| 5 | An overview of chronic myeloid leukemia and its animal models显示文摘Chronic myeloid leukemia(CML) is a form of leukemia characterized by the presence of clonal bone marrow stem cells with the proliferation of mature granulocytes(neutrophils, eosinophils, and basophils) and their precursors. CML is a type of myeloproliferative disease associated with a characteristic chromosomal translocation called the Philadelphia(Ph) chromosome or t(9;22) translocation(BCR-ABL). CML is now usually treated with targeted drugs called tyrosine kinase inhibitors(TKIs). The mechanism and natural history of CML is still unclear. Here, we summarize the present CML animal disease models and compare them with each other. Meanwhile, we propose that it is a very wise choice to establish zebrafish(Danio rerio) CML model mimics clinical CML. | 6 | SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX. | Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He | 2022 | Cellular & Molecular Immunology2022,19,1: | 4 |
| 7 | Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. | 9 | PM_(2.5)and water-soluble inorganic ion concentrations decreased faster in urban than rural areas in China显示文摘We investigated variations of PM_(2.5)and water-soluble inorganic ions chemical characteristics at nine urban and rural sites in China using ground-based observations.From 2015 to 2019,mean PM_(2.5)concentration across all sites decreased by 41.9μg/m~3with a decline of 46%at urban sites and 28%at rural sites,where secondary inorganic aerosol(SIAs)contributed to 21%(urban sites)and 17%(rural sites)of the decreased PM_(2.5).SIAs concentrations underwent a decline at urban locations,while sulfate(SO_(4)^(2–)),nitrate(NO_(3)^(–)),and ammonium(NH_(4)^(+))decreased by 49.5%,31.3%and 31.6%,respectively.However,only SO_(4)^(2–)decreased at rural sites,NO_(3)^(–)increased by 21%and NH_(4)^(+)decreased slightly.Those changes contributed to an overall SIAs increase in 2019.Higher molar ratios of NO_(3)^(–)to SO_(4)^(2–)and NH_(4)^(+)to SO_(4)^(2–)were observed at urban sites than rural sites,being highest in the heavily polluted days.Mean molar ratios of NH_(3)/NH_xwere higher in 2019 than 2015 at both urban and rural sites,implying increasing NH_xremained as free NH_(3).Our observations indicated a slower transition from sulfate-driven to nitrate-driven aerosol pollution and less efficient control of NO_(x)than SO_(2)related aerosol formation in rural regions than urban regions.Moreover,the common factor at urban and rural sites appears to be a combination of lower SO_(4)^(2–)levels and an increasing fraction of NO_(3)^(–)to PM_(2.5)under NH_(4)^(+)-rich conditions.Our findings imply that synchronous reduction in NO_(x)and NH_(3)emissions especially rural areas would be effective to mitigate NO_(3)^(–)-driven aerosol pollution. | Yangyang Zhang Aohan Tang Chen Wang Xin Ma Yunzhe Li Wen Xu Xiaoping Xia Aihua Zheng Wenqing Li Zengguo Fang Xiufen Zhao Xianlong Peng Yuping Zhang Jian Han Lijuan Zhang Jeffrey L.Collett Jr Xuejun Liu | 2022 | Journal of Environmental Sciences2022,34,12: | 2 |

| 12 | Dysregulated ceramides metabolism by fatty acid 2-hydroxylase exposes a metabolic vulnerability to target cancer metastasis显示文摘Whereas it is appreciated that cancer cells rewire lipid metabolism to survive and propagate,the roles of lipid metabolism in metastasis remain largely unknown.In this study,using esophageal squamous cell carcinoma(ESCC)as a pulmonary metastasis model,we find that the enzyme fatty acid 2-hydroxylase(FA2H),which catalyzes the hydroxylation of free fatty acids(FAs),is enriched in a subpopulation of ESCC cells with high metastatic potential,and that FA2H knockdown markedly mitigates metastatic lesions.Moreover,increased FA2H expression is positively associated with poor survival in patients with ESCC.Lipidomics analysis identifies that two dihydroceramides—Cer(d18:0/24:0)and Cer(d18:0/24:1)—are increased in FA2H-depleted metastasizing ESCC cells.Upon administration,Cer(d18:0/24:0)and Cer(d18:0/24:1)impair the formation of overt metastases in a mouse experimental metastasis model.Then,forkhead box protein C2(FOXC2)and FA2H are found to be co-upregulated in metastatic ESCC cell populations and ESCC specimens,and FA2H expression is further experimentally verified to be transcriptionally induced by FOXC2,which is boosted per se by tumour necrosis factorα(TNFα),a critical pro-metastasis cytokine in the tumour microenvironment,in metastasizing cells.Together,these results demonstrate that TNFα-FOXC2-FA2H is a novel signaling axis to promote metastasis,and its downstream dihydroceramide products could be promising drugs to intervene in metastasis. | Xuantong Zhou Furong Huang Gang Ma Wenqing Wei Nan Wu Zhihua Liu | 2022 | Signal Transduction and Targeted Therapy2022,7,11: | 2 |
