1Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. 4Influence of Al addition on solidification path and hot tearing susceptibility of Mg-2Zn-(3+0.5 x)Y-x Al alloys显示文摘Hot tearing susceptibility(HTS)of Mg-2Zn-(3+0.5 x)Y-x Al(x=0,2 and 3 at%)alloys is predicted by using modified Clyne-Davies’model(CSC^(∗)).The solidification path,solidification characteristic temperatures and dendritic coherency solid fraction have been studied by double-thermocouple thermal analysis.The solidification contraction stress vs.temperature(and time)curves are measured by using a“T”type hot tearing permanent-mold.The results reveal that the CSC^(∗)prediction values are in good agreement with the experimental results.Moreover,Al_(2)Y phase acts as the heterogeneous nucleation core ofα-Mg and significantly influences the grain size.It has been observed that minimum grain size,optimal dendritic coherency and minimum HTS are exhibited by Mg-2Zn-(3+0.5 x)Y-x Al alloy(x=2).Furthermore,when Al content was increased to 3 at%,Al_(2)Y phase exhibited a peritectic reaction and transformed into a mixed structure of Al_(2)Y and Al+Al_(3)Y phases,which increased the HTS of the alloy due to reduced fine-grained Al_(2)Y content.Guojia Zhang Yue Wang Zheng Liu Shimeng Liu 2019Journal of Magnesium and Alloys2019,7,2:4
5p53-dependent upregulation of PIG3 transcription by γ-ray irradiation and its interaction with KAP1 in responding to DNA damage显示文摘PIG3 (p53-inducible gene 3), originally identified as one of a set of genes induced by p53 before the onset of apoptosis, was assumed to contribute to early cellular response to DNA damage. Here, we studied the relation between p53 status and the increased expression of PIG3 by ionizing radiation (IR), and the related clues regarding the involvement of PIG3 in the cellular response to IR-induced DNA damage signaling. We demonstrated that the pentanucleotide microsatellite sequence was responsible for the p53-dependent induction of PIG3 transcription after irradiation, while sequence upstream of PIG3 promoter could maintain the basal level of expression which was not inducible by irradiation. The interaction of PIG3 and the KRAB-ZFP-associated protein 1 (KAP1), a DNA damage response protein, was revealed. PIG3 nucleus foci were formed 15 min after γ-ray irradiation, and which were found to partially colocalize with the phospho-KAP-1 foci as well as γ-H2AX foci. Although the lac operator tagged EGFP based reporter system revealed that PIG3 does not remodel chromatin in large scale in the cells under normal growing condition, it indeed prompted the chromatin relaxation in the cellular response to DNA damage signaling. 6Dynamic changes of postprandial plasma metabolites after intake of corn-soybean meal or casein-starch diets in growing pigs显示文摘Background: Postprandial nutrients utilization and metabolism of a certain diet is a complicated process. The metabolic feature of pigs after intake of corn-soybean meal or casein-starch diets are largely unknown. Therefore,this study was conducted to investigate the dynamic postprandial changes of plasma metabolic profile using growing pigs using metabolomics.Methods: Twenty-four growing pigs with average initial body weight(BW) about 30 kg were placed in metabolic cages and then fitted with precaval vein catheters. Pigs were fed daily 4% of initial body weight. Two experimental diets were included:(i) a starch-casein based purified diet(PD) and(ii) a common corn-soybean meal diet(CD).Plasma was collected before feeding and 0.5 h, 1 h, 2 h, 4 h, 8 h after feeding.Results: In both diets, compared to prior to feeding, the concentrations of glucose, most amino acids, metabolites such as 5-aminopentanoic acid, pipecolic acid, ornithine and 5-hydroxy-L-tryptophan were significantly increased in plasma during the first hour, whereas the concentrations of plasma triglycerides, glutamate, glycine, palmitelaidic acid, 13-HODE and oleic acid were decreased in the first hour. Compared with PD group, concentration of plasma leucine and isoleucine declined at 30 min in CD group. Plasma linoleic acid, sphingosine and many dipeptides were significantly higher in pigs fed CD.Conclusion: Most significant metabolic changes occurred during the first hour after feeding and then became relatively stable after 2 h in both diets. 8Common genetic heterogeneity of human interleukin-37 leads to functional variance显示文摘Interleukin-37(IL-37)is an inhibitory member of the IL-1 family of cytokines.We previously found that balanced selection maintains common variations of the human IL37 gene.However,the functional consequences of this selection have yet to be validated.Here,using cells expressing exogenous IL-37 variants,including IL-37 Ref and IL-37 Var1 and Var2,we found that the three variants of IL-37 exhibited different immunoregulatory potencies in response to immune stimulation.The protein level of IL-37 Var2 was found to be significantly less than that of IL-37 Ref or Var1,despite the comparable mRNA levels of all three variants.Further study showed that IL-37 Var2 was rapidly degraded by a proteasome-dependent mechanism mediated by enhanced polyubiquitination,leading to a transient upregulation of IL-37 Var2 after immune stimulation.Finally,when ectopically expressed in cells,human IL-37 Var2 exerted less inhibition on proinflammatory cytokine production than did other IL-37 variants.Conversely,purified extracellular IL-37 variant proteins demonstrated comparable inhibitory abilities in vitro.In conclusion,our study reveals that common genetic variants of IL37 lead to different immune-inhibitory potencies,primarily as a result of differences in IL-37 protein stability,suggesting the possible involvement of these variants in various human diseases.Jingjing Yan Yuling Zhang Shimeng Cheng Bin Kang Jinbiao Peng Xiaodan Zhang Meichun Yuan Wenqi Chu Wen Zhang Jiayin Shen Shuye Zhang 2017Cellular & Molecular Immunology2017,14,9:1
9Optimizing the electromagnetic wave absorption performance of designed hollow CoFe_(2)O_(4)/CoFe@C microspheres显示文摘Whereas hollow composites present some superiorities like abundant micro interfaces,outstanding impedance matching as the responses of electromagnetic wave(EMW),but versatile designs including crystal transformation,heterogeneous structures and magnetic exchange coupling to further contribution are even not designed or stressed together in previous literatures.In this article,rational design on the hollow CoFe_(2)O_(4)/CoFe@C architecture has been conducted by a sequential process of self-sacrifice by combustion,in-suit polymerization and calcination.Results of morphology observation exhibit that heterogeneous CoFe_(2)O_(4)/CoFe@C composites were generated via crystal transformation from CoFe_(2)O_(4) to CoFe alloys with encapsulated carbon,together with ultimate growth of crystal particles.As for three carbon-based architectures,relatively low-graphitization carbon layers are favorable for enhancing impedance matching and polarization relaxation,but suppressing the conductive loss essentially.Moderate carbon content endows sample S2 with the maximum magnetic saturation(Ms)of 152.4 emu g^(-1).The optimized RL of sample S3 is up to-51 dB with 30 wt%loading,and the effective absorption band(EAB)is of 5.9 GHz at the thickness of 2.17 mm,while 6.0 GHz can be reached at 2.5 mm.Therefore,this hollow multi-interfaces design definitely shed light on novel structure for new excellent absorbers.Jianwen Ge Shimeng Liu Li Liu Yu Cui Fandi Meng Yixing Li Xuefeng Zhang Fuhui Wang 2021Journal of Materials Science & Technology2021,,22:1
10Perilipin 2 and lipid droplets provide reciprocal stabilization显示文摘The lipid droplet (LD)-associated protein adipose differentiation-related protein (ADRP or PLIN2) is required for the formation and stability of the LD organelle, whereas its biological roles are still obscure. Herein, we show that PLIN2 is the most abundant protein on the lipid droplets (LDs) of mouse myoblast cell line C2C12. Both the expression of PLIN2 and the accumulation of LDs were up-regulated in a time- and dose-dependent manner when the cells were treated with oleate (OA). The protein level of PLIN2 was positively correlated with the formation of LDs, suggesting that LDs stabilize PLIN2. Furthermore, knocking out PLIN2 in C2C12 cells led to enlarged LDs and higher triacylglycerol hydrolysis activity. The isolated PLIN2 null LDs became closely contact w让h mitochondria and other cellular organelles. Additionally mitochondrial activity was suppressed by OA in PLIN2 null cells. 11Output Prediction of Helical Microfiber Temperature Sensors in Cycling Measurement by Deep Learning显示文摘The inconsistent response curve of delicate micro/nanofiber(MNF)sensors during cycling measurement is one of the main factors which greatly limit their practical application.In this paper,we proposed a temperature sensor based on the copper rod-supported helical microfiber(HMF).The HMF sensors exhibited different light intensity-temperature response relationships in single-cycle measurements.Two neural networks,the deep belief network(DBN)and the backpropagation neural network(BPNN),were employed respectively to predict the temperature of the HMF sensor in different sensing processes.The input variables of the network were the sensor geometric parameters(the microfiber diameter,wrapped length,coiled turns,and helical angle)and the output optical intensity under different working processes.The root mean square error(RMSE)and Pearson correlation coefficient(R)were used to evaluate the predictive ability of the networks.The DBN with two restricted Boltzmann machines(RBMs)provided the best temperature prediction results(RMSE and R of the heating process are 0.9705℃and 0.9969,while the values of RMSE and R of the cooling process are 0.7866℃and 0.9977,respectively).The prediction results obtained by the optimal BPNN(five hidden layers,10 neurons in each layer,RMSE=1.1266℃,R=0.9957)were slightly inferior to those obtained by the DBN.The neural network could accurately and reliably predict the response of the HMF sensor in cycling operation,which provided the possibility for the flexible application of the complex MNF sensor in a wide sensing range.Minghui CHEN Jinjin HAN Juan LIU Fangzhu ZHENG Shihang GENG Shimeng TANG Zhijun WU Jixiong PU Xining ZHANG Hao DAI 2023Photonic Sensors2023,13,3:0
12Preparation of V2O5 Porous Hollow Spheres and Study on Excellent Cycling Stability of Lithium Battery显示文摘Through one-step simple polyol-assisted (ethylene glycol) process, V2O5 hollow mi-cro-spheres about 3 micrometres in size were successfully synthesized. The structure is neat, the outer wall were porous. Testing its electrochemical properties with V2O5 hollow materials as cathode materia of lithium ion battery, the results showed that under the ratio of 1 C, the initial charge and discharge specific capacity were 236.8 mAh g^-1, 213 mAh g^-1, even if after 100 cycles, the charge and discharge specific capacity were still 220 mAh g^-1 and 219.7 mAh g^-1 respectively. 14Targeting cAMP in D1-MSNs in the nucleus accumbens, a new rapid antidepressant strategy显示文摘Studies have suggested that the nucleus accumbens(NAc)is implicated in the pathophysiology of major depression;however,the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been elucidated.Here,we identified a specific reduction of cyclic adenosine monophosphate(cAMP)in the subset of dopamine D1 receptor medium spiny neurons(D1-MSNs)in the NAc that promoted stress susceptibility,while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like behaviors.Ketamine treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons(D2-MSNs)of depressed mice,however,the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc D1-MSNs.We discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration,but not a lower dose.The fast onset property of crocin was verified through multicenter studies.Moreover,crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on D2-MSN.These findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc,and provide a potential rapid antidepressant drug candidate,crocin.Yue Zhang Jingwen Gao Na Li Peng Xu Shimeng Qu Jinqian Cheng Mingrui Wang Xueru Li Yaheng Song Fan Xiao Xinyu Yang Jihong Liu Hao Hong Ronghao Mu Xiaotian Li Youmei Wang Hui Xu Yuan Xie Tianming Gao Guangji Wang Jiye Aa 2024Acta Pharmaceutica Sinica B2024,14,2:0

16Nutritional Value Evaluation of Saccharum spontaneum L. Germplasm Resources显示文摘The contents of dry matter, crude fat, crude protein, neutral detergent fiber, acid detergent fiber, crude ash, Ca and P in 43 Saccharum spontaneum L.germplasms during the vegetative period were determined. Among them, the crude protein, crude fat, neutral detergent fiber, acid detergent fiber, P and Ca contents were treated as the judging indicators. The nutritional value evaluation was carried out with the analytic hierarchy process(AHP). The results showed A49(Guangxi)had the highest nutritional value. Its dry matter, crude protein, crude fat, crude ash,neutral detergent fiber, acid detergent fiber, Ca and P contents were 32.35%,7.20%, 4.06%, 8.07%, 75.81%, 50.72%, 0.23% and 0.16% respectively. While A3(Guangdong) had the lowest nutritional value. Its dry matter, crude protein, crude fat, crude ash, neutral detergent fiber, acid detergent fiber, Ca and P contents were32.24%, 4.62%, 0.51%, 6.38%, 46.40%, 40.02%, 0.15% and 0.09% respectively.The crude protein content differed significantly among different germplasms. A48 had the highest crude protein content(9.11%), and A14 had the lowest crude protein content(3.72%). Based on the evaluation results, the 43 Saccharum spontaneum L.germplasms were divided into 3 groups: high-nutritional value type, moderate-nutritional value type and low-nutritional value type. 17Hierarchical carbon nanosheet confined defective MoS_(x) cathode towards long-cycling zinc-ion-battery显示文摘Aqueous zinc-ion batteries(ZIBs)have attracted increasing attention due to their low cost and high safety.MoS_(2) is a promising cathode material for aqueous ZIBs due to its favorable Zn^(2+)accommodation ability.However,the structural strain and large volume changes during intercalation/deintercalation lead to exfoliation of active materials from substrate and cause irreversible capacity fading.In this work,a highly stable cathode was developed by designing a hierarchical carbon nanosheet-confined defective MoS_(x)material(CNS@MoS_(x)).This cathode material exhibits an excellent cycling stability with high capacity retention of 88.3%and~100%Coulombic efficiency after 400 cycles at 1.2 A·g^(-1),much superior compared to bare MoS_(2).Density functional theory(DFT)calculations combined with experiments illustrate that the promising electrochemical properties of CNS@MoS_(x)are due to the unique porous conductive structure of CNS with abundant active sites to anchor MoS_(x)via strong chemical bonding,enabling MoS_(x)to be firmly confined on the substrate.Moreover,this unique hierarchical complex structure ensures the fast migration of Zn^(2+)within MoS_(x)interlayer.Xiaoqi Wang Shimeng Zhang Rui Yang Shengchi Bai Jianbo Li Yu Wu Bowen Jin Xu Jin Mingfei Shao Bo Wang 2023Nano Research2023,16,7:0
18Zinc finger protein 831 promotes apoptosis and enhances chemosensitivity in breast cancer by acting as a novel transcriptional repressor targeting the STAT3/Bcl2 signaling pathway显示文摘Emerging evidence suggested that zinc finger protein 831(ZNF831)was associated with immune activity and stem cell regulation in breast cancer.Whereas,the roles and molec-ular mechanisms of ZNF831 in oncogenesis remain unclear.ZNF831 expression was significantly diminished in breast cancer which was associated with promoter CpG methylation but not mu-tation.Ectopic over-expression of ZNF831 suppressed breast cancer cell proliferation and col-ony formation and promoted apoptosis in vitro,while knockdown of ZNF831 resulted in an opposite phenotype.Anti-proliferation effect of ZNF831 was verified in vivo.Bioinformatic analysis of public databases and transcriptome sequencing both showed that ZNF831 could enhance apoptosis through transcriptional regulation of the JAK/STAT pathway.ChiP and luciferase report assays demonstrated that ZNF831 could directly bind to one specific region of STAT3 promoter and induce the transcriptional inhibition of STAT3.As a result,the attenuation of STAT3 led to a restraint of the transcription of Bcl2 and thus accelerated the apoptotic progression.Augmentation of STAT3 diminished the apoptosis-promoting effect of ZNF831 in breast cancer cell lines.Furthermore,ZNF831 could ameliorate the anti-proliferation effect of capecitabine and gemcitabine in breast cancer cell lines.Our findings demonstrate for the first time that ZNF831 is a novel transcriptional suppressor through inhibiting the expression of STAT3/Bcl2 and promoting the apoptosis process in breast cancer,suggesting ZNF831 as a novel biomarker and potential therapeutic target for breast cancer patients.Jun Fan Zhe Zhang Hongqiang Chen Dongjiao Chen Wenbo Yuan Jingzhi Li Yong Zeng Shimeng Zhou Shu Zhang Gang Zhang Jiashen Xiong Lu Zhou Jing Xu Wenbin Liu Yan Xu 2024Genes & Diseases2024,11,1:0
