1高血压病中医分型的代谢组学研究(英文)显示文摘目的:将传统中医辨证方法同现代系统生物学理论相结合,探讨原发性高血压辨证分型与基于GC/MS的血清代谢组学的关系。方法:原发性高血压辨证分为肝火亢盛、痰湿雍盛及阴虚阳亢三型。应用GC/MS测定健康人及原发性高血压病人血清内源性代谢物,并用主成分分析(PCA)、偏最小乘方分析(PLS-DA)和马氏距离(MD)分析他们的代谢谱。结果:PCA和PLS-DA分析的结果表明:健康人与高血压病人血清代谢谱有明显差异,能够被区分开,但PCA和PLS-DA不能将中医高血压的三型完全分开。利用MD不仅可以清晰地区分上述三种类型的高血压,同时还显示高血压的发展过程。结论:基于GC/MS和模式识别的代谢组学在揭示传统中医理论本质上有着广泛的应用前景。LU Yi-hong HAO Hai-ping WANG Guang-ji CHEN Hu-xiao ZHU Xuan-xuan XIANG Bing-ren HUANG Qing A Ji-Y 2007中国临床药理学与治疗学2007,12,10:54

7COOH-terminal deletion of HBx gene is a frequent event in HBV-associated hepatocellular carcinoma显示文摘AIM:To investigate the hepatitis B virus (HBV) x gene (HBx) state in the tissues of HBV-related hepatocellular carcinoma (HCC) in Chinese patients and whether there were particular HBx mutations. METHODS: HBx gene was amplified and direct sequencing was used in genomic DNA samples from 20 HCC and corresponding non-cancerous liver tissues from HBsAg-positive patients. HBV DNA integration and HBx deleted mutation were validated in 45 HCC patients at different stages by Southern blot analysis and polymerase chain reaction methods. RESULTS: The frequencies of HBx point mutations were significantly lower in HCC than their corresponding non- cancerous liver tissues (11/19 vs 18/19, P = 0.019). In contrast, deletions in HBx gene were significantly higher in HCC than their non-cancerous liver tissues (16/19 vs 4/19, P < 0.001). The deletion of HBx COOH-terminal was detected in 14 HCC tissues. A specific integration of HBx at 17p13 locus was also found in 8 of 16 HCC, and all of them also exhibited full-length HBx deletions. Integrated or integrated coexistence with replicated pattern was obtained in 45.5% (20/45) - 56.8% (25/45) tumors and 40.9% (18/45) - 52.3% (23/45) non-tumor tissues. CONCLUSION: HBx deletion, especially the COOH- terminal deletion of HBx is a frequent event in HBV-associated HCC tissues in China. HBV integration had also taken place in partial HCC tissues. 8A review on completing arsenic biogeochemical cycle: Microbial volatilization of arsines in environment显示文摘Arsenic(As) is ubiquitous in the environment in the carcinogenic inorganic forms, posing risks to human health in many parts of the world. Many microorganisms have evolved a series of mechanisms to cope with inorganic arsenic in their growth media such as transforming As compounds into volatile derivatives. Bio-volatilization of As has been suggested to play an important role in global As biogeochemical cycling, and can also be explored as a potential method for arsenic bioremediation. This review aims to provide an overview of the quality and quantity of As volatilization by fungi, bacteria, microalga and protozoans. Arsenic bio-volatilization is influenced by both biotic and abiotic factors that can be manipulated/elucidated for the purpose of As bioremediation. 10Metabonomic phenotype and identification of “heart blood stasis obstruction pattern” and “qi and yin deficiency pattern” of myocardial ischemia rat models显示文摘The traditional Chinese medicine concepts of 'Xinxueyuzuzheng (heart blood stasis obstruction pattern)' and 'Qiyinliangxuzheng (qi and yin deficiency pattern)' for myocardial ischemia rat models were constructed in the present study. Endogenous metabolites in rat plasma were analyzed using the GC/TOF-MS-based metabonomic method. Significant metabolic differences were observed between the control and two model groups, and the three groups were distinguished clearly by pattern recognition. Compared with those of the control, the levels of hydroxyproline, threonic acid, glutamine and citric acid were strikingly up or down-regulated in model rats. The metabolites contributing most to the classification between the two 'pattern' rats were identified, such as valine, serine, threonine, ornithine, hydroxyproline, lysine, 2-hydroxybutanoic acid, 3-hydroxybutanoic acid, galactofuranose and inositol. These compounds were indicated as the potential biomarkers. The results suggested that the two 'patterns' are involved in dysfunction in oxidative stress, energy metabolism and amino acid metabolism. 12Protective effect of early enteral feeding on postburn impairment of liver function and its mechanism in rats显示文摘AIM To study the protective effect of earlyenteral feeding(EEF)on the postburnimpairment of liver function and its mechanism.METHODS Wistar rats with 30% of total bodysurface area(TBSA)full-thickness burn wereemployed.The effects of EEF on the postburnchanges of gastric intramucosal pH,endotoxinlevels in portal vein,water contents of hepatictissue,blood concentrations of tumor necrosisfactor(TNF-α),plasma activities of alanineaminotransferase(ALT)and asparateaminotransferase(AST),as well as the bloodcontents of total(TB)and direct bilirubin(DB),total protein(TP)and albumin(ALB)wereserially determined within 48h postburn.RESULTS EEF could significantly improvegastric mucosal acidosis,reduce portal veinendotoxin level and water content of hepatictissue,as well as plasma concentrations of TNF-α at all timepoints after severe burns(P<0.01);postburn elevation of the plasma activities ofALT,AST and the contents of TB,DB wereeffectively prevented,whereas the plasmaconcentrations of TP and ALB were markedlyincreased 24 h and 48 h posturn in EEF groupcompared with that of the burn without EEFgroup(P<0.01).CONCLUSION EEF has significant beneficialeffects on the improvement of hepatic function in rats after severe burn,and is probably relatedwith an increase in splanchnic blood flow,reduction of the absorption of gut-originendotoxin and the consequent release ofinflammatory mediators.Zhu L Yang ZC Li A Cheng DC 2000World Journal of Gastroenterology2000,6,1:10
13Microstructures and mechanical properties of C-Mn-Cr-Nb and C-Mn-Si-Nb ultra-high strength dual-phase steels显示文摘The microstructures and mechanical properties of C-Mn-Cr-Nb and C-Mn-Si-Nb ultra-high strength dual-phase steels were studied by scanning electron microscopy(SEM),transmission electron microscopy(TEM),and tensile test.The results show that Si can promote the transformation of austenite(γ) to ferrite(α),enlarge the(α+γ) region,and increase the aging stability of martensite by inhibiting carbide precipitation.Adding Cr leads to the formation of retained austenite and martensiteaustenite(MA) constituents,as well as the decomposition of martensite during the overaging stage.Both of the steels show higher initial strain-hardening rates and two-stage strain-hardening characteristics.The C-Mn-Si-Nb steel shows the higher strain-hardening rate than the C-Mn-Cr-Nb steel in the first stage;however,there is no significant difference in the second stage.Although the tensile strength and elongation of the two steels both exceed 1000 MPa and 15%,respectively,the comprehensive mechanical properties of the C-Mn-Si-Nb steel are superior.Zhi-gang Wang A i-min Zhao Zheng-zhi Zhao Jie-yun Ye Di Tang Guo-sen Zhu 2012International Journal of Minerals,Metallurgy and Materials2012,19,10:7
14Reduced gastric acid production in burn shock period and its significance in the prevention and treatment of acute gastric mucosal lesions显示文摘AIM To investigate the changes of gastric acidproduction and its mechanism in shock period ofsevere burn in rats.METHODS A rat model with 30% TBSA full-thickness burn injury was employed and thegastric acid production,together with gastricmucosal blood flow(GMBF)and energy charge(EC)were measured serially within 48hpostburn.RESULTS The gastric acid production in theacute shock period was markedly inhibited aftersevere burn injury.At the 3rd h postburn,thegastric juice volume,total acidity and acidoutput were already significantly decreased(P<0.01),and reached the lowest point,0.63mL/L±0.20mL/L,10.81mmol/L±2.58mmol/L and 2.23mmol/h±0.73mmol/hrespectively,at the 12th h postburn.Althoughrestored to some degree 24 h after thermalinjury,the variables above were stillstatistically lower,compared with those ofcontrol animals at the 48th h postburn.TheGMBF and EC were also significantly reducedafter severe burns,consistent with the trend ofgastric acid production changes.CONCLUSION Gastric acid production,as wellas GMBF and EC was predominantly decreased in the early postburn stage,suggesting that gastricmucosal ischemia and hypoxia with resultantdisturbance in energy metabolism,but notgastric acid proper,might be the decisive factorin the pathogenesis of AGML after thermalinjury,and that the preventive use of anti-aciddrugs during burn shock period wasunreasonable in some respects.Therefore,taking effective measures to improve gastricmucosal blood perfusion as early as possiblepostburn might be more preferable for the AGMLprevention and treatment.Zhu L Yang ZC Li A Cheng DC 2000World Journal of Gastroenterology2000,6,1:7
15Cultivation-Independent Analysis of the Development of the Lactobacillus spp.Community in the Intestinal Tract of Newborn Piglets显示文摘Molecular diversity and development of the Lactobacillus community in the intestinal tract, as influenced by age and intestinal compartment, were studied in one litter of 12 conventionally raised piglets. Piglets were euthanized at each week (3 animals per time). Digesta and tissue samples from stomach, duodenum, jejunum, ileum, caecum, colon, and rectum were collected and analysed by using 16S ribosomal RNA-based methods. DGGE (denaturing gradient gel electrophoresis) profiles revealed that the Lactobacillus communities throughout the GI tract from duodenum to rectum showed good stability at same age. This indicates that fecal Lactobacillus communities can effectively represent the intestinal community. Two dominant bands were found in tissue samples of the small intestine, suggesting that the lactobacilli can adhere to the small intestinal wall. The Lactobacillus communities in different GI tract compartments developed over time. A successional change of Lactobacillus communities was observed from birth, through creep feeding to one week after weaning, showing a trend from simple to complex and back to simple. Furthermore, a clone library of Lactobacillus spp. 16S rRNA gene sequences were generated from jejunal and colonic chymes. Six dominant DGGE bands generated from jejunal chymes were matched with sequences that show 94-98% similarity to the bands derived from L. reuteri, L. delbrueckii, and L. crispatus. Seven dominant DGGE bands generated from colon chymes were matched with sequences that show 88-99% similarity to those derived from L. reuteri, L. delbrueckii, L. amylovorus/L. sobrius, and L. acidophilus. Amplicons related to L. reuteri were found in all DGGE fingerprints from jejunal digesta of age of weeks 1, 3, and 4. Amplicons related to L. amylovorus/L. sobrius were present in all DGGE fingerprints from colonic digesta of age of week 1, 3, and 4. Amplicons related to L. delbrueckii were found before weaning, L. crispatus after creep feeding before weaning and L. acidophilus after weaning. 18Age and menopausal status are important factors influencing the serum human epididymis secretory protein 4 level:a prospective cross-sectional study in healthy Chinese people显示文摘Background::Human epididymis secretory protein 4(HE4)is a new ovarian cancer biomarker.The factors influencing HE4 levels are not clear,and the reference data in China are limited.Here,we aim to evaluate the effects of menopause and age on HE4 levels and to provide a possible reference value for HE4 in healthy Chinese people.Methods::A total of 2493 healthy females aged 40 years or older were recruited from March 2013 to March 2017 with the cooperation of four medical institutions across Beijing,China.The serum levels of HE4 and cancer antigen 125(CA125)were measured by enzyme-linked immunosorbent assay.The Wilcoxon rank-sum test of variance and a stratified analysis were used to analyze the relationships among age,menopausal status,and levels of HE4 or CA125.Confidence intervals(5%-95%)were determined for reference ranges in different populations.Results::There was a statistically significant difference in median HE4 levels between the post-menopausal(n=2168)and premenopausal groups(n=325)(36.46 vs.24.04 pmol/L,Z=-14.41,P<0.001).HE4 increased significantly with age in the post-menopausal groups(H=408.18,P<0.001)but not in the pre-menopausal subjects(Z=-0.43,P=0.67).The upper 95th percentile of HE4 levels were 44.63 pmol/L for pre-menopausal women,78.17 pmol/L for post-menopausal women,and 73.3 pmol/L for all women.In the post-menopausal population,the HE4 reference ranges were 13.15 to 47.31,14.31 to 58.04,17.06 to 73.51,24.50 to 115.25,and 35.71 to 212.37 pmol/L for different age groups from forty divided by decade.The CA125 level was affected mainly by menopausal status and not age.Conclusions::Menopausal status and age were both important factors influencing the level of HE4,and age affected HE4 levels mainly in post-menopausal women.The HE4 level was higher in the post-menopausal population than in the pre-menopausal population and increased with age.Hong-Yan Cheng Lin Zeng Xue Ye Rui-Qiong M a Zhi-Jian Tang Hong-Ling Chu Yi-M ing Zhao Li-Rong Zhu Yu-Nong Gao Xiao-Hong Chang Heng Cui 2020Chinese Medical Journal2020,,11:6
