Inhibiting collagen Ⅰ production and tumor cell colonization in the lung via miR-29a-3p loading of exosome-/liposome-based nanovesicles显示文摘The lung is one of the most common sites for cancer metastasis.Collagens in the lung provide a permissive microenvironment that supports the colonization and outgrowth of disseminated tumor cells.Therefore,down-regulating the production of collagens may contribute to the inhibition of lung metastasis. It has been suggested that mi R-29 exhibits effective anti-fibrotic activity by negatively regulating the expression of collagens.Indeed, our clinical lung tumor data shows that mi R-29 a-3 p expression negatively correlates with collagen I expression in lung tumors and positively correlates with patients’outcomes. However, suitable carriers need to be selected to deliver this therapeutic mi RNA to the lungs. In this study,we found that the chemotherapy drug cisplatin facilitated mi R-29 a-3 p accumulation in the exosomes of lung tumor cells, and this type of exosomes exhibited a specific lung-targeting effect and promising collagen down-regulation.To scale up the preparation and simplify the delivery system,we designed a lung-targeting liposomal nanovesicle(by adjusting the molar ratio of DOTAP/cholesterol-mi RNAs to 4:1)to carry mi R-29 a-3 p and mimic the exosomes. This liposomal nanovesicle delivery system significantly down-regulated collagen I secretion by lung fibroblasts in vivo, thus alleviating the establishment of a pro-metastatic environment for circulating lung tumor cells. | Yan Yan Cancan Du Xixi Duan Xiaohan Yao Jiajia Wan Ziming Jiang Zhongyu Qin Wenqing Li Longze Pan Zhuoyu Gu Fazhan Wang Ming Wang Zhihai Qin | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 7
Short fixation with a 3‐rod technique for posterior hemivertebra resection in children younger than 5 years old显示文摘Importance Congenital hemivertebra is commonly treated with posterior hemivertebra resection with bilateral transpedicular fixation. However, implant‐related complications are common in children younger than 5 years old who undergo this surgical procedure. Objective To present the preliminary clinical and radiological outcomes of children younger than 5 years old treated by posterior hemivertebra resection and 3‐rod fixation technique. Methods From January 2016 to December 2017,14 consecutive patients of congenital scoliosis with 16 hemivertebrae were retrospectively reviewed, including 5 girls and 9 boys, aged between 25 and 55 months old(average,37.6 months). All patients underwent posterior hemivertebra resection with short fixation with bilateral pedicle screws and a convex lamina hook.Surgical complications and corrective outcomes were assessed based on the clinical charts and spinal radiographs with a minimum 24‐month follow‐up.Results The mean Cobb angle of the main curve was 38.4°before surgery,8.5°after surgery, and 8.7°at final follow‐up.In the compensatory cranial curve, the preoperative Cobb angle of 16.8°was corrected to 8.1°postoperatively and was 10.3°at final follow‐up.In the compensatory caudal curve, the preoperative Cobb angle of 15.9°improved to 5.3°postoperatively and was 7.8°at final follow‐up.The segmental kyphosis was corrected from 13.5°to 0.5°and was 1.1°at final follow‐up.There were no crankshaft phenomena,no proximal kyphosis, and no complications related to the instrumentation.Interpretation Posterior hemivertebra resection with instrumentation with bilateral pedicle screws and a convex lamina hook can achieve rigid fixation and deformity correction. | Dong Guo Ziming Yao Xinyu Qi Chengxin Li Xuejun Zhang | 2020 | Pediatric Investigation2020,4,2: | 4

Pyroptotic macrophages stimulate the SARS-CoV-2-associated cytokine storm显示文摘Coronavirus disease 2019(COVID-19)is an unprecedented pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).As of 22 February 2021,the worldwide pandemic has resulted in more than 110 million cases and 2.4 million deaths.1 Clinical investigation of COVID-19 patients has shown that a systemic cytokine storm can occur, especially in severe cases.2 Treatment of the SARS-CoV-2-associated cytokine storm with tocilizumab3 or anakinra4 has been shown to immediately improve the clinical outcome in most severe and critical COVID-19 patients. These data highlight the systemic cytokine storm as an important exacerbating event in severe COVID-19;however,our understanding of the molecular mechanisms involved in the initiation of the SARS-CoV-2-associated cytokine storm is limited. In the present study,we uncovered a reasonable explanation for cytokine storm initiation through the analysis of 13 autopsy samples from severe COVID-19 patients. | Jinghe Zhang Haibo Wu XiaoHong Yao Dingyu Zhang Yonggang Zhou Binqing Fu Wei Wang Heng Li Zhe Wang Ziming Hu Yong Ren Rui Sun Zhigang Tian Xiuwu Bian Haiming Wei | 2021 | Cellular & Molecular Immunology2021,18,5: | 1
Micropatterns and peptide gradient on the inner surface of a guidance conduit synergistically promotes nerve regeneration in vivo显示文摘Both of the surface topographical features and distribution of biochemical cues can influence the cell-substrate interactions and thereby tissue regeneration in vivo. However, they have not been combined simultaneously onto a biodegradable scaffold to demonstrate the synergistic role so far.In this study,a proof-of-concept study is performed to prepare micropatterns and peptide gradient on the inner wall of a poly(D,L-lactide-co-caprolactone)(PLCL)guidance conduit and its advantages in regeneration of peripheral nerve in vivo. After linear ridges/grooves of 20/40μm in width are created on the PLCL film, its surface is aminolyzed in a kinetically controlled manner to obtain the continuous gradient of amino groups, which are then transferred to CQAASIKVAV peptide density gradient via covalent coupling of glutaraldehyde. The Schwann cells are better aligned along with the stripes, and show a faster migration rate toward the region of higher peptide density.Implantation of the nerve guidance conduit made of the PLCL film having both the micropatterns and peptide gradient can significantly accelerate the regeneration of sciatic nerve in terms of rate, function recovery and microstructures, and reduction of fibrosis in muscle tissues. Moreover, this nerve conduit can also benefit the M2 polarization of macrophages and promote vascularization in vivo. | Deteng Zhang Ziming Li Haifei Shi Yuejun Yao Wang Du Pan Lu Kejiong Liang Liangjie Hong Changyou Gao | 2022 | Bioactive Materials2022,7,3: | 1
Constitutive Modeling and Dynamic Recrystallization Mechanisms of an Ultralow-carbon Microalloyed Steel During Hot Compression Tests显示文摘The hot deformation behavior of an ultralow-carbon microalloyed steel was investigated using an MMS-200 thermal simulation test machine in a temperature range of 1073-1373 K and strain rate range of 0.01-10 s-1.The results show that the flow stress decreases with increasing deformation temperature or decreasing strain rate. The strain-compensated constitutive model based on the Arrhenius equation for this steel was established using the true stress-strain data obtained from a hot compression test.Furthermore,a new constitutive model based on the Z-parameter was proposed for this steel. The predictive ability of two constitutive models was compared with statistical measures. The results indicate the new constitutive model based on the Z-parameter can more accurately predict the flow stress of an ultralow-carbon microalloyed steel during hot deformation. The dynamic recrystallization(DRX)nucleation mechanism at different deformation temperatures was observed and analyzed by transmission electron microscopy(TEM),and strain-induced grain boundary migration was observed at 1373 K/0.01 s^-1. | 李宁 HUANG Yao HAN Renheng BAO Ziming ZHU Yanqing ZHANG Hexin 赵成志 | 2020 | Journal of Wuhan University of Technology(Materials Science)2020,35,5: | 1
