1Recent advances in targeting the autotaxin-lysophosphatidate-lipid phosphate phosphatase axis in vivo显示文摘Extracellular lysophosphatidate(LPA) is a potent bioactive lipid that signals through six G-protein-coupled receptors.This signaling is required for embryogenesis,tissue repair and remodeling processes.LPA is produced from circulating lysophosphatidylcholine by autotaxin(ATX),and is degraded outside cells by a family of three enzymes called the lipid phosphate phosphatases(LPPs).In many pathological conditions,particularly in cancers,LPA concentrations are increased due to high ATX expression and low LPP activity.In cancers,LPA signaling drives tumor growth,angiogenesis,metastasis,resistance to chemotherapy and decreased efficacy of radiotherapy.Hence,targeting the ATX-LPA-LPP axis is an attractive strategy for introducing novel adjuvant therapeutic options.In this review,we will summarize current progress in targeting the ATX-LPA-LPP axis with inhibitors of autotaxin activity,LPA receptor antagonists,LPA monoclonal antibodies,and increasing low LPP expression.Some of these agents are already in clinical trials and have applications beyond cancer,including chronic inflammatory diseases.Matthew G.K. 2Antibacterial forsterite (Mg_(2)SiO_(4)) scaffold: A promising bioceramic for load bearing applications显示文摘In the current work,forsterite samples with different surface area were investigated for its antibacterial activity.Dissolution studies show that the lower degradation of forsterite compared to other silicate bioceramics,which is a desirable property for repairing bone defects.Forsterite scaffold shows superior compressive strength than the cortical bone after immersion in simulated body fluid.Bactericidal tests indicate that the forsterite had inhibition effect on the growth of clinical bacterial isolates.Forsterite may be a suitable candidate material for load bearing applications with enhanced mechanical properties and lower degradation rate.Rajan Choudhary Ankita Chatterjee Senthil Kumar Venkatraman Sivasankar Koppala Jayanthi Abraham Sasikumar Swamiappan 2018Bioactive Materials2018,3,3:3
3Evaluating and Modeling the Mechanical Properties of the Prepared PLGA/nano-BCP Composite Scaffolds for Bone Tissue Engineering显示文摘In this paper, preparation of nano-biphasic calcium phosphate (nBCP), mechanical behavior and load-bearing of poly (lactide-co-glycolide) (PLGA) and PLGA/nBCP are presented. The nBCP with composition of 63/37 (w/w) HA/β-TCP (hydroxyapatite/β-tricalcium phosphate) was produced by heating of bovine bone at 700℃. Composite scaffolds were made by using PLGA matrix and 10-50 wt% nBCP powders as reinforcement material. All scaffolds were prepared by thermally induced solid-liquid phase separation (TIPS) at -60℃ under 4 Pa (0.04 mbar) vacuum. The results of elastic modulus testing were adjusted with Ishai-Cohen and Narkis models for rigid polymeric matrix and compared to each other. PLGA/nBCP scaffolds with 30 wt% nBCP showed the highest value of yield strength among the scaffolds. In addition, it was found that by increasing the nBCP in scaffolds to 50 wt%, the modulus of elasticity was highly enhanced. However, the optimum value of yield strength was obtained at 30 wt% nBCP, and the agglomeration of reinforcing particles at higher percentages caused a reduction in yield strength. It is clear that the elastic modulus of matrix has the significant role in elastic modulus of scaffolds, as also the size of the filler particles in the matrix.M. Ebrahimian-Hosseinabadi F. 5Magnetic iron oxide nanoparticles: Synthesis and surface coating techniques for biomedical applications显示文摘Iron oxide nanoparticles are the most popular magnetic nanoparticles used in biomedical applications due to their low cost, low toxicity, and unique magnetic property. Magnetic iron oxide nanoparticles, including magnetite(Fe3O4) and maghemite(γ-Fe2O3), usually exhibit a superparamagnetic property as their size goes smaller than 20 nm, which are often denoted as superparamagnetic iron oxide nanoparticles(SPIONs) and utilized for drug delivery, diagnosis, therapy, and etc. This review article gives a brief introduction on magnetic iron oxide nanoparticles in terms of their fundamentals of magnetism, magnetic resonance imaging(MRI), and drug delivery, as well as the synthesis approaches, surface coating,and application examples from recent key literatures. Because the quality and surface chemistry play important roles in biomedical applications, our review focuses on the synthesis approaches and surface modifications of iron oxide nanoparticles. 6Biomedical applications of shape-memory polymers:how practically useful are they?显示文摘Shape-memory effect(SME) is the ability of a material to change its dimension in a predefined way in response to an external stimulus. Polymers that exhibit SME are an important class of materials in medicine, especially for minimally invasive deployment of devices. However, the rate of translation of the concept to approved products is extremely low, with mostly nitinolbased devices being approved. In this review, the general aspects of the different types of stimuli that can be used to activate SME are reviewed and sterilization issues of shape-memory polymer(SMP)-based medical devices are addressed.
