| 1 | uPAR promotes tumor-like biologic behaviors of fibroblast-like synoviocytes through PI3K/Akt signaling pathway in patients with rheumatoid arthritis显示文摘Urokinase-type plasminogen activator receptor(uPAR),is a multifunctional receptor on cell surface,widely present in endothelial cells,fibroblasts,and a variety of malignant cells.Current studies have suggested that uPAR overexpressed on synovial tissues or in synovial fluid or plasma in patients with rheumatoid arthritis(RA).However,there are limited researches regarding the role of uPAR on fibroblast-like synoviocytes of rheumatoid arthritis(RA-FLSs)and its underlying mechanisms.Here,our studies show that the expression of uPAR protein was significantly higher in fibroblast-like synoviocytes(FLSs)from RA than those from osteoarthritis or traumatic injury patients.uPAR gene silencing significantly inhibited RA-FLSs cell proliferation,restrained cell transformation from the G0/G1 phase to S phase,aggravated cell apoptosis,interfered with RA-FLSs cell migration and invasion,and reduced activation of the PI3K/Akt signaling pathway,which may be associated withβ1-integrin.Cell supernatants from uPAR gene-silenced RA-FLSs markedly inhibited the migration and tubule formation ability of the HUVECs(a human endothelial cell line).Therefore,we demonstrate that uPAR changes the biological characteristics of RA-FLSs,and affects neoangiogenesis of synovial tissues in patients with RA.All of these may be associated with theβ1-integrin/PI3K/Akt signaling pathway.These results imply that targeting uPAR and its downstream signal pathway may provide therapeutic effects in RA. | Yan Liu Yun Feng Pan You-qiu Xue Lin-kai Fang Xing-hua Guo Xin Guo Meng Liu Bi-yao Mo Meng-ru Yang Fang Liu Yun-ting Wu Nancy Olsen Song Guo Zheng | 2018 | Cellular & Molecular Immunology2018,15,2: | 9 |

| 4 | Hysteroscopic Appearence of Midsecretory Endometrium in Relation to Pinopodes Expression and the Reproductive Outcome in Infertile Women显示文摘Objective To investigate the relation between hysteroscopic finding and pinopodes expression,meanwhile to explore whether hysteroscopic finding was a predictor of reproductive outcome for infertile women. Methods A total of 79 infertile patients underwent hysteroscopic assessement of the midsecretory endometrium and hormonal measurement 7-9 d after a spontaneous ovulation assessed with transvaginal ultrasound. Pinopodes expression by scanning electron microscope was examined simultaneously in randomly selected 32 patients. All patients were followed up to determine their reproductive outcomes. Results Twenty-six (32.9%) patients were diagnosed as having 'good' midsecretory endometrium and 53 (67.1%) as 'poor' midsecretory endometrium by hysteroscopy. There were no significant differences of endometrial thickness and pattern in the 'good' group compared with the 'poor' group. Serum progesterone level in the 'good' group was apparently higher than that in the 'poor' group (P=0.025),but no correlation was observed between serum oestradiol level and hysteroscopic finding. Significant differences of the development stage and number of pinopodes were observed between the two groups (P<0.05). The pregnancy rate was significantly higher in the 'good' group (53.9%) than in the 'poor' group (26.4%) (P=0.017). | 14 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 16 | Molecular mechanism of lycopene cyclases regulating carotenoids ratio in different branches during tea leaf and flower development显示文摘Carotenoids are essential components in tea quality, contributing to leaf color and aroma. However, little information about carotenoids in different tea cultivars and their biosynthesis regulation mechanism during leaf development is known. Here we analyzed carotenoids by HPLC in the buds and leaves of 113 tea cultivars harvested on the same day. By profile clustering, carotenoids were divided into five groups. Same group cultivars displayed divergence in the total content of carotenoids but a similar molar ratio. To figure out the molecular mechanisms of this phenomenon, we further characterized all functional lycopene cyclases, which are the branch point of the carotenoid biosynthesis pathway. Two β-lycopene cyclases(CsLCYB1 and CsLCYB2) and one ε-lycopene cyclase(CsLCYE1) were cloned. Subcellular localization analysis showed that all cloned CsLCYs were localized in plastids. Enzyme activity assays in E. coli indicated both CsLCYBs catalyzed lycopene into β-carotene, and CsLCYE1 produced δ-carotene and ε-carotene. We found CsLCYB1 and CsLCYE1 predominantly expressed in leaf, while CsLCYB2was mainly expressed during flowering stages. Suppression by antisense oligonucleotides reduced CsLCYB1 and CsLCYE1 transcripts and led to reduction of both β,β-branch and β,ε-branch carotenoids in leaf. The expression levels of CsLCYB1 showed a significant positive correlation withβ,β-branch carotenoids in leaf. Our study provides carotenoid profiles of different tea cultivars, which can assist tea producers in selecting cultivars of interest. | 19 | Phylogenomic,morphological,and niche differentiation analyses unveil species delimitation and evolutionary history of endangered maples in Acer series Campestria(Sapindaceae)显示文摘Accurate species delimitation is crucial for biodiversity conservation.The Acer series Campestria comprises four species,A.campestre L.,A.miyabei Maxim.,A.miaotaiense P.C.Tsoong,and A.yangjuechi Fang&P.L.Chiu.To clarify controversies over the taxonomic status of the latter three endangered species,we undertook phylogenomic,morphological,and niche differentiation analyses in series Campestria.Our coalescent species tree of 544 and 77 single-copy nuclear genes supported series Campestria as monophyletic,with A.yangjuechi having the closest relationship with A.miaotaiense.However,in the plastome-derived tree based on 64 protein coding sequences,the four species did not cluster together,and each of them grouped with some other sympatric Acer species.Given this nuclear-cytoplasmic conflict,we hypothesize that A.yangjuechi have been subject to nuclear gene introgression and plastid(pt)capture involving another sympatric maple,that is,A.amplum Rehder.Principal component analysis and machine learning based on morphological data could not separate A.yangjuechi and A.miaotaiense,but they both could be clearly distinguished from A.miyabei.Moreover,the niche overlap tests of the two more widespread species,A.miyabei and A.miaotaiense,showed they clearly occupy distinct niches.Overall,we conclude that A.miyabei and A.miaotaiense are distinct species,while A.yangjuechi(endemic to Mt.Tianmu/East China)should be treated as a subspecies of A.miaotaiense.Our study points out that multiple lines of phylogenomic,morphological,and ecological evidence prove highly useful in species delimitation.Additionally,our results should help to inform conservation measures for endangered species of the genus Acer/series Campestria in East Asia. | Xiao-Kai Fan Jing Wu Hans Peter Comes Yu Feng Ting Wang Shu-Zhen Yang Takaya Iwasaki Hong Zhu Yun Jiang Joongku Lee Pan Li | 2023 | Journal of Systematics and Evolution2023,61,2: | 0 |
