2Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
32.5D scattering of incident plane SV waves by a canyon in layered half-space显示文摘This paper presents a 2.5D scattering of incident plane SV waves by a canyon in a layered half-space by using the indirect boundary element method (IBEM). A free field response analysis is performed to provide the displacements and stresses on the boundary of the canyon where fictitious uniform moving loads are applied to calculate the Green's functions for the displacements and stresses. The amplitudes of the loads are determined by the boundary conditions. The free field displacements are added to the fictitious uniform moving loads induced displacements and the total response is obtained. Numerical calculations are performed for a canyon with homogenous and in one layer over bedrock. 4Involvement of the Mitochondrion-dependent and the Endoplasmic Reticulum Stress-signaling Pathways in Isoliquiritigenin-induced Apoptosis of HeLa Cell显示文摘Objective Isoliquiritigenin(ISL),a licorice chalconoid,is considered to be a bioactive agent with chemopreventive potential.This study investigates the mechanisms involved in ISL-induced apoptosis in human cervical carcinoma HeLa cells.Methods Cell viability was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)assay.Apoptosis was determined by flow cytometry using an Annexin V-FITC Apoptosis Detection Kit.The intracellular ROS levels were assessed using a 2,7-dichlorofluorescein probe assay.The mitochondrial membrane potential was measured with the dual-emission potential-sensitive probe 5,5',6,6'-tetra-chloro-1,1',3,3'-tetraethyl-imidacarbocyanine iodide(JC-1).The degradation of poly-ADP-ribose polymerase(PARP)protein,the phosphorylation of PKR-like ER kinase(PERK),the phosphorylation of the α-subunit of eukaryotic initiation factor 2(eIF2α),the expression of the 78 kD glucose-regulated protein(GRP 78),and the activation of caspase-12 were analyzed via western blot analysis.Results ISL significantly inhibited the proliferation,the increase in ROS levels and apoptotic rates of HeLa cells in a concentration-dependent manner.Moreover,ISL induced mitochondrial dysfunction,caspase activation,and PARP cleavage,which displayed features of mitochondria dependent on apoptotic signals.Besides,exposure of HeLa cells to ISL triggered endoplasmic reticulum(ER)stress,as indicated by the increase in p-eIF2α and GRP78 expression,ER stress-dependent apoptosis is caused by the activation of ER-specific caspase-12.Conclusion The findings from our study suggest that ISL-induced oxidative stress causes HeLa cell apoptosis via the mitochondrion-dependent and the ER stress-triggered signaling pathways.YUAN Xuan ZHANG Bo GAN Lu WANG Zhen Hua YU Ba Cui LIU Liang Liang ZHENG Qiu Sheng WANGZhi Ping 2013Biomedical and Environmental Sciences2013,26,4:8

The amplitudes of the loads are determined by the boundary conditions. The displacements due to the free field and from the fictitious uniform moving loads have to be added to obtain the whole motion. The numerical results are carried out for the cases of a canyon in homogenous and in one layer over bedrock. The results show that the 2.5D wave scattering problem is essentially different from the 2D case, and there exist distinct differences between the wave amplification by a canyon in layered half-space and that in homogeneous half-space. 7Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 8Diffraction of SH-waves by topographic features in a layered transversely isotropic half-space显示文摘The scattering of plane SH-waves by topographic features in a layered transversely isotropic(TI) half-space is investigated by using an indirect boundary element method(IBEM).Firstly, the anti-plane dynamic stiffness matrix of the layered TI half-space is established and the free fields are solved by using the direct stiffness method.Then, Green's functions are derived for uniformly distributed loads acting on an inclined line in a layered TI half-space and the scattered fields are constructed with the deduced Green's functions.Finally, the free fields are added to the scattered ones to obtain the global dynamic responses.The method is verified by comparing results with the published isotropic ones.Both the steady-state and transient dynamic responses are evaluated and discussed.Numerical results in the frequency domain show that surface motions for the TI media can be significantly different from those for the isotropic case, which are strongly dependent on the anisotropy property, incident angle and incident frequency.Results in the time domain show that the material anisotropy has important effects on the maximum duration and maximum amplitudes of the time histories.Ba Zhenning Liang Jianwen Zhang Yanju 2017Earthquake Engineering and Engineering Vibration2017,16,1:5
9Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 11Amplification of in-plane seismic ground motion by group cavities in layered half-space (Ⅱ): with saturated poroelastic soil layers显示文摘As the continuation study on amplification of in-plane seismic ground motion by underground group cavities in layered half-space, this study extends to the case of poroelastic half-space with dry poroelastic and saturated poroelastic soil layers. The influence of poroelastic layers on the amplification of seismic ground motion is studied both in frequency domain and time domain using indirect boundary element method (IBEM). It is shown that for the example of a saturated poroelastic site in Tianjin under the excitation of Taft wave and El Centro wave, the amplification of seismic ground motion in poroelastic case is slightly smaller than that in the elastic case, and the amplification of PGA (peak ground acceleration) and its PRS (peak response spectrum) can be increased up to 38.8% and 64.6%; the predominant period of response spectra in poroelastic case becomes shorter to some extent compared with that in the elastic case. 13In-plane dynamic Green's functions for inclined and uniformly distributed loads in a multi-layered transversely isotropic half-space显示文摘The dynamic stiffness method combined with the Fourier transform is utilized to derive the in-plane Green’s functions for inclined and uniformly distributed loads in a multi-layered transversely isotropic(TI)half-space.The loaded layer is fixed to obtain solutions restricted in it and the corresponding reactions forces,which are then applied to the total system with the opposite sign.By adding solutions restricted in the loaded layer to solutions from the reaction forces,the global solutions in the wavenumber domain are obtained,and the dynamic Green’s functions in the space domain are recovered by the inverse Fourier transform.The presented formulations can be reduced to the isotropic case developed by Wolf(1985),and are further verified by comparisons with existing solutions in a uniform isotropic as well as a layered TI halfspace subjected to horizontally distributed loads which are special cases of the more general problem addressed.The deduced Green’s functions,in conjunction with boundary element methods,will lead to significant advances in the investigation of a variety of wave scattering,wave radiation and soil-structure interaction problems in a layered TI site.Selected numerical results are given to investigate the influence of material anisotropy,frequency of excitation,inclination angle and layered on the responses of displacement and stress,and some conclusions are drawn.Ba Zhenning Kang Zeqing Liang Jianwen 2018Earthquake Engineering and Engineering Vibration2018,17,2:3
143D scattering of obliquely incident plane SV waves by an alluvial valley embedded in a fluid-saturated, poroelastic layered half-space显示文摘The indirect boundary element method is used to study the 3D dynamic response of an infinitely long alluvial valley embedded in a saturated layered half-space for obliquely incident SV waves. A wave-number transform is first applied along the valley's axis to reduce a 3D problem to a2D plane strain problem. The problem is then solved in the section perpendicular to the axis of the valley. Finally, the 3D dynamic responses of the valley are obtained by an inverse wave-number transform. The validity of the method is confirmed by comparison with relevant results. 15Seismic analysis of high-speed railway irregular bridge–track system considering V-shaped canyon effect显示文摘To explore the effect of canyon topography on the seismic response of railway irregular bridge-track system that crosses a V-shaped canyon, seismic ground motions of the horizontal site and V-shaped canyon site were simulated through theoretical analysis with 12 earthquake records selected from the Pacific Earthquake Engineering Research Center(PEER) Strong Ground Motion Database matching the site condition of the bridge.Nonlinear seismic response analyses of an existing 11-span irregular simply supported railway bridge-track system were performed under the simulated spatially varying ground motions. The effects of the V-shaped canyon topography on the peak ground acceleration at bridge foundations and seismic responses of the bridge-track system were analyzed. Comparisons between the results of horizontal and V-shaped canyon sites show that the top relative displacement between adjacent piers at the junction of the incident side and the back side of the V-shaped site is almost two times that of the horizontal site, which also determines the seismic response of the fastener. The maximum displacement of the fastener occurs in the V-shaped canyon site and is 1.4 times larger than that in the horizontal site. Neglecting the effect of V-shaped canyon leads to the inappropriate assessment of the maximum seismic response of the irregular high-speed railway bridge-track system. 16Amplification of in-plane seismic ground motion by group cavities in layered half-space (Ⅰ)显示文摘Amplification of in-plane seismic ground motion by underground group cavities in layered half-space is studied both in frequency domain and time domain by using indirect boundary element method (IBEM), and the effect of cavity interval and spectrum of incident waves on the amplification are studied by numerical examples. It is shown that there may be large interaction between cavities, and group cavities with certain intervals may have significant amplification to seismic ground motion. The amplification of PGA (peak ground acceleration) and its PRS (peak response spectrum) can be increased up to 45.2% and 84.4%, for an example site in Tianjin, under the excitation of Taft wave and El Centro wave; and group cavities may also affect the spectra of the seismic ground motion. 18Scattering and diffraction of plane SH-waves by periodically distributed canyons显示文摘A new method is presented to study the scattering and diffraction of plane SH-waves by periodically distributed canyons in a layered half-space. This method uses the indirect boundary element method combined with Green's functions of uniformly distributed loads acting on periodically distributed inclined lines. The periodicity feature of the canyons is exploited to limit the discretization effort to a single canyon, which avoids errors induced by the truncation of the infinite boundary, and the computational complexity and the demand on memory can be significantly reduced. Furthermore, the total wave fields are decomposed into the free field and scattered field in the process of calculation, which means that the method has definite physical meaning. The implementation of the method is described in detail and its accuracy is verified. Parametric studies are performed in the frequency domain by taking periodically distributed canyons of semi-circular and semi-elliptic cross-sections as examples.
