2SnRK1 Phosphorylates and Destabilizes WRKY3 to Enhance Barley Immunity to Powdery Mildew显示文摘Plants recognize pathogens and activate immune responses,which usually involve massive transcriptional reprogramming.The evolutionarily conserved kinase,Sucrose non-fermenting-related kinase 1(SnRK1),functions as a metabolic regulator that is essential for plant growth and stress responses.Here,we identify barley SnRK1 and a WRKY3 transcription factor by screening a cDNA library.SnRK1 interacts with WRKY3 in yeast,as confirmed by pull-down and luciferase complementation assays.Förster resonance energy transfer combined with noninvasive fluorescence lifetime imaging analysis indicates that the interaction occurs in the barley nucleus.Transient expression and virus-induced gene silencing analyses indicate that WRKY3 acts as a repressor of disease resistance to the Bgh fungus.Barley plants overexpressing WRKY3 have enhanced fungal microcolony formation and sporulation.Phosphorylation assays show that SnRK1 phosphorylates WRKY3 mainly at Ser83 and Ser112 to destabilize the repressor,and WRKY3 non-phosphorylation-null mutants at these two sites are more stable than the wild-type protein.SnRK1-overexpressing barley plants display enhanced disease resistance to Bgh.Transient expression of SnRK1 reduces fungal haustorium formation in barley cells,which probably requires SnRK1 nuclear localization and kinase activity.Together,these findings suggest that SnRK1 is directly involved in plant immunity through phosphorylation and destabilization of the WRKY3 repressor,revealing a new regulatory mechanism of immune derepression in plants.Xinyun Han Ling Zhang Lifang Zhao Pengya Xue Ting Qi Chunlei Zhang Hongbo Yuan Lixun Zhou Daowen Wang Jinlong Qiuand Qian-Hua Shen 2020Plant Communications2020,1,4:2
3Optimization of Ultrasonic Extraction and Clean-up Protocol for the Determination of Polycyclic Aromatic Hydrocarbons in Marine Sediments by High-performance Liquid Chroma-tography Coupled with Fluorescence Detection显示文摘The procedures of ultrasonic extraction and clean-up were optimized for the determination of polycyclic aromatic hydrocarbons (PAHs) in marine sediments. Samples were ultrasonically extracted, and the extracts were purified with a miniaturized silica gel chromatographic column and analyzed with high performance liquid chromatography (HPLC) with a fluorescence detector. Ultrasonication with methanol-dichloromethane (2:1, v/v) mixture gave higher extraction efficiency than that with dichloromethane. Among the three elution solvents used in clean-up step, dichloromethane-hexane (2:3, v/v) mixture was the most satisfactory. Under the optimized conditions, the recoveries in the range of 54.82% to 94.70% with RSDs of 3.02% to 23.22% for a spiked blank, and in the range of 61.20% to 127.08% with RSDs of 7.61% to 26.93% for a spiked matrix, were obtained for the 15 PAHs studied, while the recoveries for a NIST standard reference SRM 1941b were in the range of 50.79% to 83.78% with RSDs of 5.24% to 21.38%. The detection limits were between 0.75 ng L-1 and 10.99 ng L-1for different PAHs. 6Epigenetic regulation mechanism of ABCG2 induced drug-resistant phenotype显示文摘The aim of this study is to investigate epigenetic mechanism of ABCG2 induced drug-resistance.It is not only expatiate for drug-resistance regulation mechanism in all-round,but also to provide scientific experimental basis for selecting target to reverse its drug-resistance.Apply methylation-specific PCR(MSP) to have tested methylation of ABCG2 promoter region -359 to -353 specific positions in breast cancer tissues and paired adjacent tissue of 22 cases,and test their methylation positions with MSP products for sequencing;and adopt fluorescent quantitation RT-PCR to test expression level DNMT1,DNMT3A,DNMT3B and ABCG2;to make analysis on relationship between them with statistical spearman correlation.Specific positions of ABCG2 gene promoter region of 18 cases among the 22 cases with breast cancer(18/22,82%) existed high methylation(P<0.05),MSP products sequencing proved methylation of the specific position,and mRNA expression level was relative higher in remarkable positive correlation(P<0.05).ABCG2,DNMT1,DNMT3A,DNMT3B mRNA expression levels in breast cancer tissues were obviously higher than adjacent tissues(P<0.01),and DNMT3B expression level was obviously higher than DNMT1 and DNMT3A(P<0.01) in negative correlation with ABCG2 gene expression(P=0.001).-359 to -353 positions of promoter regions of ABCG2 gene existed high methylation capable to push expression of this gene in beast cancer tissue.DNMT3B is involved in expression regulation in ABCG2 gene,and provides new scientific basis for drug-resistance target as reverse ABCG2Yuan Jianhui Zhou Jianmeng JiNana Xu Xinyun Liu Jianjun Ke Yuebin Cheng Jinquan. 7Low rate of pre-exposure prophylaxis and post-exposure prophylaxis uptake and high prevalence of transmitted drug resistance among newly diagnosed primary HIV infections in Shenzhen, China: a real-world retrospective study显示文摘Background: Understanding the characteristics of newly diagnosed primary human deficiency virus-1 (HIV-1) infection in the context of the post-antiretroviral therapy era and HIV drug prophylaxis is essential for achieving the new target of 95-95-95-95 by 2025. This study reported the characteristics of newly diagnosed primary HIV-1 infection in Shenzhen.Methods: This is a real-world retrospective study. Eighty-seven newly diagnosed primary HIV-1-infected patients were recruited from January 2021 to March 2022 at the Third People’s Hospital of Shenzhen. Demographic, epidemiological, diagnostic, drug resistance, and medical data were described and analyzed.Results: Overall, 96.6% (84/87) of the newly identified primary HIV-1-infected patients were male, including 88.5% (77/87) men have sex with men (MSM), with a median age of 29.0 years (Q_(1)-Q_(3): 24.0-34.0 years);of these, 85.1% (74/87) reported high-risk sexual behaviors with casual partners. The rate of condom usage was only 28.7% (25/87). The overall rate of pre-exposure prophylaxis (PrEP) and post-exposure prophylaxis (PEP) was 8.0% (7/87, including 4 PrEP and 3 PEP cases) around the potential exposure, although 41.4% of the patients had prior awareness of such interventions. Moreover, only 19.5% (17/87) had previously used PrEP or PEP. Of those, 58.8% (10/17) of the patients obtained drugs from the internet, and only 35.3% (6/17) reported good compliance. A total of 54.0% (47/87) of subjects were diagnosed by the HIV nucleic acid test. Acute retroviral syndrome appeared in 54.0% (47/87) of patients. The prevalence of transmitted drug resistance (TDR) mutation was 33.9% (19/56), including 6 (10.7%) against nucleoside reverse transcriptase inhibitor (NRTI) plus non-nucleoside reverse transcriptase inhibitor (NNRTI), 8 (14.3%) against NNRTI, and 5 (8.9%) against protease inhibitor (PI) only.Conclusions: Owing to the low utilization rate and incorrect usage of PrEP and PEP, massive efforts are needed to promote HIV-preventive strategies in the MSM population. 9Whole genome variants across 57 pig breeds enable comprehensive identification of genetic signatures that underlie breed features显示文摘Background:A large number of pig breeds are distributed around the world,their features and characteristics vary among breeds,and they are valuable resources.Understanding the underlying genetic mechanisms that explain across-breed variation can help breeders develop improved pig breeds.Results:In this study,we performed GWAS using a standard mixed linear model with three types of genome variants(SNP,InDel,and CNV)that were identified from public,whole-genome,sequencing data sets.We used 469 pigs of 57 breeds,and we identified and analyzed approximately 19 million SNPs,1.8 million InDels,and 18,016 CNVs.We defined six biological phenotypes by the characteristics of breed features to identify the associated genome variants and candidate genes,which included coat color,ear shape,gradient zone,body weight,body length,and body height.A total of 37 candidate genes was identified,which included 27 that were reported previously(e.g.,PLAG1 for body weight),but the other 10 were newly detected candidate genes(e.g.,ADAMTS9 for coat color).Conclusion:Our study indicated that using GWAS across a modest number of breeds with high density genome variants provided efficient mapping of complex traits.Jingya Xu Yuhua Fu Yan Hu Lilin Yin Zhenshuang Tang Dong Yin Mengjin Zhu Mei Yu Xinyun Li Yang Zhou Shuhong Zhao Xiaolei Liu 2021Journal of Animal Science and Biotechnology2021,12,2:1

11Towards a tailored indoor horticulture:a functional genomics guided phenotypic approach显示文摘As indoor horticulture gathers momentum,electric(also termed artificial)lighting systems with the ability to generate specific and tunable wavelengths have been developed and applied.While the effects of light quality on plant growth and development have been studied,authoritative and reliable sets of light formulae tailored for the cultivation of economically important plants and plant traits are lacking as light qualities employed across laboratories are inconsistent.This is due,at least in part,to the lack of molecular data for plants examined under electric lights in indoor environments.It has hampered progress in the field of indoor horticulture,in particular,the transition from small-scale indoor farming to commercial plant factories.Here,we review the effects of light quality on model and crop plants studied from a physiological,physical and biochemical perspective,and explain how functional genomics can be employed in tandem to generate a wealth of molecular data specific for plants cultivated under indoor lighting.We also review the current state of lighting technologies in indoor horticulture specifically discussing how recent narrow-bandwidth lighting technologies can be tailored to cultivate economically valuable plant species and traits.Knowledge gained from a complementary phenotypic and functional genomics approach can be harvested not only for economical gains but also for sustainable food production.We believe that this review serves as a platform that guides future light-related plant research.Claudius Marondedze Xinyun Liu Shihui Huang Cynthia Wong Xuan Zhou Xutong Pan Huiting An Nuo Xu Xuechen Tian Aloysius Wong 2018Horticulture Research2018,5,1:0
12Promoting the optoelectronic and ferromagnetic properties of Cr_(2)S_(3)nanosheets via Se doping显示文摘Doping can change the band structure of semiconductors,thereby affecting their electrical,optical,and magnetic properties.In this study,we describe the synthesis of two-dimensional(2D)Se-doped Cr_(2)S_(3)(Se-Cr_(2)S_(3))nanosheets using the chemical vapor deposition method.In these semiconductor nanosheets,the Se doping concentration can be controlled by tuning the Se/S mass ratio in the precursor.At the doping concentrations of 10.05%and 2.05%,the room temperature conductivity and mobility were increased by nearly 4 and 2 orders of magnitude,respectively.In addition,the response time of an ultrathin Se-Cr_(2)S_(3)photodetector was 200 times shorter than that of an undoped Cr_(2)S_(3)nanosheet photodetector.4.07%-Se-Cr_(2)S_(3)nanosheets show ferrimagnetic behavior with a Curie temperature of~200 K,which is 80 K higher than that of undoped Cr_(2)S_(3)nanosheets.A density functional theory calculation indicated that the Se doping can induce the formation of intercalated Cr vacancies in SeCr_(2)S_(3)and enhance its metallic characteristics.Our results demonstrated that Se-Cr_(2)S_(3)has significant potential in future electronic,optoelectronic,and spintronic devices.Xinyun Zhou Chang Liu Lingting Song Hongmei Zhang Ziwei Huang Chenglin He Bailing Li Xiaohui Lin Zucheng Zhang Shun Shi Dingyi Shen Rong Song Jia Li Xingqiang Liu Xuming Zou Le Huang Lei Liao Xidong Duan Bo Li 2022Science China(Physics,Mechanics & Astronomy)2022,65,7:0
13Multi-View & Transfer Learning for Epilepsy Recognition Based on EEG Signals显示文摘Epilepsy is a central nervous system disorder in which brain activity becomes abnormal.Electroencephalogram(EEG)signals,as recordings of brain activity,have been widely used for epilepsy recognition.To study epilep-tic EEG signals and develop artificial intelligence(AI)-assist recognition,a multi-view transfer learning(MVTL-LSR)algorithm based on least squares regression is proposed in this study.Compared with most existing multi-view transfer learning algorithms,MVTL-LSR has two merits:(1)Since traditional transfer learning algorithms leverage knowledge from different sources,which poses a significant risk to data privacy.Therefore,we develop a knowledge transfer mechanism that can protect the security of source domain data while guaranteeing performance.(2)When utilizing multi-view data,we embed view weighting and manifold regularization into the transfer framework to measure the views’strengths and weaknesses and improve generalization ability.In the experimental studies,12 different simulated multi-view&transfer scenarios are constructed from epileptic EEG signals licensed and provided by the Uni-versity of Bonn,Germany.Extensive experimental results show that MVTL-LSR outperforms baselines.The source code will be available on http://gffzz188fe103f8f1460aspvpo05kuc5xo6okc.ffgz.tsg.suse.edu.cn/didid5/MVTL-LSR.Jiali Wang Bing Li Chengyu Qiu Xinyun Zhang Yuting Cheng Peihua Wang Ta Zhou Hong Ge Yuanpeng Zhang Jing Cai 2023Computers, Materials & Continua2023,,6:0

15Ultrathin microcrystalline hydrogenated Si/Ge alloyed tandem solar cells towards full solar spectrum conversion显示文摘Thin film solar cells have been proved the next generation photovoltaic devices due to their low cost,less material consumption and easy mass production.Among them,micro-crystalline Si and Ge based thin film solar cells have advantages of high efficiency and ultrathin absorber layers.Yet individual junction devices are limited in photoelectric conversion efficiency because of the restricted solar spectrum range for its specific absorber.In this work,w e designed a nd simulated a multi-junction solar cell with its four sub-cells selectively absorbing the full solar spectrum including the ultraviolet,green,red as well as near infrared range,respectively.B y tuning the G e content,the record efficiency of 24.80%has been realized with the typical quadruple junction structure of a-Si:H/a-Si0.9Ge0.1:H/μc-Si:H/μc-Si0.5Ge0.5:H.To further reduce the material cost,thickness dependent device performances have been conducted.It can be found that the design of total thickness of 4μm is the optimal device design in balancing the thickness a nd the PCE.While the design of ultrathin quadruple junction device with total thickness of 2μm is the optimized device design regarding cost and long-term stability with a little bit more reduction in PCE.These results indicated that our solar cells combine the advantages of low cost and high stability.Our work may provide a general guidance rule of utilizing the full solar spectrum for developing high efficiency and ultrathin multi-junction solar cells.Yu Cao Xinyun Zhu Xingyu Tong Jing Zhou Jian Ni Jianjun Zhang Jinbo Pang 2020Frontiers of Chemical Science and Engineering2020,14,6:0
16Layer-structured Cr/Cr_(x)N coating via electroplating-based nitridation achieving high deuterium resistance as the hydrogen permeation barrier显示文摘Hydrogen isotope permeation through structural materials is a key issue for developing nuclear fusion energy,which will cause fuel loss and radioactive pollution.Developing ceramic coatings with high thermal shock and hydrogen resistance is an effective strategy to solve this issue.In this work,a layer-structured Cr/Cr_(x)N coating was successfully fabricated by a facile electroplatingbased nitridation technique,which is easy,facile,and applicable to coating complex-shaped substrates.The Cr/Cr_(x)N coating,composed of a bottom Fe/Cr interdiffusion zone,a middle Cr layer,and a top Cr_(x)N layer,exhibits high bonding strength,high anti-thermal-shock ability,and high deuterium permeation resistance.Its bonding strength achieves 43.6 MPa.The Cr/Cr_(x)N coating remains intact even after suffering 300 thermal shock cycles under a 600℃–water condition.Through optimizing the nitridation temperature,the Cr/Cr_(x)N coating achieves a deuterium permeation reduction factor(PRF)as high as 3599 at 500℃.Considering its scalable fabrication technique and considerable properties,the developed Cr/Cr_(x)N coating may serve as a novel high-performance hydrogen permeation barrier in various fields.Liyu ZHENG Heping LI Jun ZHOU Xinluo TIAN Zhongyang ZHENG Long WANG Xinyun WANG Youwei YAN 2022Journal of Advanced Ceramics2022,11,12:0
