| 5 | The deubiquitylase UCHL3 maintains cancer stem-like properties by stabilizing the aryl hydrocarbon receptor显示文摘Cancer stem cells(CSCs)exhibit highly aggressive and metastatic features and resistance to chemotherapy and radiotherapy.Aryl hydrocarbon receptor(AhR)expression varies among non-small cell lung cancers(NSCLCs),and the mechanisms that support abnormal AhR expression in CSCs remain elusive.Here,we identified ubiquitin carboxyl terminal hydrolase L3(UCHL3),a DUB enzyme in the UCH protease family,as a bona fide deubiquitylase of the AhR in NSCLC.UCHL3 was shown to interact with,deubiquitylate,and stabilize AhR in a manner dependent on its deubiquitylation activity.Moreover,we showed that UCHL3 promotes the stem-like characteristics and potent tumorigenic capacity of NSCLC cells.UCHL3 increased AhR stability and the binding of AhR to the promoter regions of the“stemness”genes ATP-binding cassette subfamily G member 2(ABCG2),KLF4,and c-Myc.Depletion of UCHL3 markedly downregulated the“stemness”genes ABCG2,KLF4,and c-Myc,leading to the loss of selfrenewal and tumorigenesis in NSCLCs.Furthermore,the UCHL3 inhibitor TCID induced AhR degradation and exhibited significantly attenuated efficacy in NSCLC cells with stem cell-like properties.Additionally,UCHL3 was shown to indicate poor prognosis in patients with lung adenocarcinoma.In general,our results reveal that the UCHL3 deubiquitylase is pivotal for AhR protein stability and a potential target for NSCLC-targeted therapy. | Lianlian Ouyang Bin Yan Yating Liu Chao Mao Min Wang Na Liu Zuli Wang Shouping Liu Ying Shi Ling Chen Xiang Wang Yan Cheng Ya Cao Desheng Xiao Lingqiang Zhang Shuang Liu Yongguang Tao | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 1 |
| 6 | Whole-exome mutational landscape of metastasis in patient-derived hepatocellular carcinoma cells显示文摘In order to explore the genomic basis for liver cancer metastasis,whole-exome sequencing(WES)was performed on patient-derived hepatocellular carcinoma(HCC)cell lines with differential metastatic potentials and analyzed their clonal evolution relationships.An evolutionary tree based on genomic single nucleotide polymorphism(SNP)was constructed in MegaX software.The WES data showed that the average percentage of heterogeneous mutations in each HCC cell lines was 16.55%(range,15.38%e18.17%).C:G>T:A and T:A>C:G somatic transitions were the two most frequent substitutions.In these metastatic HCC cell lines,non-silent gene mutations were found in 21.88%of known driver genes and 10 classical signaling pathways.The protein interaction network was constructed by STRING,and hub genes were found in the shared trunk mutation genes and the heterogeneous branch mutations respectively.In cBioPortal database,some of the selected hub genes were found to be associated with poor overall survival(OS)of HCC patients.Among the mutated HCC driver genes,a novel KEAP1 mutation with a homozygous frameshift truncation at the c-terminal Nrf2 binding region was detected and verified in MHCC97-H and HCC97LM3 cells.In conclusion,WES data demonstrate that HCC cell lines from tumor biopsy specimens of the same patient have obtained different metastatic potentials through repeated selection in rodents in vivo,and they do indeed have a genetic relationship at the genomic level. | Qian Zhou Zuli Li Linlan Song Di Mu Jin Wang Li Tian Yong Liao | 2020 | Genes & Diseases2020,7,3: | 1 |

| 9 | 广州市天河区结直肠癌筛查:基于粪便免疫化学检测和危险因素初步筛选出需要行结肠镜检查者显示文摘目的:探讨联合粪便免疫化学检测(FIT)和高危因素问卷调查(HRFQ)初筛出需行结肠镜检查者的结直肠癌(CRC)筛查方案应用于中国的可行性。方法:2015-2016年间,我们对中国广州市天河区45岁以上居民进行了一项CRC筛查项目。所有筛查对象均接受FIT检测和HRFQ调查,作为初筛。对于FIT阳性和/或HRFQ调查结果考虑存在高危风险的受检者,我们推荐其行进一步结肠镜检查。结果:共计10074名居民纳入CRC筛查项目,其中17.5%的受检者FIT阳性,19.4%经HRFQ调查被认为具有高危风险。经推荐,有773名受检者接受了进一步的结肠镜检查。该筛查方案对非腺瘤性息肉、低危腺瘤、高危腺瘤和CRC的总体阳性预测值分别为4.9%、11.4%、15.9%和1.6%。其中,FIT阳性结果对上述四种病变的阳性预测值分别为5.2%、15.9%、22.5%和2.5%,HRFQ阳性结果分别为4.1%、10.2%、14.3%和1.4%,FIT和HRFQ均为阳性结果的阳性预测值分别为4.5%、16.4%、23.7%和2.8%。结论:我们的两步法CRC筛查策略(FIT和HRFQ阳性者行进一步结肠镜检查)可有效发现早期CRC。鉴于对CRC和腺瘤较的高检出率和阳性预测值,该筛查方案可继续用于我们这项大规模CRC筛查项目的后续研究。 | Yi Liao Senmao Li Chunyu Chen Xuan He Feng Lin Jianping Wang Zuli Yang Ping Lan | 2018 | Gastroenterology Report2018,6,2: | 0 |

| 11 | GPR162 activates STING dependent DNA damage pathway as a novel tumor suppressor and radiation sensitizer显示文摘In the treatment of most malignancies,radiotherapy plays a significant role.However,the resistance of cancer cells to ionizing radiation(IR)is the main reason for the failure of radiotherapy,which causes tumor recurrence and metastasis.In this study,we confirmed that GPR162,an orphan receptor in the G-protein-coupled receptor family,acted as a novel radiotherapy sensitizer by interacting with the stimulator of interferon genes(STING),which targeted DNA damage responses,activated IRF3,accelerated the activation of type I interferon system,promoted the expression of chemokines including CXCL10 and CXCL4,and inhibited the occurrence and development of tumors.Interestingly,the activation of STING by overexpression of GPR162 was independent of the classical pathway of cGAS.STING inhibitors could resist the antitumor effect of overexpression of GPR162 in IR-induced mouse models.In addition,most solid tumors showed low expression of GPR162.And the higher expression of GPR162 indicated a better prognosis in patients with lung adenocarcinoma,liver cancer,breast cancer,etc.In summary,these results suggested that GPR162 may serve as a potential sensitizer of radiotherapy by promoting radiotherapy-induced STING-IFN production and increasing the expression of chemokines including CXCL10 and CXCL4 in DNA damage response,providing an alternative strategy for improving cancer radiotherapy. | Yao Long Jiaxing Guo Jielin Chen Jingyue Sun Haiyan Wang Xin Peng Zuli Wang WeiWei Lai Na Liu Long Shu Ling Chen Ying Shi Desheng Xiao Shuang Liu Yongguang Tao | 2023 | Signal Transduction and Targeted Therapy2023,8,3: | 0 |

[Method] Autumn and spring control tests were carried out in Potianyang and Miaolinyang,respectively. [Result] The rat density in autumn control area decreased from 28. 57%( outbreak) to 2. 86%,3. 81% and 6. 80% at 11,41 and 70 d after triptolide treatment,respectively. The rat density in spring control area was lower at 19 and 47 d post treatment,and the correction deratization effect was 99. 48%-99. 88%. The testicular descent rate and pregnancy rate decreased significantly after treatment of triptolide. | 13 | Controlling factors on the charging process of oil and gas in the eastern main sub-sag of the Baiyun Sag,Zhujiang River(Pearl River)Mouth Basin显示文摘The eastern main sub-sag(E-MSS)of the Baiyun Sag was the main zone for gas exploration in the deep-water area of the Zhujiang River(Pearl River)Mouth Basin at its early exploration stage,but the main goal of searching gas in this area was broken through by the successful exploration of the W3-2 and H34B volatile oil reservoirs,which provides a new insight for exploration of the Paleogene oil reservoirs in the E-MSS.Nevertheless,it is not clear on the distribution of“gas accumulated in the upper layer,oil accumulated in the lower layer”(Gas_(upper)-Oil_(lower))under the high heat flow,different source-rock beds,multi-stages of oil and gas charge,and multi-fluid phases,and not yet a definite understanding of the genetic relationship and formation mechanism among volatile oil,light oil and condensate gas reservoirs,and the migration and sequential charge model of oil and gas.These puzzles directly lead to the lack of a clear direction for oil exploration and drilling zone in this area.In this work,the PVT fluid phase,the origin of crude oil and condensate,the secondary alteration of oil and gas reservoirs,the evolution sequence of oil and gas formation,the phase state of oil and gas migration,and the configuration of fault activity were analyzed,which established the migration and accumulation model of Gas_(upper)-Oil_(lower)cocontrolled by source and heat,and fractionation controlled by facies in the E-MSS.Meanwhile,the fractionation evolution model among common black reservoirs,volatile reservoirs,condensate reservoirs and gas reservoirs is discussed,which proposed that the distribution pattern of Gas_(upper)-Oil_(lower)in the E-MSS is controlled by the generation attribute of oil and gas from source rocks,the difference of thermal evolution,and the fractionation controlled by phases after mixing the oil and gas.Overall,we suggest that residual oil reservoirs should be found in the lower strata of the discovered gas reservoirs in the oil-source fault and diapir-developed areas,while volatile oil reservoirs should be found in the deeper strata near the sag with no oil-source fault area. | Cong Chen Xiangtao Zhang Guangrong Peng Zulie Long Baojun Liu Xudong Wang Puqiang Zhai Bo Zhang | 2023 | Acta Oceanologica Sinica2023,42,3: | 0 |
