1Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
2Screening and flocculating properties of bioflocculant-producing microorganisms显示文摘Screening of bioflocculant-producing microorganisms was carried out. A strain that secreted excellent bioflocculant was isolated from municipal sewage using the spread plate technique, identified as Klebsiella sp. by the analytical profile index (API) identification system, and named A9. Several important factors that had an effect on A9’s bioflocculant-producing and flocculating activity were studied. A total of 4 g/L Kaolin suspension was used to measure the flocculating activity of the bioflocculant from A9. 8Pretreatment of copper-bearing refractory gold ores by bio-heap leaching显示文摘The refractory gold ores associated with rich copper and trapped in pyrite and quartz were studied. With conventional technique (all-sliming cyanidation), the gold recovery rate is only 51.78%. To eliminate the negative effects of copper and pyrite on cyanidation and increase the gold recovery rate, the investigation on bio-heap leaching pretreatment was made, by which Cu would be dissolved and gold would be liberated from pyrite. The experiment adopted mixed bacteria, mainly Thiobacillus ferrooxidan (named T. f1), as the bacterial catalyst for bio-preconditioning and was carried out in a PVC column with a diameter of 20 cm and a height of 1.3 m loaded with gold ores. The temperature was controlled between 28 and 30℃, the pH value was kept between 2.0-2.5,and the flux of sprinkling bacterial liquid was maintained 0.80 L/h. After 45-day's bio-oxidization, among the samples sizing from 0to 5 mm, the oxidation rates of Cu, Fe and S were respectively 44.62%, 28.16% and 25.46%, and the gold recovery rate by cyaniding increased to 80.35%.
