| 1 | Interaction of Restin with transcription factors显示文摘Restin, a member of melanoma-associated antigen superfamily gene, was first cloned from differentiated leukemia cell induced by all trans-retinoic acid, and was able to inhibit cell proliferation, but the molecular mechanism was not clear. Since Restin was localized in cell nucleus, and its homolog member, Necdin (neuronal growth suppressor factor), could interact with transcription factors p53 and E2F1, we proposed that Restin might also function as Necdin through interacting with some transcription factors. In this study, transcription factors p53, AP1, ATFs and E2Fs were cloned and used in the mammalian two-hybrid system to identify their in- teraction with Restin. The results showed that only ATF3 had a strong interaction with Restin. It is interesting to know that ATF3 was an important transcription factor for G1 cell cycle initiation in physiological stress response. | 2 | RNA-binding protein QKI regulates contact inhibition via Yes-associate protein in ccRCC显示文摘Contact inhibition adjusts organ size to the proper size and ensures the cultured cells growing to a monolayer.By regulating the downstream coordinator YAP,the evolutionarily conserved Hippo transduction pathway attunes cell growth and death in response to cell contact inhibition,polarity,self-renewal,and differentiation.Dysregulation of this pathway is involved in various diseases such as cancer.RNA-binding protein QKI regulates cell proliferation,metabolism,division,and immunity in various cancer models,but its role in cancer cell contact inhibition remains unclear.In this study,we aimed to clarify the relationship between QKI and YAP,and the role of their interaction in cell contact inhibition.We found a lower QKI expression level in sparse condition,whereas a higher expression level in confluent condition by western blot analysis and immunofluorescence assay.QKI knockdown elevated cell proliferation and invasion both in vitro and in vivo.Strikingly,the results of CCK-8 assay,colony formation assay,and transwell assay showed that the phenomenon was in accord with the expression level of pYAP and reverse with YAP.Higher levels of Wnt3a and β-catenin were also found in xenografts of QKI-knockdown clear cell renal cell carcinoma (ccRCC) CAKI-1 cells by western blot analysis and immumohistochemical staining.Finally,a positive correlation between QKI and pYAP was found in clinical specimens by immunohistochemistry.Thus,as a negative regulator of YAP,QKI attuned the cell contact inhibition,leading to inhibition of cancer cell proliferation and invasion through Wnt and GPCR pathway. | Zheng Zhu Di Wei Xi'an Li Fuli Wang Fei Yan Zibao Xing Zhao Yan Huanyu Lu Dongsheng Zhai Ziehen Ye Geng Zhang Ping Meng Yu Zheng Jiarui Yuan Zifan Lu Jianlin Yuan | 2019 | Acta Biochimica et Biophysica Sinica2019,51,1: | 2 |

| 8 | A living cell-based fluorescent reporter for high-throughput screening of anti-tumor drugs显示文摘Suppression of cellular O-linkedβ-N-acetylglucosaminylation(O-Glc NAcylation)can repress proliferation and migration of various cancer cells,which opens a new avenue for cancer therapy.Based on the regulation of insulin gene transcription,we designed a cell-based fluorescent reporter capable of sensing cellular O-Glc NAcylation in HEK293 T cells.The fluorescent reporter mainly consists of a reporter(green fluorescent protein(GFP)),an internal reference(red fluorescent protein),and an operator(neuronal differentiation 1),which serves as a'sweet switch'to control GFP expression in response to cellular OGlc NAcylation changes.The fluorescent reporter can efficiently sense reduced levels of cellular OGlc NAcylation in several cell lines.Using the fluorescent reporter,we screened 120 natural products and obtained one compound,sesamin,which could markedly inhibit protein O-Glc NAcylation in He La and human colorectal carcinoma-116 cells and repress their migration in vitro.Altogether,the present study demonstrated the development of a novel strategy for anti-tumor drug screening,as well as for conducting gene transcription studies. | Ningning Tang Ling Li Fei Xie Ying Lu Zifan Zuo Hao Shan Quan Zhang Lianwen Zhang | 2021 | Journal of Pharmaceutical Analysis2021,11,6: | 0 |
