1Identification of liver metastasis-associated genes in human colon carcinoma by mRNA profiling显示文摘Objective: Liver metastasis,which contributes substantially to high mortality,is the most common recurrent mode of colon carcinoma.Thus,it is necessary to identify genes implicated in metastatic colonization of the liver in colon carcinoma.Methods: We compared mRNA profiling in 18 normal colon mucosa(N),20 primary tumors(T) and 19 liver metastases(M) samples from the dataset GSE49355 and GSE62321 of Gene Expression Omnibus(GEO) database.Gene ontology(GO) and pathways of the identified genes were analyzed.Co-expression network and proteinprotein interaction(PPI) network were employed to identify the interaction relationship.Survival analyses based on The Cancer Genome Atlas(TCGA) database were used to further screening.Then,the candidate genes were validated by our data.Results: We identified 22 specific genes related to liver metastasis and they were strongly associated with cell migration,adhesion,proliferation and immune response.Simultaneously,the results showed that C-X-C motif chemokine ligand 14(CXCL14) might be a favorable prediction factor for survival of patients with colon carcinoma.Importantly,our validated data further suggested that lower CXCL14 represented poorer outcome and contributed to metastasis.Gene set enrichment analysis(GSEA) showed that CXCL14 was negatively related to the regulation of stem cell proliferation and epithelial to mesenchymal transition(EMT).Conclusions: CXCL14 was identified as a crucial anti-metastasis regulator of colon carcinoma for the first time,and might provide novel therapeutic strategies for colon carcinoma patients to improve prognosis and prevent metastasis.Jianling Liu Dan Wang Chaoqi Zhang Zhen Zhang Xinfeng Chen Jingyao Lian Jinbo Liu Guixian Wang Weitang Yuan Zhenqiang Sun Weijia Wang Mengjia Song Yaping Wang Qian Wu Ling Cao Dong Wang Yi Zhang 2018Chinese Journal of Cancer Research2018,30,6:1

3The BnTFL1–BnGF14nu–BnFD module regulates flower development and plant architecture in Brassica napus显示文摘Flower development and plant architecture determine the efficiency of mechanized harvest and seed yield in Brassica napus.Although TERMINAL FLOWER 1(AtTFL1)is a regulator of flower development in Arabidopsis thaliana,the function and regulatory mechanism of TFL1 orthologs in B.napus remains unclear.Six BnTFL1 paralogs in the genome of the B.napus inbred line‘K407’showed steadily increasing expression during vernalization.CRISPR/Cas-induced mutagenesis of up to four BnTFL1 paralogs resulted in early flowering and alteration of plant architecture,whereas seed yield was not altered in BnTFL1 single,double,or triple mutants.Six BnTFL1 paralogs,but not BnaA02.TFL1,showed an additive and conserved effect on regulating flowering time,total and terminal flower number,and plant architecture.BnaA10.TFL1 regulates flower development by interacting with BnaA08.FD through the protein BnaA05.GF14nu,resulting in the transcriptional repression of floral integrator and floral meristem identity genes.These findings about the regulatory network controlling flower development and plant architecture present a promising route to modifying these traits in B.napus.Jianjun Wang Chi Zhang Youpeng Chen Yanan Shao Meifang Liao Qian Hou Weitang Zhang Yang Zhu Yuan Guo Zijin Liu Christian Jung Mingxun Chen 2023The Crop Journal2023,11,6:0
4Effects of extracellular vesicle-derived noncoding RNAs on pre-metastatic niche and tumor progression显示文摘A pre-metastatic niche(PMN)is a protective microenvironment that facilitates the colonization of disseminating tumor cells in future metastatic organs.Extracellular vesicles(EVs)play a role in intercellular communication by delivering cargoes,such as noncoding RNAs(ncRNAs).The pivotal role of extracellular vesicle-derived noncoding RNAs(EV-ncRNAs)in the PMN has attracted increasing attention.In this review,we summarized the effects of EV-ncRNAs on the PMN in terms of immunosuppression,vascular permeability and angiogenesis,inflammation,metabolic reprogramming,and fibroblast alterations.In particular,we provided a comprehensive overview of the effects of EV-ncRNAs on the PMN in different cancers.Finally,we discussed the promising clinical applications of EV-ncRNAs,including their potential as diagnostic and prognostic markers and therapeutic targets.Zhuang Chen Qiming Wang Jinbo Liu Wenkang Wang Weitang Yuan Yang Liu Zhenqiang Sun Chengzeng Wang 2024Genes & Diseases2024,11,1:0
