| 1 | AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies. | Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li | 2021 | Cell Research2021,31,2: | 39 |
| 2 | Interaction of rice bZIP protein REB with the 5'-upstream region of both rice sbe1 gene and waxy gene显示文摘Rice starch branching enzyme 1 (SBE1) and granule-bound starch synthase (GBSS) catalyzed the bio-synthesis of amylopectin and amylose in rice developing en-dosperm respectively, and the genes encoding these two en-zymes, sbe1 and waxy, are mainly expressed in the en-dosperm. Within the 5 ’-upstream region of rice sbel gene, we identified a 53 bp fragment C53 which could interact with the nuclear proteins extracted from rice endosperm. We also found that fragment Ha-2 in the 5 ’-upstream region of rice waxy gene could compete with C53 for its interaction. Fur-ther experiments demonstrated that rice bZIP protein REB could interact with two ACGT elements (G-box and Hex) in the fragment C53 as well as three ACGT elements (WG1, WG2, and WG3) in the fragment Ha-2. The WG1 element could compete well with Hex element for its interaction with REB. | 3 | Introduction of antisense waxy gene into main parent lines of indica hybrid rice显示文摘Amylose content in rice endosperm is one of the key determinants of rice eating and cooking quality, and the poor quality of indica hybrid rice is closely related to the high amylose level in rice grains. In order to improve the grain quality of the indica hybrid rice by genetic engineering, an antisense fragment of rice waxy gene, driven by the 5’-franking sequences of the rice waxy gene, was successfully introduced into three major parent lines of indica hybrid rice, all contain a high amylose level in the grains, via Agrobacte-rium, and more than 100 hygromycin-resistant plants were regenerated. The analysis of PCR amplification and Southern blots indicated that the T-DNA containing the antisense waxy gene had been integrated into the genome of transgenic rice plants. Most of the primary transgenic rice plants grew normally, and the mature seeds from these transgenic plants were performed for analysis of the amylose content. | 5 | Exploration and functionalization of M1-macrophage extracellular vesicles for effective accumulation in glioblastoma and strong synergistic therapeutic effects显示文摘Glioblastoma multiforme(GBM)is a highly aggressive brain tumor with an extremely low survival rate.New and effective approaches for treatment are therefore urgently needed.Here,we successfully developed M1-like macrophage-derived extracellular vesicles(M1EVs)that overcome multiple challenges via guidance from two macrophage-related observations in clinical specimens from GBM patients:enrichment of M2 macrophages in GBM;and origination of a majority of infiltrating macrophage from peripheral blood.To maximize the synergistic effect,we further functionalized the membranes of M1EVs with two hydrophobic agents(the chemical excitation source CPPO(C)and the photosensitizer Ce6(C))and loaded the hydrophilic hypoxiaactivated prodrug AQ4N(A)into the inner core of the M1EVs.After intravenous injection,the inherent nature of M1-derived extracellular vesicles CCA-M1EVs allowed for blood-brain barrier penetration,and modulated the immunosuppressive tumor microenvironment via M2-to-M1 polarization,which increased hydrogen peroxide(H_(2)O_(2))levels.Furthermore,the reaction between H_(2)O_(2) and CPPO produced chemical energy,which could be used for Ce6 activation to generate large amounts of reactive oxygen species to achieve chemiexcited photodynamic therapy(CDT).As this reaction consumed oxygen,the aggravation of tumor hypoxia also led to the conversion of non-toxic AQ4N into toxic AQ4 for chemotherapy.Therefore,CCA-M1EVs achieved synergistic immunomodulation,CDT,and hypoxia-activated chemotherapy in GBM to exert a potent therapeutic effect.Finally,we demonstrated the excellent effect of CCA-M1EVs against GBM in cell-derived xenograft and patient-derived xenograft models,underscoring the strong potential of our highly flexible M1EVs system to support multi-modal therapies for difficult-to-treat GBM. | Xiaojun Wang Hui Ding Zongyang Li Yaonan Peng Hui Tan Changlong Wang Guodong Huang Weiping Li Guanghui Ma Wei Wei | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 3 |
| 6 | 盐酸羟考酮控释片治疗晚期癌症疼痛的临床应用(英文)显示文摘Objective: The aim of this study was to evaluate the efficacy and adverse reactions of OxyContin hydrochloride controlled release tablets in the treatment of moderate or severe pain in patients with terminal cancer and to observe any improvement on the cancer patients' quality of life. Methods: Sixty-eight patients with moderate or severe cancer pain were treated with OxyContin hydrochloride controlled release tablets. The initial dose was 5 mg/12h, or 1/2 that of the standard morphine regimen. During the course of treatment, the dosage was adjusted according to the patients' condition until the pain completely disappeared or nearly did so. Each patient received a treatment for at least 15 days. At the same time, adverse reactions, the quality of life and scores for the intensity of pain were observed and recorded [1]. Results: The final titrated dosage of OxyContin was as follows: the patients in 30 cases (44.1%) received a dosage of ≤ 30 mg/d, those in 16 cases (23.5%) received a dosage of 31 to 60 mg/d, those in 18 cases (26.5%) received a dosage of 61 to 120 mg/d and those in 4 cases (5.9%) received a dosage of ≥ 120 mg/d. The overall rate of relief from pain was 95.6%, among which the rates of excellent, effective and moderate relief were respectively 39.7%, 48.5% and 7.4%. OxyContin had mild adverse reactions and patients' quality of life was markedly improved. Conclusion: OxyContin is effective in treatment of moderate and severe cancer pain. Using RT-PCR and Southern hybridization to study OsEBP-89 tissue-specific expression, we found that its first intron (115 bp in length) of its was retained in a fraction of its transcripts of this gene in rice developing seeds. Furthermore, two OsEBP-89 cDNA clones (c89L and c89LH) were screened from a rice cDNA library. Sequence analysis revealed that the first intron was retained in c89L clone, whereas, both the first and second intron sequences were spliced in c89LH. In addition to developing seeds, the first intron unspliced transcripts of OsEBP-89 are detected in leaves and roots of rice, too. However, the ratio of the first intron unspliced to spliced OsEBP-89 transcripts varied in different tissues examined. | 18 | An ultra-porous g-C_(3)N_(4) micro-tube coupled with MXene(Ti_(3)C_(2)TX)nanosheets for efficient degradation of organics under natural sunlight显示文摘It remains as a challenge for realizing efficient photo-responsive catalysts towards largescale degradation of organic pollutants under natural sunlight.This work reports a new pore engineering strategy for creating ultra-porous g-C_(3)N_(4) micro-tubes with an unprecedentedly high specific surface area of 152.96 m^(2)/g.This is mainly associated with releasing internal vapor pressure in the autoclave where the hydrothermal treatment of the urea/melamine mixture is processed.Supported by microscopic observation,porosity measurement and spectroscopic characterization,it is found that releasing the pressure at halfway of hydrothermal process is vital for forming exfoliated rod-like precursors and the de-aggregation of these rods presents substantial benefits on the production of mesopores on g-C_(3)N_(4) microtubes during the calcination of precursors.This offers a large number of reactive sites required by photocatalytic reaction.Coupling these micro-tubes with Ti_(3)C_(2)T_(X) nanosheets via electrostatic interaction yields a 1D/2D heterojunction with a close interfacial contact.The addition of metallically conductive Ti_(3)C_(2)T_(X) nanosheets accelerates the separation between electrons and holes,and also enhances the light absorption.All these merits of structural design lead to forming a group of highly efficient catalysts demonstrating an excellent photocatalytic degradation rate of k=0.0560 min^(-1)for RhB dyes under 100 mW/cm~2 visible light radiation that micks sunlight outdoors.This laboratory valuation is further supported by an outdoor test that shows a fast degradation rate of 0.0744 min^(-1)under natural sunlight. | Zongyang Ya Qiyou Wang Jingjin Cai Peng Wang Xinyu Jiang Zhiyin Cai Shanglin Xiang Tingwei Wang Dongyu Cai | 2024 | Journal of Environmental Sciences2024,,3: | 0 |

| 20 | DICE方案与CHOP方案治疗中高度恶性非霍奇金淋巴瘤的随机对照研究(英文)显示文摘Objective: To compare efficacies and safeties of DICE and CHOP regimens in treating intermediate and high grade non-Hodgkin's lymphoma (NHL), and indicate the standard treatment for it. Methods: A total of 74 patients with moder- ately or highly malignant NHL, verified by pathology or histology, were randomized into the trial group (37 patients treated with DICE regimen) and the control group (37 patients treated with CHOP regimen). Survival rate was analyzed by Kaplan-Meier method. Chi-square test was performed between groups. Results: The complete response rate, partial response rate, and response rate were significantly higher in DICE group than in CHOP group (40.5% vs. 29.7%, 37.8% vs. 27.0%, and 78.3% vs. 56.7%, respectively, P < 0.05). The 1-, 3-, and 5-year survival rates were significantly higher in DICE group than in CHOP group (89.2% vs. 81.2%, 76.0% vs. 52.6%%, and 46.7% vs. 36.4%, respectively, P < 0.05). The major side effects, appeared with no differences (P > 0.05) in incidences between the two groups, were leukopenia, thrombocytopenia, and nausea. There were only three episodes of clinical cystitis or gross haematuria in DICE regimen. Conclusion: The results showed higher efficacy of DICE regimen over CHOP regimen.
