1Ultrasound-associated extraction of seed oil of Korean pine显示文摘Experiment on ultrasound- associated extraction of seed oil of Korean pine (Pinus koraiensis) was conducted in Northeast Forestry University, Harbin, China. The factors affecting extraction yield, such as ultrasonic frequency, extracting temperature, extracting time and the ratio of material to liquid (ratio of Korean pine seed to absolute alcohol), were analyzed under specific condition and the optimal extracting parameters were obtained as the ultrasonic frequency 32 000 Hz, the extracting temperature 80℃, the extracting time 50 rain, and the ratio of material to liquid 1: 30. 2N-AcetyI-L-cysteine and pyrrolidine dithiocarbamate inhibited nuclear factor-κB activation in alveolar macrophages by different mechanisms显示文摘Aim:To study the effects of N-acetyl-L-cysteine(NAC)and pyrrolidine dithio-carbamate(PDTC)on the phosphorylation of IκB kinase(IKK)β,IKKα,and IλBαin alveolar macrophages(AM),and to explore the pharmacological mechanisms ofNAC and PDTC as inhibitors of NF-κB activation.Methods:AM were collectedfrom bronchoalveolar lavage fluid from the patients with chronic obstructive pul-monary disease.The AM were incubated for 1.5 h with NAC and PDTC,and thenstimulated for 90 min by either tumor necrosis factor(TNF)-α or interleukin(IL)-1.Western blotting was used to detect the protein phosphorylation levels of IKKβ,IKKα,and IκBα.NF-κB activity was analyzed by using an electrophoretic mobil-ity shift assay.Results:NAC inhibited the phosphorylation of IKKβ,IKKα,andIκBα induced by TNF-α,but had no effect on the phosphorylation of IKKβ,IKKαand IκBα induced by IL-1.PDTC did not inhibit the phosphorylation of IκBαinduced by TNF-α or IL-1.Similarly,NAC inhibited the activation of NF-κBinduced by TNF-α,but had no effect on the activation of NF-κB induced by IL-1.PDTC significantly inhibited the activation of NF-κB induced by TNF-α and IL-1.The electrophoretic mobility shift assay also showed that PDTC and NAC do notdirectly inhibit NF-κB DNA binding activity in vitro.Conclusion:PDTC preventsthe degradation of IκBα via the ubiquitylation-proteasome proteolytic pathway.NAC can inhibit the processes upstream of IKK activation induced by TNF-α,which results in the decline of NF-κB activity.Ya-qing LI Zhen-xiang ZHANGI Yong-jian XU Wang NI Shi-xin CHEN Zhao YANG Dan MA 2006Acta Pharmacologica Sinica2006,27,3:29
3A Genetic Map Constructed Using a Doubled Haploid Population Derived from Two Elite Chinese Common Wheat Varieties显示文摘Genetic mapping provides a powerful tool for the analysis of quantitative trait loci (QTLs) at the genomic level. Herein, we report a new genetic linkage map developed from an F1-derived doubled haploid (DH) population of 168 lines, which was generated from the cross between two elite Chinese common wheat (Triticum aestivum L.) varieties, Huapei 3 and Yumai 57. The map contained 305 loci, represented by 283 simple sequence repeat (SSR) and 22 expressed sequence tag (EST)-SSR markers, which covered a total length of 2141.7 cM with an average distance of 7.02 cM between adjacent markers on the map. The chromosomal locations and map positions of 22 new SSR markers were determined, and were found to distribute on 14 linkage groups. Twenty SSR loci showed different chromosomal locations from those reported in other maps. Therefore, this map offers new information on the SSR markers of wheat. This genetic map provides new opportunities to detect and map QTLs controlling agronomically important traits. 4Inhibiting the expression of CD28 costimulatory molecule on human lymphocytes by special siRNA显示文摘Background The B7/CD28 pathway provides critical costimulatory signals for complete T cell activation, and members of this pathway have served as useful targets for immunotherapeutic strategies. In this study, we investigated the RNA interference (RNAi) effect induced by small interfering RNA (siRNA) targeting CD28 mRNA on human lymphocytes and its specificity.Methods According to CD28 gene sequence, we designed and synthysized three different siRNAs ( siRNA-1,siRNA-2, siRNA-3 ) containing 21 bases using SilencerTM siRNA construction kit. These siRNAs were transfected into freshly isolated human lymphocytes with Lipofectamine 2000 reagent. At 24-hour, 48-hour and 72-hour post transfection, these cells were collected and analyzed. The changes of surface expression of CD28 gene were detected by flow cytometry, and the changes of CD28 mRNA levels were determined by semiquantitative reverse transcription polymerase chain reaction (RT-PCR). The cell viability of transfected lymphocytes was determined by methyl thiazolyl tetrazolium (MTT) assay and trypan blue dye exclusion assay.Results Three siRNAs (siRNA-1, siRNA-2, siRNA-3) specifically targeting CD28 mRNA were successfully designed and constructed. Flow cytometry analysis showed that a decrease in CD28 expression was detectable at 24-hour post transfection. Different siRNA showed different inhibition effects on CD28 expression. At 48-hour post transfection, the degrees of reduction with siRNA-1, siRNA-2 and siRNA-3 were 22. 10% ± 1.63% ,73.50% ± 1.02% and 42.90% ± 0.89% respectively compared with the control ( P < 0. 001 ). Neither of the groups transfected only with siRNA or lipo showed marked reduction in CD28 expression (3.15% ± 0.75% and 4. 55% ±0. 80% ) (P >0. 05). Moreover, lymphocytes treated with siRNA-co showed no marked reduction in CD28 expression (5.07% ± 0. 96% ) (P > 0. 05 ). The results of semi-quantitative RT-PCR assay indicated CD28 mRNA level was inhibited after transfection of specific siRNAs. At least 4-fold of reduction in siRNA-2 group occurred at 48-hour post transfection compared with the control (P < 0. 001 ). MTT assay and trypan blue dye exclusion assay demonstrated that the viable cell rations of transfected lymphocytes were significantly reduced in siRNA-1, siRNA-2 and siRNA-3 groups at 48-hour post transfection (P <0.01 ). The control groups showed no marked reduction in cell viability ( P > 0.05 ).Conclusions Three different siRNAs were synthesized and transfected into lymphocytes. They could reduce the expression of CD28 and the CD28 mRNA level, siRNA-2 was the most efficient. The cell viability reduced correspondingly. Therefore, the silencing effect on CD28 mRNA induced by siRNA may contribute to costimulatory blockade. 5Synthesis of a new meso/microporous composite molecular sieve of MCM-41/mordenite显示文摘An MCM-41/mordenite composite with two- fold porous structure and stepwise-distributed acidity was prepared for the first time by using zeolite mordenite as the silica-alumina source. The composite molecular sieve has been investigated and compared with a mechanical mixture of mordenite and MCM-41 for their structure, acidity and catalytic activity by means of XRD, N2 adsorption and desorption, HRTEM, DTG, NH3-TPD and catalytic reaction. The characterization results show that the structure and property of the composite molecular sieve are quite different from those of the mechanical mixture, which might be ascribed to the incorporation of secondary building units characteristic of zeolite mordenite into the mesoporous walls of the composite which gives rise to the thicker mesoporous walls, the higher hydrothermal stability and more strong acid sites. 8Estimation of stature by using the dimensions of the right hand and right foot in Han Chinese adults显示文摘The Han Chinese people are the main ethnic group in China and the largest ethnic group in the world.The dimensions of the hands and feet have been successfully used for the estimation of stature.A total of 26,927 healthy adult subjects,comprising 13,221 men and 13,706 women,were recruited.The survey samples were chosen through random cluster sampling.The mean values were significantly higher in men than those in women for all measurements(P<0.001).All the measurements showed a statistically significant correlation with stature(P<0.01).The combination of the right hand length and the right foot length was the best predictor of stature because it had the lowest standard error of estimate.The use of multiple regression equations yielded better results than did the use of linear regression equations.The accuracy of stature prediction ranged from ±4.81 to ±6.39 cm.The present study was of great importance with regards to improving the physical anthropology database of ethnic groups in China.Xinghua Zhangi Yu Wei Lianbin Zhengi Keli Yu Dapeng Zhao Jinping Bao Yonglan Li Shunhua Lu Huanjiu Xi Guochang Xu Youfeng Wen 2017Science China(Life Sciences)2017,60,1:4

16Studies of Composition and Structure of CO_2 Corrosion Film of X56 Steel in the Medium of CO_2 and Salt Water显示文摘The X56 steel samples was corroded in the medium of salt water solution at the conditions of CO2 partial pressure POM 0.5 to 2.0 MPa, temperature 80 °C and flow rate 1.4m/s. Corrosion weigh loss, composition and structure, morphology and phase of corrosion films of the samples were investigate by SEM, EDS, XRD and XPS. The results indicated that the corrosion degree was accelerated with increasing POM ? The intense localised corrosion occurred on the surface of samples. The corrosion films mainly comprised of FeCO3 and complex phase products (Fe, Ca....)COj. There exists serious pitting on the metal substrates under the corrosion film. 17Genome-wide identification, evolution, and expression analysis of RNA-binding glycine-rich protein family in maize显示文摘The RNA‐binding glycine‐rich protein(RB‐GRP)family is characterized by the presence of a glycine‐rich domain arranged in(Gly)n‐X repeats and an RNA‐recognition motif(RRM). RB‐GRPs participate in varied physiological and biochemical processes especially in the stress response of plants. In this study, a total of 23 RB‐GRPs distributed on 10 chromosomes were identified in maize(Zea mays L.), and they were divided into four subgroups according to their conserved domain architecture. Five pairs of paralogs were identified,while none of them was located on the same chromosomal region, suggesting that segmental duplication is predominant in the duplication events of the RB‐GRPs in maize. Comparative analysis of RB‐GRPs in maize, Arabidopsis(Arabidopsis thaliana L.), rice(Oryza sativa L.), and wheat(Triticum aestivum)revealed that two exclusive subgroups were only identified in maize. Expression of eight ZmRB‐GRPs was significantly regulated by at least two kinds of stresses. In addition, cis‐elements predicted in the promoter regions of the ZmRB‐GRPs also indicated that these ZmRB‐GRPs would be involved in stress response of maize. 18Genome sequencing and characterization analysis of a Beijing isolate of chicken coronavirus infectious bronchitis virus显示文摘Avian infectious bronchitis virus (AIBV) is classified as a member of the genus coronavirus in the family coronaviridae. The enveloped virus has a positive-sense, sin-gle-stranded RNA genome of approximately 28 kilo-bases, which has a 5′ cap structure and 3′ polyadenylation tract. The complete genome sequence of infectious bronchitis virus (IBV), Beijing isolate, was determined by cloning sequencing and primer walking. The whole genome is 27733 nucleotides in length, has ten open reading frames: 5′-orf1a-orf1ab-s-3a- 3b-e-m- 6a-6b-n-3′. Alignments of the genome sequence of IBV Beijing isolate with those of two AIBV strains and one SARS coronavirus were performed respectively. The genome sequence of IBV Beijing isolate compared with that of the IBV strain LX4 (uncompleted, 19440 bp in size) was 91.2% similarity. However, the full-length genome sequence of IBV Beijing isolate was 85.2% identity to that of IBV Strain Beaudette, and was only 50.8% homology to that of SARS coronavirus. The results showed that the genome of IBV has remarkable variation. And IBV Beijing isolate is not closely related to SARS coronavirus. Phylogenetic analyses based on the whole genome sequence, S protein, M protein and N pro-tein, also showed that AIBV Beijing isolate is lone virus in group Ⅲ and is distant from SARS coronavirus. 19Nuclear translocation of OsMFT1 that is impeded by OsFTIP1 promotes drought toleranee in rice显示文摘Drought is the leading environmental threat affecting crop productivity,and plants have evolved a series of mechanisms to adapt to drought stress.The FT-interacting proteins(FTIPs)and phosphatidylethanola mine.binding proteins(PEBPs)play key roles in developmental processes,whereas their roles in the regulation of stress response are still largely unknown.Here,we report that OsFTIP1 negatively regulates drought response in rice.We showed that OsFTIP1 interacts with rice MOTHER OF FT AND TFL1(OsMFT1),a PEBP that promotes rice tolerance to drought treatment.Further studies discovered that OsMFT1 interacts with two key drought-related transcription factors,OsbZIP66 and OsMYB26,regulating their binding capacity on drought-related genes and thereby enhancing drought toleranee in rice.Interestingly,we found that OsFTIP1 impedes the nucleocytoplasmic translocation of OsMFT1,implying that dynamic modulation of drought-responsive genes by the OsMFT1-OsMYB26 and OsMFT1-OsbZIP66 complexes is integral to OsFTIP1-modulated nuclear accumulation of OsMFT1.Our findings also suggest that OsMFT1 might act as a hitherto unknown nucleocytoplasmic trafficking signal that regulates drought tolerance in rice in response to environmental signals.Ying Chen Jun Shen Liang Zhangi Haoyue Qi Lijia Yang Huanyu Wang Jiaxuan Wang Yuexing Wang Hao Du Zeng Tao Ting Zhao Pingchuan Deng Qingyao Shu Qian Qian Hao Yu Shiyong Song 2021Molecular Plant2021,14,8:2
