1Celastrol targets IRAKs to block Toll-like receptor 4-mediated nuclear factor-κB activation显示文摘OBJECTIVE: Celastrol has been established as a nuclear factor-κB(NF-κB) activation inhibitor; however, the exact mechanism behind this action is still unknown. Using text-mining technology, the authors predicted that int erleukin-1 receptor-associated kinases(IRA Ks) are potential celastrol targets, and hypothesized that targeting IRAKs might be one way that celastrol inhibits NF-κB. This is because IRAKs are key molecules for some crucial pathways to activate NF-κB(e.g., the inter leukin-1 receptor(IL-1R)/Toll- like receptor(TLR) superfamily).METHODS: The human hepatocellular cell line(Hep G2) treated with palmitic acid(PA) was used as a model for stimulating TLR4/NF-κB activation, in order to observe the potential effects of celastrol in IRAK regulation and NF-κB inhibition. The transfection of small interfering RNA was used for down-regulating TLR4, IRAK1 and IRAK4, and the Western blot method was used to detect changes in the protein expressions.RESULTS: The results showed that celastrol could effectively inhibit PA-caused TLR4-dependent NF-κB activation in the Hep G 2 cells; PA also activated IRAKs, which were inhibited by celastrol. 2Respective IL-17A production by γδ T and Th17 cells and its implication in host defense against chlamydial lung infection显示文摘The role of IL-17A is important in protection against lung infection with Chlamydiae,an obligate intracellular bacterial pathogen.In this study,we explored the producers of IL-17A in chlamydial lung infection and specifically tested the role of major IL-17A producers in protective immunity.We found thatγδT cells and Th17 cells are the major producers of IL-17A at the early and later stages of chlamydial infection,respectively.Depletion ofγδT cells in vivo at the early postinfection(p.i.)stage,when mostγδT cells produce IL-17A,failed to alter Th1 responses and bacterial clearance.In contrast,the blockade of IL-17A at the time when IL-17A was mainly produced by Th17(day 7 p.i.)markedly reduced the Th1 response and increased chlamydial growth.The data suggest that theγδT cell is the highest producer of IL-17A in the very early stages of infection,but the protection conferred by IL-17A is mainly mediated by Th17 cells.In addition,we found that depletion ofγδT cells reduced IL-1αproduction by dendritic cells,which was associated with a reduced Th17 response.This finding is helpful to understand the variable role of IL-17A in different infections and to develop preventive and therapeutic approaches against infectious diseases by targeting IL-17A.Hong Bai Xiaoling Gao Lei Zhao Ying Peng Jie Yang Sai Qiao Huili Zhao Shuhe Wang YiJun Fan Antony George Joyee Zhi Yao Xi Yang 2017Cellular & Molecular Immunology2017,14,10:7

5Anticancer potential of metabolic compounds from marine actinomycetes isolated from Lagos Lagoon sediment显示文摘Thirty-two actinomycetes strains were isolated from sediment samples from 12 different sites at Lagos Lagoon and identified using standard physiological and biochemical procedures as well as 16S rDNA gene sequence analysis. Secondary metabolites were extracted from the strains and their anticancer activity on the K562 (Human acute myelocytic leukemia), HeLa (cervical carcinoma), AGS (Human gastric), MCF- 7 (breast adenocarcinoma) and HL-60 (Human acute promyelocytic leukemia) cell lines was determined. The metabolic extracts exhibited cytotoxicity with IC50 values ranging from 0.030 mg/mL to 4.4 mg/mL. The Streptomyces bingchenggensis ULS14 extract was cytotoxic against all the cell lines tested. The bioactivity-guided extraction and purification of the metabolic extracts from this strain yielded two purified anticancer compounds: ULDF4 and ULDF5. The structures of the extracted compounds were determined using spectroscopic analyses, including electrospray ionization mass spectrophotometer and nuclear magnetic resonance (1 Dimensional and 2 Dimensional), and were shown to be structurally similar to staurosporine and kigamicin. The IC50 of ULDF4 and ULDF5 against the HeLa cell line was 0.034 mg/mL and 0.075 mg/mL, respectively. 6An unexpected large continental source of reactive bromine and chlorine with significant impact on wintertime air quality显示文摘Halogen atoms affect the budget of ozone and the fate of pollutants such as hydrocarbons and mercury.Yet their sources and significances in polluted continental regions are poorly understood.Here we report the observation of unprecedented levels(averaging at 60 parts per trillion)of bromine chloride(BrCl)at a mid-latitude site in North China during winter.Widespread coal burning in rural households and a photo-assisted process were the primary source of BrCl and other bromine gases.BrCl contributed about 55%of both bromine and chlorine atoms.The halogen atoms increased the abundance of'conventional'tropospheric oxidants(OH,HO_(2) and RO_(2))by 26%-73%,and enhanced oxidation of hydrocarbon by nearly a factor of two and the net ozone production by 55%.Our study reveals the significant role of reactive halogen in winter atmospheric chemistry and the deterioration of air quality in continental regions where uncontrolled coal combustion is prevalent.Xiang Peng Weihao Wang Men Xia Hui Chen A.R.Ravishankara Qinyi Li Alfonso Saiz-Lopez Pengfei Liu Fei Zhang Chenglong Zhang Likun Xue Xinfeng Wang Christian George Jinhe Wang Yujing Mu Jianmin Chen Tao Wang 2021National Science Review2021,8,7:2
7Crystal structure of histidine-containing phosphocarrier protein from Thermoanaerobacter tengcongensis MB4 and the implications for thermostability显示文摘Protein thermostability is an inherent characteristic of proteins from thermophilic microorganisms,and therefore enables these organisms to survive at extreme temperatures.Although it is well-known that thermostable proteins are critical for the growth of thermophilic organisms,the structural basis of protein thermostability is not yet fully understood.The histidine-containing phosphocarrier (HPr) protein,a phosphate shuttle protein in the phosphoenolpyruvate-dependent sugar transport system (PTS) of bacterial species,is an ideal model for investigating protein thermostability with respect to its small size and deficiency in disulphide bonds or cofactors.In this study,the HPr protein from Thermoanaerobacter tengcongensis (TtHPr) is cloned and purified.Crystal structure with good quality has been determined at 2.3 resolution,which provides a firm foundation for exploring the thermostable mechanism.However,it shows that the crystal structure is conserved and no clue can be obtained from this single structure.Furthermore,detailed comparison of sequence and structure with the homologs from mesoor thermophilic bacteria shows no obvious rule for thermostability,but the extra salt-bridge existing only in thermophilic bacteria might be a better explanation for thermostability of HPr.Thus,mutations are performed to interrupt the salt-bridge in HPrs in thermophilic bacteria.Using site-directed mutations and the circular dichroism method,thermostability is evaluated,and the mutational variations are shown to have a faster denaturing rate than for wild-type viruses,indicating that mutations cause instability in the HPrs.Understanding the higher-temperature resistance of thermophilic and hyperthermophilic proteins is essential to studies on protein folding and stability,and is critical in engineering efficient enzymes that can work at a high temperature.FENG ChunYan GAO Feng LIU YiWei WANG GuiHua PENG Hao MA YanHe YAN JingHua GAO George Fu 2011Science China(Life Sciences)2011,54,6:2

13Hyoscyamine-producing marine Actinomycetes from Lagos Lagoon sediment显示文摘Objective:To isolate and screen Actinoniycetes from Lagos Lagoon soil sediments for production of bioactive metabolites.Methods:Sediment samples were collected from four different locations of Lagos Lagoon and were dried for 2 weeks after which the Actinoniycetes were isolated by serial dilution using the spread plate method on starch casein and Kuster's agar supplemented with 80 ug/mL cycloheximide to prevent fungal growth.The plates were incubated at 28 C for 1-2 weeks.Isolates were selected based on their colonial characteristics as well as their Gram's reaction and subciiltured using the same media for isolation until pure cultures were obtained and incubated at 28 C for 3 d.Thereafter,they were inoculated into starch casein and Kuster's broth media and incubated for 8 d.The secondary metabolites were screened for antimicrobial activity against the following microorganisms:methicillin resistant Staphylococcus aureus.Staphylococcus aureus ATCC 29213.Escherichia coli ATCC 29522.Pseudomonas aeruginosa ATCC 27853.Candida albicans and Enterocolitis faecal is ATCC 29212.Coagulasenegative staphylococci isolated from HIV patients were also used(Staphylococcus warneri.Staphylococcus xylosus and Staphylococcus epidennidis).The antimicrobial metabolites of the Actinoniycetes isolates were identified using gas chromatography(GC).Results:Crude extracts of isolates showed antimicrobial activity against some of the test organisms.The GC data analysis showed the antibiotic profile of these isolates.Conclusions:Analysis of the crude extracts of the isolates using GC method,revealed the presence of antibiotics including an anticholinergic hyoscyamine among other conclusions.Davies Olabisi Flora Adeleye Isaac Adeyemi Wang Peng George 2015Asian Pacific Journal of Tropical Biomedicine2015,5,3:1
14Anti-infection effects of heparin on SARS-CoV-2 in a diabetic mouse model显示文摘Severe acute respiratory syndrome coronavirus 2(SARSCo V-2)infection can result in more severe syndromes and poorer outcomes in patients with diabetes and obesity.However,the precise mechanisms responsible for the combined impact of coronavirus disease 2019(COVID-19)and diabetes have not yet been elucidated,and effective treatment options for SARS-Co V-2-infected diabetic patients remain limited.To investigate the disease pathogenesis,K18-h ACE2 transgenic(h ACE2^(Tg))mice with a leptin receptor deficiency(h ACE2-Lepr^(-/-))and high-fat diet(h ACE2-HFD)background were generated.The two mouse models were intranasally infected with a 5×10^(5) median tissue culture infectious dose(TCID_(50))of SARSCo V-2,with serum and lung tissue samples collected at 3days post-infection.The h ACE2-Lepr^(-/-)mice were then administered a combination of low-molecular-weight heparin(LMWH)(1 mg/kg or 5 mg/kg)and insulin via subcutaneous injection prior to intranasal infection with1×10^(4) TCID_(50)of SARS-Co V-2.Daily drug administration continued until the euthanasia of the mice.Analyses of viral RNA loads,histopathological changes in lung tissue,and inflammation factors were conducted.Results demonstrated similar SARS-Co V-2 susceptibility in h ACE2^(Tg)mice under both lean(chow diet)and obese(HFD)conditions.However,compared to the h ACE2-Lepr^(+/+)mice,h ACE2-Lepr^(-/-)mice exhibited more severe lung injury,enhanced expression of inflammatory cytokines and hypoxia-inducible factor-1α(HIF-1α),and increased apoptosis.Moreover,combined LMWH and insulin treatment effectively reduced disease progression and severity,attenuated lung pathological changes,and mitigated inflammatory responses.In conclusion,preexisting diabetes can lead to more severe lung damage upon SARS-Co V-2 infection,and LMWH may be a valuable therapeutic approach for managing COVID-19patients with diabetes.Zhongyun Zhang Ning Zhang Xuancheng Lu Min Zhou Xiaoxiang Yan Weiqiong Gu Jingru Yang Qin Zhang Cheng Zhang Yuhuan Gong Mingjun Jia Xiaoyu Zhang Peng Ning Mei Liu Xiaoyan Li Xiaomeng Shi Wenjun Liu George FGao Guang Ning Jiqiu Wang Yuhai Bi 2023Zoological Research2023,44,6:1

16Conserved peptides enhance immune efficiency of inactive vaccines against emerging avian influenza viruses in chicken显示文摘Avian influenza viruses(AIVs) such as H5N1 and H7N9 are a great threat to poultry economics and public health. Vaccination can effectively inhibit the spread of AIV in poultry, which is also a viable strategy for controlling virus transmission from poultry to human. Adjuvants that are commonly used in current inactivated vaccines to provide stronger anti-AIV immune responses are often limited in their capacity to quantitatively induce both humoral and cellular immune responses. Herein, we assessed the levels of immune responses generated by a vaccine formulation comprising inactivated H5N1 antigen and synthetic peptides covering conserved CD4^+, CD8^+ T cell, and B cell epitopes. We found that the synthetic peptides enhanced the antibody responses against conserved influenza virus antigen M2 e. Notably, the hemagglutination inhibition test results indicated that the peptides significantly augmented the antibody responses of inactivated H5N1 antigen even in the 1/10 or 1/5 dose group, in the identical antibody level as antigen alone used at the full dose. This indicates that the peptide can significantly reduce the use of inactivated virus, lowering the cost of the vaccine. Moreover, the peptides increased the transcript levels of interleukin-4 and interferon-γcytokines in chicken peripheral blood mononuclear cells, which may facilitate both humoral and cellular immune responses. 17Gravitational wave astronomy: the current status显示文摘In the centenary year of Einstein's General Theory of Relativity, this paper reviews the current status of gravitational wave astronomy across a spectrum which stretches from attohertz to kilohertz frequencies. Sect. 1 of this paper reviews the historical development of gravitational wave astronomy from Einstein's first prediction to our current understanding the spectrum. It is shown that detection of signals in the audio frequency spectrum can be expected very soon, and that a north-south pair of next generation detectors would provide large scientific benefits. Sect. 2 reviews the theory of gravitational waves and the principles of detection using laser interferometry. The state of the art Advanced LIGO detectors are then described. These detectors have a high chance of detecting the first events in the near future. Sect. 3 reviews the KAGRA detector currently under development in Japan,which will be the first laser interferometer detector to use cryogenic test masses. Sect. 4 of this paper reviews gravitational wave detection in the nanohertz frequency band using the technique of pulsar timing. Sect. 5 reviews the status of gravitational wave detection in the attohertz frequency band, detectable in the polarisation of the cosmic microwave background, and discusses the prospects for detection of primordial waves from the big bang. The techniques described in sects. 1–5 have already placed significant limits on the strength of gravitational wave sources. Sects. 6 and 7 review ambitious plans for future space based gravitational wave detectors in the millihertz frequency band. 20Gut microbiota have blood types as human显示文摘ABO blood group system was firstly recognized by Landsteiner in 1900[1].Since then,the antigens of the ABO system (A,B and H determinants,respectively)have been shown to encompass complex carbohydrate structures [2].A and B antigen were synthesized by the sequential action of glycosyltransferases,with A and B glycosyltransferases catalyzing the addition of N-acetylgalactosamine and D-galactose to precursor H antigen,respectively.
