1Stabilization of heterochromatin by CLOCK promotes stem cell rejuvenation and cartilage regeneration显示文摘Accumulating evidence indicates an association between the circadian clock and the aging process.However,it remains elusive whether the deregulation of circadian clock proteins underlies stem cell aging and whether they are targetable for the alleviation of aging-associated syndromes.Here,we identified a transcription factor-independent role of CLOCK,a core component of the molecular circadian clock machinery,in counteracting human mesenchymal stem cell(hMSC)decay.CLOCK expression was decreased during hMSC aging.In addition,CLOCK deficiency accelerated hMSC senescence,whereas the overexpression of CLOCK,even as a transcriptionally inactive form,rejuvenated physiologically and pathologically aged hMSCs.Mechanistic studies revealed that CLOCK formed complexes with nuclear lamina proteins and KAP1,thus maintaining heterochromatin architecture and stabilizing repetitive genomic sequences.Finally,gene therapy with lentiviral vectors encoding CLOCK promoted cartilage regeneration and attenuated age-related articular degeneration in mice.These findings demonstrate a noncanonical role of CLOCK in stabilizing heterochromatin,promoting tissue regeneration,and mitigating aging-associated chronic diseases.Chuqian Liang Zunpeng Liu Moshi Song Wei Li Zeming Wu Zehua Wang Qiaoran Wang Si Wang Kaowen Yan Liang Sun Tomoaki Hishida Yanning Cai Juan Carlos lzpisua Belmonte Pedro Guillen Piu Chan Qi Zhou Weiqi Zhang Jing Qu Guang-Hui Liu 2021Cell Research2021,31,2:8

5Mechanical stress distribution and risk assessment of 110 kV GIS insulator considering Al_(2)O_(3)settlement显示文摘The growing application of gas insulated switchgear(GIS)in electric power system asks for higher reliability of epoxy(EP)insulators,thus it is important to seek effective methods to improve the operating reliability of GIS insulators.On the basis of the 110 kV GIS insulator,this study studied the aluminium oxide(Al_(2)O_(3))settlement in the curing process and the stress distribution of the horizontally laid GIS.Besides,the risk coefficient was defined to evaluate the operating reliability of insulators laid by different methods(I-V).Some conclusions are shown below:the Al_(2)O_(3)settlement in the curing process causes the insulator density to have a large variation from 2.13 to 2.32 g/cm^(3).The Al_(2)O_(3)content has a great influence on the mechanical performances of EP/Al_(2)O_(3)system.The first principle stress is localised on the upper interface between the insulator and the conductor,which is the main threat to the mechanical damage of insulator.The laying method V appears to be optimal with a pretty low risk coefficient of 31.5.In actual application,for the horizontally laid GIS,the insulator should be laid with the mould gate downward to improve its mechanical reliability.Hucheng Liang Boxue Du Jin Li Zehua Wang 2019High Voltage2019,4,1:5
6Design and test of soft-pot-tray automatic embedding system for light-economical pot seedling nursery machine显示文摘Most of the commercially-available pot seedling nursery machines are incompatible with soft-pot-trays and are labor-intensive and low in productivity.A soft-pot-tray automatic embedding system was developed in this study to achieve automatic embedding of the soft pot tray into the hard tray following sowing and covering with soil.A control system was constructed using the Arduino program development environment.An embedded-hard-tray automatic lowering mechanism and conveyor-belt-based pot-tray embedding system were designed.Dynamics analysis was conducted to derive an equation to describe the embedding process of the soft pot tray into the embedded hard tray.A prototype of the soft-pot-tray automatic embedding system was manufactured and tested.The analytical equation suggested that a minimum linear velocity of 0.86 m/s was required for a complete embedding process.The experimental results showed that the embedded-hard-tray automatic lowering mechanism was reliable and stable as the tray placement success rate was greater than 99%.The successful tray embedding rate was 100%and the seed exposure rate was less than 1%with a linear velocity of the conveyor belt of 0.92 m/s.The experiment findings agreed well with the analytical results.The proposed soft-pot-tray automatic embedding system satisfied the technical specifications for a light-economical pot seedling nursery machine.Lintao Chen Xu Ma Cong Wang Hongwei Li Zehua Li Xueshen Chen Tao Chen 2020International Journal of Agricultural and Biological Engineering2020,13,1:4

8In-situ observation of electrical tree evolution in epoxy dielectrics with internal cracks显示文摘Gas-filled internal crack might appear in thermoset materials like epoxy resin during the equipment manufacturing,which would become a vulnerable local region to initiate the electrical tree,thus prone to cause insulation failure.The withstand voltage test was carried on epoxy samples with artificial cracks based on a rod-plane electrode arrange-ment.Simultaneously,surface state variation and tree evolution with crack were observed by an optical microscope in conjunction with a charge-coupled device camera.The changes in morphology and chemical status of the crack surface were characterized by scanning electron microscopy,laser Raman spectrometer and energy dispersive spec-trometer,respectively.It was found that the erosion and tree started from the borderline of crack under a relatively low electric field strength;however,the area near the electrode had relatively little damage.The breaking of epoxy molecular chains coarsens the crack surface and further forms deep channels on a micro-level,which is the forerunner of the electrical tree inception.Based on these,the initiation mechanisms of the crack-induced electrical tree and the reasons for the erosion near the borderline have been revealed.This study provides a train thought for the polymer degradation opening up into the initial tree channel during tree evolution processes.Zehua Wu Huidong Tian Sijia Zhu He Li Qingyu Wang Zongliang Xie Peng Liu Zongren Peng 2021High Voltage2021,6,2:3
9A grouting simulation method for quick-setting slurry in karst conduit:The sequential flow and solidification method显示文摘It is difficult to temporally and spatially track and characterize the slurry viscosity in flowing water during grouting simulation.In this study,a sequential flow and solidification(SFS)method considering the spatial-temporal evolution of slurry viscosity in flowing water in karst conduit is proposed.First,a time-dependent model for the threshold function of slurry viscosity is established.During the grouting process,the spatial-temporal evolution of slurry viscosity is revealed by tracking the diffusion behavior of the slurry injected at different times.This method is capable of describing the gradual solidification process of the slurry during grouting.Furthermore,a physical model of grouting in a karst conduit is developed.Second,the effectiveness of the SFS method in grouting simulation is verified by the experiment of grouting conduit in flowing water.The SFS method enables real-time monitoring of fluid velocity and pressure during grouting in flowing water and provides a feasible calculation method for revealing the grouting plugging mechanism in complex karst conduits at different engineering scales.In addition,it can be used to guide the design of grouting tests in flowing water,improve cost efficiency,and provide theoretical basis for optimizing grouting design and slurry selection.Zhenhao Xu Dongdong Pan Shucai Li Yichi Zhang Zehua Bu Jie Liu 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,2:3
10Hepatitis B virus evades immune recognition via RNA adenosine deaminase ADAR1-mediated viral RNA editing in hepatocytes显示文摘HBV is considered as a“stealth”virus that does not invoke interferon(IFN)responses;however,the mechanisms by which HBV bypasses innate immune recognition are poorly understood.In this study,we identified adenosine deaminases acting on RNA 1(ADAR1),which is a key factor in HBV evasion from IFN responses in hepatocytes.Mechanically,ADAR1 interacted with HBV RNAs and deaminated adenosine(A)to generate inosine(I),which disrupted host immune recognition and thus promoted HBV replication.Loss of ADAR1 or its deficient deaminase activity promoted IFN responses and inhibited HBV replication in hepatocytes,and blocking the IFN signaling pathways released the inhibition of HBV replication caused by ADAR1 deficiency.Notably,the HBV X protein(HBx)transcriptionally promoted ADAR1 expression to increase the threshold required to trigger intrinsic immune activation,which in turn enhanced HBV escape from immune recognition,leading to persistent infection.Supplementation with 8-azaadenosine,an ADAR1 inhibitor,efficiently enhanced liver immune activation to promote HBV clearance in vivo and in vitro.Taken together,our results delineate a molecular mechanism by which HBx promotes ADAR1-derived HBV immune escape and suggest a targeted therapeutic intervention for HBV infection.Liyuan Wang Yang Sun Xiaojia Song Zehua Wang Yankun Zhang Ying Zhao Xueqi Peng Xiaodong Zhang Chunyang Li Chengjiang Gao Nailin Li Lifen Gao Xiaohong Liang Zhuanchang Wu Chunhong Ma 2021Cellular & Molecular Immunology2021,18,8:2
11Signaling pathways in obesity:mechanisms and therapeutic interventions显示文摘Obesity is a complex,chronic disease and global public health challenge.Characterized by excessive fat accumulation in the body,obesity sharply increases the risk of several diseases,such as type 2 diabetes,cardiovascular disease,and nonalcoholic fatty liver disease,and is linked to lower life expectancy.Although lifestyle intervention(diet and exercise)has remarkable effects on weight management,achieving long-term success at weight loss is extremely challenging,and the prevalence of obesity continues to rise worldwide.Over the past decades,the pathophysiology of obesity has been extensively investigated,and an increasing number of signal transduction pathways have been implicated in obesity,making it possible to fight obesity in a more effective and precise way.In this review,we summarize recent advances in the pathogenesis of obesity from both experimental and clinical studies,focusing on signaling pathways and their roles in the regulation of food intake,glucose homeostasis,adipogenesis,thermogenesis,and chronic inflammation.We also discuss the current antiobesity drugs,as well as weight loss compounds in clinical trials,that target these signals.The evolving knowledge of signaling transduction may shed light on the future direction of obesity research,as we move into a new era of precision medicine.Xue Wen Bohan Zhang Beiyi Wu aitao Xiao Zehua Li Ruoyu Li Xuewen Xu Tao Li 2022Signal Transduction and Targeted Therapy2022,7,9:2

13Secure Cloud Architecture for 5G Core Network显示文摘Service-based architecture(SBA)is a profound advancement in the novel 5G Core network(5GC).Existing studies show that SBA can benefit from cloud computing to achieve extensibility,modularity,reusability,and openness.It also brings security problems(e.g.,hypervisor hijacking,and malware injection).To provide secure 5G services,we propose a service-based cloud architecture called Mimicloud for 5GC based on dynamic and heterogeneous techniques.Mimicloud provides flexible reconfiguration mechanisms to protect containers and eliminate all attack knowledge obtained from adversaries.We use multiple containers to execute crucial services and ensure security with crosscheck.Mimicloud employs heterogeneous components to prevent multiple containers from being breached through the same vulnerabilities.Experimental results show that Mimicloud can effectively strengthen the security of the 5GC.The performance overhead is analyzed in order to demonstrate its scalability.LI Lingshu WU Jiangxing HU Hongchao LIU Wenyan GUO Zehua 2021Chinese Journal of Electronics2021,30,3:2
14Enhanced weathering input from South Asia to the Indian Ocean since the late Eocene显示文摘nhanced silicate weathering induced by the uplift of the Himalayan-Tibetan Plateau(HTP)has been considered as the major cause of pCO_(2) decline and Cenozoic cooling.However,this hypothesis remains to be validated,largely due to the lack of a reliable reconstruction of the HTP weathering flux.Here,we present a 37-million-year record of the difference in the seawater radiogenic neodymium isotopic composition(△ε_(Nd))of Ocean Drilling Program(ODP)sites and Fe-Mn crusts between the northern and central Indian Ocean,which indicates the contribution of regional weathering input from the South Asian continent to the Indian Ocean.The results show a long-term increase in △ε_(Nd) and thus provide the first critical evidence of enhanced South Asian weathering input since the late Eocene.The evolution coincided well with major pulses of surface uplift in the HTP and global climatic transitions.Our foraminiferal eNd record suggests that tectonic uplift and silicate weathering in South Asia,especially in the Himalayas,might have played a significant role in the late Cenozoic cooling.Zehua Song Shiming Wan Christophe Colin Christian France-Lanord Zhaojie Yu Arnaud Dapoigny Hualong Jin Mengjun Li Jin Zhang Debo Zhao Xuefa Shi Anchun Li 2023Science Bulletin2023,,3:2

17Electrochemical detection of methyl-paraoxon based on bifunctional cerium oxide nanozyme with catalytic activity and signal amplification effect显示文摘A new electrochemical sensor for organophosphate pesticide(methyl-paraoxon)detection based on bifunctional cerium oxide(CeO_(2))nanozyme is here reported for the first time.Methyl-paraoxon was degraded into p-nitrophenol by using CeO_(2) with phosphatase mimicking activity.The CeO_(2) nanozymemodified electrode was then synthesized to detect p-nitrophenol.Cyclic voltammetry was applied to investigate the electrochemical behavior of the modified electrode,which indicates that the signal enhancement effect may attribute to the coating of CeO_(2) nanozyme.The current research also studied and discussed the main parameters affecting the analytical signal,including accumulation potential,accumulation time,and pH.Under the optimum conditions,the present method provided a wider linear range from 0.1 to 100 mmol/L for methyl-paraoxon with a detection limit of 0.06 mmol/L.To validate the proof of concept,the electrochemical sensor was then successfully applied for the determination of methyl-paraoxon in three herb samples,i.e.,Coix lacryma-jobi,Adenophora stricta and Semen nelumbinis.Our findings may provide new insights into the application of bifunctional nanozyme in electrochemical detection of organophosphorus pesticide.Yuzhou Sun Jinchao Wei Jian Zou Zehua Cheng Zhongming Huang Liqiang Gu Zhangfeng Zhong Shengliang Li Yitao Wang Peng Li 2021Journal of Pharmaceutical Analysis2021,11,5:1
18Bioinspired molecules design for bilateral synergistic passivation in buried interfaces of planar perovskite solar cells显示文摘Trap-mediated energy loss in the buried interface with non-exposed feature constitutes one of the serious challenges for achieving high-performance perovskite solar cells(PSCs).Inspired by the adhesion mechanism of mussels,herein,three catechol derivatives with functional Lewis base groups,namely 3,4-Dihydroxyphenylalanine(DOPA),3,4-Dihydroxyphenethylamine(DA)and 3-(3,4-Dihydroxyphenyl)propionic acid(DPPA),were strategically designed.These molecules as interfacial linkers are incorporated into the buried interface between perovskite and SnO_(2) surface,achieving bilateral synergetic passivation effect.The crosslinking can produce secondary bonding with the undercoordinated Pb^(2+) and Sn^(4+) defects.The PSCs treated with DOPA exhibited the best performance and operational stability.Upon the DOPA passivation,a stabilized power conversion efficiency(PCE)of 21.5%was demonstrated for the planar PSCs.After 55 days of room-temperature storage,the unencapsulated devices with the DOPA crosslinker could still maintain 85%of their initial performance in air under relative humidity of-15%.This work opens up a new strategy for passivating the buried interfaces of perovskite photovoltaics and also provides important insights into designing defect passivation agents for other perovskite optoelectronic devices,such as light-emitting diodes,photodetectors,and lasers.Bin Wang Junjie Ma Zehua Li Gangshu Chen Qiang Gu Shuyao Chen Yiqiang Zhang Yanlin Song Jingbo Chen Xiaodong Pi Xuegong Yu Deren Yang 2022Nano Research2022,15,2:1
