| 1 | Targeting the alternative bile acid synthetic pathway for metabolic diseases显示文摘The gut microbiota is profoundly involved in glucose and lipid metabolism,in part by regulating bile acid(BA)metabolism and affecting multiple BA-receptor signaling pathways.BAs are synthesized in the liver by multi-step reactions catalyzed via two distinct routes,the classical pathway(producing the 12α-hydroxylated primary BA,cholic acid),and the alternative pathway(producing the non-12α-hydroxylated primary BA,chenodeoxycholic acid).BA synthesis and excretion is a major pathway of cholesterol and lipid catabolism,and thus,is implicated in a variety of metabolic diseases including obesity,insulin resistance,and nonalcoholic fatty liver disease.Additionally,both oxysterols and BAs function as signaling molecules that activate multiple nuclear and membrane receptor-mediated signaling pathways in various tissues,regulating glucose,lipid homeostasis,inflammation,and energy expenditure.Modulating BA synthesis and composition to regulate BA signaling is an interesting and novel direction for developing therapies for metabolic disease.In this review,we summarize the most recent findings on the role of BA synthetic pathways,with a focus on the role of the alternative pathway,which has been under-investigated,in treating hyperglycemia and fatty liver disease.We also discuss future perspectives to develop promising pharmacological strategies targeting the alternative BA synthetic pathway for the treatment of metabolic diseases. | Wei Jia Meilin Wei Cynthia Rajani Xiaojiao Zheng | 2021 | Protein & Cell2021,12,5: | 17 |
| 2 | Gut microbiota alterations are distinct for primary colorectal cancer and hepatocellular carcinoma显示文摘Colorectal cancer(CRC)and hepatocellular carcinoma(HCC)are the second and third most common causes of death by cancer,respectively.The etiologies of the two cancers are either infectious insult or due to chronic use of alcohol,smoking,diet,obesity and diabetes.Pathological changes in the composition of the gut microbiota that lead to intestinal inflammation are a common factor for both HCC and CRC.However,the gut microbiota of the cancer patient evolves with disease pathogenesis in unique ways that are affected by etiologies and environmental factors.In this review,we examine the changes that occur in the composition of the gut microbiota across the stages of the HCC and CRC.Based on the idea that the gut microbiota are an additional“lifeline”and contribute to the tumor microenvironment,we can observe from previously published literature how the microbiota can cause a shift in the balance from normal→inflammation→diminished inflammation from early to later disease stages.This pattern leads to the hypothesis that tumor survival depends on a less proinflammatory tumor microenvironment.The differences observed in the gut microbiota composition between different disease etiologies as well as between HCC and CRC suggest that the tumor microenvironment is unique for each case. | Wei Jia Cynthia Rajani Hongxi Xu Xiaojiao Zheng | 2021 | Protein & Cell2021,12,5: | 14 |
| 3 | Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. | 4 | Comparative efficacy and safety of Chinese herbal injections combined with transcatheter hepatic arterial chemoembolization in treatment of liver cancer: a bayesian network Meta-analysis显示文摘OBJECTIVE: To address the optimal Chinese herbal injections(CHIs) against liver cancer, the present network Meta-analysis is designed to investigate the comparative efficacy and safety of different CHIs.METHODS: Several electronic databases were searched up to June 1 st, 2017. The quality assessment was conducted and network Meta-analysis was performed to compare the efficacy and safety of different CHIs plus transcatheter hepatic arterial chemoembolization(TACE). Primary outcomes were 1-year and 2-year survival rate, the secondary outcomes includes the clinical effective rate, performance status and the adverse reactions(ADRs). Data analysis was applied Stata 13.0 and Win BUGS 1.4 software.RESULTS: A total of 105 randomized controlled trials(RCTs) were identified for inclusion in this analysis, with data for 7683 patients and 13 CHIs. The results suggested that Javanica oil emulsion,Huachansu injection plus TACE were more favorable for 1-year and 2-year survival rate than other CHIs. | 5 | Calorie restriction and its impact on gut microbial composition and global metabolism显示文摘Calorie restriction (CR)is a dietary regimen that reduces calorie intake without incurring malnutrition or a reduction in essential nutrients.It has long been recognized as a natural strategy for promoting health, extending longevity,and prevents the development of metabolic and age-related diseases.In the present review,we focus on the general effect of CR on gut microbiota composition and global metabolism.We also propose mechanisms for its beneficial effect.Results showed that probiotic and butyrate-producing microbes increased their relative abundance,whereas proinflammatory strains exhibited suppressed relative abundance following CR.Analyses of the gut microbial and host metabolisms revealed that most host microbial co-metabolites were changed due to CR.Examples of dramatic CR-induced changes in host metabolism included a decrease in the rate of lipid biosynthesis and an increase in the rates of fatty acid catabolism,p-oxidation,glycogenolysis,and gluconeogenesis.The observed phenotypes and the further verification of the direct link between gut microbiota and metabolome may benefit patients that are at risk for developing metabolic disease.Thus,improved gut microbiota composition and metabolome are potential biomarkers for determining the effectiveness of dietary interventions for age-related and metabolic diseases. | Xiaojiao Zheng Shouli Wang Wei Jia | 2018 | Frontiers of Medicine2018,12,6: | 3 |
| 6 | An Estimation Method for Multi-channel EEG Data Based on Canonical Correlation Analysis显示文摘Electroencephalogram(EEG) signal is often contaminated by electronic noise as well as movement artifacts. This paper presented an algorithm based on Canonical correlation analysis(CCA) to estimate multichannel EEG data. Different from previous studies, in which CCA was mainly used to detect the invariant features specific to each brain state, in this paper, the canonical variates computed by CCA were used to reconstruct the multi-channel EEG data. Firstly, two data sets, EEG signals and the reference signals based on prior knowledge were constructed. Next, canonical variates were computed by pro jecting the two data sets onto basis vectors. Finally,a least squares solution was used to estimate the multichannel EEG data. The experiment results suggested that the algorithm is capable of reconstructing the actual specific components with high quality. | 10 | Ultrathin poly(cyclocarbonate-ether)-based composite electrolyte reinforced with high-strength functional skeleton显示文摘Composite polymer electrolytes(CPEs)are considered to be the most promising to break through the performance and safety limitations of traditional lithium-ion batteries because of their excellent electrochemical and mechanical properties.Aiming at the performance limitations of the most common polyether matrix such as poly(ethylene oxide)(PEO),a novel poly(cyclocarbonate-ether)polymer matrix was prepared by in-situ thermal curing,the weaker interaction between its C=O bond and Li^(+)can promote the rapid transport of Li^(+).Adding ionic liquid and active filler LLZTO to the matrix can synergistically reduce the crystallinity of matrix and promote the dissociation of lithium salts.In addition,a 3D functional skeleton made of polyacrylonitrile(PAN)and lithium fluoride(LiF)can greatly improve the mechanical strength of polymer matrix after cold pressing,and Li F is also conducive to interface stability.The thickness of the optimal sample(VP6L/CPL)was only 25μm,and its ionic conductivity,lithium ion transference number,and electrochemical stability window were as high as 7.17×10^(-4)S cm^(-1)(25℃),0.54 and 5.4 V,respectively,while the mechanical strength reaches 6.1 MPa,which can fully inhibit the growth of lithium dendrites.The excellent electrochemical performance and mechanical strength enable the assembled Li|VP6L/CPL|Li battery to be continuously charged for over 200 h and cycled stably for more than 2300 h,and Li|VP6L/CPL|LFP battery can be stably cycled for more than 400 and 550 cycles at 1 C(40℃)and 0.5 C(25℃),respectively. | Xiaojiao Zheng Da Xu Ning Fu Zhenglong Yang | 2023 | Journal of Energy Chemistry2023,,6: | 1 |

| 16 | Hot Deformation Behaviors in Ti-6Al-4V/(TiB+TiC)Composites显示文摘Thermal compression testing was investigated using the Gleeble 3800 thermal simulator,and thermal deformation behavior of particle-reinforced titanium matrix composites(TMCs)was studied under deformation temperatures of 750-900°C,strain rates of 0.001-1 s^(-1),and experimental deformation of 60%.According to obtained flow stress curves,the hot deformation characteristics were analyzed.Based on the Arrhenius hyperbolic sinusoidal model,the constitutive equation at high temperature was established.Based on the theory of dynamic material models,a hot processing map of TMCs at high temperature was established,and the peak region of power dissipation rate and the instability region in the hot processing map were both determined.At the same time,the corresponding microstructures in the peak power dissipation rate and rheological instability regions were observed.The results showed that flow stress decreased with increasing deformation temperature and increased with increasing strain rate.The thermal deformation activation energy of titanium matrix composites was 301.8 kJ/mol.The Ti-6Al-4V/(TiB+TiC)composites possessed only one instability zone under high-temperature compression at a strain of 0.5,with corresponding temperatures at 750-840°C and strain rates at 0.1-1 s^(-1).The optimal thermal deformation parameters included corresponding temperatures of 830-880°C and strain rates of 0.001-0.05 s^(-1).The microstructures corresponding to optimal hot working parameters in processing maps were more homogeneous than the microstructures in the instability zone,including the distribution uniformity of reinforcement and the degree of dynamic recrystallization,and no instability phenomena including abnormal grain growth,microcracks or intensive fracture of reinforcements were found,indicating that the hot processing map had a positive guiding effect on the option of desirable material thermal-working parameters. | Xuejian Lin Hongjun Huang Fuyu Dong Yue Zhang Xiaoguang Yuan Bowen Zheng Xiaojiao Zuo | 2021 | Acta Metallurgica Sinica(English Letters)2021,34,12: | 0 |
