2Characteristic dysbiosis of gut microbiota of Chinese patients with diarrhea-predominant irritable bowel syndrome by an insight into the pan-microbiome显示文摘Background:Irritable bowel syndrome(IBS)is reported associated with the alteration of gut microbial composition termed as dysbiosis.However,the pathogenic mechanism of IBS remains unclear,while the studies of Chinese individuals are scarce.This study aimed to understand the concept of dysbiosis among patients with Chinese diarrhea-predominant IBS(IBS-D),as a degree of variance between the gut microbiomes of IBS-D population and that of a healthy population.Methods:The patients with IBS-D were recruited(assessed according to the Rome III criteria,by IBS symptom severity score)from the Outpatient Department of Gastroenterology of Peking University Third Hosp让al,and volunteers as healthy controls(HCs)were enrolled,during 2013.The 16S rRNA sequences were extracted from fecal samples.Ribosomal database project resources,basic local alignment search tool,and SparCC software were used to obtain the phylotype composition of samples and the internal interactions of the microbial community.Herein,the non-parametric test,Wilcoxon rank-sum test was carried out to find the statistical significance between HC and IBS-D groups.All the P values were adjusted to q values to decrease the error rate.Results:The study characterized the gut microbiomes of Chinese patients with IBS-D,and demonstrated that the dysbiosis could be characterized as directed alteration of the microbiome composition leading to greater disparity between relative abundance of two phyla,Bacteroidetes(Z=4.77,q=1.59×10^-5)and Firmicutes(Z=-3.87,q=5.83×10^-4).Moreover,it indicated that the IBS symptom features were associated with the dysbiosis of whole gut microbiome,instead of one or several certain genera even they were dominating.Two genera,Bacteroides and Lachnospiracea incertae sedis,were identified as the core genera,meanwhile,the non-core genera contribute to a larger pan-microbiome of the gut microbiome.Furthermore,the dysbiosis in patients with IBS-D was associated with a reduction of network complexity of the interacted microbial community(HC us.IBS-D:639 vs.154).The disordered metabolic functions of patients with IBS-D were identified as the potential influence of gut microbiome on the host(significant difference with q<0.01 between HC and IBS-D).Conclusions:This study supported the view of the potential influence of gut microbiome on the symptom of Chinese patients with IBS-D,and further characterized dysbiosis in Chinese patients with IBS-D,thus provided more pathological evidences for IBS-D with the further understanding of dysbiosis.Zhe Wang Cong-Min Xu Yi-Xuan Liu Xiao-Qi Wang Lu Zhang Mo Li Shi-Wei Zhu Zhong-Jie Xie Pei-Hong Wang Li-Ping Duan Huai-Qiu Zhu 2019Chinese Medical Journal2019,,8:15
3Exosomes derived from bone marrow mesenchymal stem cells protect the injured spinal cord by inhibiting pericyte pyroptosis显示文摘Mesenchymal stem cell(MSC)transplantation is a promising treatment strategy for spinal cord injury,but immunological rejection and possible tumor formation limit its application.The therapeutic effects of MSCs mainly depend on their release of soluble paracrine factors.Exosomes are essential for the secretion of these paracrine effectors.Bone marrow mesenchymal stem cell-derived exosomes(BMSC-EXOs)can be substituted for BMSCs in cell transplantation.However,the underlying mechanisms remain unclear.In this study,a rat model of T10 spinal cord injury was established using the impact method.Then,30 minutes and 1 day after spinal cord injury,the rats were administered 200μL exosomes via the tail vein(200μg/mL;approximately 1×106 BMSCs).Treatment with BMSC-EXOs greatly reduced neuronal cell death,improved myelin arrangement and reduced myelin loss,increased pericyte/endothelial cell coverage on the vascular wall,decreased bloodspinal cord barrier leakage,reduced caspase 1 expression,inhibited interleukin-1βrelease,and accelerated locomotor functional recovery in rats with spinal cord injury.In the cell culture experiment,pericytes were treated with interferon-γand tumor necrosis factor-α.Then,Lipofectamine 3000 was used to deliver lipopolysaccharide into the cells,and the cells were co-incubated with adenosine triphosphate to simulate injury in vitro.Pre-treatment with BMSC-EXOs for 8 hours greatly reduced pericyte pyroptosis and increased pericyte survival rate.These findings suggest that BMSC-EXOs may protect pericytes by inhibiting pyroptosis and by improving blood-spinal cord barrier integrity,thereby promoting the survival of neurons and the extension of nerve fibers,and ultimately improving motor function in rats with spinal cord injury.All protocols were conducted with the approval of the Animal Ethics Committee of Zhengzhou University on March 16,2019.Yan Zhou Lu-Lu Wen Yan-Fei Li Kai-Min Wu Ran-Ran Duan Yao-Bing Yao Li-Jun Jing Zhe Gong Jun-Fang Teng Yan-Jie Jia 2022Neural Regeneration Research2022,17,1:15
4Endovascular treatment with or without intravenous alteplase for acute ischaemic stroke due to basilar artery occlusion显示文摘Background and purpose It remains controversial if endovascular treatment(EVT)can improve the outcome of patients with acute basilar artery occlusion(BAO).This study aims to compare the functional outcomes between EVT with and without intravenous thrombolysis(IVT)first in patients who had acute ischaemic stroke(AIS)due to BAO.Methods Patients who had AIS with BAO who underwent EVT within 24 hours of onset were enrolled in this multicentre cohort study,and the efficacy and safety were compared between IVT+EVT and direct EVT.The primary outcome was 90-day functional independence.All outcomes were assessed with adjusted OR(aOR)from the multivariable logistic regression.In addition,a meta-analysis was performed on all recently published pivotal studies on functional independence after EVT in patients with BAO.Results Of 310 enrolled patients with BAO,241(78%)were treated with direct EVT and 69(22%)with IVT+EVT.Direct EVT was associated with a worse functional outcome(aOR,0.46(95%CI 0.24 to 0.85),p=0.01).IVT+EVT was associated with a lower percentage of patients who needed≥3 passes of stent retriever(10.14%vs 20.75%).The meta-analysis regression revealed a potential positive correlation between bridging with IVT first and functional independence(r=0.14(95%CI 0.05 to 0.24),p<0.01).Conclusions This study showed that compared with direct EVT,EVT with IVT first was associated with better functional outcomes in patients with BAO treated within 24 hours of onset.The meta-analysis demonstrated similar favourable efficacy of IVT first followed by EVT in patients with BAO.Ximing Nie David Wang Yuehua Pu Yufei Wei Qixuan Lu Hongyi Yan Xin Liu Lina Zheng Jingyi Liu Xinxuan Yang Yarong Ding Dacheng Liu Wanying Duan Zhe Zhang Zhonghua Yang Miao Wen Weibin Gu Xinyi Hou Xinyi Leng Yuesong Pan Zhongrong Miao Liping Liu 2022Stroke & Vascular Neurology2022,7,3:10

6The influence of nanosized precipitates on Portevin-Le Chatelier bands and surface roughness in AlMgScZr alloy显示文摘The influence of different precipitate-dislocation interactions,namely dislocation shearing and bypassing mechanisms,on PLC bands and the resultant surface roughness in AlMgScZr alloy was investigated.Three-dimensional surface roughness was quantitatively measured by confocal microscopy.We find that the introduction of shearable precipitates increases the stress amplitude,decreases the PLC bands number and surface roughness.However,the stress amplitude decreases,the PLC bands number and surface roughness increase with shearable precipitates turning to nonshearable precipitates.By analyzing the precipitation strengthening mechanisms quantitatively,the influence of precipitates on PLC bands and the resultant surface roughness was explored.Furthermore,our study demonstrates that the shearable precipitates can decrease the surface roughness by decreasing the number of PLC bands,which is instructive for designing structural materials with desirable mechanical property and surface quality.Yaoxiang Duan Han Chen Zhe Chen Lei Wang Mingliang Wang Jun Liu Fengguo Zhang Haowei Wang 2021Journal of Materials Science & Technology2021,,28:3

8Clinical characteristic and fecal microbiota responses to probiotic or antidepressant in patients with diarrhea-predominant irritable bowel syndrome with depression comorbidity: a pilot study显示文摘To the Editor: Irritable bowel syndrome (IBS) is a prevalent functional gastrointestinal disorder that presents as abdominal pain with altered bowel habits. The pathophysiologic mechanism of IBS is not well understood, although many hypotheses have been proposed, including visceral hypersensitivity, gastrointestinal dysmotility, lowgrade inflammation of the intestinal mucosa, and dysfunction of the brain-gut interaction. Dysfunction of the braingut interaction is thought to be involved in IBS because a considerable proportion of patients with IBS have some form of psychologic comorbidity, such as depression or anxiety. In addition, mental and psychologic problems increase the risk for IBS and the symptom severity of IBS. Recently, changes in the gut microbiota have also been suggested to contribute to both IBS and depression. 10Preliminary study on detection and cleaning of parasitic bunches显示文摘To establish a nuclear resonant scattering beamline at the High Energy Photon Source(HEPS),it is essential to develop tools for detection and cleaning of parasitic bunches,for meeting the stringent demands on bunch purity.To this end,a novel time-correlated single-photon counting system was implemented at the electron storage ring of the Beijing Electron–Positron Collider II(BEPCII).The purity deterioration process over a week-long operation was recorded by the system.In this study,the mechanism of impurity growth was analyzed by numerical methods and validated on measurements.The agreement between the experimental results and the cal-culation was fairly good.Two main sources of parasitic bunches,pre-accelerators and the Touschek scattering were confirmed.A bunch-cleaning technique,based on a sinu-soidal signal mixed with a pseudo-square wave,was also developed and implemented,and its capability to improve the bunch purity to the level of 10–7 was experimentally demonstrated.We present the experimental setup,princi-ple,and measurement results of a system for detection and cleaning of parasitic bunches.Jun He Yan-Feng Sui Yan-Hua Lu Di Yin Zhe Duan Sai-Ke Tian De-Chong Zhu Ying Zhao Jun-Hui Yue Jian-She Cao 2021Nuclear Science and Techniques2021,32,10:2
11On the robustness of EEG tensor completion methods显示文摘During the acquisition of electroencephalographic(EEG) signals, data may be missing or corrupted by noise and artifacts. To reconstruct the incomplete data, EEG signals are firstly converted into a three-order tensor(multi-dimensional data) of shape time × channel × trial. Then, the missing data can be efficiently recovered by applying a tensor completion method(TCM).However, there is not a unique way to organize channels and trials in a tensor, and different numbers of channels are available depending on the EEG setting used, which may affect the quality of the tensor completion results. The main goal of this paper is to evaluate the robustness of EEG completion methods with several designed parameters such as the ordering of channels and trials, the number of channels, and the amount of missing data. In this work, the results of completing missing data by several TCMs were compared. To emulate different scenarios of missing data, three different patterns of missing data were designed.Firstly, the amount of missing data on completion effects was analyzed, including the time lengths of missing data and the number of channels or trials affected by missing data. Secondly, the numerical stability of the completion methods was analyzed by shuffling the indices along channels or trials in the EEG data tensor. Finally, the way that the number of electrodes of EEG tensors influences completion effects was assessed by changing the number of channels. Among all the applied TCMs, the simultaneous tensor decomposition and completion(STDC) method achieves the best performance in providing stable results when the amount of missing data or the electrode number of EEG tensors is changed. In other words, STDC proves to be an excellent choice of TCM, since permutations of trials or channels have almost no influence on the complete results. The STDC method can efficiently complete the missing EEG signals. 12Stress Relaxation and Sensitivity Weight for Bi-Directional Evolutionary Structural Optimization to Improve the Computational Efficiency and Stabilization on Stress-Based Topology Optimization显示文摘Stress-based topology optimization is one of the most concerns of structural optimization and receives much attention in a wide range of engineering designs.To solve the inherent issues of stress-based topology optimization,many schemes are added to the conventional bi-directional evolutionary structural optimization(BESO)method in the previous studies.However,these schemes degrade the generality of BESO and increase the computational cost.This study proposes an improved topology optimization method for the continuum structures considering stress minimization in the framework of the conventional BESO method.A global stress measure constructed by p-norm function is treated as the objective function.To stabilize the optimization process,both qp-relaxation and sensitivity weight scheme are introduced.Design variables are updated by the conventional BESO method.Several 2D and 3D examples are used to demonstrate the validity of the proposed method.The results show that the optimization process can be stabilized by qp-relaxation.The value of q and p are crucial to reasonable solutions.The proposed sensitivity weight scheme further stabilizes the optimization process and evenly distributes the stress field.The computational efficiency of the proposed method is higher than the previous methods because it keeps the generality of BESO and does not need additional schemes.Chao Ma Yunkai Gao Yuexing Duan Zhe Liu 2021Computer Modeling in Engineering & Sciences2021,,2:2

19Twist2 is a valuable prognostic biomarker for colorectal cancer显示文摘AIM: To investigate the significance of Twist2 for colorectal cancer (CRC). METHODS: In this study, 93 CRC patients were included who received curative surgery in Eastern Hepatobiliary Surgery Hospital from January 1999 to December 2010. Records of patients' clinicopathological characteristics and follow up data were reviewed. Formalin-fixed, paraffin-embedded tissue blocks were used to observe the protein expression of Twist2 and E-cadherin by immunohistochemistry. Two independent pathologists who were blinded to the clinical information performed semiquantitative scoring of immunostaining. A total score of 3-6 (sum of extent + intensity) was considered as Twist2-positive expression. The expression of E-cadherin was divided into two levels (preserved and reduced). An exploratory statistical analysis was conducted to determine the association between Twist2 expression and clinicopathological parameters, as well as E-cadherin expression. Furthermore, the variables associated with prognosis were analyzed by Cox's proportional hazards model. Kaplan-Meier analysis was used to plot survival curves according to different expression levels of Twist2. RESULTS: Twist2-positive expression was observed in 66 (71.0%) samples and mainly located in the cytoplasm. Forty-three (46.2%) samples showed reduced expression of E-cadherin. There were no significant correlations between Twist2 expression and any of the clinicopathological parameters. However, Twist2-positive expression was significantly associated with reduced expression of E-cadherin (P=0.040). Multivariate analysis revealed that bad M-stage [hazard ratio (HR)=7.694, 95%CI: 2.927-20.224,P < 0.001] and Twist2-positive (HR=5.744, 95%CI: 1.347-24.298,P=0.018) were the independent risk factors for poor overall survival (OS), while Twist2-positive (HR=3.264, 95%CI: 1.455-7.375, P=0.004), bad N-stage (HR=2.149, 95%CI: 1.226-3.767, P=0.008) and bad M-stage (HR=10.907, 95%CI: 4.937-24.096, P < 0.001) were independently associated with poor disease-free survival (DFS). Survival curves showed a definite trend for Twist2-negative patients to have longer OS and DFS than Twist2-negative patients, not only overall, but also for patients in different stages, especially for DFS of patients in stage Ⅲ (P=0.033) and Ⅳ (P=0.026).
