| 1 | Regulation and function of signal transducer and activator of transcription 3显示文摘Signal transducer and activator of transcription 3(STAT3), a member of the STAT family, is a key regulator of many physiological and pathological processes. Significant progress has been made in understanding the transcriptional control, posttranslational modification, cellular localization and functional regulation of STAT3. STAT3 can translocate into the nucleus and bind to specific promoter sequences, thereby exerting transcriptional regulation. Recent studies have shown that STAT3 can also translocate into mitochondria, participating in aerobic respiration and apoptosis. In addition, STAT3 plays an important role in inflammation and tumorigenesis by regulating cell proliferation, differentiation and metabolism. Conditional knockout mouse models make it possible to study the physiological function of STAT3 in specific tissues and organs. | 2 | Broussonin E suppresses LPS-induced inflammatory response in macrophages via inhibiting MAPK pathway and enhancing JAK2-STAT3 pathway显示文摘Macrophages play an important role in inflammation, and excessive and chronic activation of macrophages leads to systemic inflammatory diseases, such as atherosclerosis and rheumatoid arthritis. In this paper, we explored the anti-inflammatory effect of broussonin E, a novel phenolic compound isolated from the barks of Broussonetia kanzinoki, and its underlying molecular mechanisms. We discovered that Broussonin E could suppress the LPS-induced pro-inflammatory production in RAW264.7 cells, involving TNF-α, IL-1β, IL-6, COX-2 and iNOS. And broussonin E enhanced the expressions of anti-inflammatory mediators such as IL-10, CD206 and arginase-1(Arg-1) in LPS-stimulated RAW264.7 cells. Further, we demonstrated that broussonin E inhibited the LPS-stimulated phosphorylation of ERK and p38 MAPK. Moreover, we found that broussonin E could activate janus kinase(JAK) 2, signal transducer and activator of transcription(STAT) 3. Downregulated pro-inflammatory cytokines and upregulated anti-inflammatory factors by broussonin E were abolished by using the inhibitor of JAK2-STAT3 pathway, WP1066. | 3 | Expression of p-STAT3 and vascular endothelial growth factor in MNNG-induced precancerous lesions and gastric tumors in rats显示文摘AIM: To investigate the dynamic expression of p-signal transducer and activator of transcription 3(STAT3) and vascular endothelial growth factor(VEGF) in the formation of gastric tumors induced by drinking water containing N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Wistar rats.METHODS: One hundred and twenty Wistar rats were randomly divided into two groups(60 in each group): Control group and Model group. The rats in each group were then randomly divided into three groups(20 in each group): C/M15, C/M25 and C/M40(15, 25 and 40 represent the number of feeding weeks from termination). Rats in the control group received normal drinking water and rats in the model group received drinking water containing 100 μg/m L MNNG. Stomach tissues were collected at the end of the 15 th, 25 th and 40 th week, respectively, for microscopic measurement using hematoxylin and eosin staining. The expression of p-STAT3 and VEGF in different pathological types of gastric tissue, including normal, inflammation, atrophy, hyperplasia and gastric stromal tumor, was observed by immunohistochemistry and Western blot, and the corelation between p-STAT3 and VEGF was analyzed. RESULTS:(1) The expression of p-STAT3 in tissue with gastritis, atrophy, dysplasia and gastric stromal tumor were significantly increased in the model group compared with the control group(2.5 ± 1.0, 2.75 ±0.36, 6.2 ± 0.45, 5.67 ± 0.55 vs 0.75 ± 0.36, P = 0.026, 0.035, 0.001, 0.002, respectively); the expression of p-STAT3 in tissue with dysplasia was higher than that in samples with gastritis or atrophy(6.2 ± 0.45 vs 2.5 ± 1.0, P = 0.006; 6.2 ± 0.45 vs 2.75 ± 0.36, P = 0.005, respectively); however, the expression of p-STAT3 in gastritis and atrophy was not significantly different(P > 0.05);(2) the expression of VEGF in tissue with gastritis, atrophy, dysplasia and gastric stromal tumor was significantly increased in the model group compared with normal gastric mucosa; and the expression of VEGF in tissue with dysplasia was higher than that in tissue with inflammation and atrophy(10.8 ± 1.96 vs 7.62 ± 0.25, P = 0.029; 10.8 ± 1.96 vs 6.26 ± 0.76, P = 0.033, respectively); similarly, the expression of VEGF in tissue with gastritis and atrophy was not significantly different(P > 0.05); and(3) the expression of VEGF was positively correlated with p-STAT3. | 4 | Interleukin-22 ameliorates acute severe pancreatitisassociated lung injury in mice显示文摘AIM: To investigate the potential protective effect of exogenous recombinant interleukin-22(r IL-22) on L-arginine-induced acute severe pancreatitis(SAP)-associated lung injury and the possible signaling pathway involved.METHODS: Balb/c mice were injected intraperitoneally with L-arginine to induce SAP. Recombinant mouse IL-22 was then administered subcutaneously to mice. Serum amylase levels and myeloperoxidase(MPO) activity in the lung tissue were measured after the L-arginine administration. Histopathology of the pancreas and lung was evaluated by hematoxylin and eosin(HE) staining. Expression of B cell lymphoma/leukemia-2(Bcl-2), Bcl-x L and IL-22RA1 m RNAs in the lung tissue was detected by real-time PCR. Expression and phosphorylation of STAT3 were analyzed by Western blot. RESULTS: Serum amylase levels and MPO activity in the lung tissue in the SAP group were significantly higher than those in the normal control group(P < 0.05). In addition, the animals in the SAP group showed significant pancreatic and lung injuries. The expression of Bcl-2 and Bcl-x L m RNAs in the SAP group was decreased markedly, while the IL-22RA1 m RNA expression was increased significantly relative to the normal control group(P < 0.05). Pretreatment with PBS did not significantly affect the serum amylase levels, MPO activity or expression of Bcl-2, Bcl-x L or IL-22RA1 m RNA(P > 0.05). Moreover, no significant differences in the degrees of pancreatic and lung injuries were observed between the PBS and SAP groups. However, the serum amylase levels and lung tissue MPO activity in the r IL-22 group were significantly lower than those in the SAP group(P < 0.05), and the injuries in the pancreas and lung were also improved. Compared with the PBS group, r IL-22 stimulated the expression of Bcl-2, Bcl-x L and IL-22RA1 m RNAs in the lung(P < 0.05). | 5 | Recent updates of precision therapy for gastric cancer: Towards optimal tailored management显示文摘Signaling pathways of gastric carcinogenesis and gastric cancer progression are being avidly studied to seek optimal treatment of gastric cancer. Among t h e m, h e p a t o c y t e g r o w t h f a c t o r( H G F) / c- M E T, phosphoinositide 3-kinase(PI3K)/Akt/mammalian target of rapamycin(m TOR) and janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3) pathways have been widely investigated. Their aberrant expression or mutation has been significantly associated with advanced stage or poor prognosis of gastric cancer. Recently, aberrations of immune checkpoints including programmed cell death-1/programmed cell death ligand-1(PD-1/PD-L1) have been suggested as an important step in the formation of a microenvironment favorable for gastric cancer. Accomplishments in basic research have led to the development of novel agents targeting these signaling pathways. However, phase Ⅲ studies of selective anti-HGF/c-MET antibodies and m TOR inhibitor failed to show significant benefits in terms of overall survival and progression-free survival. Few agents directly targeting STAT3 have been developed. However, this target is still critical issue in terms of chemoresistance, and SH2-containing protein tyrosine phosphatase 1 might be a significant link to effectively inhibit STAT3 activity. Inhibition of PD-1/PD-L1 showed durable efficacy in phase Ⅰ studies, and phase Ⅲ evaluation is warranted. Therapeutic strategy to concurrently inhibit multiple tyrosine kinases is a reasonable option, however, lapatinib needs to be further evaluated to identify good responders. Regorafenib has shown promising effectiveness in prolonging progression-free survival in a phase Ⅱ study. | 8 | Signal transducer and activator of transcription 3 promotes the Warburg effect possibly by inducing pyruvate kinase M2 phosphorylation in liver precancerous lesions显示文摘BACKGROUND Study shows that signal transducer and activator of transcription 3(STAT3) can increase the Warburg effect by stimulating hexokinase 2 in breast cancer and upregulate lactate dehydrogenase A and pyruvate dehydrogenase kinase 1 in myeloma. STAT3 and pyruvate kinase M2(PKM2) can also be activated and enhance the Warburg effect in hepatocellular carcinoma. Precancerous lesions are critical to human and rodent hepatocarcinogenesis. However, the underlying molecular mechanism for the development of liver precancerous lesions remains unknown. We hypothesized that STAT3 promotes the Warburg effect possibly by upregulating p-PKM2 in liver precancerous lesions in rats.AIM To investigate the mechanism of the Warburg effect in liver precancerous lesions in rats.METHODS A model of liver precancerous lesions was established by a modified Solt-Farber method. The liver pathological changes were observed by HE staining and immunohistochemistry. The transformation of WB-F344 cells induced with Nmethyl-N'-nitro-N-nitrosoguanidine and hydrogen peroxide was evaluated by the soft agar assay and aneuploidy. The levels of glucose and lactate in the tissue and culture medium were detected with a spectrophotometer. The protein levels of glutathione S-transferase-π, proliferating cell nuclear antigen(PCNA), STAT3,and PKM2 were examined by Western blot and immunofluorescence.RESULTS We found that the Warburg effect was increased in liver precancerous lesions in rats. PKM2 and p-STAT3 were upregulated in activated oval cells in liverprecancerous lesions in rats. The Warburg effect, p-PKM2, and p-STAT3 expression were also increased in transformed WB-F344 cells. STAT3 activation promoted the clonal formation rate, aneuploidy, alpha-fetoprotein expression,PCNA expression, G1/S phase transition, the Warburg effect, PKM2 phosphorylation, and nuclear translocation in transformed WB-F344 cells.Moreover, the Warburg effect was inhibited by stattic, a specific inhibitor of STAT3, and further reduced in transformed WB-F344 cells after the intervention for PKM2.CONCLUSION The Warburg effect is initiated in liver precancerous lesions in rats. Therefore, a new com?posite ultrasonic transducer array with three acoustic beam incidence modes is developed. The space model of the array is also established to obtain the defect reflection point location. And the crack ultrasound image is thus formed through a series of small cubical elements expanded around the point locations by using the projection of binariza?tion values extracted from the received ultrasonic echo signals. Laboratory experiments are performed on a pipeline sample with di erent types of cracks to verify the e ectiveness and performance of the proposed technique. From the image, the presence of small cracks can be clearly observed, in addition to the sizes and orientations of the cracks. | 11 | Bear Bile Powder Inhibits Growth of Hepatocellular Carcinoma via Suppressing STAT3 Signaling Pathway in Mice显示文摘Objective: To evaluate the inhibitory effect of bear bile powder(BBP) on hepatocellular carcinoma(HCC) growth in vivo and investigate the underlying mechanisms. Methods: A HCC xenograft mouse model was developed by producing with huh7 cells. After 5 days following xenograft implantation, ten HCC xenograft mice were given intra-gastric administration with 10 mg/(kg·d) dose of BBP or saline for 3 weeks. Tumor growth in HCC xenograft mice was evaluated by measuring the tumor weight and volume. Cell apoptosis, proliferation or tumor angiogenesis were examined via immunohistochemical(IHC) staining for transferase-mediated deoxyuridine triphosphate-biotin nick end labeling(TUNEL), proliferating cell nuclear antigen(PCNA) or cluster of differentiation 31(CD31), respectively. Phosphorylation of signal transducer and activator of transcription 3(STAT3) were determined by Western blot. The m RNA and protein expressions of Bcl-2, Bax, Cyclin D1 and Cyclin-dependent kinase 4(CDK4) in HCC tumor tissues were respectively determined by reverse transcription polymerase chain reaction(RT-PCR) and Western blot. The protein expression of vascular endothelial growth factor A(VEGF-A) in tumor tissues was examined by IHC staining. Results: BBP treatment led to a significant decrease on tumor volume and tumor weight in HCC mice(P<0.05) and had no effect on the change of body weight. In addition, BBP profoundly promoted cell apoptosis, inhibited cell proliferation and intratumoral microvessel density in HCC tumor tissues(P<0.05). Moreover, BBP treatment remarkably suppressed the STAT3 phosphorylation and modulated the expression of critical target genes including Bcl-2, Bax, Cyclin D1, CDK4 and VEGF-A in HCC mice. | 12 | Impact of high glucose on metastasis of colon cancer cells显示文摘AIM:To investigate the possible mechanism of how glucose promotes invasion and metastasis of colon cancer cells.METHODS:CT-26 rat colorectal cancer cells were cultured in different concentrations of glucose environments(10,20,and 30 mmol/L).Wound healing assay and transwell chamber invasion assay were utilized to test the migration and invasion,respectively.In order to understand the role of signal transducer and activator of transcription 3(STAT3)in the process,STAT3 inhibitors,including Stattic(an STAT3 specific inhibitor)and small interfering RNA targeting STAT3,were used to block STAT3 function to evaluate their impact on CT-26 cell motion.To verify whether STAT3 and matrix metalloproteinase-9(MMP-9)protein expression is associated with glucose-induced cell movement,Western blot was used to compare the differences in the expression of MMP-9 and STAT3 in cells incubated with and without STAT3 inhibitors in high glucose condition.RESULTS:In both wound healing and invasion assays,the migration and invasion of CT-26 cells increased gradually with the increase in glucose concentration.However,the glucose-induced migration and invasion were obviously inhibited by STAT3 inhibitors(P<0.05).Similarly,in Western blot assessment,both MMP-9and STAT3 expression increased under a high glucose environment and the highest expression was achieved when 30 mmol/L glucose was used.However,in cells treated with 30 mmol/L mannitol,either MMP-9 or STAT3expression did not increase(P>0.05).When STAT3inhibitors were added in the 30 m M glucose group,not only STAT3 but also MMP-9 expression decreased significantly(P<0.05).CONCLUSION:Our study provides evidence that glucose can promote both migration and invasion of CT-26 cells,and that the STAT3-induced MMP-9 signal pathway is involved in this process. | Cheng-Yao Lin Chih-Hui Lee Chien-Cheng Huang Song-Tay Lee How-Ran Guo Shih-Bin Su | 2015 | World Journal of Gastroenterology2015,21,7: | 6 |

| 15 | Proton pump inhibitor: The dual role in gastric cancer显示文摘Proton pump inhibitors(PPIs) are one of the most frequently used medications for upper gastrointestinal diseases. However, a number of physicians have raised concern about the serious side effects of long-term use of PPIs, including the development of gastric cancer. Recent epidemiological studies have reported a significant association between long-term PPI intake and the risk of gastric cancer, even after successful Helicobacter pylori eradication. However, the effects of PPIs on the development of pre-malignant conditions such as atrophic gastritis or intestinal metaplasia are not fully known, suggesting the need for comprehensive and confirmative studies are needed in the future. Meanwhile,several experimental studies have demonstrated the effects of PPIs in reducing chemoresistance in gastric cancer cells by modulating the acidic microenvironment, cancer stemness and signal transducer and activator of transcription 3(STAT3) signaling pathway. The inhibitory effects of PPIs on STAT3 activity may overcome drug resistance and enhance the efficacy of conventional or targeted chemotherapeutic agents. | 16 | Principle and Experimental Verification of Novel Dual Driving Face Rotary Ultrasonic Motor显示文摘Existing rotary ultrasonic motors operating in extreme environments cannot meet the requirements of good environmental adaptability and compact structure at same time,and existing ultrasonic motors with Langevin transducers show better environmental adaptability,but size of these motors are usually big due to the radial arrangement of the Langevin transducers.A novel dual driving face rotary ultrasonic motor is proposed,and its working principle is experimentally verified.The working principle of the novel ultrasonic motor is firstly proposed.The 5th in-plane flexural vibration travelling wave,excited by the Langevin transducers around the stator ring,is used to drive the rotors.Then the finite element method is used in the determination of dimensions of the prototype motor,and the confirmation of its working principle.After that,a laser Doppler vibrometer system is used for measuring the resonance frequency and vibration amplitude of the stator.At last,output characteristics of the prototype motor are measured,environmental adaptability is tested and performance for driving a metal ball is also investigated.At room temperature and 200 V(zero to peak) driving voltage,the motor's no-load speed is 80 r/min,the stalling torque is 0.35 N·m and the maximum output power is 0.85 W.The response time of this motor is 0.96 ms at the room temperature,and it decreases or increases little in cold environment.A metal ball driven by the motor can rotate at 210 r/min with the driving voltage 300 V(zero to peak).Results indicate that the prototype motor has a large output torque and good environmental adaptability.A rotary ultrasonic motor owning compact structure and good environmental adaptability is proposed,and lays the foundations of ultrasonic motors' applications in extreme environments. | LU Xiaolong HU Junhui YANG Lin ZHAO Chunsheng | 2013 | Chinese Journal of Mechanical Engineering2013,26,5: | 4 |
| 17 | Implementing wavelet transform with SAW elements显示文摘In the design of the finger-overlap envelope according to the envelope of wavelet func-tion, it is concluded that the pulse-response function of the interdigital transducer (IDT) for surface acoustic wave (SAW) is identical to the wavelet function. SAW type of the wavelet-transform ele-ment is manufactured. A new method of using two wavelet-transform elements to manufacture the reconstruction element of the wavelet transform is proposed. The sources of the element error are analyzed, and the methods for reducing the error are put forward. | 18 | Butein effects in colitis and interleukin-6/signal transducer and activator of transcription 3 expression显示文摘AIM: To evaluate the effects of butein on inflammatory cytokines, matrix metalloproteinase-9(MMP-9), andcolitis in interleukin(IL)-10-/- mice.METHODS: To synchronize colitis, 8- to 10-wk-old IL-10-/- mice were fed pellet-chow containing piroxicam for 2 wk. Subsequently, phosphate-buffered saline or butein(1 mg/kg per day, ip) was injected for 4 wk. Histologic scores, inflammatory cytokines, MMP-9 and phosphorylated signal transducer and activator of transcription 3(p STAT3) expressions were analyzed in IL-10-/- mice and in Colo 205 cells.RESULTS: Butein reduced the colonic inflammatory score by > 50%. Expression levels of IL-6, IL-1β, interferon(IFN)-γ and MMP-9 were decreased in the colons of mice exposed to butein, whereas other inflammatory cytokines(IL-17 A, IL-21 and IL-22) were unchanged. Immunohistochemical staining for p STAT3 and MMP-9 was significantly decreased in the buteintreated groups compared with the controls. | 19 | STAT3及XRCC4基因多态性与肝细胞癌易感性的关系研究显示文摘目的:探讨转录激活因子3(STAT3)及X线修复交叉互补基因4(XRCC4)基因多态性与中国人群肝细胞癌(HCC)易感性的关系。方法采用病例对照研究,以确诊的200例乙型肝炎病毒(HBV)感染相关 HCC 为肝癌组,207例性别及年龄匹配的 HBsAg阳性患者为非肝癌组。采用PCR-限制性长度多态性技术检测STAT3 rs2292152和XRCC4 rs1805377多态性位点的基因型,Logistic回归分析比较不同基因型与 HCC易感风险的关系。结果(1)STAT3 rs2293152位点3种基因型(CC、CG和GG型)在肝癌组分布频率分别为17%(34/200)、49.5%(99/200)和33.5%(67/200),在非肝癌组中分别为18.4%(38/207)、56.5%(117/207)和25.1%(52/207),各基因型在两组之间的差异无统计学意义(P>0.05);以CC基因型作参照,携带rs2293152 GG型的个体 HCC患病风险差异无统计学意义(OR=1.440,95% CI=0.800~2.592,P=0.224)。(2)XRCC4 rs1805377位点3种基因型(AA、GA和GG 型)在肝癌组分布频率分别为61%(122/200)、30%(60/200)和9%(18/200),在非肝癌组中分别为59.9%(124/207)、31.9%(66/207)和8.2%(17/207),各基因型在两组之间的差异无统计学意义(P>0.05);以 AA 基因型作参照,携带rs1805377 GG型的个体 HCC 患病风险差异无统计学意义(OR=1.076,95% CI=0.530~2.185,P=0.839)。结论 STAT3 rs2292152和XRCC4 rs1805377多态性位点可能与中国人群HBV 相关HCC 易感性无密切关系。 | 刘懿 余德才 潘明洁 毕永春 周乙华 | 2014 | 国际检验医学杂志2014,35,7: | 3 |
| 20 | Effects of boundary conditions on vibrating mode of acoustic logging dipole transducer显示文摘Acoustic transducer is an important part of acoustic well logging tool. In this paper,ANSYS software package is used to design acoustic dipole transducer and simulate vibrating mode of the dipole transducer in different mechanical boundary conditions. The results show that boundary conditions influence the number of vibrating mode in the same frequency band and the frequency value of the same vibrating mode. Several acoustic dipole transducers are designed according to the results of numerical simulation and laboratory measurements. The basic frequency of vibrating mode of experi-ment has good agreement with that of simulation.
