| 1 | Soil microbial biomass carbon and nitrogen in forest ecosystems of Northeast China:a comparison between natural secondary forest and larch plantation显示文摘Aims Natural secondary forest(NSF)and larch plantation are two of the predominant forest types in Northeast China.However,how the two types of forests compare in sustaining soil quality is not well understood.This study was conducted to determine how natural secondary forest and larch plantation would differ in soil microbial biomass and soil organic matter quality.Methods Microbial biomass carbon(MBC),microbial biomass nitrogen(MBN),soil organic carbon(SOC)and total nitrogen(TN)in the 0-to 15-cm and 15-to 30-cm soil layers were investigated by making chemical and biological measurements in the montane region of eastern Liaoning Province,Northeast China,during the growing season of 2008 in stands of NSF and Larix olgensis plantation(LOP).Important Findings We found that soil MBC and MBN were significantly lower in the LOP than in the NSF.Both MBC and MBN declined significantly with increasing soil depth in the two types of stands.The ratios of MBC to SOC(MBC/SOC)and MBN to TN(MBN/TN)were also significantly lower in the LOP than in the NSF.Moreover,the values of MBC,MBC/SOC,and MBN/TN significantly varied with time and followed a similar pattern during the growing season,all with an apparent peak in summer.Our results indicate that NSF is better in sustaining soil microbial biomass and nutrients than larch plantation in the temperate Northeast China.This calls for cautions in large-scale conversions of the native forests to coniferous plantations as a forest management practice on concerns of sustaining soil productivity. | Kai Yang Jiaojun Zhu Min Zhang Qiaoling Yan Osbert Jianxin Sun | 2010 | Journal of Plant Ecology2010,3,3: | 44 |
| 2 | Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. | 3 | Rice CYP703A3, a cytochrome P450 hydroxylase, is essential for development of anther cuticle and pollen exine显示文摘Anther cuticle and pollen exine act as protective envelopes for the male gametophyte or pollen grain, but the mechanism underlying the synthesis of these lipidic polymers remains unclear. Previously, a tapetum‐expressed CYP703A3, a putative cytochrome P450 fatty acid hydroxylase, was shown to be essential for male fertility in rice(Oryza sativa L.). However,the biochemical and biological roles of CYP703A3 has not been characterized. Here, we observed that cyp703a3‐2 caused by one base insertion in CYP703A3 displays defective pollen exine and anther epicuticular layer, which differs from Arabidopsis cyp703a2 in which only defective pollen exine occurs. Consistently, chemical composition assay showed that levels of cutin monomers and wax components were dramatically reduced in cyp703a3‐2 anthers. Unlike the wide range of substrates of Arabidopsis CYP703A2, CYP703A3 functions as an in‐chain hydroxylase only for a specific substrate, lauric acid, preferably generating 7‐hydroxylated lauric acid. Moreover, chromatin immunoprecipitation and expression analyses revealed that the expression of CYP703A3 is directly regulated by Tapetum Degeneration Retardation, a known regulator of tapetum PCD and pollen exine formation. It resulted from the northeastward subduction of the Early Palaeozoic Qilan oceanic crust beneath the Alxa block. High-pressure metamorphism, which occurred during the subduction, progressed through three stages: the initial stage of medium T-high P,the main stage of temperature decrease and pressure increase, and the lag stage of pressure decrease and temperature increase. | 5 | Targets and molecular mechanisms of triptolide in cancer therapy显示文摘Triptolide(TPL/TL) is a natural drug with novel anticancer effects. Preclinical studies indicated that TPL inhibits cell proliferation, induces cell apoptosis, inhibits tumor metastasis and enhances the effect of other therapeutic methods in various cancer cell lines. Multiple molecules and signaling pathways, such as caspases, heat-shock proteins, NF-κB, and deoxyribonucleic acid(DNA) repair-associated factors, are associated with the anti-cancer effect. TPL also improves chemoradiosensitivity in cancer therapy. Phase I trials indicate the potential clinical value of TPL use. | 6 | Effects of Saccharomyces cerevisiae fermentation products on performance and rumen fermentation and microbiota in dairy cows fed a diet containing low quality forage显示文摘Background: A possible option to meet the increased demand of forage for dairy industry is to use the agricultural byproducts, such as corn stover. However, nutritional value of crop residues is low and we have been seeking technologies to improve the value. A feeding trial was performed to evaluate the effects of four levels of Saccharomyces cerevisiae fermentation product(SCFP; Original XP; Diamond V) on lactation performance and rumen fermentation in mid-lactation Holstein dairy cows fed a diet containing low-quality forage. Eighty dairy cows were randomly assigned into one of four treatments: basal diet supplemented with 0, 60, 120, or 180 g/d of SCFP per head mixed with 180, 120, 60, or 0 g of corn meal, respectively. The experiment lasted for 10 wks, with the first 2 weeks for adaptation.Results: Dry matter intake was found to be similar(P > 0.05) among the treatments. There was an increasing trend in milk production(linear, P ≤ 0.10) with the increasing level of SCFP supplementation, with no effects on contents of milk components(P > 0.05). Supplementation of SCFP linearly increased(P < 0.05) the N conversion, without affecting rumen pH and ammonia-N(P > 0.05). Increasing level of SCFP linearly increased(P < 0.05) concentrations of ruminal total volatile fatty acids, acetate, propionate, and butyrate, with no difference in molar proportion of individual acids(P > 0.05). The population of fungi and certain cel ulolytic bacteria(Ruminococcus albus, R. flavefaciens and Fibrobacter succinogenes)increased linearly(P < 0.05) but those of lactate-utilizing(Selenomonas ruminantium and Megasphaera elsdeni) and lactate-producing bacteria(Streptococcus bovis) decreased linearly(P ≤ 0.01) with increasing level of SCFP. The urinary purine derivatives increased linearly(P < 0.05) in response to SCFP supplementation, indicating that SCFP supplementation may benefit for microbial protein synthesis in the rumen.Conclusions: The SCFP supplementation was effective in maintaining milk persistency of mid-lactation cows receiving diets containing low-quality forage. The beneficial effect of SCFP could be attributed to improved rumen function; 1)microbial population shift toward greater rumen fermentation efficiency indicated by higher rumen fungi and cel ulolytic bacteria and lower lactate producing bacteria, and 2) rumen microbial fermentation toward greater supply of energy and protein indicated by greater ruminal VFA concentration and increased N conversion. Although many approaches, including various nanosystems, have been developed to promote the distribution of chemotherapeutics in the brain tumor, the delivery efficiency and the possible damage to the normal brain function still greatly restrict the clinical application of the nanocarriers.Therefore, it is urgent and necessary to discover more safe and effective BBB penetration and gliomatargeting strategies. In the present study, menthol, one of the strongest BBB penetration enhancers screened from traditional Chinese medicine, was conjugated to casein, a natural food protein with brain targeting capability. Then the conjugate self-assembled into the nanoparticles to load anti-cancer drugs.The nanoparticles were characterized to have appropriate size, spheroid shape and high loading drug capacity. Tumor spheroid penetration experiments demonstrated that penetration ability of mentholmodified casein nanoparticles(M-CA-NP) into the tumor were much deeper than that of unmodified nanoparticles. In vivo imaging further verified that M-CA-NPs exhibited higher brain tumor distribution than unmodified nanoparticles. The median survival time of glioma-bearing mice treated with HCPT-MCA-NPs was significantly prolonged than those treated with free HCPT or HCPT-CA-NPs. HE staining ofthe organs indicated the safety of the nanoparticles. | 9 | Analysis and application in controlling surrounding rock of support reinforced roadway in gob-side entry with fully mechanized mining显示文摘In order to optimize gob-side entry in fully-mechanized working face in moderate-thick-coal seams, we adopt a new attempt to pack roadside by pumping ordinary concrete, which is very important for the development of gob-side entry technology. The concrete has a long initial setting time and a low initial strength. So it is difficult to control the surrounding rock. In this paper, we analyze the effect of using roadside cable to reinforce supporting in gob-side entry surrounding rock controlling based on elas-tic-plastic and material mechanics knowledge. And then we propose a scheme that cable is used to reinforce roadside supporting and a single hydraulic prop is used as the temporary supporting in gob side. Using the numerical simulation software FLAC2D, we numerically simulated supporting scheme. Results of both the 2D modeling and the industrial test on No.3117 face in Jingang Mine prove that the scheme is feasible. | 11 | The polyketide synthase OsPKS2 is essential for pollen exine and Ubisch body patterning in rice显示文摘Lipid and phenolic metabolism are important for pollen exine formation. In Arabidopsis, polyketide synthases(PKSs) are essential for both sporopollenin biosynthesis and exine formation. Here, we characterized the role of a polyketide synthase(OsPKS2) in male reproduction of rice(Oryza sativa). Recombinant OsPKS2 catalyzed the condensation of fatty acyl-CoA with malonyl-CoA to generate triketide and tetraketide α-pyrones, the main components of pollen exine. Indeed, the ospks2 mutant had defective exine patterning and was male sterile. However, the mutant showed no significant reduction in sporopollenin accumulation. Compared with the WT(wild type), ospks2 displayed unconfined and amorphous tectum and nexine layers in the exine, and less organized Ubisch bodies. Like the pksb/lap5 mutant of the Arabidopsis ortholog, ospks2 showed broad alterations in the profiles of anther-related phenolic compounds.However, unlike pksb/lap5, in which most detected phenolics were substantially decreased, ospks2 accumulated higher levels of phenolics. | 12 | Drug targeting through platelet membrane-coated nanoparticles for the treatment of rheumatoid arthritis显示文摘The effective drug treatment of rheumatoid arthritis (RA) is hindered by poordelivery efficiency to the diseased site and the serious side effects caused bywide-spread drug distribution. Traditional drug-targeting strategies, such asligand modification, are complex, laborious, and inefficient. Inspired by theintrinsic relationship between platelets and RA, platelet-mimetic nanoparticles(PNPs) were developed for targeted drug delivery in RA. Through plateletreceptor-mediated adhesion, an intact platelet membrane was coated onto poly(lactic-co-glycolic add) nanopartides, endowing the resulting PNPs with variousfunctional receptors. By coating with platelet membranes, the nanoparticleswere stabilized and had a better circulation profile, providing a benefit forpassive targeting. In vitro binding of PNPs to inflamed endothelium, and in vivoaccumulation in joints of a collagen-induced arthritis (CIA) mouse model of RAwere significantly improved via P-selectin and GVPI recognition, indicating thatthe PNPs could effectively target to RA tissues through multiple mechanisms,similar to natural platelets. Moreover, FK506, a model drug, was loaded into thePNPs and used to treat RA. Pharmacodynamic studies demonstrated that theFK506-PNPs had a notable anti-arthritic effect in CIA mice. | 15 | Local orthogonal transform for a class of acoustic waveguides显示文摘There are some curved interfaces in acoustic waveguides. To compute wave propagation along the waveguides with some marching methods, flattening of the internal interfaces is needed. In this paper, a local orthogonal coordinate transform and an equation transform are constructed to solve the two-dimensional Helmholtz equation for the waveguides bounded by a flat top, a flat bottom and two curved internal interfaces with three layered media. The curved internal interfaces are flattened by the local orthogonal coordinate transform, and the corresponding transformed Helmholtz equation can be solved by some marching methods. This treatment can be extended in multilayered medium waveguides. The one-way reformulation based on the Dirichlet-to-Neumann (DtN) map is then used to reduce the boundary value problem to an initial value problem. Numerical implementation of the resulting operator Riccati equation uses a large range step method to discretize the range variable and a truncated local eigenfunction expansion to approximate the operators. | 16 | Large range step method for acoustic waveguide with two layer media显示文摘A numerical method is developed for solving the two-dimensional Helmholtz equation in a two-layer region bounded by a flat top, a flat bottom and a curved interface. A better local orthogonal transformation is used to flatten the curved interface of the waveguide. The one-way re-formulation based on the Dirichlet-to-Neumann (DtN) map is then used to reduce the boundary value problem to an initial value problem. Numerical implemetation of the resulting operator Riccati equation uses a large range step method for discretizing the range variable and a truncated local eigenfunction expansion for approximating the operators. | 17 | Preparation and hydrogen storage properties of MgH2-trimesic acid-TM MOF(TM=Co,Fe) composites显示文摘Two kinds of metal-organic frameworks(MOFs) based on Co(Ⅱ) and Fe(Ⅱ) as metal ions and trimasic acid(TMA) as organic linker were synthesized. They were used to prepare corresponding Mg H2-TM MOF(TM = Co, Fe) composites via ball-milling. X-ray diffraction analyses show the formation of Mg2 Co and α-Fe phases in Mg H2-TMMOF composites after decomposition. Both of the well dispersed Mg2Co andα-Fe nanoparticles exhibit considerable catalytic efficiency in accelerating the sorption kinetics of Mg H2.The dehydrogenated MgH2-Fe MOF composite shows faster hydriding kinetics than the pure Mg H2 and Mg H2-CoMOF. Meanwhile, the apparent dehydrogenation activation energy(Ed) of the Mg H2-Co MOF and Mg H2-Fe MOF composites are 151.3 ± 9.4 and 142.3 ± 6.5 kJ/mol H2, both of which are lower than that of pure Mg H2(181.4 ± 9.2 kJ/mol H2). | 20 | Evolutionary trace analysis of eukaryotic DNA topoisomerase I superfamily: Identification of novel antitumor drug binding site显示文摘The studies of novel inhibitors of DNA topoisomerase I (Topo I) have already be-come very promising in cancer chemotherapy. Identifying the new drug-binding residues is playing an important role in the design and optimization of Topo I inhibitors. The designed com-pounds may have novel scaffolds, thus will be helpful to overcome the toxicities of current camptothecin (CPT) drugs and may provide a solution to cross resistance with these drugs. Mul-tiple sequence alignments were performed on eukaryotic DNA topoisomerase I superfamily and thus the evolutionary tree was constructed. The Evolutionary Trace method was applied to iden-tify functionally important residues of human Topo I. It has been demonstrated that class-specific hydrophobic residues Ala351, Met428, Pro431 are located around the 7,9-position of CPT, indi-cating suitable substitution of hydrophobic group on CPT will increase antitumor activity. The conservative residue Lys436 in the superfamily is of particular interest and new CPT derivatives designed based on this residue may greatly increase water solubility of such drugs. It has also been demonstrated that the residues Asn352 and Arg364 were conservative in the superfamily, whose mutation will render CPT resistance. As our molecular docking studies demonstrated they did not make any direct interaction with CPT, they are important drug-binding site residues for future design of novel non-camptothecin lead compounds.
