1Growth performance and physiological parameters of the second generation selected and control groups of Pinctada martensii显示文摘In May of 2007,the second generation selected (SS) and control (SC) groups of pearl oyster Pinctada martensii were established by selecting 10% breeders with the largest and mean shell length,respectively,from the first generation selected group.Growth performance of the SS and SC groups were compared on the basis of measurement data at Days 8,18,60,95,195 and 365.On Day 365,100 individuals (60.0–75.0 mm at shell length) were sampled from each group and then subjected to the experiment where physiological parameters such as filtrate rate,oxygen consumption and ammonia excretion were measured at 15,20,25 and 30°C.The results show that the SS group had significantly larger mean shell length and shell height than the SC group at Days 8,18,60,95,195 and 365 (P <0.05).The genetic gains at different ages varied from 6.0% to 17.0% for shell length and 5.7% to 14.6% for shell height,respectively.At 15,20,25 and 30 ° C,the SS groups had significantly larger filtrate rate than the SC group (P <0.05).At 15 and 25 °C,the differences in oxygen consumption rate between the SS and SC groups were not significant (P >0.05).At 20 and 30 °C,however,the oxygen consumption rate of the SS group was significantly larger than the SC group (P <0.05).At 15,20,25 and 30 °C,there were no significant differences in ammonia excretion rate between the SS and SC groups (P >0.05).The present results indicate that there existed considerable genetic variability in the base population and a further selection could be likely fruitful.Mass selection for faster growth might produce animals that had higher intake of metabolizable energy by virtue of faster filtrating behavior.DENG Yuewen YU Ziniu DU Xiaodong WANG Qingheng FU Shao 2011Acta Oceanologica Sinica2011,30,2:10
2Genomic landscape and genetic manipulation of the black soldier fly Hermetia illucens,a natural waste recycler显示文摘The black soldier fly(BSF),Hermetia illucens(Diptera:Stratiomyidae),is renowned for its bioconversion of organic waste into a sustainable source of animal feed.We report a high-quality genome of 1.1 Gb and a consensus set of 16,770 gene models for this beneficial species.Compared to those of other dipteran species,the BSF genome has undergone a substantial expansion in functional modules related to septic adaptation,including immune system factors,olfactory receptors,and cytochrome P450s.We further profiled midgut transcriptomes and associated microbiomes of BSF larvae fed with representative types of organic waste.We find that the pathways related to digestive system and fighting infection are commonly enriched and that Firmicutes bacteria dominate the microbial community in BSF across all diets.To extend its potential practical applications,we further developed an efficient CRISPR/Cas9-based gene editing approach and implemented this to yield flightless and enhanced feeding capacity phenotypes,both of which could expand BSF production capabilities.Our study provides valuable genomic and technical resources for optimizing BSF lines for industrialization.Shuai Zhan Gangqi Fang Minmin Cai Zongqing Kou Jun Xu Yanghui Cao Liang Bai Yixiang Zhang Yongmao Jiang Xingyu Luo Jian Xu Xia Xu Longyu Zheng Ziniu Yu Hong Yang Zhijian Zhang Sibao Wang Jeffery K.Tomberlin Jibin Zhang Yongping Huang 2020Cell Research2020,30,1:9
3The role of β18-β19 loop structure in insecticidal activity of Cry1Ac toxin from Bacillus thuringiensis显示文摘The β18-β19 loop in domain III of Cry1Ac toxin is unique among Bacillus thuringiensis Cry proteins. In this study, the role of the loop structure in insecticidal activity of Cry1Ac toxin was investigated. Alanine scanning mutations within the loop were initially generated and most mutants were over-expressed and reduced toxicity at different degrees, except mutant N546A that showed almost 2 times enhanced toxicity against Helicoverpa armigera larvae. Further mutagenic analysis of N546 revealed that a charged amino acid in this position would cause very unfavorable influence on insecticidal activity. In addition, the deletion of N546 led to protein instability because of destruction of the loop integrity. Besides, mutant W544F was much more toxic than W544Y, indicating that hydrophobic nature of the position was important for maintaining the stability and activity of Cry1Ac protein. 4Analysis of the efficacy of transcatheter arterial infusion chemotherapy in the treatment of pancreatic carcinoma显示文摘Objective:To evaluate the clinical efficacy of infusion of gemcitabine(GEM) and fluorouracil(5-FU)through the celiac artery and superior mesenteric artery in the treatment of pancreatic carcinoma(PC).Methods:We analyzed 20 patients diagnosed clinically or pathologically with PC,without metastases,who had an estimated survival duration of>3 months in our department from May 2009 to December 2014.Nine patients were treated directly without surgical resection of the tumor,while the other 11 patients were treated after surgery.In all patients,the femoral artery was punctured using the Seldinger technique,and a catheter was placed in the opening of the celiac artery or the superior mesenteric artery.We administered 500 mg/m2 GEM and 500 mg/m2 5-FU.Observational data included data on clinical efficacy and survival rates during the follow-up period of 3-72 months.Results:Twenty patients were treated 85 times with transcatheter arterial infusion chemotherapy(TAI).The survival rates were 80%,40%,35%,20%,10%,and 5% at 3,6,12,24,and 72 months,respectively.Conclusion: TAI chemotherapy with GEM and 5-FU may be a therapeutic option for the treatment of PC.Chunhui Nie Yuelin Zhang Guanhui Zhou Tanyang Zhou Tongyin Zhu Baoquan Wang Shengqun Chen Hongliang Wang Ziniu Yu Li Jing Junhui Sun 2021Journal of Interventional Medicine2021,4,1:4

17Tetrapioid induction of Crassostrea hongkongensis and C.sikamea by inhibiting the polar body 1 release in diploid fertilized eggs显示文摘The production of an all-triploid population by mating tetrapioid males with diploid females is the best and most fundamental method for the large-scale production of triploid oysters.Obtaining a stable tetrapioid population is essential for guaranteed production in industrialized triploid cultivation.C.hongkongensis and C.sikamea are important oyster breeding species in southern China,and have great economic value.However,there are not any published data on inducing tetrapioid C.hongkongensis or C.sikamea.Therefore,we investigated tetrapioid induction in these two oyster species by inhibiting the PB1 release in diploid fertilized eggs using Cytochalasin B(CB)under 31℃,15%o salinity.The results confirmed that the optimal tetrapioid induction conditions for C.hongkongensis were a CB concentration of 0.50 mg/L with induction starting at 9.0 min after fertilization,and stopping at 21.0 min after fertilization;the induction efficiency index reached 0.123 under these conditions.The optimal tetrapioid induction conditions for C.sikamea were a CB concentration of 0.50 mg/L,with induction starting at 7.5 min after fertilization and stopping at 18 min after fertilization;the induction efficiency index could be as high as 0.281 under these conditions.However,we confirmed that the tetrapioid rate decreased with larval growth,and no tetrapioids were detected in the juvenile period of either C.hongkongensis or C.sikamea.This may be attributed to the very low survival of the tetrapioid larvae induced by this method,especially as most tetrapioid larvae died during the first three days.In summary,it is simple to directly induce tetrapioid C.hongkongensis and C.sikamea larvae by inhibiting the PB1 release of diploid zygotes,but the low survival rate makes it challenging to obtain viable juvenile tetrapioids.Yanping Qin Zohaib Noor Xingyou Li Yuehuan Zhang Ziniu Yu Haitao Ma Jun Li Yinyin Zhou Riguan Mo 2021Marine Life Science & Technology2021,3,4:1
