| 2 | Relationship between phenotypes of cell-function differentiation and pathobiological behavior of gastric carcinomas显示文摘AIM To reveal the correlation between thefunctional differentiation phenotypes of gastriccarcinoma cells and the invasion and metastasisby a new way of cell-function classification.METHODS Surgically resected specimens of361 gastric carcinomas (GC) were investigatedwith enzyme-, mucin-, and tumor-related markerimmunohistochemist ry. According to thedirection of cell-function differentiation,stomach carcinomas were divided into fivefunctionally differentiated types.iation type (AFDT): there were 82 (22.7%)patients including 76 (92.7%) aged 45 years.Sixty-nine (84.1%) cases belonged to theintestinal type. Thirty-eight (46.3%) expressedCD44v6 and 9 (13.6%) of 66 male patientsdeveloped liver metastasis. The 5-year survivalrate of patients in this group (58.5%) was higherMucin secreting function differentiation type(MSFDT): 54 (15%) cases. Fifty-three (98.1%)tumors had penetrated the serosa, 12 (22.2%)expressed ER and 22 (40.7%) expressedCD44v6. The postoperative 5-year survival ratefunction differentiation type (AMPFDT): therewere 180 (49.9%) cases, including 31 (17.2%)aged yanger than 45 years. The tumor was morecommon in women (62, 34.4%,) and expressedmore frequently estrogen receptors (ER) ( 129,81.7%) than other types (P<0.01). Ovarymetastasis was found in 12 (19.4%) out of 62female subjects. The patients with this type GChad the lowest 5-year survival rate (24.7%)differentiation type (SFDT): 13 (3.6%) cases.Nine (69.2%) tumors of this type derived fromAPUD system, the other 4 (30.7%) were ofdifferent histological differentiation. Sixty percent of the patients survived at least five years.(8.9%) cases. Nineteen (59.4%) cases hadlymph node metastases but no one with liver orovary metastasis. The 5-year survival rate was28.1%.CONCLUSION This new cell-functionclassification of GC is helpful in indicating thecharacteristics of invasion and metastasis of GCwith different cell-function differentiationphenotypes. | 3 | Multispherical interactions and their effects on the Tibetan Plateau's earth system: a review of the recent researches显示文摘The Tibetan Plateau(TP) is a regional Earth system showing very strong interactions among its lithosphere, hydrosphere, cryosphere, biosphere, atmosphere, and anthrosphere. hese interactions manifest TP's impact on surrounding regions and relect TP's response to the global change. Quantifying the multispherical interactions is critically important to understand the TP environment. Our recent years researches including the ongoing program entitled ‘Tibetan Multi-Spheres Interactions and heir Resource-Environment Signiicance(TIMI)', the completed program entitled. ‘Paleo-Altitudes of Tibetan Plateau and Environment(PATE)', as well as the other relating projects have focused on multidisciplinary research approaches and emphasized on three major pathways: Eurasia-Indian plates collision on deep-Earth dynamics, uplit impact on Earth's mantle–crust dynamics, and contemporary interface on land surface and atmospheric dynamics. Our researches have taken in situ measurement as priority and developed several platforms of data acquisition and analysis, including the platforms of water-phase transformations, and ecosystem observations. Our ield investigations have been conducted to obtain data about stratum, paleontology, paleoenvironment, genetic diferentiation of animals and plants. We have developed conceptual and mathematical models for crust uplit formation, paleoclimate, glacial melt,water–air interface lux, vegetation climate, and soil erosion. We have also assessed the anthropogenic impacts on environment. | 11 | Barometric altimetry system as virtual constellation applied in CAPS显示文摘This work describes the barometric altimetry as virtual constellation applied to the Chinese Area Positioning System (CAPS), which uses the transponders of communication satellites to transfer navigation messages to users. Barometric altimetry depends on the relationship of air pressure varying with altitude in the Earth’s atmosphere. Once the air pressure at a location is measured the site altitude can be found. This method is able to enhance and improve the availability of three-dimensional positioning. The difficulty is that the relation between barometric pressure and altitude is variable in different areas and under various weather conditions. Hence, in order to obtain higher accuracy, we need to acquire the real-time air pressure corresponding to an altimetric region’s reference height. On the other hand, the altimetry method will be applied to satellite navigation system, but the greatest difficulty lies in how to get the real-time air pressure value at the reference height in the broad areas overlaid by satellite navigation. We propose an innovational method to solve this problem. It is to collect the real-time air pressures and temperatures of the 1860 known-altitude weather observatories over China and around via satellite communication and to carry out time extrapolation forecast uniformly. To reduce data quantity, we first partition the data and encode them and then broadcast these information via navigation message to CAPS users’ receivers. Upon the interpolations being done in receivers, the reference air pressure and temperature at the receiver’s nearby place is derived. Lastly, combing with the receiver-observed real air pressure and temperature, the site’s altitude can be determined. The work is presented in the following aspects: the calculation principle, formulae, data collection, encoding, prediction, interpolation method, navigation message transmission together with errors causes and analyses. | 13 | Modeling soil conservation, water conservation and their tradeoffs: A case study in Beijing显示文摘Natural ecosystems provide society with important goods and services. With the rapid increase in human populations and excessive utilization of natural resources, humans frequently enhance the production of some services at the expense of the others. Although the need for tradeoffs between conservation and development is urgent, the lack of efficient methods to assess such tradeoffs has impeded progress. Three land use strategy scenarios (development scenario, plan trend scenario and conservation scenario) were created to forecast potential changes in ecosystem services from 2007 to 2050 in Beijing, China. GIS-based techniques were used to map spatial and temporal distribution and changes in ecosystem services for each scenario. The provision of ecosystem services differed spatially, with significant changes being associated with different scenarios. Scenario analysis of water yield (as average annual yield) and soil retention (as retention rate per unit area) for the period 2007 to 2050 indicated that the highest values for these parameters were predicted for the forest habitat under all three scenarios. Annual yield/retention of forest, shrub, and grassland ranked the highest in the conservation scenario. Total water yield and soil retention increased in the conservation scenario and declined dramatically in the other two scenarios, especially the development scenario. The conservation scenario was the optimal land use strategy, resulting in the highest soil retention and water yield. Our study suggests that the evaluation and visualization of ecosystem services can effectively assist in understanding the tradeoffs between conservation and development. | 15 | Biochar: A review of its impact on pesticide behavior in soil environments and its potential applications显示文摘Biochar is produced from the pyrolysis of carbon-rich plant- and animal-residues under low oxygen and high temperature conditions and has been increasingly used for its positive role in soil compartmentalization through activities such as carbon sequestration and improving soil quality. Biochar is also considered a unique adsorbent due to its high specific surface area and highly carbonaceous nature. Therefore, soil amendments with small amounts of biochar could result in higher adsorption and, consequently, decrease the bioavailability of contaminants to microbial communities, plants, earthworms, and other organisms in the soil. However, the mechanisms affecting the environmental fate and behavior of organic contaminants, especially pesticides in biochar-amended soil, are not well understood. The purpose of this work is to review the role of biochar in primary processes, such as adsorption–desorption and leaching of pesticides. | 16 | High-resolution records of Holocene from the Shuangchi Maar Lake in Hainan Island显示文摘The results of sedimentary characteristics, grain-size, clay mineral, C/N and pollen analysis reveal the evolution history of the Shuangchi Maar Lake of Hainan Island as well as the short-term climate variations in tropical areas. It is testified that the Maar sediment system was sensitive in recording the rapid climate changes in the Holocene. These multidisciplinary results demonstrate that the thermal maximum in the Holocene climate lasted from 7200 to 2700 aBP, with several abrupt ephemeral vibrations occurring between 7250-6120 aBP and at around 4460, 3850 and 2700 aBP. In the late Holocene, particularly at ~2700 aBP, a distinct environmental change indicating an important climate transition in the tropical eastern Asia was recorded, probably responding to the enhancement of ENSO phenomena in the Pacific Ocean. | 17 | Comparison of 5-hydroxytryptophan signaling pathway characteristics in diarrhea-predominant irritable bowel syndrome and ulcerative colitis显示文摘AIM: To study differences in the visceral sensitivity of the colonic mucosa between patients with diarrheapredominant irritable bowel syndrome(IBS-D) and those with ulcerative colitis(UC) in remission and to relate these differences with changes in the 5-hydroxytryptophan(5-HT) signaling pathway. METHODS: Gastrointestinal symptoms were used to determine the clinical symptom scores and rectal visceral sensitivity of patients with IBS-D and patients with UC in remission. Blood levels of 5-HT and5-hydroxyindoleacetic acid(5-HIAA) were measured using an HPLC-electrochemical detection system. The levels of 5-HT 3 receptor(3R), 4R, and 7R m RNAs in colonic biopsy samples were detected using reverse transcription-polymerase chain reaction. The protein expression of TPH1 was analyzed by Western blot and immunohistochemistry.RESULTS: Abdominal pain or discomfort, stool frequency, and the scores of these symptoms in combination with gastrointestinal symptoms were higher in the IBS-D and UC groups than in the control groups. However, no significant differences were observed between the IBS-D and UC remission groups. With respect to rectal visceral sensitivity, the UC remission and IBS-D groups showed a decrease in the initial perception threshold, defecating threshold and pain threshold. However, these groups exhibited significantly increased anorectal relaxation pressure. Tests examining the main indicators of the 5-HT signaling pathway showed that the plasma 5-HT levels, 5-HIAA concentrations, TPH1 expression in the colonic mucosa, and 5-HT3 R and 5-HT5 R expression were increased in both the IBS-D and the UC remission groups; no increases were observed with respect to 5-HT7 R expression.CONCLUSION: The IBS-D and UC groups showed similar clinical symptom scores, visceral sensitivity, and levels of serotonin signaling pathway indicators in the plasma and colonic mucosa. However, the pain threshold and 5-HT7 R expression in the colonic mucosa were significantly different between these groups. | 18 | Response of runoff and soil loss to reforestation and rainfall type in red soil region of southern China显示文摘To evaluate the long-term effects of reforestation types on soil erosion on degraded land, vegetation and soil properties under conventional sloping farmland (CSF) and three different reforestation types including a Pinus massoniana secondary forest (PSF), an Eucommia ulmoides artificial economic forest (EEF) and a natural succession type forest (NST), were investigated at runoff plot scale over a six-year period in a red soil region of southern China. One hundred and thirty erosive rainfall events generating runoff in plots were grouped into four rainfall types by means of K-mean clustering method. Erosive rainfall type I is the dominant rainfall type. The amount of runoff and the soil loss under erosive rainfall type III were the most, followed by rain-fall type II, IV and I. Compared with CSF treatment, reforestation treatments decreased the average annual runoff depth and the soil loss by 25.5%-61.8% and 93.9%- 96.2% during the study period respectively. Meanwhile, runoff depth at PSF and EEF treatments was significantly lower than that in NST treatment, but no significant difference existed in soil erosion modulus among the three reforestation treatments. This is mainly due to the improved vegetation properties (i.e., vegetation coverage, biomass of above- and below-ground and litter-fall mass) and soil properties (i.e., bulk density, total porosity, infiltration rate and organic carbon content) in the three reforestation treatments compared to CSF treatment. | 19 | Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. | 20 | Effects of baicalin in CD4 + CD29 + T cell subsets of ulcerative colitis patients显示文摘AIM:To evaluate the role of baicalin in ulcerative colitis(UC) with regard to the CD4+CD29+ T helper cell,its surface markers and serum inflammatory cytokines.METHODS:Flow cytometry was used to detect the percentage of CD4+CD29+ cells in patients with UC.Real time polymerase chain reaction was used to detect expression of GATA-3,forkhead box P3,T-box expressed in T cells(T-bet),and retinoic acid-related orphan nuclear hormone receptor C(RORC).Western blotting was used to analyze expression of nuclear factor-κB(NF-κB) p65,phosphorylation of NF-κB(p-NF-κB) p65,STAT4,p-STAT4,STAT6 and p-STAT6.The concentrations of interferon-γ(IFN-γ),interleukin(IL)-4,IL-5,IL-6,IL-10 and TGF-β in serum were determined by ELISA assay.RESULTS:The percentages of CD4+CD29+ T cells were lower in treatment with 40 and 20 μmol/L baicalin than in the treatment of no baicalin.Treatment with 40 or 20 μmol/L baicalin significantly upregulated expression of IL-4,TGF-β1 and IL-10,increased p-STAT6/STAT6 ratio,but downregulated expression of IFN-γ,IL-5,IL-6,RORC,Foxp3 and T-bet,and decreased ratios of T-bet/GATA-3,p-STAT4/STAT4 and p-NF-κB/NF-κB compared to the treatment of no baicalin.CONCLUSION:The results indicate that baicalin regulates immune balance and relieves the ulcerative colitis-induced inflammation reaction by promoting proliferation of CD4+CD29+ cells and modulating immunosuppressive pathways. | Feng-Yan Yu Shao-Gang Huang Hai-Yan Zhang Hua Ye Hong-Gang Chi Ying Zou Ru-Xi Lv Xue-Bao Zheng | 2014 | World Journal of Gastroenterology2014,20,41: | 18 |
