| 5 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 6 | Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. | 7 | Selective logging enhances ecosystem multifunctionality via increase of functional diversity in a Pinus yunnanensis forest in Southwest China显示文摘Background:The impacts of selective logging on ecosystem multifunctionality(EMF)remain largely unexplored.In this study,we analyzed the response of nine variables related to four ecosystem functions(i.e.nutrient cycling,soil carbon stocks,decomposition,and wood production)to five selective logging intensities in a Pinus yunnanensisdominated forest.We included a control group with no harvest to evaluate the potential shifts in EMF of the P.yunnanensis forests.We also assessed the relationship between above-and belowground biodiversity and EMF under these different selective logging intensities.Additionally,we evaluated the effects of biotic and abiotic factors on EMF using a structural equation modeling(SEM)approach.Results:Individual ecosystem functions(EFs)all had a significant positive correlation with selective logging intensity.Different EFs showed different patterns with the increase of selective logging intensity.We found that EMF tended to increase with logging intensity,and that EMF significantly improved when the stand was harvested at least twice.Both functional diversity and soil moisture had a significant positive correlation with EMF,but soil fungal operational taxonomic units(OTUs)had a significant negative correlation with EMF.Based on SEM,we found that selective logging improved EMF mainly by increasing functional diversity.Conclusion:Our study demonstrates that selective logging is a good management technique from an EMF perspective,and thus provide us with potential guidelines to improve forest management in P.yunnanensis forests in this region.The functional diversity is maximized through reasonable selective logging measures,so as to enhance EMF. | Xiaobo Huang Shuaifeng Li Jianrong Su | 2020 | Forest Ecosystems2020,7,4: | 3 |

| 9 | Drug-eluting intraocular lens with sustained bromfenac release for conquering posterior capsular opacification显示文摘Cataract is the leading cause of visual impairment,and posterior capsular opacification(PCO)is the most common long-term complication of modern cataract surgery,which can cause severe visual impairment after surgery.The proliferation,migration,and epithelial-mesenchymal transition(EMT)of residual lens epithelial cells(LECs)stimulated by growth factors and cytokines,are the key pathological mechanisms involved in the development of PCO.This study demonstrated that non-steroidal anti-inflammatory drug(NSAID),bromfenac,was capable of effectively inhibiting cell migration,overexpression of EMT markers,such as fibronectin(FN),matrix metalloproteinase 2(MMP2),α-smooth muscle actin(α-SMA),and transcription factor Snail,and extracellular signal-regulated kinase(ERK)/glycogen synthase kinase-3β(GSK-3β)signaling induced by transforming growth factor-β2(TGF-β2)in vitro.The inhibitory effect of bromfenac on TGF-β2-induced EMT was also verified on a primary lens epithelial cell model using human anterior capsules.Furthermore,based on ultrasonic spray technology,we developed a drug-eluting intraocular lens(IOL)using poly(lactic-co-glycolic acid)(PLGA)with sustained bromfenac release ability for the prevention of PCO development.In the rabbit models of cataract surgery,bromfenac-eluting IOL exhibited remarkable PCO prevention and inflammation suppression effects with excellent biocompatibility.In conclusion,bromfenac can inhibit TGF-β2-induced cell migration and the EMT of LECs via ERK/GSK-3β/Snail signaling.The present study offers a novel approach for preventing PCO through PLGA-based drug sustained-release IOLs. | Xiaobo Zhang Kairan Lai Su Li Jing Wang Jiayong Li Wei Wang Shuang Ni Bing Lu Andrzej Grzybowski Jian Ji Haijie Han Ke Yao | 2022 | Bioactive Materials2022,7,3: | 2 |

In this paper, V2 I channel measurements at 5.92 GHz are conducted in typical urban and highway scenarios.The frequency and bandwidth of transmission, as well as the deployment of the RSU(roadside unit) and the OBU(on board unit), are selected by considering the recommendation proposed by 3 GPP TR 36.885. Then,based on the measured data, the key channel characteristic parameters of the V2 I channel are extracted,including path loss, root-mean-square delay spread,stationarity distance, and Doppler spread, etc. Also,the statistical characteristics of the parameters, including time-varying and Doppler characteristics, are investigated and characterized.
