which was 1.5 and 1.6 times greater than the climate mean,respectively.This study reveals a significant relationship between the moisture source with three moisture transport processes,i.e.,trajectory density,moisture content,and moisture uptake of air-particles.A broad anomalous anticyclonic circulation over the Indo-Northwestern Pacific(Indo-NWP)provides a favorable environment to enhance the moisture transport from the IND and WPO into the YRV.In the 2020 mei-yu season,a record-breaking Indo-NWP anomalous anticyclonic circulation contributed to a higher trajectory density as well as higher moisture content and moisture uptake of air-particles from the IND and WPO regions.This collectively resulted in unprecedented moisture transport from source origins,thus contributing to the mei-yu flood over the YRV in 2020.Lixia ZHANG Dan ZHAO Tianjun ZHOU Dongdong PENG Chan XIAO 2021Advances in Atmospheric Sciences2021,38,12:4
11Building metallurgical bonding interfaces in an immiscible Mo/Cu system by irradiation damage alloying (IDA)显示文摘For the immiscible Mo/Cu system with a positive heat of mixing(△H_m > 0), building metallurgical bonding interfaces directly between immiscible Mo and Cu and preparing Mo/Cu laminar metal matrix composites(LMMCs) are very difficult. To solve the problem, a new alloying method for immiscible systems, which is named as irradiation damage alloying(IDA), is presented in this paper. The IDA primarily consists of three steps. Firstly, Mo is damaged by irradiation with multi-energy(186, 62 keV) Cu ion beams at a dose of 2 × 10^(17) ions/cm^2. Secondly, Cu layers are superimposed on the surfaces of the irradiation-damaged Mo to obtain Mo/Cu laminated specimens. Thirdly, the irradiation damage induces the diffusion alloying between Mo and Cu when the laminated specimens are annealed at 950℃ in a protective atmosphere.Through IDA, Mo/Cu LMMCs are prepared in this paper. The tensile tests carried out for the Mo/Cu LMMCs specimens show that the Mo/Cu interfaces constructed via IDA have high normal and shear strengths.Additionally, the microstructure of the Mo/Cu interface is characterized by High Resolution Transmission Electron Microscopy(HRTEM), X-ray diffraction(XRD) and Energy Dispersive X-ray(EDX) attached in HRTEM. The microscopic characterization results show that the expectant diffusion between Mo and Cu occurs through the irradiation damage during the process of IDA. Thus a Mo/Cu metallurgical bonding interface successfully forms. Moreover, the microscopic test results show that the Mo/Cu metallurgical interface is mainly constituted of crystalline phases with twisted and tangled lattices, and amorphous phase is not observed. 12Adsorption of uranium(Ⅵ) onto amidoxime-functionalized ultrahigh molecular weight polyethylene fibers from aqueous solution显示文摘Amidoxime-functionalized ultra-high molecular weight polyethylene fibers(UHMWPEF-AO) were used to absorb uranium U(VI) from aqueous solutions.In this paper,we study effects of pH,initial U(VI) concentration,contact time,and temperature on U(VI) adsorption by UHMWPEF-AO.The adsorption process agrees well with pseudo-second-order and Langmuir model.UHMWPEFAO exhibits excellent adsorptive performance for U(VI)with a maximum adsorption capacity of 176.12 mg/g at pH4 and 298 K.The structures of UHMWPEF-AO and U(VI)-loaded UHMWPEF-AO are characterized by FT-IR and nano-CT.U(VI)-loaded UHMWPEF-AO is sintered after adsorption process to recycle absorbed U(VI).Powders collected after sintering process are examined by scanning electron microscopy and X-ray diffraction.These results indicate that UHMWPEF-AO is a promising candidate to remove U(VI) from uranium aqueous solutions.Chun-Yu Xie Shi-Pei Jing Yu Wang Xiao Lin Hong-Liang Bao Cheng-Zhi Guan Chan Jin Jian-Qiang Wang 2017Nuclear Science and Techniques2017,28,7:3
13Performance of BCC-CSM Models with Different Horizontal Resolutions in Simulating Extreme Climate Events in China显示文摘In this study,the performance of the Beijing Climate Center(BCC)Climate System Model version 1.1(BCCCSM1.1)(280-km resolution)and the BCC-CSM1.1 m(110-km resolution)in simulating extreme climate events over China in the last 40 years is compared.Both models capture the main spatial distribution features of heavy precipitation(R95T),the number of consecutive wet days(CWD),the annual count of days with precipitation≥1 mm(R1),the maximum consecutive 5-day precipitation(Rx5),and the numbers of frost days(FD)and summer days(SU).The BCC-CSM1.1 m has a better ability to simulate the detailed distribution of extreme climate events than the BCCCSM1.1,including R95T,CWD,R1,and the simple precipitation intensity index(SDII).However,the BCCCSM 1.1 m does not show an improvement in simulating the number of days with extreme precipitation(R90N),the number of consecutive dry days(CDD),the heat wave duration index(HWDI),the warm day frequency(TX90 P),and cold night frequency(TN10P).This indicates that the simulation of the R95T,CWD,R1,and SDII climate events is more sensitive to the resolution of the model.The improved BCC-CSM1.1 m is used to explore the projection of extreme climate change in China during the 21 st century under the RCP4.5(Representative Concentration Pathways)and RCP8.5 scenarios.The results show that extreme precipitation will increase dramatically over North and Southwest China in the late 21 st century.The CWD index will decrease on the Tibetan Plateau and in northeastern and central China and will increase in other parts of China;R1 will increase in northern China and decrease in southern China;Rx5 will increase dramatically in southern China;FD will decrease and SU will increase over China in the late 21 st century under both emission scenarios,with larger amplitudes in RCP8.5.Linxiao WEI Xiaoge XIN Chan XIAO Yonghua LI Yao WU Hongyu TANG 2019Journal of Meteorological Research2019,33,4:3

15HMGB1 from Astrocytes Promotes EAE by Influencing the Immune Cell Infiltration-Associated Functions of BMECs in Mice显示文摘High mobility group box 1(HMGB1)has been reported to play an important role in experimental autoimmune encephalomyelitis(EAE).Astrocytes are important components of neurovascular units and tightly appose the endothelial cells of microvessels by their perivascular endfeet and directly regulate the functions of the blood-brain barrier.Astrocytes express more HMGB1 during EAE while the exact roles of astrocytic HMGB1 in EAE have not been well elucidated.Here,using conditional-knockout mice,we found that astrocytic HMGB1 depletion decreased morbidity,delayed the onset time,and reduced the disease score and demyelination of EAE.Meanwhile,there were fewer immune cells,especially pathogenic T cells infiltration in the central nervous system of astrocytic HMGB1 conditional-knockout EAE mice,accompanied by up-regulated expression of the tight-junction protein Claudin5 and down-regulated expression of the cell adhesion molecules ICAM1 and VCAM1 in vivo.In vitro,HMGB1 released from astrocytes decreased Claudin5 while increased ICAM1 and VCAM1 expressed by brain microvascular endothelial cells(BMECs)through TLR4 or RAGE.Taken together,our results demonstrate that HMGB1 derived from astrocytes aggravates EAE by directly influencing the immune cell infiltration-associated functions of BMECs.Junyu Shi Yifan Xiao Na Zhang Mengya Jiao Xuhuan Tang Chan Dai Chenchen Wang Yong Xu Zheng Tan Feili Gong Fang Zheng 2022Neuroscience Bulletin2022,38,11:3

17FTIR Spectroscopy in Cultural Heritage Studies: Non-destructive Analysis of Chinese Handmade Papers显示文摘Chinese handmade papers have been the carriers of paper-based documents, paintings and calligraphies, which are of great importance for historical and cultural heritage studies. This research developed the attenuated total reflectance-Fourier transform infrared(ATR-FTIR) spectroscopic technique for the analysis of Chinese handmade papers. The test was applied to Chinese handmade papers taken from bark, hemp and bamboo groups, followed by papers from ancient books and arts collected in library and museum. Our study indicated tliat FTIR was an efficient analytical method to identify fiber types of library/museum papers of unknown origin. Moreover, information about degree of crystallinity and state of deterioration of cellulosic fibers was learned from FTIR spectra. 18S6B-2 Matrine Inhibits Itching by Lowering the Activity of Calcium Channel显示文摘Sophorae Flavescentis Radix(SFR)is a medicinal herb with many functions that are involved in anti-inflammation,antinociception,and anticancer.SFR is also used to treat a variety of itching diseases.Matrine(MT)is one of the main constituents in SFR and also has the effect of relieving itching,but the antipruritic mechanism is still unclear.Here,we investigated the effect of MT on antipruritus.In acute and chronic itch models,MT significantly inhibited the scratching behavior not only in acute itching induced by histamine(His),chloroquine(CQ)and compound 48/80 with a dose-depended manner,but also in the chronic pruritus models of Atopic dermatitis(AD)and Acetone-ether-water(AEW)in mice.Furthermore,MT can be detected in the blood after intraperitoneal injection(i.p.)and subcutaneous injection(s.c.).Finally,electrophysiological and calcium image results show that MT inhibits the excitatory synaptic transmission from dorsal root ganglion(DRG)to the dorsal horn of the spinal cord by suppressing presynaptic N-type calcium channels.Taken together,we believe that MT is a novel drug candidate in treating pruritus diseases,especially for histamine-independent and chronic pruritus,which might be attributed to inhibition of presynaptic N-type calcium channels.GENG Xiao SHI Hao YE Fan DU Han QIAN Lin-nan GU Le-ying WU Guan-yi ZHU Chan YANG Yan WANG Chang-ming ZHOU Yuan YU Guang LIU Qin DONG Xin-zhong YU Lei 唐宗湘 2018神经药理学报2018,8,4:2
19Real-time simultaneous recording of electrophysiological activities and dopamine overflow in the deep brain nuclei of a non-human primate with Parkinson’s disease using nano-based microelectrode arrays显示文摘Parkinson’s disease(PD)is characterized by a progressive degeneration of nigrostriatal dopaminergic neurons.The precise mechanisms are still unknown.Since the neuronal communications are inherently electrical and chemical in nature,dual-mode detection of PD-related neuroelectrical and neurochemical information is essential for PD research.Subthalamic nucleus(STN)highfrequency stimulation(HFS)can improve most symptoms of PD patients and decrease the dosage of antiparkinsonian drugs.The mechanism of STN-HFS for PD still remains elusive.In this study,a silicon-based dual-mode microelectrode array(MEA)probe was designed and fabricated,and systematic dual-mode detection methods were established.The recording sites were modified using Pt nanoparticles and Nafion to improve the signal-to-noise(SNR)ratio.To evaluate its applicability to PD research,in vivo electrophysiological and electrochemical detection was performed in normal and hemiparkinsonian models,respectively.Through comparison of the dual-mode signals,we demonstrated the following in a PD monkey:(1)the maximum dopamine concentration in the striatum decreased by 90%;(2)the spike firing frequency increased significantly,especially in the region of the cortex;(3)the spectrogram analysis showed that much power existed in the 0–10 Hz frequency band;and(4)following repeated subthalamic nucleus high-frequency stimulation trials,the level of DA in the striatum increased by 16.5μM,which led to a better elucidation of the mechanism of HFS.The dual-mode MEA probe was demonstrated to be an effective tool for the study of neurological disorders.Song Zhang Yilin Song Mixia Wang Guihua Xiao Fei Gao Ziyue Li Guoxian Tao Ping Zhuang Feng Yue Piu Chan Xinxia Cai 2018Microsystems & Nanoengineering2018,4,1:2
