The Glutamatergic Postrhinal Cortex–Ventrolateral Orbitofrontal Cortex Pathway Regulates Spatial Memory Retrieval显示文摘A deficit in spatial memory has been taken as an early predictor of Alzheimer’s disease(AD) or mild cognitive impairment(MCI). The uncinate fasciculus(UF) is a long-range white-matter tract that connects the anterior temporal lobe with the orbitofrontal cortex(OFC)in primates. Previous studies have shown that the UF impairment associated with spatial memory deficits may be an important pathological change in aging and AD, but its exact role in spatial memory is not well understood. The pathway arising from the postrhinal cortex(POR) and projecting to the ventrolateral orbitofrontal cortex(vlOFC)performs most of the functions of the UF in rodents.Although the literature suggests an association between spatial memory and the regions connected by the POR–vlOFC pathway, the function of the pathway in spatialmemory is relatively unknown. To further illuminate the function of the UF in spatial memory, we dissected the POR–vlOFC pathway in mice. We determined that the POR–vlOFC pathway is a glutamatergic structure, and that glutamatergic neurons in the POR regulate spatial memory retrieval. We also demonstrated that the POR–vlOFC pathway specifically transmits spatial information to participate in memory retrieval. These findings provide a deeper understanding of UF function and dysfunction related to disorders of memory, as in MCI and AD. | Xinyang Qi Zhanhong Jeff Du Lin Zhu Xuemei Liu Hua Xu Zheng Zhou Cheng Zhong Shijiang Li Liping Wang Zhijun Zhang | 2019 | Neuroscience Bulletin2019,35,3: | 7

Impaired Parahippocampal Gyrus-Orbitofrontal Cortex Circuit Associated with Visuospatial Memory Deficit as a Potential Biomarker and Interventional Approach for Alzheimer Disease显示文摘The parahippocampal gyrus-orbitofrontal cortex(PHG-OFC)circuit in humans is homologous to the postrhinal cortex(POR)-ventral lateral orbitofrontal cortex(vlOFC)circuit in rodents. Both are associated with visuospatial malfunctions in Alzheimer’s disease(AD).However, the underlying mechanisms remain to be elucidated. In this study,we explored the relationship between an impaired POR-vlOFC circuit and visuospatial memory deficits through retrograde tracing and in vivo local field potential recordings in 5XFAD mice, and investigated alterations of the PHG-OFC circuit by multi-domain magnetic resonance imaging(MRI)in patients on the AD spectrum. We demonstrated that an impaired glutamatergic POR-vlOFC circuit resulted in deficient visuospatial memory in 5XFAD mice. Moreover,MRI measurements of the PHG-OFC circuit had an accuracy of 77.33%for the classification of amnestic mild cognitive impairment converters versus non-converters.Thus,the PHG-OFC circuit explains the neuroanatomical basis of visuospatial memory deficits in AD,thereby providing a potential predictor for AD progression and a promising interventional approach for AD. | Lin Zhu Zan Wang Zhanhong Du Xinyang Qi Hao Shu Duan Liu Fan Su Qing Ye Xuemei Liu Zheng Zhou Yongqiang Tang Ru Song Xiaobin Wang Li Lin Shijiang Li Ying Han Liping Wang Zhijun Zhang | 2020 | Neuroscience Bulletin2020,36,8: | 2
Differential transcriptomic landscapes of multiple organs from SARS-CoV-2 early infected rhesus macaques显示文摘SARS-CoV-2 infection causes complicated clinical manifestations with variable multi-organ injuries,how-ever,the underlying mechanism,in particular immune responses in different organs, remains elusive. In this study, comprehensive transcriptomic alterations of 14 tissues from rhesus macaque infected with SARS-CoV-2 were analyzed.Compared to normal controls,SARS-CoV-2 infection resulted in dysregulation of genes involving diverse functions in various examined tissues/organs,with drastic transcriptomic changes in cerebral cortex and right ventricle.Intriguingly, cerebral cortex exhibited a hyperinflammatory state evidenced by sig-nificant upregulation of inflammation response-related genes.Meanwhile, expressions of coagulation,angio-genesis and fibrosis factors were also up-regulated in cerebral cortex.Based on our findings, neuropilin 1(NRP1),a receptor of SARS-CoV-2,was significantly elevated in cerebral cortex post infection, accompanied by active immune response releasing inflammatory factors and signal transmission among tissues, which enhanced infection of the central nervous system(CNS)in a positive feedback way, leading to viral encephalitis.Overall, our study depicts a multi-tissue/organ tran-scriptomic landscapes of rhesus macaque with early infection of SARS-CoV-2,and provides important insights into the mechanistic basis for COVID-19-asso-ciated clinical complications. | Chun-Chun Gao Man Li Wei Deng Chun-Hui Ma Yu-Sheng Chen Yong-Qiao Sun Tingfu Du Qian-Lan Liu Wen-Jie Li Bing Zhang Lihong Sun Si-Meng Liu Fengli Li Feifei Qi Yajin Qu Xinyang Ge Jiangning Liu Peng Wang Yamei Niu Zhiyong Liang Yong-Liang Zhao Bo Huang Xiao-Zhong Peng Ying Yang Chuan Qin Wei-Min Tong Yun-Gui Yang | 2022 | Protein & Cell2022,13,12: | 1

Aminoacyl-tRNA Synthetase-based Prognosis Model and Exploration of Potential Mechanisms in Hepatocellular Carcinoma显示文摘Background and Aims:Aminoacyl-tRNA synthetases(ARSs)participate in tumor initiation and progression but their involvement in hepatocellular carcinoma(HCC)is not clear. This study aimed to investigate the prognostic value and underlying mechanisms of ARS in HCC.Methods:Data were obtained from The Cancer Genome Atlas(TCGA),International Cancer Genome Consortium,Gene Expression Omnibus and Human Protein Atlas databases. The prognostic model was constructed with the use of Cox regression and least absolute shrinkage and selection operator regression.Kaplan-Meier survival analysis, enrichment analysis, single sample gene set enrichment analysis and tumor mutation burden calculation were performed with R to evaluate the model and explore the underlying mechanism.Wilcoxon tests were used for comparisons between groups.Results:Aspartyl-tRNA synthetase 2(DARS2),tyrosyl-tRNA synthetase 1(YARS1)and cysteinyl-tRNA synthetase 2(CARS2)were identified as prognostic biomarkers and enrolled in model construction. The area under receiver operating characteristic curve of the model was 0.775.The model was used to assign patients from TCGA into low-and high-risk groups. Those in the high-risk group had a worse prognosis(p<0.001).The clinical significance of the model was tested in different clinical subgroups.Genetic mutation analysis had a higher TP53 mutation frequency in high-risk group.Enrichment analysis and study of immune-related cells and molecules found that the high-risk group was characterized by immune-cell infiltration and immunosuppression states. Conclusions:A novel ARS family-based model of HCC prognosis was constructed.TP53 mutation frequency and immune-suppressive status accounted for a worse prognosis in patients included in the high-risk group. | Yixun Jin Xinyang Huang Zhuoxin Wang Hui Xu Qi Wang Lifu Wang | 2023 | Journal of Clinical and Translational Hepatology2023,11,4: | 0
