1Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16

3Endovascular treatment with or without intravenous alteplase for acute ischaemic stroke due to basilar artery occlusion显示文摘Background and purpose It remains controversial if endovascular treatment(EVT)can improve the outcome of patients with acute basilar artery occlusion(BAO).This study aims to compare the functional outcomes between EVT with and without intravenous thrombolysis(IVT)first in patients who had acute ischaemic stroke(AIS)due to BAO.Methods Patients who had AIS with BAO who underwent EVT within 24 hours of onset were enrolled in this multicentre cohort study,and the efficacy and safety were compared between IVT+EVT and direct EVT.The primary outcome was 90-day functional independence.All outcomes were assessed with adjusted OR(aOR)from the multivariable logistic regression.In addition,a meta-analysis was performed on all recently published pivotal studies on functional independence after EVT in patients with BAO.Results Of 310 enrolled patients with BAO,241(78%)were treated with direct EVT and 69(22%)with IVT+EVT.Direct EVT was associated with a worse functional outcome(aOR,0.46(95%CI 0.24 to 0.85),p=0.01).IVT+EVT was associated with a lower percentage of patients who needed≥3 passes of stent retriever(10.14%vs 20.75%).The meta-analysis regression revealed a potential positive correlation between bridging with IVT first and functional independence(r=0.14(95%CI 0.05 to 0.24),p<0.01).Conclusions This study showed that compared with direct EVT,EVT with IVT first was associated with better functional outcomes in patients with BAO treated within 24 hours of onset.The meta-analysis demonstrated similar favourable efficacy of IVT first followed by EVT in patients with BAO.Ximing Nie David Wang Yuehua Pu Yufei Wei Qixuan Lu Hongyi Yan Xin Liu Lina Zheng Jingyi Liu Xinxuan Yang Yarong Ding Dacheng Liu Wanying Duan Zhe Zhang Zhonghua Yang Miao Wen Weibin Gu Xinyi Hou Xinyi Leng Yuesong Pan Zhongrong Miao Liping Liu 2022Stroke & Vascular Neurology2022,7,3:10
4Enhanced Expression of miR-425 Promotes Esophageal Squamous Cell Carcinoma Tumorigenesis by Targeting SMAD2显示文摘Esophageal squamous cell carcinoma(ESCC) is one of the most common and deadly cancers in the world. Currently, clinical therapy of ESCC remains limited and the five-year survival rate is poor. The function of miR-425 has been reported in multiple human cancers.However, the tumorigenic role and clinical significance of miR-425 in ESCC remains unclear. We found that enhanced expression of miR-425 in ESCC cell lines not only promoted cell proliferation and colony formation, but also increased cellular metastasis. Furthermore, we revealed the mechanism that miR-425 inhibited the expression of SMAD2 by targeting the second binding site in the 30-untranslated region(30-UTR) in ESCC. This mode of action influenced not only SMAD2 mRNA expression but also protein expression. In addition, we detected the expression of miR-425 in ESCC tissues and plasma. Moreover, we analyzed the relationship between miR-425 expression and SMAD2 m RNA expression. We found that miR-425 was overexpressed in ESCC tissues and the plasma relative to adjacent normal tissues and plasma of healthy individuals. Furthermore, there was a negative correlation between miR-425 expression and SMAD2. 5Changes of Petroleum Acid Distribution Characterized by FT-ICR MS in Heavy Acidic Crude Oil after True Boiling Point Distillation显示文摘The molecular transformations of carboxylic acids in heavy acidic SL crude before and after true boiling point distillation were examined by ultra-high resolution negative-ion electrospray ionization(ESI) Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR MS). The acid class(heteroatom number), type(z numbers) and carbon number distributions were positively characterized. It was found out that the total acid number(TAN) of SL crude decreased after true boiling point distillation, and the abundance of O2 class in mass spectra was also found to be reduced from 67.6% to 34.5% in SL TBP mixed crude as measured by MS spectra, indicating to a potential carboxylic acid decomposition. However, it was interesting that the carboxylic acids type distribution in both oils was almost the same although their relative abundance in SL TBP mixed crude turned to be much lower, suggesting that various petroleum carboxylic acid types have the similar thermal decomposition reaction behavior. Furthermore, for each O2 type of acids in SL TBP mixed crude, the abundance of carboxylic acids with carbon number higher than 35 was reduced greatly, especially for those with carbon number higher than 60, the mass peaks of which were nearly totally removed, indicating that the large carboxylic acid molecules in heavy fractions decomposed more significantly because of longer heating time during the true boiling point distillation process. 6Spatial-temporal Evolution and Determinants of the Belt and Road Initiative: A Maximum Entropy Gravity Model Approach显示文摘The spatial interaction model is an effective way to explore the geographical disparities inherent in the Belt and Road Initiative(BRI) by simulating spatial flows. The traditional gravity model implies the hypothesis of equilibrium points without any reference to when or how to achieve it. In this paper, a dynamic gravity model was established based on the Maximum Entropy(MaxEnt) theory to estimate and monitor the interconnection intensity and dynamic characters of bilateral relations. In order to detect the determinants of interconnection intensity, a Geodetector method was applied to identify and evaluate the determinants of spatial networks in five dimensions. The empirical study clearly demonstrates a heterogeneous and non-circular spatial structure. The main driving forces of spatial-temporal evolution are foreign direct investment, tourism and railway infrastructure construction, while determinants in different sub-regions show obvious spatial differentiation. Southeast Asian countries are typically multi-island area where aviation infrastructure plays a more important role. North and Central Asian countries regard oil as a pillar industry where power and port facilities have a greater impact on the interconnection. 8Multi-stage Dynamic Post-disaster Recovery Strategy for Distribution Networks Considering Integrated Energy and Transportation Networks显示文摘To improve the resilience of distribution networks(DNs),a multi-stage dynamic recovery strategy is proposed in this paper,which is designed for post-disaster DN considering an integrated energy system(IES)and transportation network(TN).First,the emergency response quickly increases the output of gas turbines(GTs)in the natural gas network(NGN),and responsively reconfigures the DN in microgrids,to maximize the amount of loads to be restored.The single-commodity flow model is adopted to construct spanning tree constraints.Then,in the second stage of energy storage recovery,mobile energy storage systems(MESSs)are deployed to cover the shortages of power demands,i.e.,to further restore the loads after evaluating the load recovery situation.The Floyd algorithm based dynamic traffic assignment(DTA)is selected to obtain the optimal path of the MESSs.In the third stage,the outputs of various post-disaster recovery measures are adjusted to achieve an economically optimized operation.Case studies demonstrate the effectiveness of the proposed dynamic post-disaster recovery strategy.Xinyi Jiang Jian Chen Ming Chen Zhen Wei 2021CSEE Journal of Power and Energy Systems2021,7,2:6
9Vertical displacement loading tides and self-attraction and loading tides in the Bohai, Yellow, and East China Seas显示文摘The loading tides are calculated by means of the Green's function method based on a high-resolution regional ocean tide model, the TOPO7.0 global ocean tide model, and the Gutenberg-Bullen A Earth model. The results show that the maximal amplitude of M2 vertical displacement loading (VDL) tide in the Bohai, Yellow, and East China Seas exceeding 28mm appears 150km off the Zhejiang coast; the second maximum exceeding 20mm appears in Inchon Bay; and the third maximum exceeding 14mm is located in the northeast of the North Yellow Sea. The maximal amplitudes of S2 VDL tide at the above three locations exceed 10, 8, and 4mm, respectively. The maximal amplitudes of the K1 and O1 VDL tides, exceeding 13 and 10 mm respectively, appear near the central and north Ryukyu Islands; the amplitudes tend to decease toward the inward areas. The phases of semidiurnal VDL tides are basically opposite to those of corresponding ocean tides. The phases of diurnal VDL tides are basically opposite to those of corresponding ocean tides in the most part of the East China Sea and the eastern part of the South Yellow Sea. This anti-phase relationship generally does not hold in the rest parts of the Bohai and Yellow Seas. The distribution patterns of self-attraction and loading (SAL) tides are very similar to those of VDL tides. The SAL tides have amplitudes about 1.2-1.7 times of the corresponding VDL tides and their phases are basically opposite to the corresponding VDL tides. 11Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
12Cotidal charts and tidal power input atlases of the global ocean from TOPEX/Poseidon and JASON-1 altimetry显示文摘The global distributions of eight principal tidal constituents, M_2 , S_2 , K_1 , O_1 , N_2 , K_2 , P_1 , and Q_1 , are derived using TOPEX/Poseidon and JASON-1(T/P-J) satellite altimeter data for 16 a. The intercomparison of the derived harmonics at 7000 subsatellite track crossover points shows that the root mean square (RMS) values of the tidal height differences of the above eight constituents range from 1.19 cm to 2.67 cm, with an average of about 2 cm. The RMS values of the tidal height differences between T/P-J solutions and the harmonics from ground measurements at 152 tidal gauge stations for the above constituents range from 0.34 cm to 1.08 cm, and the relative deviations range from 0.031 to 0.211. The root sum square of the RMS differences of these eight constituents is 2.12 cm, showing the improvement of the present model over the existing global ocean tidal models. Based on the obtained tidal model the global ocean tidal energetics is studied and the global distribution of the tidal power input density by tide-generating force of each constituent is calculated, showing that the power input source regions of semidiurnal tides are mainly concentrated in the tropical belt between 30 S and 30 N, while the power input source regions of diurnal tides are mainly concentrated off the tropic oceans. The global energy dissipation rates of the M_2 , S_2 , K_1 , O_1 , N_2 , P_1 , K_2 and Q_1 tides are 2.424, 0.401, 0.334, 0.160, 0.113, 0.035, 0.030 and 0.006 TW, respectively. 13Statistical Description of Debris Particle Size Distribution in Electrical Discharge Machining显示文摘Gap debris as discharge product is closely related to machining process in electrical discharge machining(EDM). A lot of recent researches have focused on the relationship among debris size, surfaces texture, remove rate, and machining stability. The study on statistical distribution of debris size contributes to the research, but it is still superficial currently. In order to obtain the distribution law of the debris particle size, laser particle size analyzer(LPSA) combined with scanning electron microscope(SEM) is used to analyze the EDM debris size. Firstly, the heating dried method is applied to obtain the debris particles. Secondly, the measuring range of LPSA is determined as 0.5-100 μm by SEM observation, and the frequency distribution histogram and the cumulative frequency distribution scattergram of debris size are obtained by using LPSA. Thirdly, according to the distribution characteristic of the frequency distribution histogram, the statistical distribution functions of lognormal, exponentially modified Gaussian(EMG), Gamma and Weibull are chosen to achieve curve fitting of the histogram. At last, the distribute law of the debris size is obtained by fitting results. Experiments with different discharge parameters are carried out on an EDM machine designed by the authors themselves, and the machining conditions are tool electrode of red-copper material, workpiece of ANSI 1045 material and working fluid of de-ionized water. The experimental results indicate that the debris sizes of all experiment sample truly obey the Weibull distribution. 14A novel intronic circular RNA,circGNG7,inhibits head and neck squamous cell carcinoma progression by blocking the phosphorylation of heat shock protein 27 at Ser78 and Ser82显示文摘Background:There is increasing evidence that circular RNAs(circRNAs)play a significant role in pathological processes including tumorigenesis.In contrast to exonic circRNAs,which are the most frequently reported circRNAs in cancer so far,the studies of intronic circRNAs have been greatly lagged behind.Here,we aimed to investigate the regulatory role of intronic circRNAs in head and neck squamous cell carcinoma(HNSCC).Methods:We conducted whole-transcriptome sequencing with four pairs of primary tumor tissues and adjacent normal tissues from HNSCC patients.Then,we characterized circGNG7 expression in HNSCC tissues and cell lines and explored its association with the prognosis of HNSCC patients.We also identified interactions between circGNG7 and functional proteins,which alter downstream signaling that regulate HNSCC progression.Results:In this study,we identified a new intronic circRNA,circGNG7,and validated its functional roles in HNSCC progression.CircGNG7 was predominately localized to the cytoplasm,and its expression was downregulated in both HNSCC tissues andCAL27,CAL33,SCC4,SCC9,HN6,and HN30 cells.Low expression of circGNG7 was significantly correlated with poor prognosis in HNSCC patients.Consistent with this finding,overexpression of circGNG7 strongly inhibited tumor cell proliferation,colony formation,in vitro migration,and in vivo tumor growth.Mechanistically,the expression of circGNG7 in HNSCC cells was regulated by the transcription factor SMAD family member 4(SMAD4).Importantly,we discovered that circGNG7 could bind to serine residues 78 and 82 of the functional heat shock protein 27(HSP27),occupying its phosphorylation sites and hindering its phosphorylation,which reduced HSP27-JNK/P38 mitogen-activated protein kinase(MAPK)oncogenic signaling.Downregulation of circGNG7 expression in HNSCC increased HSP27-JNK/P38 MAPK signaling and promoted tumor progression.Conclusions:Our results revealed that a new intronic circRNA,circGNG7,functions as a strong tumor suppressor and that circGNG7/HSP27-JNK/P38 MAPK signaling is a novel mechanism by which HNSCC progression can be controlled.Houyu Ju Zhenrong Hu Dongliang Wei Jinyun Huang Xinyi Zhang Mengyu Rui Zhi Li Xiaomeng Zhang Jingzhou Hu Wei Guo Guoxin Ren 2021Cancer Communications2021,41,11:4
15V2X-Communication Assisted Interference Minimization for Automotive Radars显示文摘With the development of automated driving vehicles, more and more vehicles will be fitted with more than one automotive radars, and the radar mutual interference will become very significant. Vehicle to everything (V2X) communication is a potential way for coordinating automotive radars and reduce the mutual interference. In this paper, we analyze the positional relation of the two radars that interfere with each other, and evaluate the mutual interference for different types of automotive radars based on Poisson point process (PPP). We also propose a centralized framework and the corresponding algorithm, which relies on V2X communication systems to allocate the spectrum resources for automotive radars to minimize the interference. The minimum spectrum resources required for zero-interference are analyzed for different cases. 17Adaptive cavity-enhanced dual-comb spectroscopy显示文摘Resolution and bandwidth are critical for cavity-enhanced dual-comb spectroscopy(CE-DCS). Here, we pioneer an adaptive approach in CE-DCS to improve the broadband as well as the resolution. Postcorrections to dualcomb interferograms adaptively compensate the relative phase jitters of the optical frequency combs and result in both a mode-resolved spectral resolution and a signal-to-noise ratio of 440:1 in 1 s. Meanwhile, an adaptive combcavity locking scheme exploits more than 90% of the comb modes, covering 340 cm-1(10 THz) at 6450 cm-1. For a single dual-comb interferogram, more than 40,000 comb teeth spaced by 250 MHz are measured in less than7.5 ms, contributing to a noise equivalent absorption per spectral element of 2 × 10-10 cm-1·Hz-1∕2. 18Surface Oxygen Injection in Tin Disulfide Nanosheets for Efficient CO_(2) Electroreduction to Formate and Syngas显示文摘Surface chemistry modification represents a promising strategy to tailor the adsorption and activation of reaction intermediates for enhancing activity.Herein,we designed a surface oxygen-injection strategy to tune the electronic structure of SnS_(2) nanosheets,which showed effectively enhanced electrocatalytic activity and selectivity of CO_(2) reduction to formate and syngas(CO and H_(2)).The oxygen-injection SnS_(2) nanosheets exhibit a remarkable Faradaic efficiency of 91.6%for carbonaceous products with a current density of 24.1 mA cm^(−2) at−0.9 V vs RHE,including 83.2%for formate production and 16.5%for syngas with the CO/H_(2) ratio of 1:1.By operando X-ray absorption spectroscopy,we unravel the in situ surface oxygen doping into the matrix during reaction,thereby optimizing the Sn local electronic states.Operando synchrotron radiation infrared spectroscopy along with theoretical calculations further reveals that the surface oxygen doping facilitated the CO_(2) activation and enhanced the affinity for HCOO*species.This result demonstrates the potential strategy of surface oxygen injection for the rational design of advanced catalysts for CO_(2) electroreduction.Tao Chen Tong Liu Tao Ding Beibei Pang Lan Wang Xiaokang Liu Xinyi Shen Sicong Wang Dan Wu Dong Liu Linlin Cao Qiquan Luo Wei Zhang Wenkun Zhu Tao Yao 2021Nano-Micro Letters2021,13,11:3

20Flotillin1 promotes EMT of human small cell lung cancer via TGF-β signaling pathway显示文摘Objective:Small cell lung carcinoma(SCLC)is considered one of the most aggressive types of lung cancer due to its rapid growth and early metastasis.No tumor markers or therapeutic targets have been demonstrated to be specific or effective in SCLC to date.This study aims to evaluate the potential of Flotillin1(Flot1)as a target of SCLC treatment.Methods:Flot1 expression level in the tissue of SCLC and other tissue of lung disease was detected using immunohistochemical staining.Transwell and Matrigel assays were employed to examine migration and invasion of cancer cells.Flow cytometry and xCELLigence system were used to evaluate cell apoptosis and cell viability,respectively.Expression levels of Flot1,epithelialmesenchymal transition(EMT)marker E-cadherin,vimentin,cyclinD1,TGF-β-Smad2/3,and p-AKT were examined using Western blot.Furthermore,xenograft tumor in nude mice was used to evaluate the growth and metastasis of NCI-H446 cells in vivo.Results:Our results demonstrated that Flot1 is highly expressed in SCLC samples and that its expression correlates strongly with clinical stage,distant metastasis,and poor survival.The knockdown of Flot1 decreased the growth,migration,and invasiveness of SCLC cells and reversed EMT phenotype in vitro and in vivo,while enhanced Flot1 expression exhibited the opposite behavior.Gene expression profile analysis demonstrated that Flot1-regulated genes frequently mapped to the AKT and TGF-β-Smad2/3pathways.Our results further revealed that Flot1 affected the progression of SCLC via regulation of EMT progression.Conclusions:These findings indicated an oncogenic role of Flot1 via promoting EMT in SCLC and suggested its potential as a tumor marker and prognostic indicator.Lianmei Zhao Jie Li Yueping Liu Wei Zhou Yanan Shan Xinyi Fan Xinliang Zhou Baoen Shan Yongmei Song Qimin Zhan 2018Cancer Biology & Medicine2018,15,4:3
