Inflammasomes as therapeutic targets in human diseases显示文摘Inflammasomes are protein complexes of the innate immune system that initiate inflammation in response to either exogenous pathogens or endogenous danger signals.Inflammasome multiprotein complexes are composed of three parts:a sensor protein,an adaptor, and pro-caspase-1.Activation of the inflammasome leads to the activation of caspase-1,which cleaves pro-inflammatory cytokines such as IL-ip and IL-18,leading to pyroptosis.Effectors of the inflammasome not only provide protection against infectious pathogens, but also mediate control over sterile insults.Aberrant inflammasome signaling has been implicated in the development of cardiovascular and metabolic diseases, cancer, and neurodegenerative disorders. Here,we review the role of the inflammasome as a double-edged sword in various diseases, and the outcomes can be either good or bad depending on the disease,as well as the genetic background. We highlight inflammasome memory and the two-shot activation process. We also propose the M-and N-type inflammation model, and discuss how the inflammasome pathway may be targeted for the development of novel therapy. | Yangxin Li Hui Huang Bin Liu Yu Zhang Xiangbin Pan Xi-Yong Yu Zhenya Shen Yao-Hua Song | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 3
Stem cell-derived exosomes repair ischemic muscle injury by inhibiting the tumor suppressor Rbl-mediated NLRP3 inflammasome pathway显示文摘Dear Editor,Lower-limb ischemia is a serious clinical condition affecting many patients world-wide and there is no effective therapy.Ischemia activates the NLRP3 inflammasome, which triggers tissue damage by releasing inflammatory cytokines including IL-1β and IL-18.1 However, the molecular mechanisms underlying activation of the NLRP3 inflammasome remain largely unknown. | Yanli Wang Wenping Xie Bin Liu Hui Huang Wei Luo Yu Zhang Xiangbin Pan Xi-Yong Yu Zhenya Shen Yangxin Li | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 3
A fully biodegradable polydioxanone occluder for ventricle septal defect closure显示文摘Ventricular septal defect(VSD)is one of the commonest congenital heart diseases(CHDs).Current occluders for VSD treatment are mainly made of nitinol, which has the risk of nickel allergy, persistent myocardial abrasion and fatal arrythmia.Herein,a fully biodegradable polydioxanone(PDO)occluder equipped with a shape line and poly-L-lactic acid PLLA membranes is developed for VSD closure without the addition of metal marker.PDO occluder showed great mechanical strength, fatigue resistance, geometry fitness, biocompatibility and degradability. In a rat subcutaneous implantation model,PDO filaments significantly alleviated inflammation response, mitigated fibrosis and promoted endothelialization compared with nitinol. The safety and efficacy of PDO occluder were confirmed in a canine VSD model with 3-year follow-up,demonstrating the biodegradable PDO occluder could not only effectively repair VSD,induce cardiac remodeling but also address the complications associated with metal occluders.Furthermore,a pilot clinical trial with five VSD patients indicated that all the occluders were successfully implanted under the guidance of echocardiography and no adverse events occurred during the 3-month follow-up.Collectively,the fully bioresorbable PDO occluder is safe and effective for clinical VSD closure and holds great promise for the treatment of structural CHDs. | Zefu Li Pengxu Kong Xiang Liu Shuyi Feng Wenbin Ouyang Shouzheng Wang Xiaopeng Hu Yongquan Xie Fengwen Zhang Yuxin Zhang Rui Gao Weiwei Wang Xiangbin Pan | 2023 | Bioactive Materials2023,,6: | 1
Performance of Cooperative Spatial Modulation with AF Protocol in Wireless Relay Network显示文摘By combing Spatial modulation(SM) with the Amplify-and-forward(AF) cooperative communication, a cooperative SM scheme with AF(AF-SM) protocol is presented, and the corresponding Bit error rate(BER)performance is investigated. Based on the performance analysis, the error probability of antenna index estimation(Pa) and the error probability of symbol estimation(Pd),which constitutes the overall average BER, are derived, respectively. As a result, tightly approximate closedform expressions of Paand Pdare attained, respectively.Using these expressions, the closed-form overall average BER is achieved. During the analysis, the computational complexity of AF-SM detector is also provided in terms of the complex addition and multiplication numbers.Besides, the asymptotic BER at high SNR and the corresponding diversity gain are further derived, and the resultant diversity gain of Nr+ 1 is obtained for the system with Nrreceive antennas. Simulation results show that our theoretical analysis is valid, and provides good performance evaluation method for cooperative SM system. Moreover, the BER performance can be effectively improved as Nrincreases due to higher diversity gain. | YU Xiangbin PAN Qing WANG Cheng | 2019 | Chinese Journal of Electronics2019,28,1: | 1
Influence of Sc Addition on Precipitation Behavior and Properties of Al-Cu-Mg Alloy显示文摘In this study, the Al-5.10Cu-0.65 Mg alloys without and with 0.2 wt% Sc addition were prepared to unveil the effect of Sc on the precipitate behavior and properties by atomic-scale scanning transmission electron microscopy. The results show that addition of Sc is able to significantly suppress the formation of the peak-hardening θ’ and S precipitates but in favor of stabilizing Guinier–Preston–Bagaryatsky(GPB) zones during isothermal aging at 180 ℃. The generation of unique GPB precursors dictating the evolution of the subsequent peak-hardening GPB zones is highly correlated with the insufficient vacancies and dragging effect of Sc to Cu in the Al-Cu-Mg-Sc alloy, thereby restricting the nucleation of θ′ and S precipitates owing to the decreased available Cu solutes. In property, 0.2 wt% Sc addition can not only enhance the corrosion resistance but also the electrical conductivity of the peak-hardening alloy. In addition, the peak-hardening Al-Cu-Mg-Sc alloy is also featured with increased strength without losing the ductility compared to its solid solution state. Further analyses indicate that the improved properties are essentially attributed to the formation of densely distributed strain-free GPB zones and smaller precipitate-free zones. The obtained results could provide a potential design strategy of high-performance alloys by Sc addition. | Xiangbin Han Shuangbao Wang Bo Wei Shuai Pan Guizhen Liao Weizhou Li Yuezhou Wei | 2022 | Acta Metallurgica Sinica(English Letters)2022,35,6: | 1
Ribosome biogenesis in disease:new players and therapeutic targets显示文摘The ribosome is a multi-unit complex that translates mRNA into protein.Ribosome biogenesis is the process that generates ribosomes and plays an essential role in cell proliferation, differentiation, apoptosis, development, and transformation. The mTORC1,Myc,and noncoding RNA signaling pathways are the primary mediators that work jointly with RNA polymerases and ribosome proteins to control ribosome biogenesis and protein synthesis.Activation of mTORC1 is required for normal fetal growth and development and tissue regeneration after birth.Myc is implicated in cancer development by enhancing RNA Pol II activity, leading to uncontrolled cancer cell growth. The deregulation of noncoding RNAs such as microRNAs,long noncoding RNAs,and circular RNAs is involved in developing blood, neurodegenerative diseases, and atherosclerosis. We review the similarities and differences between eukaryotic and bacterial ribosomes and the molecular mechanism of ribosome-targeting antibiotics and bacterial resistance. We also review the most recent findings of ribosome dysfunction in COVID-19 and other conditions and discuss the consequences of ribosome frameshifting,ribosome-stalling,and ribosome-collision.We summarize the role of ribosome biogenesis in the development of various diseases.Furthermore,we review the current clinical trials, prospective vaccines for COVID-19,and therapies targeting ribosome biogenesis in cancer, cardiovascular disease, aging, and neurodegenerative disease. | Lijuan Jiao Yuzhe Liu Xi-Yong Yu Xiangbin Pan Yu Zhang Junchu Tu Yao-Hua Song Yangxin Li | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 1
Performance Analysis and Power Allocation for Cooperative SSK System with Receive Correlation in Rayleigh Fading Channel显示文摘In this paper, the performance analysis of cooperative space shift keying(SSK)system with dual hop amplify and forward(AF)in receive correlated Rayleigh channel is presented.By means of the performance analysis,a closed form approximate expression of the average bit error rate(BER)is derived for the performance evaluation. With this expression,in a high signal to noise ratio(SNR)region,a tight closed form asymptotic BER of the system is also derived. It can simplify the calculation of average BER and provide effective evaluation for asymptotic performance.By minimizing this asymptotic BER expression,a suboptimum power allocation(PA)scheme is developed, and the closed form PA coefficients are obtained.Simulation indicates that the suboptimal PA scheme outperforms the conventional equal PA scheme, and its performance is very close to that of the exhaustive search based optimal PA scheme but with low complexity. Moreover, the system performance under the spatially correlated channel is worse than that in spatially independent channel due to the effect of spatial correlation. | YU Xiangbin LI Qiyishu HU Yaping Pan Qing | 2019 | Transactions of Nanjing University of Aeronautics and Astronautics2019,36,4: | 0
Outcomes of Self-Expanding Transcatheter Pulmonary Valves:Extended Follow-Up of a Prospective Trial显示文摘Background:The Venus-P valve was the first self-expanding valve used world-wide for transcatheter pulmonary valve replacement(TPVR)in patients with severe pulmonary regurgitation(PR).We intended to report the extended follow-up results from the prospective trial(No.NCT02590679).Methods:A total of 38 patients with severe PR(mean age 24.2±13.2)were included.Follow-up data were obtained after implanted at 1,6,and 12 months and yearly after. The frame geometry was assessed on post-implant computer tomography(CT)scanning by calculating the non-circularity[circularity ratio(minimum diameter/maximum diameter)<0.9]and under-expansion[expansion ratio(derived external valve area/nominal external valve area)<0.9).Adverse events(all-cause mortality, reintervention, valve dysfunction, stent fracture and endocarditis)were recorded. Results:All valves were implanted successfully with normal function at discharge.Geometric CT analysis showed underexpanded valve was detected in 22 patients(63%)and non-circular valve was seen in 16 patients(46%).During a median follow-up of 4.8 years(range 0.3-8.1),there were 1 death and 1 surgical explant, both resulting from endocarditis.Five-year freedom from valve dysfunction and stent fracture were 84.8%(95%CI 74.8-94.7)and 83.5%(95%CI 73.8-93.2).Endocarditis occurred in 3 patients at a median time of 7 months.Stent fracture was more common in patients with non-circularity stents. Conclusion:TPVR using Venus-P valve is associated with favorable outcomes at 5 years.Non-circular shapes in the valve level may have a higher risk of stent fracture. | Jingnan Zhang Junyi Wan Yihang Li Yu Han Jiahua Pan Fang Fang Shiliang Jiang Xiangbin Pan Gejun Zhang | 2023 | Congenital Heart Disease2023,18,2: | 0
Performance Analysis of AF Relaying Aided Space Shift Keying System with Imperfect Channel Estimation显示文摘Space shift keying (SSK) is a spectrally efficient and low-complexity technique that only uses antenna index to convey information.Combining SSK with cooperative communication, the transmission reliability of SSK system can be improved effectively. In this paper, considering imperfect channel information, the performance of cooperative SSK system with amplify-and-forward (AF) relaying protocol is investigated, and the effect of estimation error on the performance is analyzed.According to the performance analysis, the probability density function and moment generating function of effective signal-to-noise ratio are derived, respectively.Using these results, the closed-form expression of average bit error rate (BER) can be achieved.Meanwhile, the asymptotically approximated BER and the corresponding diversity order analysis are presented for the performance evaluation.By computer simulations, the validness of the presented theoretical analysis is verified, and the theoretical BERs with different estimation errors are shown to be close to those of the corresponding simulations. | Pan Qing Yu Xiangbin Hu Yaping Wang Xi Wang Cheng | 2018 | Transactions of Nanjing University of Aeronautics and Astronautics2018,35,6: | 0

The sex determination gene doublesex is required during adulthood to maintain sexual orientation显示文摘Gender differences in behavior are prevalent among animal kingdoms. The causal links among a particular gene, neural circuitry, and gendered behaviors have long been of particular interest in neuroscience. The fruit fly, Drosophila melanogaster, is a wonderful animal model with fruitful genetic tools to tackle these questions(Guo et al., 2019). It has been found that two sex determination genes, fruitless(fru) and doublesex(dsx), jointly govern the sex differences in the nervous system that regulate the male-specific courtship behavior(Burtis and Baker, 1989;Ito et al., 1996;Ryner et al. | Qionglin Peng Jiangtao Chen Xiangbin Su Rong Wang Caihong Han Yufeng Pan | 2022 | Journal of Genetics and Genomics2022,49,2: | 0
