| 1 | Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. | 2 | CRISPR/Cas9-mediated correction of human genetic disease显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated(Cas) protein 9 system(CRISPR/Cas9) provides a powerful tool for targeted genetic editing. Directed by programmable sequence-specific RNAs,this system introduces cleavage and double-stranded breaks at target sites precisely. Compared to previously developed targeted nucleases, the CRISPR/Cas9 system demonstrates several promising advantages, including simplicity, high specificity,and efficiency. Several broad genome-editing studies with the CRISPR/Cas9 system in different species in vivo and ex vivo have indicated its strong potential, raising hopes for therapeutic genome editing in clinical settings. Taking advantage of non-homologous end-joining(NHEJ) and homology directed repair(HDR)-mediated DNA repair, several studies have recently reported the use of CRISPR/Cas9 to successfully correct disease-causing alleles ranging from single base mutations to large insertions. | 3 | Comparison of Effects of Mechanical Stretching on Osteogenic Potential of ASCs and BMSCs显示文摘Mechanical forces play critical roles in the development and remodeling processes of bone.As an alternative cell source for bone engineering,adipose-derived stem cells(ASCs)should be fully investigated for their responses to mechanical stress.Similarly,the osteogenic potential,stimulated by mechanical stress,should be compared with bone marrow stromal cells(BMSCs),which have been clinically used for bone tissue engineering.In this study,ASCs and BMSCs were osteogenic-induced for 48 hours,and then subjected to uniaxial mechanical stretching for 2 or 6 hours.Cell orientation,osteogenic regulatory genes,osteogenic genes and ALP activities were measured and compared between ASCs and BMSCs.ASCs could align in a perpendicular way to the direction of stretching stress,while BMSCs did not present a specific alignment.Both 2 and 6 hours mechanical stretching could enhance the mRNA expression of Osx and Runx2 in BMSCs and ASCs,while OCN mRNA only increased in ASCs after 6 hours mechanical loading.Mechanical stretching enhanced the BMP-2 mRNA expression in ASCs,while only after 6 hours of mechanical loading significantly increased the BMP-2 gene expression in BMSCs.Significant differences only exist between ASCs and BMSCs loaded at 2 hours of mechanical stretching.It is concluded that ASCs are more rapid responders to mechanical stress,and have greater potential than BMSCs in osteogenesis when stimulated by mechanical stretching,indicating their usefulness for bone study in a rat model. | Brian E. | 4 | A facile ionic liquid approach to prepare cellulose fiber with good mechanical properties directly from corn stalks显示文摘It is very difficult to directly spin the lignocellulose without pretreatment.Ionic liquids(ILs)are promising solvent to dissolve lignocellulose to prepare cellulose fiber.However,the degree of cellulose polymerization(DP)is reduced when lignocellulose is dissolved in ILs,and the lignin removal rate is low.The elongation at break and tensile strength of the fibers obtained by spinning the lignocellulose dissolved in ILs are poor.In this paper,preparing cellulose fiber directly from lignocellulose based on dissolving corn stalk via[C4mim]Cl-L-arginine binary system is achieved.It shows that the removal rate of lignin can reach 92.35%and the purity of cellulose can reach 85.32%after corn stalk was dissolved at 150℃C for 11.5 h when the mass fraction of arginine is 2.5%.The elongation at break of fiber reached 10.12%and the tensile strength reached 420 MPa.It is mainly due to the fact that L-arginine not only inhibits the degradation of cellulose but also promotes the delignination.Without any pulping or pretreatment,preparing cellulose fibers via direct dissolution and extrusion may provide a simple and effective way to prepare many novel cellulose materials. | Jiming Yang Xingmei Lu Yanqiang Zhang Junli Xu Yongqing Yang Qing Zhou | 2020 | Green Energy & Environment2020,5,2: | 2 |

| 6 | Modeling the effect of water vaporization and salt precipitation on reservoir properties due to carbon dioxide sequestration in a depleted gas reservoir显示文摘Vaporization of water during both gas reservoir development and CO2 geological sequestration in saline formations can cause salt precipitation with rapid loss of formation porosity and permeability.Water vaporization and precipitation of halite around a single well from stage production to CO2 injection are studied to investigate the effect on reservoir properties in a gas reservoir.This paper identifies and quantifies post-flood dry zones and permeability changes in depleted gas reservoir after CO2 exposure with the comparison in gas production period by performing the numerical simulation with the compositional simulator.Simulation results indicate that water vaporization and salt precipitation occur during gas production,and can be intensified by CO2 injection.Dry supercritical CO2 injection vaporizes the brine promoting brine concentration and halite precipitation.The simulation indicates a drying area with a radius of 78 m around wellbore after CO2 injection.Porosity reduces with the most scope of 58%,and permeability can be decreased by up to 93.9%due to salt precipitation.And the injectivity is damaged by 99.77%at the end of injection period based on maximum permeability reduction.Moreover,six factors are investigated to conduct the influence analysis,showing that higher salinity brine,higher injection rate,higher irreducible water saturation,lower initial permeability,higher temperature and with capillary flow are conditions enhancing the salt precipitation due to dry CO2 injection. | Yong Tang Ruizhi Yang Xingmei Kang | 2018 | Petroleum2018,4,4: | 1 |

| 10 | Polydopamine functionalized mesoporous silica as ROS-sensitive drug delivery vehicles for periodontitis treatment by modulating macrophage polarization显示文摘Periodontitis is recognized as the major cause of tooth loss in adults, posing an adverse impact on systemic health. In periodontitis, excessive production of reactive oxygen species (ROS) at the inflamed site culminates in periodontal destruction. In this study, a novel ROS-responsive drug delivery system based on polydopamine (PDA) functionalized mesoporous silica nanoparticles was developed for delivering minocycline hydrochloride (MH) to treat periodontitis. The outer PDA layer and the inner MH of the nanoparticles acted as ROS scavengers and anti-inflammatory agents, respectively. Under the synergistic action of PDA and MH, macrophages were polarized from the pro-inflammatory M1 to the anti-inflammatory M2 phenotype. The in vitro experiments provided convincing evidence that PDA could scavenge ROS effectively, and the expression of pro-inflammatory cytokines was attenuated and the secretion of anti-inflammatory cytokines was enhanced through M1 to M2 polarization of macrophages with the cooperation of MH. In addition, the results obtained from the periodontitis rat models demonstrated that the synergetic effect of PDA and MH prevented alveolar bone loss without causing any adverse effect. | 12 | MgO-laden biochar enhances the immobilization of Cd/Pb in aqueous solution and contaminated soil显示文摘Great progress has been made in recent years to alleviate the heavy metal pollution,but the development of low-cost and eco-friendly adsorbents is still challenging.In this work,MgO-loaded biochar,as a potential adsorbent,was successfully synthesized via copyrolysis of corn straw and MgCl2∙6H2O at 600℃and used for heavy metal immobilization in aqueous solution and contaminated soil.Nemerow pollution index and potential ecological risk index methods were also used to assess the potential ecological risk of the metals in soil after remediation.The results showed MgO-laden biochar exhibited a much higher Cd2+adsorption capacity than the pristine biochar.Pseudo-second-order and Langmuir/Langmuir-Freundlich equations could describe the adsorption kinetics and isotherm of Cd2+well.The maximum adsorption capacity of Cd2+could reach to 1058.8 mg g−1 estimated by Langmuir-Freundlich equation.Cd2+adsorption on MgO-laden biochar composite mainly involved the hydrolyzation of MgO,ionization of Mg(OH)2 and precipitation of Cd2+and OH−.In addition,complexes with oxygen-containing groups and adsorption by Mg(OH)2 also enhanced Cd immobilization.The result of soil-leaching test showed that the concentrations of TCLP-leaching(Toxicity Characteristic Leaching Procedure)Cd and Pb reduced by 22.4%and 29.0%,respectively,after 4%of MgO-laden biochar amendment,and the integrated pollution index and potential ecological risk decreased by 28.9%and 28.5%,respectively.These results suggest that MgO-laden corn straw biochar may be promising for application as a low-cost adsorbent for wastewater treatment and soil remediation. | Yan Wang Lu Wang Zhangtao Li Dong Yang Jianming Xu Xingmei Liu | 2021 | Biochar2021,3,2: | 0 |
