| 1 | Role and effect of vein-transplanted human umbilical cord mesenchymal stem cells in the repair of diabetic foot ulcers in rats显示文摘Diabetic foot ulcer(DFU)is one of diabetic complications,which is frequently present and tormented in diabetes mellitus.Most multipotent mesenchymal stromal cells(MSCs)are capable of immune evasion,providing an allogeneic,ready-to-use,cell product option for therapeutic applications.The beneficial effect of MSCs for the treatment of a variety of traumatic injuries,such as open wounds,has been extensively explored.In this study,a rat DFU model was used to simulate the pathophysiology of clinical patients and to investigate the localization of human umbilical cord mesenchymal stem cells(hUC-MSCs)after intravenous transplantation and its role in DFU healing,so as to evaluate the potential of hUC-MSCs in the treatment of DFU.The diabetic rat model was established by streptozotocin injection,which was used to create full-thickness foot dorsal skin wounds to mimic DFU by a 6-mm skin biopsy punch and a Westcott scissor.The hUC-MSCs were transplanted through femoral vein,and the ulcer cicatrization situation and the fate of hUC-MSCs were evaluated.Our data suggest that intravenously transplantated hUC-MSCs have the ability to migrate and locate to the wound tissue and are helpful to wound healing in DFU rats,partly by regulating inflammation,trans-differentiation and providing growth factors that promote angiogenesis,cell proliferation and collagen deposition.Herein,we demonstrate that hUC-MSC transplantation is able to accelerate DFU healing in rats and transplantation of exogenous stem cells may be a potential strategy for clinical application in DFUs. | Rongfeng Shi Weishuai Lian Yinpeng Jin Chuanwu Cao Shilong Han Xiaohu Yang Suming Zhao Maoquan Li Hui Zhao | 2020 | Acta Biochimica et Biophysica Sinica2020,52,6: | 13 |
| 2 | Regulation of phenylpropanoid biosynthesis by MdMYB88 and MdMYB124 contributes to pathogen and drought resistance in apple显示文摘MdMYB88 and MdMYB124 have been demonstrated to be responsible for lignin accumulation in apple under drought stress.In this study,using a metabolomic approach,we identified differentially accumulated phenylpropanoid and flavonoid metabolites in MdMYB88/124 transgenic RNAi plants under control and long-term drought stress conditions in apple roots.We confirmed the regulation of phenylalanine by MdMYB88 and MdMYB124 via UPLC-MS in apple roots under both control and drought conditions.Using Electrophoretic Mobility Shift Assay(EMSA)and ChIPquantitative PCR(qPCR)analyses,we found that MdMYB88 positively regulates the MdCM2 gene,which is responsible for phenylalanine biosynthesis,through binding to its promoter region.Under long-term drought conditions,MdMYB88/124 RNAi plants consistently accumulated increased amounts of H2O2 and MDA,while MdMYB88 and MdMYB124 overexpression plants accumulated decreased amounts of H2O2 and MDA.We also examined the accumulation of metabolites in the phenylpropanoid biosynthesis pathway in the leaves of MdMYB88 and MdMYB124 transgenic apple plants after long-term drought stress.We found that metabolites responsible for plant defense,including phenylpropanoids and flavonoids,accumulated less in the RNAi plants but more in the overexpression plants under both control and drought conditions.We further demonstrated that MdMYB88/124 RNAi plants were more sensitive to Alternaria alternata f.sp.mali and Valsa mali,two pathogens that currently severely threaten apple production.In contrast,MdMYB88 and MdMYB124 overexpression plants were more tolerant to these pathogens.The cumulative results of this study provided evidence for secondary metabolite regulation by MdMYB88 and MdMYB124,further explained the molecular roles of MdMYB88 and MdMYB124 in drought resistance,and provided information concerning molecular aspects of their roles in disease resistance. | Dali Geng Xiaoxia Shen Yinpeng Xie Yusen Yang Ruiling Bian Yuqi Gao Pengmin Li Liying Sun Hao Feng Fengwang Ma Qingmei Guan | 2020 | Horticulture Research2020,7,1: | 5 |
| 3 | Regulatory effect of salicylic acid and methyl jasmonate supplementation on ergosterol production in Hericium erinaceus mycelia显示文摘We evaluated the biomass and ergosterol content of Hericium erinaceus mycelium, and extracellular enzyme activities in H. erinaceus liquid culture following salicylic acid(SA) and methyl jasmonic acid(Me JA)supplementation. The optimal SA concentration was100 lmoláL-1, where the highest ergosterol content of 2.33 mgág-1was obtained following 6-day cultivation with100 lmoláL-1SA supplementation, and which was significantly higher than the unsupplemented control(p \ 0.01). Following 4-day supplementation with50 lmoláL-1Me JA, the highest ergosterol content obtained was 1.988 mgág-1, which was 25.8 % higher than the unsupplemented control. | 4 | Apple SERRATE negatively mediates drought resistance by regulating MdMYB88 and MdMYB124 and microRNA biogenesis显示文摘The function of serrate(SE)in miRNA biogenesis in Arabidopsis is well elucidated,whereas its role in plant drought resistance is largely unknown.In this study,we report that MdSE acts as a negative regulator of apple(Malus×domestica)drought resistance by regulating the expression levels of MdMYB88 and MdMYB124 and miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,mdm-miR319,and mdm-miR399.MdSE interacts with MdMYB88 and MdMYB124,two positive regulators of apple drought resistance.MdSE decreases the transcript and protein levels of MdMYB88 and MdMYB124,which directly regulate the expression of MdNCED3,a key enzyme in abscisic acid(ABA)biosynthesis.Furthermore,MdSE is enriched in the same region of the MdNECD3 promoter where MdMYB88/MdMYB124 binds.Consistently,MdSE RNAi transgenic plants are more sensitive to ABA-induced stomatal closure,whereas MdSE OE plants are less sensitive.In addition,under drought stress,MdSE is responsible for the biogenesis of mdm-miR399,a negative regulator of drought resistance,and negatively regulates miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,and mdm-miR319,which are positive regulators of drought resistance.Taken together,by revealing the negative role of MdSE,our results broaden our understanding of the apple drought response and provide a candidate gene for apple drought improvement through molecular breeding. | Xuewei Li Pengxiang Chen Yinpeng Xie Yan Yan Liping Wang Huan Dang Jing Zhang Lingfei Xu Fengwang Ma Qingmei Guan | 2020 | Horticulture Research2020,7,1: | 4 |

| 6 | Genome-wide identification of droughtresponsive microRNAs in two sets of Malus from interspecific hybrid progenies显示文摘Drought stress can negatively impact apple fruit quality and yield.Apple microRNAs(miRNAs)participate in apple tree and fruit development,as well as in biotic stress tolerance;however,it is largely unknown whether these molecules are involved in the drought response.To identify drought-responsive miRNAs in Malus,we first examined the drought stress tolerance of ten F1 progenies of R3(M.×domestica)×M.sieversii.We performed Illumina sequencing on pooled total RNA from both drought-tolerant and drought-sensitive plants.The sequencing results identified a total of 206 known miRNAs and 253 candidate novel miRNAs from drought-tolerant plants and drought-sensitive plants under control or drought conditions.We identified 67 miRNAs that were differentially expressed in drought-tolerant plants compared with drought-sensitive plants under drought conditions.Under drought stress,61 and 35 miRNAs were differentially expressed in drought-tolerant and drought-sensitive plants,respectively.We determined the expression levels of seven out of eight miRNAs by stem-loop qPCR analysis.We also predicted the target genes of all differentially expressed miRNAs and identified the expression of some genes.Gene Ontology analyses indicated that the target genes were mainly involved in stimulus response and cellular and metabolic processes.Finally,we confirmed roles of two miRNAs in apple response to mannitol.Our results reveal candidate miRNAs and their associated mRNAs that could be targeted for improving drought tolerance in Malus species,thus providing a foundation for understanding the molecular networks involved in the response of apple trees to drought stress. | Chundong Niu Haiyan Li Lijuan Jiang Mingjia Yan Cuiying Li Dali Geng Yinpeng Xie Yan Yan Xiaoxia Shen Pengxiang Chen Jun Dong Fengwang Ma Qingmei Guan | 2019 | Horticulture Research2019,6,1: | 2 |

| 14 | Experimental study on the effect of hydrodynamic conditions on flocculation and settling properties of fine-grain sediment显示文摘The flocculation of cohesive sediment particles is a function of the collision efficiency of sediment particles,which is mainly influenced by local flow hydrodynamics.A detailed study on local hydrodynamic characteristics in a novel stirred tank was done to measure the flow field of a turbulent flow.Instantaneous flow velocity fields were obtained and processed,and the average flow field was computed.Particle image velocimetry(PIV)results measuring the hydrodynamic characteristics(expressed as turbulent kinetic energy(TKE),local dissipation rate of TKE,velocity gradient or shear rate,turbulent shear stress,etc.)were analyzed and discussed.Particle tracking and automated image processing techniques were utilized to analyze the impact of the hydrodynamic conditions on the floc and settling properties.The differences in floc properties under the two turbulent shear states(i.e.,steady-state(SS)and unsteady-state(US))hydrodynamic conditions were examined.The results show that floc size and floc size distribution(FSD)of pure kaolin clay flocs are significantly influenced by the local dissipation rate of TKE and the turbulent intensity states.The floc size,df,84,at local dissipation rate of TKE of 11×10^(-4),29×10^(-4),44×10^(-4),83×10^(-4),and 142×10^(-4)m^(2)/s^(3)is 97,108,107,124,and 150 mm,respectively.It was found that by increasing the local dissipation rate of TKE,the FSD was skewed left.The US shear condition generated larger flocs(and in higher proportions)relative to the SS condition.The settling velocity and flux settling velocities of flocs formed under SS conditions are higher than those formed under US conditions.The fractal dimension of SS flocs also is higher than the fractal dimension of US flocs.These results highlight a significant impact of the states of hydrodynamics on floc properties such as floc size and FSD as well as the settling and morphological properties of flocs. | Bismark Odum Chunyang Xu Yongping Chen Yinpeng Yao Yan Zhou | 2023 | International Journal of Sediment Research2023,38,6: | 0 |
| 15 | Optimal Configuration of an Off-Grid Hybrid Wind-Hydrogen Energy System: Comparison of Two Systems显示文摘Due to the uncertainty of renewable energy power generation and the non-linearity of load demand,it becomes complicated to determine the capacity of each device in hybrid renewable energy power generation systems.This work aims to optimize the capacity of two types of the off-grid hybrid wind-hydrogen energy system.We considered the maximum profit of the system and the minimum loss of power supply probability as optimization goals.Firstly,we established steady-state models of the wind turbine,alkaline electrolyzer,lead-acid battery,and proton exchange membrane fuel cell in matrix laboratory software to optimize the capacity.Secondly,we analyzed the operating mode of the system and determined two system structures(system contains batteries whether or not).Finally,according to the wind speed and load in the sample area,we compared the economics of the two systems and selected the optimal configuration for the area.In the same calculation example data,the non-dominated sorting genetic algorithm-II(NSGA-II)is used to optimize the capacity of each device in the two systems.The results showed that the profit of the without battery-equipped system is 32.38%higher than another system.But the power supply reliability is the opposite.To avoid the contingency of the calculation results,we used the traditional genetic algorithm(GA)and ant colony optimization(ACO)to calculate the same example.The results showed that NSGA-II is significantly better than GA and ACO in terms of iteration steps and calculation results.The required architecture for the System-I composes of 3 numbers of 10 kW wind turbines,61 sets of 12 V·240 Ah leadacid batteries,8 kW electrolytic cell,and 6 kW PEMFC.The net profit and LPSP are ¥44,315 and 0.01254 respectively.The required architecture for the System-II composes of 2 numbers of 10 kW wind turbines,24 kW electrolytic cells,and 18 kW PEMFC.Net profit and LPSP are ¥58,663 and 0.03244,respectively.This paper provided two schemes for the optimal configuration of the hybrid wind-hydrogen energy system in islanding mode,which provided a theoretical basis for practical engineering applications. | Zekun Wang Yan Jia Yingjian Yang Chang Cai Yinpeng Chen | 2021 | Energy Engineering2021,118,6: | 0 |

| 17 | Adipose-derived stem cells show hepatic differentiation potential and therapeutic effect in rats with acute liver failure显示文摘Hepatocyte transplantation contributes to the repair of liver damage,but hepatocyte resources are limited,making it difficult for this to become a routine treatment.Previous studies have confirmed that mesenchymal stem cells(MSCs)can be induced to differentiate into hepatocyte-like cells(HLCs)by adding different cytokine combinations in vitro,and they then play some roles of hepatocytes.Our previous studies found that the differentiation ability of stem cells is closely related to the origin of the tissue.To identify the mesenchymal stem cells that are most suitable for hepatic differentiation and the treatment of liver failure,we use a three-phase induction process in which human adipose-derived stem cells(hADSCs)and umbilical cord mesenchymal stem cells(hUCMSCs)are induced to differentiate towards HLCs in vitro,and rats with acute liver failure(ALF)induced by D-gal are cured by MSCs and MSC-derived HLCs(MSCs-HLC),respectively.We find that hADSCs are stronger than hUCMSCs in hepatic differentiation ability,and they have a better curative effect when using hADSCs-HLC or jointly using hADSCs and hADSCs-HLC,which has positive significance for hepatocyte regeneration,recovery of liver function and reduction of systemic inflammatory reaction,finally improving the survival rate of rats with acute liver failure. | Yinpeng Jin Rongfeng Shi Tangkai Qi Qiang Li Cong Chen Shane Gao Fengjuan Gao Danjing Yang Guangxi Sun Jianqing Xu Qingchun Fu Jun Xu Xiaoyan Zhang | 2023 | Acta Biochimica et Biophysica Sinica2023,55,4: | 0 |
