4Phytophthora sojae Effector PsAvh240 Inhibits Host Aspartic Protease Secretion to Promote Infection显示文摘Plants secrete defense molecules into the extracellular space (the apoplast) to combat attacking microbes. However, the mechanisms by which successful pathogens subvert plant apoplastic immunity remain poorly understood. In this study, we show that PsAvh240, a membrane-localized effector of the soybean pathogen Phytophthora sojae, promotes P. sojae infection in soybean hairy roots. We found that PsAvh240 interacts with the soybean-resistant aspartic protease GmAP1 in planta and suppresses the secretion of GmAP1 into the apoplast. By solving its crystal structure we revealed that PsAvh240 contain six a helices and two WY motifs. The first two a helices of PsAvh240 are responsible for its plasma membrane-localization and are required for PsAvh240's interaction with GmAP1. The second WY motifs of two PsAvh240 molecules form a handshake arrangement resulting in a handshake-like dimer. This dimerization is required for the effector's repression of GmAP1 secretion. 6Nitric oxide alleviates aluminum-induced oxidative damage through regulating the ascorbateglutathione cycle in roots of wheat显示文摘The possible association with nitric oxide(NO) and ascorbate-glutathione(As A-GSH) cycle in regulating aluminum(Al) tolerance of wheat(Triticum aestivum L.) was investigated using two genotypes with different Al resistance. Exposure to Al inhibited root elongation, and triggered lipid peroxidation and oxidation of As A to dehydroascorbate and GSH to glutathione disulfide in wheat roots. Exogenous NO significantly increased endogenous NO levels, and subsequently alleviated Al-induced inhibition of root elongation and oxidation of As A and GSH to maintain the redox molecules in the reduced form in both wheat genotypes. Under Al stress, significantly increased activities and gene transcriptional levels of ascorbate peroxidase, glutathione reductase, and dehydroascorbate reductase,were observed in the root tips of the Al-tolerant genotype Jian-864. Nitric oxide application enhanced the activity and gene transcriptional level of these enzymes in both wheat genotypes. g-Glutamylcysteine synthetase was not significantly Researaffected by Al or NO, but NO treatments increased the activity of glutathione peroxidase and glutathione S-transferase to a greater extent than the Al-treated wheat seedlings. 7A disintegrin and metalloproteinase with thrombospondin motif 1(ADAMTS1) expression increases in acute aortic dissection显示文摘Acute aortic dissection(AAD) is a life-threatening cardiovascular disease caused by progressive medial degeneration of the aortic wall. A disintegrin and metalloproteinase with thrombospondin motifs 1(ADAMTS1) is a recently identified extracellular metalloproteinase participating in the development of vascular disease, such as atherosclerosis. In the present study, we found that ADAMTS1 was significantly elevated in blood samples from AAD patients compared with patients with acute myocardial infarction and healthy volunteers. Based on these findings, we established an AAD model by infusing angiotensin II in older mice. AAD was successfully developed in aorta tissues, with an incidence of 42% after 14 days in the angiotensin II group. Macrophage and neutrophil infiltration was observed in the media of the aorta, and ADAMTS1 overexpression was found in the aorta by Western blot and immunohistochemistry. Double immunofluorescence staining showed the expression of ADAMTS1 in macrophages and neutrophils. Consistent with the upregulation of ADAMTS1 in aortic dissection tissues, versican(a proteoglycan substrate of ADAMTS1) was degraded significantly more in these tissues than in control aortic tissues. 8ZrC-ZrB_2-SiC ceramic nanocomposites derived from a novel single-source precursor with high ceramic yield显示文摘For the first time, ZrC-ZrB_2-SiC ceramic nanocomposites were successfully prepared by a single-source-precursor route, with allylhydridopolycarbosilane(AHPCS),triethylamine borane(TEAB),and bis(cyclopentadienyl) zirconium dichloride(Cp_2 ZrCl_2) as starting materials. The polymer-to-ceramic transformation and thermal behavior of obtained single-source precursor were characterized by means of Fourier transform infrared spectroscopy(FT-IR) and thermal gravimetric analysis(TGA). The results show that the precursor possesses a high ceramic yield about 85% at 1000 ℃.The phase composition and microstructure of formed ZrC-ZrB_2-SiC ceramics were investigated by means of X-ray diffraction(XRD) and high resolution transmission electron microscopy(HRTEM).Meanwhile, the weight loss and chemical composition of the resultant ZrC-ZrB_2-SiC nanocomposites were investigated after annealing at high temperature up to 1800 ℃. 9Utilization of Ogura CMS germplasm with the clubroot resistance gene by fertility restoration and cytoplasm replacement in Brassica oleracea L显示文摘Clubroot disease,a major plant root disease caused by Plasmodiophora brassicae,has become one of the most destructive diseases among cultivated cruciferous vegetables.However,clubroot-resistant Brassica oleracea materials are rare.A few clubroot-resistant cabbage varieties are available on the market,but all are Ogura cytoplasmic male sterile(CMS)types.Therefore,in this study,to reutilize the clubroot-resistant Ogura CMS germplasm of cabbage,a new fertility-restored Ogura CMS material,16Q2-11,was used as a bridge to transfer the clubroot resistance(CR)gene from the Ogura CMS cytoplasm to the normal cytoplasm by a two-step method(a fertility restoration and cytoplasm replacement method).In the first cross for fertility restoration of Ogura CMS clubroot-resistant cabbage(FRCRC),16Q2-11 was used as a restorer to cross with Ogura CMS materials containing the CR gene CRb2.Eleven Rfo-positive progenies were generated,of which four contained CRb2:F8-514,F8-620,F8-732 and F8-839.After inoculation with race 4 of P.brassicae,these four CRb2-positive individuals showed resistance.Furthermore,F8-514 and F8-839 were then used as male parents in the second cross of FRCRC to cross with cabbage inbred lines,resulting in the successful introgression of the CRb2 gene into the inbred lines.All offspring produced from this step of cross,which had a normal cytoplasm,showed a high resistance to race 4 of P.brassicae and could be utilized for the breeding of clubrootresistant cabbage varieties in the future.This is the first time that the Ogura CMS restorer has been used to restore the fertility of Ogura CMS clubroot-resistant cabbages,which could improve germplasm diversity in cabbage and provide a reference method for using CMS germplasm in Brassica crops.Wenjing Ren Zhiyuan Li Fengqing Han Bin Zhang Xing Li Zhiyuan Fang Limei Yang Mu Zhuang Honghao Lv Yumei Liu Yong Wang Hailong Yu Yangyong Zhang 2020Horticulture Research2020,7,1:7
10Comparison of efficacy between HLA6/6-and HLA3/6-matched haploidentical hematopoietic stem cell transplant in T-cell-replete transplants between parents and children显示文摘To compare the efficacy of HLA6/6-matched haploidentical hematopoietic stem cell transplant(haplo-HSCT) with that of HLA3/6-matched HSCT in T-cell-replete transplants, we recruited 27 consecutive recipients from multiple centers who received HLA6/6-matched haplo-HSCT from a parent or child donor between February 2010 and May 2016. A matched-pair analysis was designed. For each recipient from the study cohort, two recipients were randomly selected from the control cohort and matched(according to patient age, patient sex, disease type, disease status, donor age, donor sex, and recipient-donor relationship). No significant differences were found in hematopoietic recovery. The incidence of grade II–IV and III–IV acute graft versus host disease was similar(18.5% vs. 31.5%, P=0.216; 11.1% vs. 9.3%, P=0.792) in the HLA6/6 and HLA3/6 groups, respectively. The3-year cumulative incidence of relapse was 14.8% and 17.0%(P=0.800). The 3-year cumulative incidence of nonrelapse mortality was 12.1% and 17.6%(P=0.751). The estimated 3-year disease-free survival was 73.1% and 65.5%(P=0.489). The estimated 3-year overall survival was 74.7% and 74.0%(P=0.946). The data suggested the efficacy and safety of the HLA6/6-and the HLA3/6-matched haplo-HSCT between parents and children are comparable. 11Low frequency vibration control of railway vehicles based on ahigh static low dynamic stiffness dynamic vibration absorber显示文摘In order to control the low frequency vibration of railway vehicles, a vertical two degrees of freedom(2DOF) low frequency dynamic vibration absorber(DVA) based on acceleration is proposed. Parameters of the dynamic vibration absorber are put forth to control the low frequency vibration of car body bouncing and pitching. Next, the acceleration power spectrum density(PSD)and ride quality of the car body are calculated based on the pseudo excitation method(PEM) and covariance algorithm,respectively. According to the requirement of 2DOF low frequency DVA, the isolators with high static low dynamic stiffness(HSLDS) are designed. A high-speed train dynamic model containing HSLDS isolators is established to validate the effects on the car body vibration. The results reveal that the 2D low frequency DVA can significantly reduce the vibration of the car body bouncing and pitching. 14A Fairness Resource Allocation Algorithm for Coverage and Capacity Optimization in Wireless Self-Organized Network显示文摘To achieve the higher resource efficiency, Coverage and Capacity Optimization(CCO) as an important role of the network self-healing and self-optimization, has become a focus topic in wireless Self-Organized Network(SON). In this paper, a novel CCO scheme is proposed to maximize utility function of the integrated coverage and capacity. It starts with the analysis on the throughput proportional fairness(PF) algorithm and then proposes the novel Coverage and Capacity Proportional Fairness(CCPF) allocation algorithm along with a proof of the algorithms convergence. This proposed algorithm is applied in a coverage capacity optimization scheme which can guarantee the reasonable network capacity by the coverage range accommodation. Next, we simulate the proposed CCO scheme based on telecom operators' real network data and compare with three typical resource allocation algorithms: round robin(RR), proportional fairness(PF) and max C/I. 15Confinement and antenna effect for ultrasmall Y_(2)O_(3):Eu^(3+) nanocrystals supported by MOF with enhanced near-UV light absorption thereby enhanced luminescence and excellently multifunctional applications显示文摘A novel host-guest luminous system with enhanced near-UV light absorption thereby enhanced luminescence are designed based on the synergism of quantum confinement,spatial confinement,and antenna effect,where ultrasmall Y_(2)O_(3):Eu^(3+)nanocrystals are fixed inside MOF(Eu/Y-BTC)as supporting structure.The Eu/Y-BTC not only limits the size and leads to lattice distortion of Y_(2)O_(3):Eu^(3+)nanocrystals and controls the distance between nanocrystals,but also promotes the light absorption and emission.The significantly red-shifted and broadened charge transfer band of Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)leads to the excellent applications of Y_(2)O_(3):Eu^(3+)in white light-emitting diodes(LEDs).Our results show that white light with superior color quality(CRI>90)and extremely high luminous efficacy(an LER of 335 lm/W)could be achieved using Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)as red phosphor.The Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)also improves the photoelectric performance of dye-sensitized solar cells(DSSCs),not only because Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)has a large specific surface area and the adsorption amount of the dye is increased,but also because the valence band position of Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)is 2.41 eV,which can provide an additional energy level between the TiO2 and dye,promoting electron transfer.For these advantageous features,the multifunctional Y_(2)O_(3):Eu^(3+)/(Eu/Y-BTC)composite product will open new avenues in white LEDs and DSSCs.Hongyi Xu Wenjing Yu Kai Pan Guofeng Wang Peifen Zhu 2021Nano Research2021,14,3:5
16Numerical simulations of rip currents off arc-shaped coastlines显示文摘The rip currents induced by waves off arc-shaped coastlines are seriously harmful to humans, but understanding of their characteristics is lacking. In this study, the FUNWAVE model was used to calculate the wave-induced currents in the Haller experiment and the ideal arc-shaped coast similar to Sanya Dadonghai, Hainan Province,China. The results showed that the FUNWAVE model has considerable ability to simulate the rip currents, and it was used to further simulate rip currents off arc-shaped coastlines to investigate their characteristics. The rip currents were found to be stronger as the curvature of arc-shaped coastline increased. Coastal beach slope exerts a significant influence on rip currents; in particular, an overly steep or overly mild slope is not conducive to creating rip currents. Furthermore, the rip currents were found to become weaker as the size of arc-shaped coast decreased. 17Nano-hydroxyapatite accelerates vascular calcification via lysosome impairment and autophagy dysfunction in smooth muscle cells显示文摘Vascular calcification(VC)is a common characteristic of aging,diabetes,chronic renal failure,and atherosclerosis.The basic component of VC is hydroxyapatite(HAp).Nano-sized HAp(nHAp)has been identified to play an essential role in the development of pathological calcification of vasculature.However,whether nHAp can induce calcification in vivo and the mechanism of nHAp in the progression of VC remains unclear.We discovered that nHAp existed both in vascular smooth muscle cells(VSMCs)and their extracellular matrix(ECM)in the calcified arteries from patients.Synthetic nHAp had similar morphological and chemical properties as natural nHAp recovered from calcified artery.nHAp stimulated osteogenic differentiation and accelerated mineralization of VSMCs in vitro.Synthetic nHAp could also directly induce VC in vivo.Mechanistically,nHAp was internalized into lysosome,which impaired lysosome vacuolar H+-ATPase for its acidification,therefore blocked autophagic flux in VSMCs.Lysosomal re-acidification by cyclic-3′,5′-adenosine monophosphate(cAMP)significantly enhanced autophagic degradation and attenuated nHAp-induced calcification.The accumulated autophagosomes and autolysosomes were converted into calcium-containing exosomes which were secreted into ECM and accelerated vascular calcium deposit.Inhibition of exosome release in VSMCs decreased calcium deposition.Altogether,our results demonstrated a repressive effect of nHAp on lysosomal acidification,which inhibited autophagic degradation and promoted a conversion of the accumulated autophagic vacuoles into exosomes that were loaded with undissolved nHAp,Ca^(2+),Pi and ALP.These exosomes bud off the plasma membrane,deposit within ECM,and form calcium nodules.Vascular calcification was thus accelerated by nHAP through blockage of autophagic flux in VSMCs.Qi Liu Yi Luo Yun Zhao Pingping Xiang Jinyun Zhu Wangwei Jing Wenjing Jin Mingyao Chen Ruikang Tang Hong Yu 2022Bioactive Materials2022,7,2:5
18Calculation of Depth to Detachment and Its Significance in the Kuqa Depression显示文摘The depth to detachment level is a critical factor affecting the quality of structural modeling in fold and thrust belts. There are several detachment levels developed in the Kuqa depression. Based on the excess-area diagram, this paper concerns mainly the calculation of the depth to detachment in the Kuqa depression. The result demonstrates that the detachment levels are situated in different strata in varying zones, such as the Paleogene Kugeliemu Formation, the Paleozoic and the crystalline basement. 19CD147 regulates antitumor CD8^(+)T-cell responses to facilitate tumor-immune escape显示文摘Negative regulation of antitumor T-cell-immune responses facilitates tumor-immune escape.Here,we show that deletion of CD147,a type I transmembrane molecule,in T cells,strongly limits in vivo tumor growth of mouse melanoma and lung cancer in a CD8^(+)T-cell-dependent manner.In mouse tumor models,CD147 expression was upregulated on CD8^(+)tumor-infiltrating lymphocytes(TILs),and CD147 was coexpressed with two immune-checkpoint molecules,Tim-3 and PD-1.Mining publicly available gene-profiling data for CD8^(+)TILs in tumor biopsies from metastatic melanoma patients showed a higher level of CD147 expression in exhausted CD8^(+)TILs than in other subsets of CD8^(+)TILs,along with expression of PD-1 and TIM-3.Additionally,CD147 deletion increased the abundance of TILs,cytotoxic effector function of CD8^(+)T cells,and frequency of PD-1^(+)CD8^(+)TILs,and partly reversed the dysfunctional status of PD-1^(+)Tim-3^(+)CD8^(+)TILs.The cytotoxic transcription factors Runx3 and T-bet mediation enhanced antitumor responses by CD147^(-/-)CD8^(+)T cells.Moreover,CD147 deletion in T cells increased the frequency of TRM-like cells and the expression of the T-cell chemokines CXCL9 and CXCL10 in the tumor microenvironment.Analysis of tumor tissue samples from patients with non-small-cell lung cancer showed negative correlations between CD147 expression on CD8^(+)TILs and the abundance of CD8^(+)TILs,histological grade of the tumor tissue samples,and survival of patients with advanced tumors.Altogether,we found a novel function of CD147 as a negative regulator of antitumor responses mediated by CD8^(+)TILs and identified CD147 as a potential target for cancer immunotherapy.Yatong Chen Jing Xu Xiaodong Wu Hui Yao Zhou Yan Ting Guo Wenjing Wang Peixiao Wang Yu Li Xiangmin Yang Hao Li Huijie Bian Zhi-Nan Chen 2021Cellular & Molecular Immunology2021,18,8:4
20Clinical characteristics of 310 SARS-CoV-2 Omicron variant patients and comparison with Delta and Beta variant patients in China显示文摘Although the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant has spread,data on the clinical characteristics of infected patients are limited.In this study,the demographic,clinical characteristics,and laboratory data of 310 SARS-CoV-2 Omicron variant patients treated at Haihe Hospital of Tianjin were collected and analyzed.Information on these patients was compared to 96 patients with the Delta variant of concern(VOC)and 326 patients with the Beta VOC during the previous coronavirus disease 2019(COVID-19)outbreak in Harbin.Of the 310 patients infected with the Omicron variant,the median age was 35 years.Most patients were clinically classified as mild(57.74%),and the most common symptoms were cough(48.71%),fever(39.35%),and sore throat(38.26%).The results for different vaccination groups in the Omicron group showed that the median of“SARS-CoV-2 specific IgG”after 2 or 3 doses of vaccination was higher than the unvaccinated group(all Ps<0.05).Older age was associated with a higher proportion of moderate cases and lower asymptomatic and mild cases based on clinical classifications.Compared to the Delta and Beta groups,the median age of the Omicron group was younger.The total number of asymptomatic patients and mild patients in the Omicron virus group was higher than the Delta and Beta groups(60.97%vs.54.17%vs.47.55%).This study presented the clinical characteristics of the first group of patients infected with the Omicron variant in Tianjin,China,and compared their clinical features with patients infected by the Delta and Beta variants,which would increase our understanding of the characteristics of SARS-CoV-2 Omicron variant.Wei Yang Songliu Yang Lei Wang Yuxin Zhou Yu Xin Hongxu Li Wenjing Mu Qi Wu Lei Xu Mingyan Zhao Changsong Wang Kaijiang Yu 2022Virologica Sinica2022,37,5:4
