| 1 | Depression compromises antiviral innate immunity via the AVP-AHI1-Tyk2 axis显示文摘Depression is a serious public-health issue.Recent reports have suggested higher susceptibility to viral infections in depressive patients.However,how depression affects antiviral innate immune signaling remains unknown.Here,we revealed a reduction in expression of Abelson helper integration site 1(AHI1)in the peripheral blood mononuclear cells(PBMCs)and macrophages from the patients with major depressive disorder(MDD),which leads to attenuated antiviral immune response.We found that depression-related arginine vasopressin(AVP)induces reduction of AHI1 in macrophages.Further studies demonstrated that AHI1 is a critical stabilizer of basal type-I-interferon(IFN-I)signaling.Mechanistically,AHI1 recruits OTUD1 to deubiquitinate and stabilize Tyk2,while AHI1 reduction downregulates Tyk2 and IFN-I signaling activity in macrophages from both MDD patients and depression model mice.Interestingly,we identified a clinical analgesic meptazinol that effectively stimulates AHI1 expression,thus enhancing IFN-I antiviral defense in depression model mice.Our study promotes the understanding of the signaling mechanisms of depression-mediated antiviral immune dysfunction,and reveals meptazinol as an enhancer of antiviral innate immunity in depressive patients. | Hong-Guang Zhang Bin Wang Yong Yang Xuan Liu Junjie Wang Ning Xin Shifeng Li Ying Miao Qiuyu Wu Tingting Guo Yukang Yuan Yibo Zuo Xiangjie Chen Tengfei Ren Chunsheng Dong Jun Wang Hang Ruan Miao Sun Xingshun Xu Hui Zheng | 2022 | Cell Research2022,32,10: | 3 |
| 2 | Sieving carbons promise practical anodes with extensible low-potential plateaus for sodium batteries显示文摘Non-graphitic carbons are promising anode candidates for sodium-ion batteries,while their variable and complicated microstructure severely limits the rational design of high-energy carbon anodes that could accelerate the commercialization of sodium-ion batteries,as is the case for graphite in lithium-ion batteries.Here,we propose sieving carbons,featuring highly tunable nanopores with tightened pore entrances,as high-energy anodes with extensible and reversible low-potential plateaus(<0.1 V).It is shown that the tightened pore entrance blocks the formation of the solid electrolyte interphase inside the nanopores and enables sodium clustering to produce the plateau.Theoretical and spectroscopic studies also show that creating a larger area of sodiophilic pore surface leads to an almost linearly increased number of sodium clusters,and controlling the pore body diameter guarantees the reversibility of sodium cluster formation,producing a sieving carbon anode with a record-high plateau capacity of 400 mAh g.More excitingly,this approach to preparing sieving carbons has the potential to be scalable for modifying different commercial porous carbons. | Qi Li Xiangsi Liu Ying Tao Jianxing Huang Jun Zhang Chunpeng Yang Yibo Zhang Siwei Zhang Yiran Jia Qiaowei Lin Yuxuan Xiang Jun Cheng Wei Lv Feiyu Kang Yong Yang Quan-Hong Yang | 2022 | National Science Review2022,9,8: | 3 |

| 5 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. | 6 | Versatile subtypes of pericytes and their roles in spinal cord injury repair,bone development and repair显示文摘Vascular regeneration is a challenging topic in tissue repair. As one of the important components of the neurovascular unit(NVU),pericytes play an essential role in the maintenance of the vascular network of the spinal cord. To date, subtypes of pericytes have been identified by various markers, namely the PDGFR-β, Desmin, CD146, and NG2, each of which is involved with spinal cord injury(SCI) repair. In addition, pericytes may act as a stem cell source that is important for bone development and regeneration, whilst specific subtypes of pericyte could facilitate bone fracture and defect repair. One of the major challenges of pericyte biology is to determine the specific markers that would clearly distinguish the different subtypes of pericytes, and to develop efficient approaches to isolate and propagate pericytes. | 7 | Recent progress on the hydrogen storage properties of ZrCo-based alloys applied in International Thermonuclear Experimental Reactor(ITER)显示文摘Under the development of International Thermonuclear Experimental Reactor(ITER) system aimed at realizing the controllable fusion reaction to solve the energy crisis fundamentally, there is an urgent need to find an appropriate material for tritium handling. ZrCo alloy is considered to be a promising candidate for the storage and delivery of hydrogen isotopes due to the favorable characteristics such as low plateau pressure for absorption, high dissociation pressure at moderate temperature and better ability of trapping ~3He. However, the hydrogen induced disproportionation and the slower recovery/deliverty rate of ZrCo-based alloys have limited their further application in ITER system. This review summarizes the efforts towards enhancing the hydrogen storage properties of ZrCo-based alloys including element substitution, surface modification, disproportionation mechanism investigation and the isotope effect study. Element substitution and surface modification play positive role to improve the anti-disproportionation ability and kinetic property of the alloys. However, the ZrCo-based alloys require to be further modified by more attempts such as new composition, novelty modification method or catalyst addition in order to better satisfy the application demands for tritium handling. | 8 | Human Cytomegalovirus Influences Host circRNA Transcriptions during Productive Infection显示文摘Human cytomegalovirus(HCMV)is a double-strand DNA virus widely infected in human.Circular RNAs(circ RNAs)are non-coding RNAs with most functions of which keep unknown,and the effects of HCMV productive infection on host circ RNA transcriptions remain unclear.In this study,we profiled 283 host circ RNAs that significantly altered by HCMV productive infection in human embryonic lung fibroblasts(HELF)by RNA deep sequencing and bioinformatics analysis.Among these,circ SP100,circ MAP3 K1,circ PLEKHM1,and circ TRIO were validated for their transcriptions and sequences.Furthermore,characteristics of circ SP100 were investigated by RT-q PCR and northern blot.It was implied that circ SP100 was produced from the sense strand of the SP100 gene containing six exons.Kinetics of circ SP100 and SP100 m RNA were significantly different after infection:circ SP100 levels increased gradually along with infection,whereas SP100 m RNA levels increased in the beginning and dropped at 24 h post-infection(hpi).Meanwhile,a total number of 257 proteins,including 10 HCMV encoding proteins,were identified potentially binding to cytoplasmic circ SP100 by RNA antisense purification(RAP)and mass spectrometry.Enrichment analysis showed these proteins were mainly involved in the spliceosome,protein processing,ribosome,and phagosome pathways,suggesting multiple functions of circ SP100 during HCMV infection. | Jingui Deng Yujing Huang Qing Wang Jianming Li Yanping Ma Ying Qi Zhongyang Liu Yibo Li Qiang Ruan | 2021 | Virologica Sinica2021,36,2: | 1 |

| 10 | Real time observation of the photocleavage of single DNA molecules显示文摘A method for real time observation of photo-cleavage of stretched λDNA at single molecular level by a fluorescent microscope coupled with CCD is developed. DNA molecules stained with YOYO-1 are stretched by the mo-lecular combing technique and fixed on a modified slide. Then the process of photocleavage and relaxation of DNA under radiation of blue light is observed. We speculate that the conformation change of stretched DNA and the effect of water are likely to facilitate the effect of YOYO photocleav-age DNA molecules. | 12 | Establishment and validation of psoriasis evaluation models显示文摘Psoriasis is a common inflammatory skin disease that seriously affects the patient's quality of life.The diagnosis of psoriasis is mainly based on clinical and pathological features,and the assessment depends on the psoriasis area and severity index(PASI).However,there are few reliable and accurate evaluation methods to assess lesion severity and therapeutic effects.This work identified 17 model genes from GEO datasets and established 6 psoriasis evaluation models by LASSO regression,linear regression,and random forest separately.Models were trained and evaluated in different GEO datasets.All 6 models accurately classified psoriatic lesions and non-lesional skin in training and testing data,and showed good AUC.In biologics-treated samples,the model scores were positively correlated with the severity of lesions and negatively correlated with treatment length.Thus,models have the potential to assess the therapeutic effects.In addition,the expression of model genes was examined in keratinocytes,skin of IMQ-induced psoriatic mice,and lesions of psoriasis patients.The RNA and protein levels of model genes increased in cytokine-stimulated keratinocytes and psoriatic lesions as expected.This work provides new methods to assess the lesion severity and therapeutic effects of biologies in psoriasis. | Yibo Hu Ling Jiang Li Lei Liping Luo Haoran Guo Ying Zhou Jinhua Huang Jing Chen Qinghai Zeng | 2022 | Fundamental Research2022,2,1: | 0 |
