| 1 | Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. | 3 | Mechanical properties and enhancement mechanisms of titanium-graphene nanocomposites显示文摘Molecular dynamics(MD)simulations of the titanium-graphene nanocomposites(TiGNCs)under uniaxial tension are carried out to investigate the mechanical properties and reinforcement mechanism of graphene in composites.It is found that introduction of mechanically robust graphene limits the strain-induced dislocation and araorphization and thereby highly improves the mechanical properties of metallic titanium that are greatly affected by the crystal stacking orientation of graphene and titanium layers.The thickness of titanium layers,interface interaction and working temperature play an important role in the mechanical strength and elastic moduli of composites.The results show the mechanical properties of TiGNCs are monotonically enhanced with reduction of the titanium layer thickness and working temperature,and the Young5s modulus obtained by MD simulation are higher than that predicted by the rule of mixture(ROM)due to consideration of interfacial interaction in computational calculation.In addition,once the critical thickness of titanium layer is reached,graphene wrinkles are induced in composites because of Poisson's effect induced large lateral compression stress in the interface region.This study provides helpful insights into fundamental understanding reinforcing mechanism of graphene and ultimately contribute to the optimal design and performance of mechanically robust graphene-based metallic composites. | Wanhong Tang Jie Zhang Jianyang Wu Jinyou Shao Peng Ding Guozhen Hou Xiaoming Chen | 2020 | Acta Mechanica Sinica2020,36,4: | 7 |
| 4 | Preparation and evaluation of a new releasable PEGylated tumor necrosis factor-α(TNF-α) conjugate for therapeutic application显示文摘To design a releasable PEGylated TNF-α(rPEG-TNF-α ), a cathepsin B-sensitive dipeptide (Val-Cit moiety) was inserted into conventional PEG-modified TNF- (PEG-TNF- ), facilitating its clinical use for anti-tumor therapy. Comparative pharmaco- kinetic and pharmacodynamic studies showed that the half-lives of both PEGylated forms of TNF-α were ~60-fold greater than that of unmodified TNF-α . In addition, the in vitro bioactivity of rPEG-TNF-α was greater than that of PEG-TNF-α with the same degree of PEG modification. Release of TNF-α from rPEG-TNF-α in vitro was dependent on the presence of cathepsin B and was inhibited by a cathepsin B inhibitor. Despite the potent cytotoxicity of unmodified TNF-α against normal cells, its PEGylated forms at higher TNF-α concentrations showed low cytotoxic activity against these cells. In contrast, both forms of PEGylated TNF-α showed potent cytotoxic activity against the B16 and L929 cell lines, with rPEG-TNF-α being 5- and 9- fold more potent, respectively, than PEG-TNF-α . | 5 | The long non-coding RNA expression profile of Coxsackievirus A16 infected RD cells identified by RNA-seq显示文摘Coxsackievirus A16(CVA16) is one of major pathogens of hand, foot and mouth disease(HFMD) in children. Long non-coding RNAs(Inc RNAs) have been implicated in various biological processes,but they have not been associated with CVA16 infection. In this study, we comprehensively characterized the landscape of Inc RNAs of normal and CVA16 infected rhabdomyosarcoma(RD)cells using RNA-Seq to investigate the functional relevance of Inc RNAs. We showed that a total of 760 Inc RNAs were upregulated and 1210 Inc RNAs were downregulated. Out of these dysregulated Inc RNAs, 43.64% were intergenic, 22.31% were sense, 15.89% were intronic, 8.67% were bidirectional, 5.59% were antisense, 3.85% were s RNA host Inc RNAs and 0.05% were enhancer. Six dysregulated Inc RNAs were validated by quantitative PCR assays and the secondary structures of these Inc RNAs were projected. Moreover, we conducted a bioinformatics analysis of an Inc RNAs(ENST00000602478) to elucidate the diversity of modification and functions of Inc RNAs. In summary, the current study compared the dysregulated Inc RNAs profile upon CVA16 challenge and illustrated the intricate relationship between coding and Inc RNAs transcripts. | 11 | Volatile organic compound emissions from straw-amended agricultural soils and their relations to bacterial communities:A laboratory study显示文摘A laboratory study was conducted to investigate volatile organic compound(VOC) emissions from agricultural soil amended with wheat straw and their associations with bacterial communities for a period of 66 days under non-flooded and flooded conditions. The results indicated that ethene, propene, ethanol, i-propanol, 2-butanol, acetaldehyde, acetone,2-butanone, 2-pentanone and acetophenone were the 10 most abundant VOCs, making up over 90% of the total VOCs released under the two water conditions. The mean emission of total VOCs from the amended soils under the non-flooded condition(5924 ng C/(kg·hr)) was significantly higher than that under the flooded condition(2211 ng C/(kg·hr)). One 'peak emission window' appeared at days 0–44 or 4–44, and over 95% of the VOC emissions occurred during the first month under the two water conditions. Bacterial community analysis using denaturing gradient gel electrophoresis(DGGE) showed that a relative increase of Actinobacteria, Bacteroidetes, Firmicutes and γ-Proteobacteria but a relative decrease of Acidobacteria with time were observed after straw amendments under the two water conditions. Cluster analysis revealed that the soil bacterial communities changed greatly with incubation time, which was in line with the variation of the VOC emissions over the experimental period. Most of the above top 10 VOCs correlated positively with the predominant bacterial species of Bacteroidetes, Firmicutes and Verrucomicrobia but correlated negatively with the dominant bacterial species of Actinobacteria under the two water conditions.
